Background: Chronic inflammation in the brain has known to be associated with the development of a various neurological diseases including dementia. In general, the characteristic of neuro-inflammation is the activated microglia over the brain where the pathogenesis occurs. Pro-inflammatory repertoires, interleukin-1${\beta}$ (IL-1${\beta}$) and nitric oxide (NO), are the main causes of neuro-degenerative disease, particularly in Alzheimer's disease (AD) which is caused by neuronal destruction. Those pro-inflammatory repertoires may lead the brain to chronic inflammatory status, and thus we hypothesized that chronic inflammation would be inhibited when pro-inflammatory repertoires are to be well controlled by inactivating the signal transduction associated with inflammation. Methods: In the present study, we examined whether biphenyl dimethyl dicarboxylate (DDB), an active compound from Schizandra chinensis Baillon, inhibits the NO production by a direct method using Griess reagent and by RT-PCR in the gene expression of inducible nitric oxide synthase (iNOS) and IL-1${\beta}$. Western blots were also used for the analysis of NF-${\kappa}B$ and I${\kappa}B$. Results: In the study, we found that DDB effectively inhibited IL-1${\beta}$ as well as NO production in BV-2 microglial cell, and the translocation of NF-${\kappa}B$ was comparably inhibited in the presence of DDB comparing those to the positive control, lipopolysaccharide. Conclusion: The data suggested that the DDB from Schizandra chinensis Baillon may play an effective role in inhibiting the pro-inflammatory repertoires which may cause neurodegeneration and the results imply that the compound suppresses a cue signal of the microglial activation which can induce the brain pathogenesis such as Alzheimer's disease.
Objective : The aim of this study was to analyse the latest trend of medical science articles about Bojungikki-Tang published in Japan. Methods : We searched medical science articles published from 2003 to 2008 in Japan on the web site, CiNii and then these articles were reviewed and classified according to experimental subject and method. Results : Fifty-two articles, related to Bojungikki-Tang were searched and these were classified into three classes, pharmacological signaling studies, clinical case studies and the others. As a result of classification, clinical case studies were most abundant by 38 cases, pharmacological signaling studies were 10 and the others were 4. This result suggested that the study of Bojungikki-Tang was mainly focused on clinical study rather than experimental study. Among the clinical case studies, clinical studies related to immune disease took the most greatest number and number of other clinical studies were evenly distributed over the various disease. Immune intensification effect of Bojungikki-Tang can be thought continuous with reinforce the middle Jiao Qi in company with improvement effect on growth hormone failure and male infertility. A fact that the one prescription, Bojungikki-Tang could be used to treat various diseases reflects a viewpoint of Oriental medicine that is based on the respect for and understanding of individual constitution and disposition while paying attention to the gender difference and individuality. The clinical usefulness of Bojungikki-Tang on obstructive sleep apnea syndrome and visceroptosis concerns with ascend Yang and treat Qi sinking. Pharmacological signaling studies were mainly focused on diseases that were treated Bojungikki-Tang in clinic, however, there was one study that experimented on changes of numerous gene expression according to the treatment of Bojungikki-Tang using DNA array. Conclusion : The therapeutic system of Oriental medicine is different in various aspects form that of Western medicine and appropriate harmonization of east and west is expected to improve the quality of medical care. In this regard, continuous investigation and analysis about herbal medical science papers in Japan can also be useful to make progress in Korean traditional medicine.
