Mitochondrial DNA variation and phylogeography of native Mongolian goats

  • Objective: Mongolia is one of a few countries that supports over 25 million goats, but genetic diversity, demographic history, and the origin of goat populations in Mongolia have not been well studied. This study was conducted to assess the genetic diversity, phylogenetic status and population structure of Mongolian native goats, as well as to discuss their origin together with other foreign breeds from different countries using hypervariable region 1 (HV1) in mtDNA. Methods: In this study, we examined the genetic diversity and phylogenetic status of Mongolian native goat populations using a 452 base-pair long fragment of HVI of mitochondrial DNA from 174 individuals representing 12 populations. In addition, 329 previously published reference sequences from different regions were included in our phylogenetic analyses. Results: Investigated native Mongolian goats displayed relatively high genetic diversities. After sequencing, we found a total of 109 polymorphic sites that defined 137 haplotypes among investigated populations. Of these, haplotype and nucleotide diversities of Mongolian goats were calculated as 0.997±0.001 and 0.0283±0.002, respectively. These haplotypes clearly clustered into four haplogroups (A, B, C, and D), with the predominance of haplogroup A (90.8%). Estimates of pairwise differences (Fst) and the analysis of molecular variance values among goat populations in Mongolia showed low genetic differentiation and weak geographical structure. In addition, Kazakh, Chinese (from Huanghuai and Leizhou), and Arabian (Turkish and Baladi breeds) goats had smaller genetic differentiation compared to Mongolian goats. Conclusion: In summary, we report novel information regarding genetic diversity, population structure, and origin of Mongolian goats. The findings obtained from this study reveal that abundant haplogroups (A to D) occur in goat populations in Mongolia, with high levels of haplotype and nucleotide diversity.

Microsatellite makers를 이용한 마품종 간의 평가 및 유전적 다양성 (Assessment of Genetic Diversity of Horse Breeds Using Microsatellite Makers)

  • 본 연구는 마품종 간의 유전적 배경과 다양성을 분석하기 위해 제주마, 경주마, 더러브렛, 몽고마, 쿼터호스, 일본마를 기초 축으로 생물체 집단 간의 유전적 배경 및 관련성을 연구하는데 도움 되는 microsatellite marker를 사용하여 13종의 좌위에 대한 개체별 유전자형을 분석하여 대립 유전자별 빈도에 있어 품종별 차이를 보이는 microsatellite loci들을 나타냈다. 전체 개체들의 유전적 다양성을 나타내는 유전자 좌위별 품종별 기대 이형질성은 0.669-0.869를 나타냈고, 전체 집단 간의 유전자 분화정도는 0.061-0.219를 나타냈다. 이러한 대립 유전자 빈도를 근거로 하여 DISPAN 프로그램을 이용하여 집단 간 표준 유전적 거리를 도식하여 유전자 좌위별 유전적 거리는 0.137-0.414를 나타내어 마품종간의 유전적 다양성과 관련성을 고찰할 수 있게 되었다. 따라서 초위성체 유전자 표지를 이용하여 집단 내의의 혈통분석 및 분자 진화학을 연구하는데 도움이 될 것으로 사료된다.

한의학과 개인맞춤의학에 대한 소고; 변증논치에 근거한 '증 기반 개인맞춤의학' (A Review on Korean Medicine and Personalized Medicine: Syndrome-based Personalized Medicine on the Basis of Syndrome Differentiation and Treatment)

  • Objectives: This study aimed to review the characteristics of personalized medicine and Korean medicine, and the correlation between personalized medicine and Korean medicine. Methods: We investigated various studies in PubMed, Scopus and domestic Korean medicine journals. In addition, we discussed the topic based on literature. Results: Western medicine developed as evidence-based medicine. However, its limitations are being reached, so a new paradigm of medicine is needed. As a result, personalized medicine has appeared. Recently, through the development of human genomics, personalized medicine has been researched on the basis of individual genetic characteristics. Korean medicine has developed with a unique holistic approach and treats not the disease itself but the patient's body. Its characteristic is well expressed through syndrome differentiation and treatment. Syndrome differentiation represents the nature of person-centered medicine and becomes the root of personalized medicine. Conclusions: Compared with genome-based personalized medicine of Western medicine, Korean medicine could be classified as syndrome-based personalized medicine. It would be great to apply this characteristic to clinical practices.

