Study on Production of Cloned Animals by Recycling Nuclear Transplantation II. Improved Second Generation Cloning of Rabbit Embryos Using Donor Nuclei with Synchronized Cell Cycles (반복핵이식에 의한 복제동물 생산에 관한 연구 II. 토끼에서 공핵배의 세포주기 조절에 의한 제2세대 복제배의 생산효율 개선)

  • large scale production of cloned embryos requires the technology of multiple generation nuclear transplantation(NT) using NT embryos as the subsequent donor nuclei. The purposes of this study were producing the second generation cloned rabbit embryos, and also to determine the electrofusion rate and in vitro developmental potential comparatively in the cloned embryos of the first and second NT generation. The embryos of 16-cell stage were collected from the mated does by flushing oviducts with Dulbecco's phosphate buffered saline(D-PBS) containing 10% fetal calf serum(FCS) at 47 hours after hCG injection In the first generation NT, the nuclear donor embryos were synchronized in the phase of Gi /S transition of 32-cell stage. The first generation NT embryos which were developed to 8-cell were synchronized in Gi /S transition phase of the following 16-cell stage and used as donor nuclei for second generation Synchronization of the cell cycle of blastomeres was induced, first, using an inhibitor of microtuble polymerization, colcemid for 10 hours to arrest blastomeres in M phase, and secondly, using a DNA synthesis inhibitor, aphidicolin for 1.5 to 2 hours to arrest them in Gi /S transition boundary. The recipient cytoplasms were obtained by removing the nucleus and the first polar body from the oocytes collected at 14 hours after hCG injection. The separated donor blastomeres were injected into the enucleated recipient oocytes by micromanipulation and were electrofused by electrical stimulation of three pulses for 60 $\mu$sec at 1.25 kV /cm in 0.28 M rnannitol solution The fused oocytes were co-cultured with a monolayer of rabbit oviductal epithelial cells in M-199 solution containing 10% FCS for 120 hours at 39$^{\circ}C$ in a 5% $CO_2$ incubator. Following in vitro culture of the first and second generation cloned embryos to blastocyst stage, they were stained with Hoechst 33342 dye for counting the number of blastomeres by fluorescence microscopy. The results obtained were summarized as follows: 1. The electrofusion rate was found to be similar as 79.4 and 91.5% in the first and second generation NT rabbit embryos, respectively. 2. The in vitro developmental potential to blastocyst stage of the second generation NT embryos (23.3%) was found significantly(p<0.05) lower, compared with that of the first generation NT embryos (56.8%). 3. The mean blastomeres counts of embryos developed to blastosyst stage following in vitro culture for 120 hours and also their daily cell cycles during the culture period were decreased significantly (p<0.05) to 104.3 cells and 1.33 cylces in the second NT generation, compoared with 210.4 cells and 1.54 cycles in the first NT generation, respectively.

Effect of Family Characteristics of the Baby Boomer Generation on the Level of Preparation for Old Age (베이비부머 세대의 가족 특성이 노후준비수준에 미치는 영향)