The main pathological hallmark of Alzheimer's disease is the deposition of amyloid-beta ($A{\beta}$) peptides in the brain. $A{\beta}$ has been widely used to mimic several aspects of Alzheimer's disease. However, several characteristics of amyloid-induced Alzheimer's disease pathology are not well established, especially in mice. The present study aimed to develop a new Alzheimer's disease model by investigating how $A{\beta}$ can be effectively aggregated using prokaryotes and eukaryotes. To express the $A{\beta}42$ complex in HEK293 cells, we cloned the $A{\beta}42$ region in a tandem repeat and incorporated the resulting construct into a eukaryotic expression vector. Following transfection into HEK293 cells via lipofection, cell viability assay and western blotting analysis revealed that exogenous $A{\beta}42$ can induce cell death and apoptosis. In addition, recombinant His-tagged $A{\beta}42$ was successfully expressed in Escherichia coli BL21 (DE3) and not only readily formed $A{\beta}$ complexes, but also inhibited the proliferation of SH-SY5Y cells and E. coli. For in vivo testing, recombinant His-tagged $A{\beta}42$ solution ($3{\mu}g/{\mu}l$ in $1{\times}PBS$ containing $1mM\;Ni^{2+}$) was injected stereotaxically into the left and right lateral ventricles of the brains of C57BL/6J mice (n = 8). Control mice were injected with $1{\times}PBS$ containing $1mM\;Ni^{2+}$ following the same procedure. Ten days after the sample injection, the Morris water maze test confirmed that exogenous $A{\beta}$ caused an increase in memory loss. These findings demonstrated that $Ni^{2+}$ is capable of complexing the 50-kDa amyloid and that intracerebroventricular injection of $A{\beta}42$ can lead to cognitive impairment, thereby providing improved Alzheimer's disease models.
Objective : This study was designed to investigate the effect of the water extract of Gamisamgibopae-tang(GMSGBPT), an oriental herbal formulation, on the growth of NCI-H460 and A549 human non-small-cell lung cancer cell lines. Methods : Cytotoxicity and cell morphology were evaluated by MTT assay and inverted microscope, respectively. Apoptosis was detected using agarose gel electrophoresis and flow cytometer. The expression levels of mRNAs and proteins of target genes were determined by RT-PCR and western blot analyses, respectively Result and Conclusion : We found that exposure of A549 cells to GMSGBPT resulted in the growth inhibition in a dose-dependent manner as measured by MTT assay, but GMSGBPTdid not affect the growth of NCI-H460 cells. The anti-proliferative effect of GMSGBPT treatment in A549 cells was associated with morphological changes, formation of apoptotic bodies and DNA fragmentation, and flow cytometry analysis confirmed that GMSGBPT treatment increased the populations of apoptotic-sub G1 phase. Growth inhibition and apoptotic cell death by GMSGBPT were connected with a up-regulation of cyclin-dependent kinase inhibitor p21 (WAF1/CIP1) mRNA and protein in a tumor suppressor p53-independent fashion. However GMSGBPT treatment did not affect other growth regulation-related genes such as early growth response-1 (Egr-1), nonsteroidal anti-inflammatory drug (NSAID)-activated gene-1 (NAG-1), inducible nitric oxide synthase (iNOS), cyclooxygenases (COXs), telomere-regulatory factors in A549 orNCI-H460 cells. Taken together, these findings partially provide novel insights into the possible molecular mechanism of the anti-cancer activity of GMSGBPT.
Bloomless cucumber fruits are commercially produced by grafting onto the pumpkin stocks (Cucurbita moschata) to restricted silicon ($SiO_2$) absorption. Inhibition of silicon absorption in bloomless stocks is conferred by a mutant allele of the CmLsi1 homologous to Lsi1 in rice. In this study, we characterized the Lsi1 homologs in pumpkin (C. moschata) and its cold-tolerant wild relative C. ficifolia ('Heukjong') in order to develop a DNA marker for selecting a bloomless trait and to establish the molecular basis for breeding bloomless stock cultivars of C. ficifolia. A Cleaved amplified polymorphic sequence (CAPS) marker (CM1-CAPS) was designed based on a non-sysnonymous single nucleotide polymorphism (SNP, C>T) of the CmLsi1 mutant-type allele, and its applicability for Marker-assisted selection (MAS) was confirmed by evaluating three bloom and five bloomless pumpkin stock cultivars. Quantitative RT-PCR of the CmLsi1 for these stock cultivers implied that expression level of the CmLsi1 gene does not appear to be associated with the bloom/bloomless trait and may differ depending on plant species and tissues. A full length cDNA of the Lsi1 homolog [named CfLsi1($B^+$)] of 'Heukjong' (C. ficifolia), was cloned and sequence comparison between CmLsi1($B^+$) and CfLsi1($B^+$) revealed that there exists total 24 SNPs, of which three were non-synonymous. Phylogenetic analysis of CfLsi1($B^+$) and Lsi1 homologs further revealed that CfLsi1($B^+$) is closesly related to Nodulin 26-like intrinsic proteins (NIPs) and most similar to CpNIP1 of C. pepo than C. moschata.