Cultural conditions affect somatic embryogenesis in Catharanthus roseus L. (G.) Don

  • We established an efficient plant regeneration system for Catharanthus roseus L. (G.) Don through somatic embryogenesis. Embryogenic callus was induced from hypocotyl of seed germinated in vitro. Somatic embryogenesis in Catharanthus has been categorized into three distinct stages: (1) initiation and proliferation of embryo; (2) maturation, and; (3) germination or plantlet conversion. Beside plant growth regulators, various stages of embryogenesis were screened for their response to a wide variety of factors (pH, gelrite, light, sugar alcohols, polyethyleneglycol and amino acids), which affect embryogenesis. All of the tested factors had a small to marked influence on embryogeny and eventual conversion to plantlets. The plantlets were acclimatized successfully in a greenhouse. To our knowledge, this is the first report describing a detailed study of various cultural factors which regulate embryogenesis in C. roseus. The results discussed in this paper may be used in mass propagation to produce medicinal raw material, and the embryo precursor cells could be used in genetic modification programmes that aim to improve the alkaloid yield as well.

Effect of Nitrogen Split Application Methods on Development of Vascular Bundle and Yield Components of Rice Cultivars

  • This experiment was conducted to evaluate the effect of split application of nitrogen(N) on development of vascular bundle(VB) and yield components of rice. Two cultivars were used in this study; IR58, an indica type and Shinunbongbyeim a japonica type. The number and total cross sectional area of the VB in the peduncle and leaf blade were more and bigger in N split application than 100 percent basal fertilizer. Nitrogen split application at necknode differentiation stage increased the number and size of the VB. Nitrogen split application resulted in increased panicle number with application of N before transplanting and at tillering stage; increased spikelets number with N application at necknode differentiation stage; and increased spikelet fertility and 1000 grain weight with N application at necknode differentiation and heading stages. Grain yield increased 7-10% in N split as compared to all basal application. The total cross sectional area of VB in peduncle closely correlated with the number of spikelets per panicle. Nitrogen management can have an impact on spikelet differentiation through more and bigger VB and increase grain yield potential.

StrokeMed: an integrated literature database for stroke and the differentiation of stroke syndrome

  • Complex diseases, such as stroke and cancer, have two or more genetic influences and are affected by environmental factors, which complicate them. Due to the complex characteristics of these diseases, we must search and study comprehensive literature-based article resources. Some disease-related literature databases have been developed through specialized journal issues or major websites. Most of them, however, are scattered throughout a website, and users encounter difficulties in finding accurate and comprehensive information easily and quickly. We developed StrokeMed, an integrated literature database for stroke and the differentiation of stroke syndrome. The system allows users to explore PubMed search results, categorized by MeSH (Medical Subject Headings), and the differentiation of stroke syndrome in Oriental medicine. StrokeMed collects data from important sites, such as PubMed, Scirus, and Scopus, automatically to maintain higher-quality and updated content. Currently, the system indexes more than 20,000 PubMed abstracts that are related to stroke, stroke etiology, and Oriental medicine. The system provides valuable literature information to the scientific and medical fields in stroke.

Genotype Fingerprinting, Differentiation and Association between Morphological Traits and SSR Loci of Soybean Landraces

  • Fifty-nine Korean soybean (Glycine max L. Merr.) landrace accessions were tested for genotype fingerprinting, differentiation and association between morphological traits and SSR profile. Using 8 SSR loci, 59 varieties were divided into 55 groups, and only 4 pairs of varieties were not uniquely identified. The resolving power of SSR for soybean genotyping was much higher than that of the morphological traits that were studied. Identification efficiency also differed among SSR loci. Those loci with higher numbers of alleles distinguished varieties more effectively. Genetic differentiation values of the soybean landraces varied from 0.57 to 0.82 with a mean of 0.68. The number of alleles detected by the 8 loci ranged from 3 to 8. and the effective number of alleles ranged from 2.3 to 5.1. In a study of the association of SSR alleles with morphological traits, some alleles seemed to be related with some specific morphological traits. Comparison of two kinds of dendrograms which were derived from SSR markers and quantitative traits indicated that the dendrograms were not consistent. Considering the correlation between single SSR locus and qualitative traits governed by major genes, the data suggest that alleles of microsatellite loci be more closely related to some traits determined by major genes than those determined by minor genes.
