피부 섬유아세포에서 밤나무 잎 추출물의 항산화 및 항노화 효능 (Antioxidant and Anti-aging Effects of Extracts from Leaves of Castanea crenata Siebold & Zucc. in Human Dermal Fibroblast)

  • 현재 식품공전에 식품원료로 사용이 가능한 것으로 등록된 국내 산림지역 자생 식물을 식품산업에 활용하고자 국내 자생식물 45종을 선별하였고, 본 연구팀의 선행연구에서 45종의 항산화 활성을 평가한 결과, 다른 종들과 비교하여 우수한 항산화 활성을 보인 밤나무 잎을 본 연구에서 사용하였다. 본 연구에서는 hydrogen peroxide로 산화적 스트레스를 유도한 상태에서 밤나무 잎 추출물의 세포 보호효과, 세포내 항산화 효과 및 항노화 효과를 관찰하였다. 세포 보호효과를 측정한 결과 밤나무 잎 추출물의 농도 유의적으로 세포를 보호하는 효과를 확인하였고, 특히 $100{\mu}g/mL$에서는 양성대조군으로 사용한 항산화 물질인 ascorbic acid 수준까지 세포를 보호하는 효과를 나타내었다. $H_2$-DCFDA 염색을 통한 세포내 항산화 효과를 확인한 결과, 형광현미경과 형광흡광도 측정에서 모두 밤나무 잎 추출물이 농도 유의적으로 세포내 ROS를 저감을 확인하였고, 세포 보호효과와 마찬가지로 농도 $100{\mu}g/mL$에서는 ascorbic acid와 비슷한 수준을 보였다. SA-${\beta}$-galactosidase 염색을 통하여 관찰한 세포노화 억제 효과는 ROS 생성 억제 효과와 유사한 경향으로 밤나무 잎 추출물의 농도 유의적으로 세포노화 억제를 관찰되었다. 이상의 결과를 종합하여 볼 때 밤나무 잎 추출물은 산화적 스트레스로 인한 세포노화를 억제하는 효능을 보였으며, 이는 천연물에서 유래한 항산화 및 항노화 활성 첨가제로 활용도가 매우 넓을 것으로 판단된다.

제주 연안해수로부터 한천 분해 효소 및 자일란 분해 효소를 생산하는 Catenovulum jejuensis A28-5의 동정 및 특성 규명 (Identification and Characterization of an Agarase- and Xylanse-producing Catenovulum jejuensis A28-5 from Coastal Seawater of Jeju Island, Korea)

  • Strain A28-5는대한민국 제주도 연안의 해수샘플로부터 고체배지내 xylan과 agar를 분해하는 균주로 분리되었다. Strain A28-5는그람 음성균으로 한 개의 polar flagella로 운동성을 갖는 $Na^+$ 이온 요구성 균주로 분석되었다. 또한 ampilcillin과 thiostreptone 등의 항생제에 내성을 보였다. Genome 내 G+C content는 43.96%이고, Menaquinone-7 (MK-7)을 predominant quinone으로 함유하고 있었다. Strain A28-5의 세포벽을 구성하는 주요 지방산은 $C_{16:1}$ ${\omega}7c/iso-C_{15:0}$ 2-OH (23.32%), $C_{16:0}$ (21.83%), $C_{18:1}$ ${\omega}7c$ (17.98%)였다. strain A28-5의 16S rRNA gene sequence는 Catenovulum agarivorans YM01와 가장 높은 상동성(98.94%)을 보였으며, Neighbor-Joining phylogenetic tree 제작을 통해서 Catenovulum agarivorans YM01와 가장 높은 근연관계를 보이는 것을 증명하였다. Catenovulum agarivorans YM01과의 DNA-DNA hybridization 분석을 통하여 A28-5을 Catenovulum 속의 신종으로 분류하여Catenovulum jejuensis A28-5로 명명하였다. 이 균주의 액체배양으로부터 준비된 두 종류의 조효소를 이용한 xylan 또는 agarose와의 효소반응액을 Thin layer chromatography로 분석하여 각각 tetramer와 hexamer의 xylooligosaccharides와 (neo)agarooligosacchardes가 생산되는 것을 확인하였다.

