Difference in Susceptibility of Tyrosine Residue to Oxidative Iodination between a Thioredoxin Box Region and a Hormonogenic Region

  • Peptide fragments, isolated from proteolytic cleavage of thyroglobulin at specific sites, were examined for the iodination of tyrosine residues. The 50 kDa polypeptide, which was prepared from digestion of bovine thyroglobulin and continuous preparative SDS-PAGE, was subjected to reduction with DTT and alkylation with iodoacetic acid to generate S-car-boxymethylated peptide derivative, which was further hydrohysed by endoproteinase-Asp-N. Peptide products were separated by RP-HPLC, and each fraction was analyzed by LC/ESI-MS and MALDI-MS analyses. Based on the specificity of endoproteinase-Asp-N andthe mass spectra data, a peptide fragment turned out to correspond to a peptide, DALCCVKCPEGSYFQ (1438-1452), characterized by the presence of a thioredoxin box (CVKC) and a tyrosine residue. In addition, another peptide fragment (1453-1465) containing a thioredoxin box (CIPC) and a tyrosine residue was also observed. However, any evidence of iodination of the tyrosine residue present in these peptides was not provided. Meanwhile, tyrosine residues in the peptides, DVEEALAGKYLAGRFA (1366-1381) and DYSGLLLAFQVFLL (1290-1303) were found to be iodinated; mono- or diiodinated tyrosine residues, characteristic of a hormogenic site, existed in both peptides. In addition, the tyrosine residue in the peptide (1218-1252), corresponding to a hormonogenic site was also iodinated. Thus, there was a sharp difference of the susceptibility to oxidative iodination between the tyrosine residue in a hormonogenic site and that in a thioredoxin region. From these results, it is suggested that polypeptide region adjacent to tyrosine residues may govern the susceptibility of tyrosine to oxidative iodination.

  • There are many possible disturbance sources on such a spacecraft, but reaction wheel assembly (RWA) which is generally used for spacecraft attitude control is anticipated to be the largest. These effects on degradation of performance of spacecraft such as attitude stability. In reaction wheel, disturbance caused by imbalance and speed error. It is hard to emulate speed error disturbance because it is not coincide with wheel frequency. This paper concentrates on emulating and analyzing the speed error disturbance. Firstly, classify the causes that lead to speed error disturbance which generate RPM fluctuation. Secondly, simulated with disturbance driver module and reaction wheel assembly which are developed by Spacecraft Control Lab. Experimental investigations have been carried out to test the disturbance emulator module as a disturbance generator for RWA. Measurements and test have been conducted on various fault. Frequency analysis of test data show that speed error disturbance effects on wheel settling wheel speed or fluctuation type.

  • The purpose of this study is to design and implement web-based quality cost management system to measure the performance of quality improvement activities in the business firms. The designed web-based quality cost management system which is also known as WQCMS (Web-based Q-Cost Management System) utilized newly classified objective performance indicator items to measure quality related activities. Well-classified performance measurement Indicator item lists which are played key role in the system was researched and standardized according to size and business categories of enterprise. Its code standards are based from fundamental PAF Model. Developed WQCMS have ability to collect and analyze quality data generated from various different departments in the inside or outside of the enterprise without any limitations, if end-users are able to access wide area network. It provides the capability to integrate quality information from database and to generate various easy analysis reports to management's needs using built-in analysis tool modules with real-time. The proposed system was developed using Microsofi's .Net technology, ASP.NET and MS-SQL Server 2000. By web-enabling Q-cost management system, the effectiveness of the system management and utilization was realized by easiness of information Integration and economical efficiency.

NEW RESULTS TO BDD TRUNCATION METHOD FOR EFFICIENT TOP EVENT PROBABILITY CALCULATION

  • A Binary Decision Diagram (BDD) is a graph-based data structure that calculates an exact top event probability (TEP). It has been a very difficult task to develop an efficient BDD algorithm that can solve a large problem since its memory consumption is very high. Recently, in order to solve a large reliability problem within limited computational resources, Jung presented an efficient method to maintain a small BDD size by a BDD truncation during a BDD calculation. In this paper, it is first identified that Jung's BDD truncation algorithm can be improved for a more practical use. Then, a more efficient truncation algorithm is proposed in this paper, which can generate truncated BDD with smaller size and approximate TEP with smaller truncation error. Empirical results showed this new algorithm uses slightly less running time and slightly more storage usage than Jung's algorithm. It was also found, that designing a truncation algorithm with ideal features for every possible fault tree is very difficult, if not impossible. The so-called ideal features of this paper would be that with the decrease of truncation limits, the size of truncated BDD converges to the size of exact BDD, but should never be larger than exact BDD.

Genetic Distances and Variations of Three Geographic Hairtail Populations Identified by PCR Analysis

  • In the present study, muscle tissues were obtained separately from individuals from Atlantic hairtail population (AHP), Gunsan hairtail population (GHP) and Chinese hairtail population (CHP), respectively. The seven decamer primers were used to generate the shared loci, specific, unique shared loci to each population and shared loci by the three hairtail populations. Here, averagely, a decamer primer generated 64.7 amplified products per primer in the AHP population, 55.7 in GHP population and 56.4 in CHP population. The number of unique shared loci to each population and number of shared loci by the three populations generated by genetic analysis using 7 decamer primers in AHP, GHP and CHP population. 119 unique shared loci to each population, with an average of 17 per primer, were observed in the AHP population, and 28 loci, with an average of 4 per primer, were observed in the CHP population. The hierarchical dendrogram point out three main branches: cluster 1 (ATLANTIC 01 ~ ATLANTIC 07), cluster 2 (GUNSAN 08 ~ GUNSAN 14) and cluster 3 (CHINESE 15 ~ CHINESE 21). The shortest genetic distance displaying significant molecular difference was between individuals' CHINESE no. 16 and CHINESE no. 18 (0.045). In the long run, individual no. 01 of the AHP population was most distantly related to CHINESE no. 19 (genetic distance = 0.430). Consequently, PCR analysis generated on the genetic data displayed that the geographic AHP population was widely separated from CHP population, while individuals of CHP population were fairly closely related to those of GHP population.