세계 각국은 자국의 자생식물을 활용하여 다양한 산업 활용에 집중하고 있으며 국내에서도 자생식물을 활용한 화장품 원료 소재 개발이 절실히 필요한 실정이다. 이에 본 연구는 특유의 기후와 환경에 의해 울릉도 특유의 식물분포를 갖는 울릉도 자생식물 후보군을 선정하고 이를 소재로 개발하여 화장품 원료로의 활용 가능성을 시험하였다. 섬초롱, 술패랭이, 해국에서 캘러스를 유도하고 대량배양 하였다. 대량배양하여 얻은 울릉도 자생식물 캘러스를 열수 및 에탄올 추출하여 COX-2 발현 비교, collagen 합성능 측정, wound healing 등 다양한 효능을 시험하였다. HPLC 분석을 통하여 열수 및 에탄올 추출물의 유효성분에 차이를 보임을 확인하였으며 또한 효능평가에서도 차이를 보였다. 술패랭이, 해국 캘러스 에탄올 추출물을 처리하였을 경우 항염 관련 단백질인 COX-2의 발현을 감소시키고, 모든 에탄올추출물은 wound healing assay를 통해 상처 치유능을 보임을 확인하였다. 이를 통해 울릉도 자생식물 캘러스 추출물이 자연친화적, 친환경적이며, 또한 국내 자생하는 식물을 활용한 소재로써 항염 및 상처치료 관련 제품에 기여할 것이다.
대만산 배추 (Brassica campestris M.)로부터 분리한 BcHSPI7.6 cDNA(내열성 유전자)를 pBKSl-l vector에 subcloning하므로서, NPTII 유전자와 P35S-HSPI7.6 cDNA를 가지는 pBKH4 재조합 플라스미드를 제작하였다. 이들 플라스미드를 갖는 A. tumefaciens LBA4404로서 담배잎 단편을 24시간 동안 공배양하므로서 감염시켰으며, 이들 유전자로 형질전환된 shoot는 $100\;{\mu\textrm{g}}/ml$의 가나마이신을 첨가한 MS-n/B 배지에서 선발하였다. 담배((Nicotiana. tabacum)의 열에 대한 치사온도는 $50^{\circ}$에서 15분 이상이었으며, BcHSPI7.6 cDNA를 갖는 형질전환된 식물체는 이 온도에서 내열성을 나타내었다. 식울체의 형질전환 여부는 ${\alpha}^{_32}P$로 표지한 BcHSPI7.6 cDNA 단편을 probe로 이용해서 Southern blot hybridization을 실시하므로서 확인하였다. BcHSPI7.6 cDNA의 발현정도는 Northern blot 분석과 이중면역확산 방법으로 확인하였다. 본 연구에서, 담배에 도입한 BcHSPI7.6 cDNA는 내열성과 관련이 있는 유전자로서, HSPI7.6 단백질은 식물체를 열에 의한 손상으로부터 방지해 주는 protector 역할을 하는 것 으로 사료된다.
Salmoenella는 대표적인 intracellular pathogen으로 숙주의 면역 세포인 macrophage 내에서 살아남아 이들을 매개로 숙주의 몸 전체를 이동해 가면서 전신성 감염을 일으킨다. 살모넬라는 숙주 세포 내부의 이러한 극한 환경을 극복하기 위해서 다양한 방어 기작을 가진다. 본 연구에서는 복합적인 스트레스가 작용하는 정지기 Salmonella에서 특이적으로 발현되는 단백질에 주목하였다. 정지기 상태의 Salmonella에서 약 20 kDa의 단백질이 특이적으로 많이 발현되었으며, 세포질 분획을 통해 이 단백질이 세포질 부분에 존재함을 알 수 있었다. MALDI-TOF 분석을 통해 이 단백질이 $\b{D}NA$ binding $\b{p}rotein$ in $\b{s}tationary$ phase (Dps) 단백질임을 확인하였다. Dps 단백질은 스트레스가 주어진 상황에서 DNA에 비특이적으로 결합하여 DNA가 안정한 형태를 유지하도록 하여 스트레스로부터 염색체를 보호하는 역할을 하는 것으로 알려져 있다. 이후의 연구를 위하여 과발현하여 정제한 Dps 단백질을 토끼에 주사하여 Dps 특이적인 항체를 제조하였다. dps 발현에 영향을 미치는 조절자 단백질을 알기 위하여 다양한 S. typhimrium 돌연변이주들 내에서의 Dps 단백질양을 조사하였다.