국내 산림식물 11종의 항산화 활성 (Antioxidant Activity of 11 Species in Korean Native Forest Plants)

  • This study aimed to investigate antioxidant activities from 11 forest plants, and determine their total phenolics, flavonoids and proantocyanidins contents. In addition, the antioxidant activities were correlated with antioxidant compounds. Among the samples, Cornus officinalis, Castanea crenata, Lindera erythrocarpa, Carpinus laxiflora and Pourthiaea villosa showed significantly higher 2,2-diphenyl-1-picrylhydrazyl (DPPH) ($IC_{50}=21.12{\sim}28.93{\mu}g/mL$) and 2,2'-azino-bis(3-ethylbenzothia zoline-6-sulfonic acid) (ABTS) ($IC_{50}=28.18{\sim}52.55{\mu}g/mL$) radical scavenging ability with reducing power ($IC_{50}=59.91{\sim}93.64{\mu}g/mL$) than other plants; and C. crenata, L. erythrocarpa and Rubus coreanus showed strong nitric oxide (NO) inhibition activity (${\geq}60%$). In addition, L. erythrocarpa, C. laxiflora and P. villosa showed higher oxygen radical absorbance capacity (ORAC) values (${\geq}1,100{\mu}M$ TE/g sample) than other samples. High total phenolic contents were observed in C. crenata (429.11 mg GAE/g), L. erythrocarpa (437.11 mg GAE/g), C. laxiflora (408.67 mg GAE/g) and P. villosa (404.11 mg GAE/g). The DPPH and ABTS radical scavenging activity with reducing power were significantly correlated with total phenolic contents ($R^2=0.71{\sim}0.79$), but total phenolic contents were not correlated with NO inhibition and ORAC ($R^2=0.35{\sim}0.43$). Therefore, these results suggested that C. officinalis, C. crenata, L. erythrocarpa, C. laxiflora and P. villosa are potential natural antioxidative candidate ingredients.

간호사를 위한 종양유전교육 프로그램의 효과 (The Effect of the Cancer Genetic Education Program for Nurses)

  • Purpose: Advancing genetic knowledge for oncology nurses is especially important in Korea because physicians have launched to incorporate genetic risk assessment and genetic testing into their practice. The purpose of this paper was to identify the effect of the first academic cancer genetic risk assessment and counseling course for Korean nurses. Methods: Thirty-five nurses were recruited and educated from June 8 to 14, 2006 in Seoul, Korea. Two measurement tools were used: 'knowledge about the hereditary breast and ovarian cancer (HBOC)' and 'knowledge about the cancer genetics'. Results: Students' score of knowledge about HBOC at pre-education was $12.22{\pm}2.23$ and after education, it increased to $13.62{\pm}1.76$. This change was statistically significant (t=-3.253, p=.003). The score of knowledge about cancer genetics at pre-education was $11.31{\pm}3.44$, and after education it has increased to $16.17{\pm}1.94$. It also was statistically significant (t=-6.92, p=.000). Conclusion: This program was effective to be a starting point for establishing genetic educational planning for the oncology nurses in Korea. This academically-based course is recognized as valuable by oncology nurses. With this new knowledge, nurses can begin to expand their role in delivering comprehensive cancer care services in Korea.

Genetic Structure of the Jellyfish Rhopilema esculentum (Scyphozoa: Rhizostomatidae) in Korean Coastal Waters

  • The edible jellyfish Rhopilema esculentum occurs in waters throughout northeastern Asia, including in Korea, China, and Japan. In Korean waters, R. esculentum has appeared in two regions (Gangwha and Muan). Based on the appearance of young medusae and coastal distribution records, these two regions may be key R. esculentum breeding sites. In the present study, we investigate and compare the genetic structure of R. esculentum in the two regions using mitochondrial sequences (16S ribosomal RNA and cytochrome c oxidase subunit I). The genetic diversity of the R. esculentum population at Ganghwa exceeded that of the population at Muan. Despite considerable geographic separation (400 km) between the two regions(Gangwha and Muan), our haplotype network suggests that the Gangwha and Muan populations of R. esculentum are related. The simple and monotonous genetic structure of the Muan population shows that R. esculentum emergence is relatively recent. In contrast, the Gangwha population shows evolution. Moreover, jellyfish of the Gangwha population are genetically diverse and remain constant despite environmental fluctuations in the Han River. The Gangwha area is considered to be the old origin of R. esculentum in Korea.

  • Evasion, approach and predation are examples of innate behaviour that are fundamental for the survival of animals. Uniting these behaviours is the assessment of threat, which is required to select between these options. Far from being comprehensive, we give a broad review over recent studies utilising optic techniques that have identified neural circuits and genetic identities underlying these behaviours.

Evaluation of Genetic Effects of Demographic Bottleneck in Muzzafarnagri Sheep from India Using Microsatellite Markers

  • Genetic variability is an important component in the ability of populations to adapt in the face of environmental change. Severe human impacts reduced Muzzafarnagri sheep of India from 500,000 in 1972 to 10,989 in 1973-74. Here we report for the first time the effect of this population decline on levels of genetic variability at 13 FAO recommended ovine microsatellite loci and contrast levels of variability to that in a breed from the same geographical region, which differed in numbers, by an order of magnitude (Marwari sheep). Of the 13 loci, 100% were polymorphic in both breeds. A high degree of genetic variation was observed within populations in terms of both allele diversity (number of alleles per locus, >4) and gene diversity (expected heterozygosity, >0.5), which implied that there is still a substantial amount of genetic diversity at the nuclear loci in a declining population. Nevertheless, overall low number of alleles per locus and relatively less abundance of low frequency alleles in Muzzafarnagri sheep suggested that genetic variability has been comparatively reduced in this population. Bottleneck analysis indicated that a genetic bottleneck did not occur during the most recent decline. In addition, we found that the differentiation among populations was moderate ($F_{ST}$= 11.8%). This study on assessment of genetic effects of the population declines in ovines is a step towards identification of genetically impoverished or healthy populations, which could prove to be a useful tool to facilitate conservation planning in this important species of small ruminants.