  • The purpose of this study was to investigate the effect of family characteristics of the baby boomer generation on the level of their later life preparation. This study randomly sampled a birth cohort between 1955 and 1963 consisting of participants living in Seoul, South Korea with a spouse and a child/children. This study investigated a total of 455 subjects from March 2 to March 20, 2016. The study findings are summarized as follows. This study analyzed the characteristics of baby boomers' family relationships and their effects on their later life preparation. It was found that 55.4% of the subjects had at least 2 children, and 44.6% had 1 child. Concerning their children's marital status, 33.8% had married children and 66.2% had unmarried children. The level of their children's support was found at 2.82 points (standard deviation [SD] = .64), which is slightly higher than the mean value of 2.5. The spouse satisfaction among the subjects was found to be 3.59 (.79) which is higher than the mean value of 3. The general status of later life preparation was 2.70 (SD = .44), which is lower than the mean value of 3. In the sub-dimension, the level of social preparation was 2.98 points (SD = .61); the level of economic preparation was 2.60 (SD = .64); and the level of physical preparation was 2.53 (SD = .45). All the values were lower than the mean value of 3. The overall status of later life preparation of the subjects in this study was low. The physical preparation level was particularly low. To analyze the factors that affect baby boomers' later life preparation, a hierarchical regression analysis was implemented. As a result, a significant effect was found in specific factors, such as spouse satisfaction (${\beta}$ = .32, p < .001), age (${\beta}$ = .26, p < .001), number of children (${\beta}$ = -.18, p <.001), health status (${\beta}$ = .18, p < .001), gender (${\beta}$ = -.11, p < .05), household income (${\beta}$ = .10, p < .05), and children's marital status (${\beta}$ = .10, p < .05). That is, the higher the spousal satisfaction, the older the age, and the lower the number of children, the higher the levels of later life preparation. Further, a higher level of later life preparation was observed in women, those with higher household incomes, and those with married children.

Wind power forecasting based on time series and machine learning models (시계열 모형과 기계학습 모형을 이용한 풍력 발전량 예측 연구)

  • Wind energy is one of the rapidly developing renewable energies which is being developed and invested in response to climate change. As renewable energy policies and power plant installations are promoted, the supply of wind power in Korea is gradually expanding and attempts to accurately predict demand are expanding. In this paper, the ARIMA and ARIMAX models which are Time series techniques and the SVR, Random Forest and XGBoost models which are machine learning models were compared and analyzed to predict wind power generation in the Jeonnam and Gyeongbuk regions. Mean absolute error (MAE) and mean absolute percentage error (MAPE) were used as indicators to compare the predicted results of the model. After subtracting the hourly raw data from January 1, 2018 to October 24, 2020, the model was trained to predict wind power generation for 168 hours from October 25, 2020 to October 31, 2020. As a result of comparing the predictive power of the models, the Random Forest and XGBoost models showed the best performance in the order of Jeonnam and Gyeongbuk. In future research, we will try not only machine learning models but also forecasting wind power generation based on data mining techniques that have been actively researched recently.

  • Objectives : This study was conducted to investigate the effect of 1-year administration of second-generation antipsychotics (SGAs) on the platelet activity in patients with schizophrenia through a retrospective review of the medical records. Methods : The mean platelet component (MPC) value was used as an index of the platelet activity. The included subjects (N=24) were the patients who were confirmed to have taken SGAs continuously for one year after the first MPC measurement had been performed. The change of MPC was verified through a paired sample t-test. Results : The result revealed that the mean MPC value was significantly decreased from 26.5±1.4 g/dL to 25.6±1.8 g/dL after 1-year administration of SGAs from the time of the first MPC measurement. Conclusions : This study suggests that 1-year administration of SGAs may be related with increased platelet activity, and that close monitoring for risks such as cerebrovascular/cardiovascular or thromboembolic diseases may be necessary during SGAs treatment in clinical practice.

  • Excessive heat generation at the implant-bone interface may cause irreversible bone damage and loss of osseointegration. The effect of heat generation in vitro at the implant surface caused by abutment reduction with high-speed dental turbine was examined. Titanium-alloy abutments connected to a titanium alloy screw-implant embedded in an acrylic-resin block in a $37^{\circ}C$ water bath were prepared. Temperature changes were recorded via embedded thermocouples at the cervix and apex of the implant surface. Analysis of variance for repeated measures was used to compare seven treatment groups. Fifty seconds of continuous cutting with air and water coolant caused a mean temperature increase of $1.24^{\circ}C$ at apex and $5.77^{\circ}C$ at cervix. Similar intermittent cutting caused increase of $2.50^{\circ}C$ at apex and $1.64^{\circ}C$ at cervix. But, continuous cutting with air coolant caused a mean temperature increase of $6.47^{\circ}C$ at apex and $5.77^{\circ}C$ at cervix. Similar intermittent cutting caused increase of $6.47^{\circ}C$ at apex and $5.77^{\circ}C$ at cervix. Preparation of implant abutment does not lead to detrimental effect on peri-implant tissues provided that adequate cooling. However, without water cooling, extreme overheating could be provoked, reaching the critical temperature that would lead to irreversible bone damage within only a few seconds.

