유전자 치료에서 PET의 역할 (Role of PET in Gene Therapy)

  • In addition to the well-established use of positron emission tomography (PET) in clinical oncology, novel roles for PET are rapidly emerging in the field of gene therapy. Methods for controlled gene delivery to living bodies, made available through advances in molecular biology, are currently being employed in animals for research purposes and in humans to treat diseases such as cancer. Although gene therapy is still in its early developmental stage, it is perceived that many serious illnesses could be treated successfully by the use of therapeutic gene delivery. A major challenge for the widespread use of human gene therapy is to achieve a controlled and effective delivery of foreign genes to target cells and subsequently, adequate levels of expression. As such, the availability of noninvasive imaging methods to accurately assess the location, duration, and level of transgene expression is critical for optimizing gene therapy strategies. Current endeavors to achieve this goal include methods that utilize magnetic resonance imaging, optical imaging, and nuclear imaging techniques. As for PET, reporter systems that utilize genes encoding enzymes that accumulate positron labeled substrates and those transcribing surface receptors that bind specific positron labeled ligands have been successfully developed. More recent advances in this area include improved reporter gene constructs and radiotracers, introduction of potential strategies to monitor endogenous gene expression, and human pilot studies evaluating the distribution and safety of reporter PET tracers. The remarkably rapid progress occurring in gene imaging technology indicates its importance and wide range of application. As such, gene imaging is likely to become a major and exciting new area for future application of PET technology.

조직 특이 발현 Sodium Iodide Symporter 유전자 이입에 의한 방사성옥소 간암세포 치료와 광학영상을 이용한 치료효과 평가 (Radioiodine Therapy of Liver Cancer Cell Following Tissue Specific Sodium Iodide Symporter Gene Transfer and Assessment of Therapeutic Efficacy with Optical Imaging)

  • 목적: 조직 특이 프로모터를 이용하면 특정 암조직내에서만 원하는 치료유전자를 발현시킬 수 있다. 나트륨 옥소 공동 수송체(sodium iodide symporter: NIS) 유전자는 옥소를 섭취하는 특성을 가져 방사성옥소를 이용한 치료용 유전자로 사용될 수 있다. 광학 영상용 유전자인 luciferase (Luc) 유전자를 세포에 이입하면 비침습적으로 유전자가 이입된 세포의 상태를 평가할 수 있다. 본 연구는 간암 특이성을 나타내는 AFP 프로모터에 의해 발현이 조절되는 NIS유전자와 CMV프로모터에 의해 발현되는 Luc유전자를 간암세포에 이입하여 NIS유전자 이입에 의한 방사성옥소 유전자치료의 효과를 알아보고, 종양사멸 정도를 광학 리포터 유전자 발현으로 알아보고자 하였다. 대상 및 방법: AFP enhancer와 GSTP 프로모터를 연결하여 AFP프로모터를 제작하였으며 이를 NIS유전자와 연결하였다. 또한 CMV 프로모터에 조절 받는 Luc 유전자를 동시에 삽입하여 AFP-NIS-CMV-Luc 유전자 발현 벡터를 생산하였다. 실험 대상 세포주로는 간암세포주인 HepG2와 Huh-7 세포와 사람 대장암세포주인 HCT-15 세포를 이용하였다. AFP-NIS-CMV-Luc 발현벡터를 Liposome을 이용해 실험대상 세포주 내로 이입하였으며, 방사성옥소 섭취율과 방사성옥소의 유출량을 측정하였다. 또한 Luciferase 발현 정도를 luminometer로 측정하였으며, clonogenic assay를 통하여 I-131에 대한 세포주에 따른 사멸효과 차이를 알아보았다. AFP-NIS-CMV-Luc 유전자 이입 세포주를 누드마우스에 대퇴부 피하에 주입하여 I-131 축적여부를 감마카메라 영상을 획득하였다. 결과: AFP-NIS-CMV-Luc 유전자 발현 벡터를 제작하였다. AFP-NIS-CMV-Luc 유전자가 이입된 HepG2와 Huh-7 세포의 방사성옥소 섭취율은 유전자 이입이 되지 않은 대조군 HepG2와 Huh-7 세포에 비하여 높았으며, $KClO_4$를 처리시 옥소 섭취가 저해되었다. 대장암 세포주인 HCT-15세포에 AFP-NIS-CMV-Luc유전자를 이입 시 방사성옥소의 섭취률은 증가되지 않았다. 30분간 방사성옥소를 섭취시킨 AFP-NIS-Luc 유전자가 이입된 HepG2와 Huh-7 세포에서의 방사성옥소의 유출반감기는 약 4분과 6분으로 각각 나타났다. AFP-NIS-CMV-Luc 유전자가 이입된 HepG2, Huh-7세포의 Luc 유전자의 발현은 241, 441 $RLU/2\;{\times}\;10^5$ cells로 나타났으며, 대조군 HepG2와 Huh구세포에서의 Luc 유전자의 발현은 74, $RLU/2\;{\times}\;10^5$ cells로 나타났다. HCT-15 세포는 AFP-NIS-CMV-Luc 유전자 이입에 따라 I-131에 의한 세포 사멸능이 증가되지 않았으나, HepG2 및 Huh-7 세포는 FP-NIS-CMV-Luc 유전자 이 입에 따라 I-131에 의한 세포 사멸능이 증가되었으며, Huh-7세포의 경우 0.5mCi의 I-131을 투여한 경우 모든 세포가 사멸하였다. AFP-NIS-CMV-Luc 유전자가 이입된 Huh-7 세포수가 많을수록 방사성옥소 섭취율이 증가하며 luciferase활성도도 높게 나타났다. AFP-NIS-CMV-Luc 유전자가 이입된 Huh-7 세포를 이식한 누드마우스에 I-131 감마카메라 영상에서 종양이식부위에 방사능 축적을 관찰 할 수 있었다. 결론: AFP프로모터의 의하여 NIS유전자가 발현되며, CMV프로모터에 의한 Luc 유전자가 발현되는 벡터를 제작하였으며, 이 벡터를 이입한 경우 간암세포에서만 I-131의 세포 독성이 증가하는 효과를 나타내었다. 또한 Luc유전자를 이용하여 비침습적인 광학 영상으로 세포사멸 효과를 확인할 수 있었다. 간암특이 프로모터에 조절되는 치료 유전자와 광학리포터 유전자를 한 벡터에 동시에 이입하면 간암 특이 유전자 치료와 그 치료효과를 비침습적으로 평가할 수 있을 것으로 생각된다.