국내 종양유전상담 간호사를 위한 단기 교육프로그램 개발 (The First Korean Cancer Genetic Counseling Program for Nurses)

  • Genetic knowledge for oncology nurses is important in Korea because oncologists are incorporating genetic counseling and genetic testing into their practice. The purpose of this paper is to describe our method of developing the first academic cancer genetic risk assessment and counseling course for Korean nurses. A one-week (non-credit) cancer genetics counseling program was constructed for master's level Korean oncology nurses. The course emphasized basic genetic concepts and principles the genetics of cancer; hereditary cancer syndromes; family history assessments; pedigree construction; risk calculation; surveillance recommendations and treatment options ethical, legal, social, and psychological issues inherent in genetic testing. The goals of this program are to: 1) provide a comprehensive knowledge base for nurses who are currently expanding their scope of practice into the genetic counseling role 2) introduce this knowledge to nurses who want to use it in their practice; and 3) provide cancer genetic knowledge and resources to Korean nursing faculty who plan to incorporate this knowledge into existing master's courses. This academically-based course is recognized as valuable by nurses, nursing faculty, and physicians. With this new knowledge nurses can begin toexpand their role in delivering comprehensive cancer care services.

Genetic diversity and population structure of endangered Neofinetia falcata (Orchidaceae) in South Korea based on microsatellite analysis

  • Population genetic assessment is essential for the conservation and management of endangered and rare plants. Neofinetia falcata is endangered epiphyte orchid and protected by law in Korea. In Korea, this species is only found on islands in the South Sea of Korea (including Jeju-do) and the southern coast of the Korean Peninsula. We developed nine microsatellite makers to assess the genetic diversity and population genetic structure of three populations of N. falcata. The genetic diversity at the species level was low, which can be attributed to inbreeding or fragmentation into small, isolated populations. A recent bottleneck was detected in one population, likely due to overcollection. N. falcata exhibited moderated levels of differentiation among populations, with the three populations were divided into two clusters based on genetic structure. The genetic diversity and structure of N. falcata are affected by restricted gene flow by pollen or seeds due to isolation and geographic distance. Strategies for in situ and ex situ conservation of this species are been proposed based on the results of our study.

EVALUATION OF GENETIC TOXICITY FROM ENVIRONMENTAL POLLUTANTS IN DAPHNIA MAGNA AND CHIRONOMUS TENTANS FOR APPLICATION IN ECOLOGICAL RISK ASSESSMENT

  • The genetic toxicity of environmental pollutants, namely, nonylphenol (NP), bisphenol A (BPA) and chloropyriphos (CP) was investigated in aquatic sentinel species, freshwater crustacean, Daphnia magna, and larva of aquatic midge, Chironomus tentans, using Comet assay. Physiological effect of such pollutants was also investigated by studying the specimens' rates of reproduction, growth and survival. Acute toxicity results showed that, as expected, Daphnia was more sensitive than Chironomus to chemical exposure. The order of acute toxicity was CP > NP > BPA in D. magna and NP > CP > BPA in C. tentans. BPA may exert a genotoxic effect on D. magna and C. tentans, given that DNA strand breaks increased in both species exposed to this compound, whereas NP- and CP-induced DNA damage occurred only in C. tentans. In vivo genotoxic data obtained in aquatic sentinel species could provide valuable information for freshwater quality monitoring. The experiments with NP-exposed D. magna showed that the pollutant has long-term effects on reproduction, whereas no short-term effect on DNA integrity was found, being an example of a false-negative result from the biomarkers perspective. This result could be interpreted that other mechanism than genetic alteration might be involved in NP-induced reproduction failure in D. magna. False-positive results from the genotoxic biomarker obtained in BPA-exposed D. magna and in NP-exposed C. tentans make it difficult to use DNA integrity as an early warning biomarker. However, as the mere presence of genotoxic compounds, which are potentially carcinogenic, is of high concern to human and ecosystem health, it could also be important to rapidly and effectively detect genotoxic compounds in the aquatic system in ways that do not necessarily accompany a higher level of alteration. Considering the potential of D. magna and C. tentans as bioindicator species, and the importance of genotoxic biomarkers in ecotoxicity monitoring, DNA damage in these species could provide useful information for environmental risk assessment.