Higher-order Spectral Method for Regular and Irregular Wave Simulations

  • In this study, a nonlinear wave simulation code is developed using a higher-order spectral (HOS) method. The HOS method is very efficient because it can determine the solution of the boundary value problem using fast Fourier transform (FFT) without matrix operation. Based on the HOS order, the vertical velocity of the free surface boundary was estimated and applied to the nonlinear free surface boundary condition. Time integration was carried out using the fourth order Runge-Kutta method, which is known to be stable for nonlinear free-surface problems. Numerical stability against the aliasing effect was guaranteed by using the zero-padding method. In addition to simulating the initial wave field distribution, a nonlinear adjusted region for wave generation and a damping region for wave absorption were introduced for wave generation simulation. To validate the developed simulation code, the adjusted simulation was carried out and its results were compared to the eighth order Stokes theory. Long-time simulations were carried out on the irregular wave field distribution, and nonlinear wave propagation characteristics were observed from the results of the simulations. Nonlinear adjusted and damping regions were introduced to implement a numerical wave tank that successfully generated nonlinear regular waves. According to the variation in the mean wave steepness, irregular wave simulations were carried out in the numerical wave tank. The simulation results indicated an increase in the nonlinear interaction between the wave components, which was numerically verified as the mean wave steepness. The results of this study demonstrate that the HOS method is an accurate and efficient method for predicting the nonlinear interaction between waves, which increases with wave steepness.

Assessment of Performance on the Asian Dust Generation in Spring Using Hindcast Data in Asian Dust Seasonal Forecasting Model (황사장기예측자료를 이용한 봄철 황사 발생 예측 특성 분석)

  • This study investigated the prediction skill of the Asian dust seasonal forecasting model (GloSea5-ADAM) on the Asian dust and meteorological variables related to the dust generation for the period of 1991~2016. Additionally, we evaluated the prediction skill of those variables depending on the combination of the initial dates in the sub-seasonal scale for the dust source region affecting South Korea. The Asian dust and meteorological variables (10 m wind speed, 1.5 m relative humidity, and 1.5 m air temperature) from GloSea5-ADAM were compared to that from Synoptic observation and European Centre for medium range weather forecasts reanalysis v5, respectively, based on Mean Bias Error (MBE), Root Mean Square Error (RMSE), and Anomaly Correlation Coefficient (ACC) as evaluation criteria. In general, the Asian dust and meteorological variables in the source region showed high ACC in the prediction scale within one month. For all variables, the use of the initial dates closest to the prediction month led to the best performances based on MBE, RMSE, and ACC, and the performances could be improved by adjusting the number of ensembles considering the combination of the initial date. ACC was as high as 0.4 in Spring when using the closest two initial dates. In particular, the GloSea5-ADAM shows the best performance of Asian dust generation with an ACC of 0.60 in the occurrence frequency of Asian dust in March when using the closest initial dates for initial conditions.

  • I believe that establishing the purpose of research on the MZ generation's communication is an important first step in understanding this generation's unique communication style and analyzing its influence. Research on the MZ generation's communication identifies the communication characteristics of the MZ generation and the communication tools and platforms used by the MZ generation, such as social media and messenger apps, and analyzes how they differ from the existing generation. I can understand. It has been shown that the MZ generation can live happily in modern society without a complicated philosophy or a clear philosophy of life. This does not mean that life is meaningless or confusing. The MZ generation can be satisfied with simple and concrete solutions to the meaning of life, and can live without the need to completely systematize everything. In other words, their lives are not as complicated as those of previous generations and can have a variety of meanings. In other words, it does not necessarily need to be defined as a philosophical system. Although this paper cannot clearly divide the lives of the MZ generation into one philosophical system, it was nevertheless possible to see that the lives of each member of the MZ generation can have many meanings, and this meaning includes the MZ generation's unique purposes, values, I could see that they were looking for a sense of ability and a sense of self-worth.