  • The MPC(Moisture Protected Computer) system is completely unique. It was recently invented in Korea and it is a technological break-through in the field of computer systems. It will be instrumental in the development of a complete moisture air-tight capability and will work well in field environments. The MPC includes the following: 1) An enclosed design. 2) Moisture proof computer enclosure joints. 3) System dust protection & an impact buffing system. 4) A normal temperature maintenance system when the temperature inside the computer is low. 5) The ability to generate heat and a radiate system inside the computer 6) An automatic power input sensing controller device. 7) A safety device in case of mis-operation. 8) A proper admission procedure for various tests. Because of the above, and as a result of temperature property experiments, there aren't any operational problems. The result of this treatise could provide very important data for verification of cases involving: using strong materials to protect against explosions, searching for ways to make the size and weight small, and for making repairs easier. As a result, it could achieve price and function competitiveness in advanced countries such as the USA and in Europe.

Opinion-Mining Methodology for Social Media Analytics

  • Social media have emerged as new communication channels between consumers and companies that generate a large volume of unstructured text data. This social media content, which contains consumers' opinions and interests, is recognized as valuable material from which businesses can mine useful information; consequently, many researchers have reported on opinion-mining frameworks, methods, techniques, and tools for business intelligence over various industries. These studies sometimes focused on how to use opinion mining in business fields or emphasized methods of analyzing content to achieve results that are more accurate. They also considered how to visualize the results to ensure easier understanding. However, we found that such approaches are often technically complex and insufficiently user-friendly to help with business decisions and planning. Therefore, in this study we attempt to formulate a more comprehensive and practical methodology to conduct social media opinion mining and apply our methodology to a case study of the oldest instant noodle product in Korea. We also present graphical tools and visualized outputs that include volume and sentiment graphs, time-series graphs, a topic word cloud, a heat map, and a valence tree map with a classification. Our resources are from public-domain social media content such as blogs, forum messages, and news articles that we analyze with natural language processing, statistics, and graphics packages in the freeware R project environment. We believe our methodology and visualization outputs can provide a practical and reliable guide for immediate use, not just in the food industry but other industries as well.

Target segmentation in non-homogeneous infrared images using a PCA plane and an adaptive Gaussian kernel

  • We propose an efficient method of extracting targets within a region of interest in non-homogeneous infrared images by using a principal component analysis (PCA) plane and adaptive Gaussian kernel. Existing approaches for extracting targets have been limited to using only the intensity values of the pixels in a target region. However, it is difficult to extract the target regions effectively because the intensity values of the target region are mixed with the background intensity values. To overcome this problem, we propose a novel PCA based approach consisting of three steps. In the first step, we apply a PCA technique minimizing the total least-square errors of an IR image. In the second step, we generate a binary image that consists of pixels with higher values than the plane, and then calculate the second derivative of the sum of the square errors (SDSSE). In the final step, an iteration is performed until the convergence criteria is met, including the SDSSE, angle and labeling value. Therefore, a Gaussian kernel is weighted in addition to the PCA plane with the non-removed data from the previous step. Experimental results show that the proposed method achieves better segmentation performance than the existing method.

Characteristics Analysis of Flex Link according to Mold Clamping Force in Injection Molding Machine (형체력에 따른 사출성형기 플렉스링크의 특성 분석)

  • Reproducibility of injection molding machines are studied at the study of this time. We applied computer aided engineering program so it could generate clamping force, about 1,500 kN, to the nozzle center part of flex link in tie-bar and at this time, we made sure condition of stress distribution and transformation quantity in flex link. The result of computer aided engineering transformation quantity was confirmed that transformation of top area was 247~257 kN and bottom areas was 273~279 kN and also was confirmed that the stresses are distributed in a range of 57~750 $N/mm^2$ from top to the bottom of the surface. This time we could confirm the condition of transformation quantity and stress distribution by enforcing the previously used structure analysis of flex link. And we utilized the reference data to establish important point of section for non destructive test overhaul.

Zinc-induced Apoptosis in C6 glial Cells via Generation of Hydrogen Peroxide($H_2O_2$) (신경교세포주 C6 glial에서 Zinc의 Hydrogen Peroxide($H_2O_2$) 생성을 통한 세포고사)

  • Zinc is known to generate reactive oxygen species (ROS) including superoxide anion and hydrogen peroxide ($H_2O_2$), which eventually contribute to cytotoxicity in a variety of cell types. Here in, we demonstrated that zinc decreased the viability of C6 glial cells in a time and dose-dependent manner, which was revealed as apoptosis characterized by ladder-pattern fragmentation of genomic DNA. chromatin condensation and DNA fragmentation in Hoechst dye staining. Zinc-induced apoptosis of C6 glial cells was prevented by the addition of catalase and antioxidants including reduced glutathione (GSH), N-acetyl-L-cysteine (NAC) and pyrrolidinedithiocarbamate (PDTC). Wefurther confirmed that zinc decreased intrac-ellular levels of GSH and generated $H_2O_2$in C6 glial cells. Moreover, antioxidants also decreased the generation of zinc-induced $H_2O_2$ in C6 glial cells. These data indicated that zinc-induced the apoptotic death of C6 glial cells via generation of reactive oxygen species such as $H_2O_2$.