방풍통성산(防風通聖散)이 고지방사료(高脂肪飼料) 식이(食餌)로 유발(誘發)된 비만(肥滿)생쥐에서 비만유전자(肥滿遺傳子) 및 관련인자(關聯因子)에 미치는 영향(影響) (The Inhibitory Effects of Bangpuangtongseoung-san on the Obese gene and Obese Inhibitory about Obese-mouse induced by High Fat Diet)

  • Objectives : In this study, the aim was to investigate the inhibitory effects of Bangpungtong-seoung-san on the obese gene and obese inhibitory about obese-mouse induced by high fat diet. Methods : In order to investigate the effects of Bangpuangtongseoung-san(BPTS) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BPTS extract) and fed for 13weeks. Results : 1. The change of body weight and the final increase of body weight were decreased significantly. 2. The amount of the adipocyte in body weight was decreased significantly. 3. In primary adipocytes, ${\beta}3AR$ gene expression was increased significantly and leptin gene expression was decreased in $100{\mu}g/ml$ density significantly. 4. In primary adipocytes, the amount of TNF-${\alpha}$ was decreased in $100{\mu}g/ml$ density significantly and the amount of leptin was decreased but did not show significance. 5. In adipocytes tissue, the expression of leptin was decreased and the expression of ${\beta}3AR$ was increased. Conclusions : These results suggest that BPTS may inhibit the expression of the obese gene mad obese inhibitory about obese-mouse induced by high fat diet.