Cerebellar Activation Related to Various Tasks Using fMRI (다양한 임무 부여시 기능적 자기공명영상에서 관찰된 소뇌의 활성화)

  • Purpose : Although it's been known for half a century that unique structures have evolved in the cerebellum and they then became greatly enlarged in the human brain, the function of these structures still remains unknown. The purpose of this study was to assess cerebellar activation during motor, sensory, word generation, listening comprehension, and working memory tasks with using functional magnetic resonance imaging (fMRI). Materials and Methods : Eleven healthy right-handed subjects (Male: female, 6:5, mean age: 27.4years) were imaged on a Siemens 1.5T scanner. Whole brain functional maps were acquired using BOLD EPI sequences in the axial plane. Each paradigm consisted of five epochs of activation vs. the control condition. The activation tasks consisted of left finger complex movement, sensory stimulation of the left hand, word generation, listening comprehension, and working memory tasks. The reference function was a boxcar waveform. The activation maps were thresholded at p = 0.001. SPM 5 evaluated the activated areas and responses within the cerebellum. Results : Cerebellar activation was observed on motor task, word generation task, and working memory task. There were 949 activated areas and the mean fitted and adjusted response was 0.68 during the motor task. There were 319 activated areas and the mean fitted and adjusted response was 0.15 during the word generation task. There were 330 activated areas and the mean fitted and adjusted response was 0.26 during the working memory task. Conclusion : Our results suggest that the cerebellum is involved in a variety of functional tasks, including motor, word generation, and working memory tasks. However, during the motor task, the cerebellum showed a large activated area and a high response. Cerebellar function can be evaluated by fMRI.

DTM GENERATION OF RADARSAT AND SPOT SATELLITE IMAGERY USING GROUND CONTROL POINTS EXTRACTED FROM SAR IMAGE

  • Ground control points(GCPs) can be extracted from SAR data given precise orbit for DTM generation using optic images and other SAR data. In this study, we extract GCPs from ERS SAR data and SRTM DEM. Although it is very difficult to identify GCPs in ERS SAR image, the geometry of optic image and other SAR data are able to be corrected and more precise DTM can be constructed from stereo optic images. Twenty GCPs were obtained from the ERS SAR data with precise Delft orbit information. After the correction was applied, the mean values of planimetric distance errors of the GCPs were 3.7m, 12.1 and -0.8m with standard deviations of 19.9m, 18.1, and 7.8m in geocentric X, Y, and Z coordinates, respectively. The geometries of SPOT stereo pair were corrected by 13 GCPs, and r.m.s. errors were 405m, 705m and 8.6m in northing, easting and height direction, respectively. And the geometries of RADARS AT stereo pair were corrected by 12 GCPs, and r.m.s. errors were 804m, 7.9m and 6.9m in northing, easting and height direction, respectively. DTMs, through a method of area based matching with pyramid images, were generated by SPOT stereo images and RADARS AT stereo images. Comparison between points of the obtained DTMs and points estimated from a national 1 :5,000 digital map was performed. For DTM by SPOT stereo images, the mean values of distance errors in northing, easting and height direction were respectively -7.6m, 9.6m and -3.1m with standard deviations of 9.1m, 12.0m and 9.1m. For DTM by RADARSAT stereo images, the mean values of distance errors in northing, easting and height direction were respectively -7.6m, 9.6m and -3.1m with standard deviations of 9.1m, 12.0m and 9.1m. These results met the accuracy of DTED level 2