조혈촉진 cytokine인 Granulocyte colony stimulating factor (G-CSF)는 골수세포를 자극하여 granulocyte로 증식, 분화시키는 기능을 가지며, 현재 아주 고가의 치료제로 사용되고 있다. 인간 G-CSF (hG-CSF)를 아직 시도되지 않은 누에 유래 세포주인 BM5 세포에서 발현시키고 생산 효율을 높이기 위해 hG-CSF cDNA를 변형하였다. hG-CSF의 cDNA의 endoplasmic reticulum (ER) signal sequence 부분을 누에의 소포체에서 분비되는 단백질인 prophenoloxidase (PPAE), protein disulfide isomerase (PDI)와 bombyxin (BX)에서 유래한 누에특이 ER signal sequence로 대체한 hG-CSF의 cDNA 함유 벡터를 구축하였다. 이들 벡터를 사용하여 형질전환한 BM5 세포의 배양액에 분비된 G-CSF 단백질을 western blot으로 분석하여 발현을 확인하였다. 누에특이 ER signal sequence들로 대체된 hG-CSF cDNA를 포함하는 벡터에 의한 hG-CSF 단백질 생산이 인간 G-CSF cDNA가 든 벡터에 의한 hG-CSF의 생산보다 월등히 효율적이었다. 또한, PPAE-signal sequence를 포함하는 hG-CSF 단백질은 배양배지에서 형질전환 4일 후에 최고에 달하였고, 7 일째까지 비슷한 양이 배지 내에서 검출되었다. 이상의 결과는 인간유래 유전자가 곤충세포 내에서 발현 될 때 인간유래 유전자 보다는 곤충 유전자발현 시스템에 맞게 변형했을 경우 더 효율적인 단백질 발현을 얻을 수 있음을 보여 준다.
피부의 생태계는 미생물에게 다양한 형태의 서식처를 제공하며, 광범위한 미생물들이 살고 있다. 숙주인 사람은 이들과 공생관계를 이루고 있으며, 이들은 숙주에 많은 긍정적인 영향을 미친다. 피부에 분포하는 미생물들의 다양한 대사물질은 피부세포에 영향을 미치고, 피부장벽 기능, 노화방지 및 항염증에 광범위한 효능을 나타내는 것으로 알려져 있다. 본 연구에서는 사람의 피부에서 신규한 Sporichthyaceae bacterium strain K-07을 분리하였고, 해당 미생물의 16S rRNA 분석결과 Sporichthya속의 미생물과 상동성이 93.4%인 것으로 신규속(genus)으로 확인되었다. 그리고 신규로 분리된 K-07 배양액을 처리하였을 때, HaCaT cell에 어떤 변화가 나타나는지에 대한 분석을 실시하였다. 분리된 신규 미생물 strain K-07의 16S rRNA sequence 분석결과 상동성이 93.4% 이하로 확인되었고, 보고되지 않은 새로운 종으로 확인되었다. Filaggrin, cluadin1, claudin4, ${\alpha}SMase$, 및 CerS3 / HAS3 및 aquaporin3 / IL-6 및 TNF-${\alpha}$ / TSLP 및 TARC를 대상으로 strain K-07 배양액을 처리하여 변화를 관찰하였다. 그 결과 filaggrin, cluadin1, claudin4, ${\alpha}$SMase, 및 CerS3 / HAS3 및 aquaporin3에서는 음성 대조군 대비 우수한 증가 효과가 나타남을 확인하였다. 그리고 IL-6 및 TNF-${\alpha}$ / TSLP 및 TARC에 대해서도 우수한 억제능을 나타내는 것을 확인하였다. 결론적으로 신규 미생물 strain K-07 배양액은 피부 장벽활성 증진에 매우 효과적인 작용을 하며, 염증 억제에 우수한 효능을 나타내는 것으로 확인되므로 피부에 효과적인 소재로 사용될 수 있을 것이다.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