소 수정란에서 Green Fluorescent Protein 유전자 검색 및 PCR에 의한 성감별 (Screening of Green Fluorescent Protein Gene and Sexing by PCR in Bovine Embryos)

  • The efficiency of transgenic livestock production could be improved by early screening of transgene-integration and sexing of embryos at preimplantational stages before trasferring them into recipients. We examined the effciency of multiplex PCR analysis for the simultaneous confirmation of the trasgene and sex during the preimplantational development of bovine embryos and the possibility of green fluorescent protein(GFP) gene as a non-invasive marker for the early screening of transgenic embryos. The GFP gene was microinjected into the male pronuclei of bovine zygotes produced in vitro. The injected zygotes were co-cultured in TCM-199 containing 10% FCS with boving oviductal epithelial cells in a 5% CO2 incubator. Seventeen(13.0%) out of 136 gene-injected bovine zygotes developed by multiplex PCR analysis and the expression of GFP was detected by observing green fluorescence in embryos under a fluorescent microscope. Eight(67%) of 12 embryos at 2-cell to blastocyst stage were positive in the PCR analysis, but only two(11.8%) of 17 blastocysts expressed the GFP gene. Their sex was determined as 7 female and 5 male embryos by the PCR analysis. The results indicate that the screening of GFP gene and sex in bovine embryos by PCR analysis and fluorescence detection could be a promisible method for the preselection of transgenic embryos.

Polymorphisms of Transmembrane Channel-like 1 Gene are Associated with Kawasaki Disease in Korean Population

  • Kawasaki disease (KD) is believed to be infectious but etiology and the mechanism of development remain elusive. The aim of this study was to investigate the association between transmembrane channel-like 1 (TMC1) gene and KD. One hundred nine KD patients and 424 normal controls were enrolled. Of all KD patients, 34 developed coronary artery lesions (CALs). Eleven single nucleotide polymorphisms (SNPs) within TMC1 gene were selected and SNP genotyping was performed by the direct sequencing. Genotype frequencies were analyzed with the SNPAnalyzer, Helixtree, and SNPStats programs. In the present study, six SNPs (rs7851577, rs10781105, rs2589615, rs1663743, rs1373628, and rs1373626) were significantly associated with the risk of KD. In further haplotype analysis, one haplotype (CGGACCCT) showed a significant association between KD and control groups. These results suggest that TMC1 gene may be a susceptibility gene for KD in Korean population.

Significant Gene Selection Using Integrated Microarray Data Set with Batch Effect

  • In microarray technology, many diverse experimental features can cause biases including RNA sources, microarray production or different platforms, diverse sample processing and various experiment protocols. These systematic effects cause a substantial obstacle in the analysis of microarray data. When such data sets derived from different experimental processes were used, the analysis result was almost inconsistent and it is not reliable. Therefore, one of the most pressing challenges in the microarray field is how to combine data that comes from two different groups. As the novel trial to integrate two data sets with batch effect, we simply applied standardization to microarray data before the significant gene selection. In the gene selection step, we used new defined measure that considers the distance between a gene and an ideal gene as well as the between-slide and within-slide variations. Also we discussed the association of biological functions and different expression patterns in selected discriminative gene set. As a result, we could confirm that batch effect was minimized by standardization and the selected genes from the standardized data included various expression pattems and the significant biological functions.

Altered Gene Expression Profiles in the Lungs of Streptozotocin-induced Diabetic Mice

  • Diabetes mellitus is a common heterogeneous metabolic disorder, characterized by deposition of extracellular matrix, oxidative stress, and vascular dysfunction, thereby leading to gradual loss of function in multiple organs. However, little attention has been paid to gene expression changes in the lung under hyperglycemic conditions. In this study, we found that diabetes inuced histological changes in the lung of streptozotocin-induced diabetic mice. Global gene expression profiling revealed a set of genes that are up- and down-regulated in the lung of diabetic mice. Among these, expression of Amigo2, Adrb2, and Zbtb16 were confirmed at the transcript level to correlate significantly with hyperglycemia in the lung. We further evaluated the effect of human umbilical cord-derived perivascular stem cells (PVCs) on these gene expression in the lung of diabetic mice. Our results show that administration of PVC-conditioned medium significantly suppressed Amig2, Adrb2, and Zbtb16 upregulation in these mice, suggesting that these genes may be useful indicators of lung injury during hyperglycemia. Furthermore, PVCs offer a promising alternative cell therapy for treating diabetic complications via regulation of gene expression.

Lactosylceramide Mediates the Expression of Adhesion Molecules in TNF-${\alpha}$ and IFN ${\gamma}$-stimulated Primary Cultured Astrocytes

  • Here we have investigated how lactosylceramide (LacCer) modulates gene expression of adhesion molecules in TNF-${\alpha}$ and IFN ${\gamma}$ (CM)-stimulated astrocytes. We have observed that stimulation of astrocytes with CM increased the gene expression of ICAM-1 and VCAM-1. D-Threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) and N-butyldeoxynojirimycin (NBDNJ), inhibitors of glucosylceramide synthase (GLS) and LacCer synthase (galactosyltransferase, GalT-2), inhibited the gene expression of ICAM-1 and VCAM-1 and activation of their gene promoter induced by CM, which were reversed by exogenously supplied LacCer. Silencing of GalT-2 gene using its antisense oligonucleotides also attenuated CM-induced ICAM-1 and VCAM-1 expression, which were reversed by LacCer. PDMP treatment and silencing of GalT-2 gene significantly reduced CM-induced luciferase activities in NF-${\kappa}B$, AP-1, GAS, and STAT-3 luciferase vectors-transfected cells. In addition, LacCer reversed the inhibition of NF-${\kappa}B$ and STAT-1 luciferase activities by PDMP. Taken together, our results suggest that LacCer may play a crucial role in the expression of ICAM-1 and VCAM-1 via modulating transcription factors, such as NF-${\kappa}B$, AP-1, STAT-1, and STAT-3 in CM-stimulated astrocytes.

Characterization of a Novel Gene in the Extended MHC Region of Mouse, NG29/Cd320, a Homolog of the Human CD320

  • Background: The MHC region of the chromosome contains a lot of genes involved in immune responses. Here we have investigated the mouse NG29/Cd320 gene in the centrometrically extended MHC region of chromosome 17. Methods: We cloned the NG29 gene by RT-PCR and confirmed the tissue distribution of its gene expression by northern blot hybridization. We generated the NG29 gene expression constructs and polyclonal antibody against the NG29 protein to perform the immunofluorescence, immunoprecipitation and flow cytometric analysis. Results: The murine NG29 gene and its human homologue, the CD320/8D6 gene, were similar in the gene structure and tissue expression patterns. We cloned the NG29 gene and confirmed its expression in plasma membrane and intracellular compartments by transfecting its expresssion constructs into HEK 293T cells. The immunoprecipitation studies with rabbit polyclonal antibody raised against the NG29-NusA fusion protein indicated that NG29 protein was a glycoprotein of about 45 kDa size. A flow cytometric analysis also showed the NG29 expression on the surface of Raw 264.7 macrophage cell line. Conclusion: These findings suggested that NG29 gene in mouse extended MHC class II region was the orthologue of human CD320 gene even though human CD320/8D6 gene was located in non-MHC region, chromosome 19p13.

폐암세포주에 대한 p53 및 $p16^{INK4a}$의 복합종양억제유전자요법의 효과 (Enhanced Growth Inhibition by Combined Gene Transfer of p53 and $p16^{INK4a}$ in Adenoviral Vectors to Lung Cancer Cell Lines)

  • 배경 : 대표적인 종양억제유전자인 p53 및 p16은 세포주기 조절 및 자연괴멸 유도에서 서로 다른 역할을 하고 있으며 폐암에서는 동시에 비활성화 되어 있는 경우가 흔하다. 이 종양억제유전자들 동시에 이입시 서로 상호작용을 통해 효과가 증진되리라 기대되고 있다. 방법 : 본 연구에서는 p53 및 p16을 아데노바이러스 벡터를 이용하여 폐암세포주에 동시에 이입하여 그 상호작용을 관찰하였다. 복합투여시 세포성장곡선을 분석하여 isobologra을 이용한 상호작용을 검증하고 세포주기의 변화 및 체외종양형성능의 변화도 관찰하였다. 결과 : Isobologram을 이용한 분석에서 adeno-virus-p53와 adenovirus-p16의 복합투여는 NCI H358에서는 상승적인 성장억제를, NCI H23에서는 부가적인 성장 억제를 보였다. 유세포분석기를 이용한 세포주기의 분석 및 체외종양형성능 검사에서도 p53 및 p16의 복합투여시 단독 투여보다 강한 억제효과를 보였다. 결론 : 이러한 상승적인 p53 및 p16의 상호작용은 이 복합요법이 새로운 유전자치료법으로 발전할 수 있는 가능성을 보였다.
