  • We have studied on the preparation of $TiCo_xFe_{1-x}$(x=0.50~1.00) system alloy, the characteristics of $TiCo_xFe_{1-x}$(x=0.50~1.00) system alloy by X-ray diffractometer (XRD), pressure composition temperature (PCT) curve, scanning electron microscopy (SEM) and the $H_2-N_2$ gas mixture separation of $TiCo_xFe_{1-x}$(x=0.50~1.00)- stainless steel (SS) composite membranes. The formation of $TiCo_xFe_{1-x}$(x=0.50~1.00) system alloys with cubic crystal same as TiCo was confirmed by X-ray diffractometer. $TiCo_xFe_{1-x}$(x=0.50~1.00) system alloys showed the hysteresis at $120^{\circ}C$. As the Fe content of $TiCo_xFe_{1-x}$(x=0.50~1.00) system alloys increased, the hysteresis was increased both range x=0.90~1.00 and x=0.55~0.60, and the range x=0.55~0.90 gave decreased hysteresis. $TiCo_{0.55}Fe_{0.45}$ alloy was the one showed the lowest hysteresis among them. The lowest value of hydrogen permeation pressure of $TiCo_xFe_{1-x}$(x=0.50~1.00)-SS composite membrane was $TiCo_{0.55}Fe_{0.45}$-SS composite membrane with the value of 2.5 atm at $120^{\circ}C$; otherwise, $TiCo_{0.90}Fe_{0.10}$-SS composite had the highest pressure value among the membranes with the value of 10 atm. $TiCo_{0.55}Fe_{0.45}$-SS composite membrane was the best to separate the $H_2-N_2$ gas mixture excellently among the $TiCo_xFe_{1-x}$(x=0.50~1.00)-SS composite membranes since $TiCo_{0.55}Fe_{0.45}$ had the least hysteresis, and hydrogen permeation pressure was the lowest with value of 2.5 atm.

  • Recently, the studies about rock physics model (RPM) in shale reservoir are widely performed. In shale reservoir, the degree of the maturity can be estimated by kerogen and GOR (Gas-Oil Ratio). The researches on the rock physics model of shale reservoir with the amount of kerogen have been actively carried out but not with GOR. Thus, in this study, we analyzed the changes in seismic velocity and density, and AVO (Amplitude Variation with Offset) response depending on changes in GOR and the amount of kerogen. Since the shale consists of plate-like particles, it has vertical transverse isotropy (VTI). Therefore we estimated the seismic velocity and density by using Backus averaging method and analyzed AVO responses based on these estimated properties. The results of analysis showed that the changes in the velocity with the GOR variation are small but the velocity changes with the variation in kerogen amount are relatively larger. In case, GOR 180 (Litre/Litre) which is boundary between heavy oil and light oil, when volume fraction of kerogen increased from 5% to 35%, the P-wave velocity normal to the layering increased 51%. That is, it helps estimating maturity of kerogen through the velocity. Meanwhile, when rates of oil-gas mixture are large, the effect of GOR variation on the velocity change became larger. In case volume fraction of kerogen is 5%, the P-wave velocity normal to the layering was estimated $1.46km/s^2$ in heavy oil (GOR 40) but $1.36km/s^2$ in light oil (GOR 300). The AVO responses analysis showed class 4 regardless of the GOR and amount of kerogen because variation of poisson's ratio is small. Therefore, shale reservoir has possibility to have class 4.

  • This research was focused on the selective separation of $CO_2$ or $CH_4$ from mixture of these gases, by controlling the size of pore or pore gate. Pitch based activated carbon fibers(ACF) were used as adsorbents. The size of pore gate was controlled by the molecule having similar size to that of pore opening. After the adsorption of adsorbate on pore surface, planar molecules such as benzene and naphthalene covered the pore gate. The slow release of adsorbate from the pores covered by planar molecules makes apertures between planar molecules covering pore gate and this structure can be fixed by rapid pyrolysis. The control of pore gate using benzene as covering molecules could not accomplished due to the simultaneous volatilization of benzene and adsorbate$(CO_2)$ caused by similar temperatures of benzene volatilization and adsorbate desorption. Therefore we replaced benzene with naphthalene looking for the stability at a $CO_2$ desorption temperature. The naphthalene molecule was adsorbed on the ACF up to 15% of ACF weight and showed no desorption until $100^{\circ}C$, indicating that the molecule could be used as a good cover molecule. Naphthalene could cover almost all the pore gate, reducing BET surface area from 753 $m^2/g$ to 0.7 $m^2/g$. A mixed gas$(CO_2:CH_4=50:50)$ was adsorbed on the naphthalene treated OG-7A ACF. The amount of $CO_2$ adsorption increased with total pressure, whileas thai of $CH_4$ was not so much influenced on the pressure, indicating that $CO_2$ made more compounds on the ACF surface along with total pressure increase. The most $CO_2$ and the least $CH_4$ were adsorbed in the condition of 0.4 atm, resulting in the highly pure $CH_4$ left in ACF.

  • Background: Epigallocatechin-3-gallate (EGCG) is the major catechin in green tea, and has shown antiproliferative, antiangiogenic, antimetastatic and cell cycle pertubation activity in various tumor models. Hypoxia can be induced because angiogenesis is insufficient for highly proliferating cancer. Hypoxia-inducible factor-1$\alpha$ (HIF-1$\alpha$) and its downstream target, vascular endothelial growth factor (VEGF), are important for angiogenesis, tumor growth and metastasis. The aim of this study was to determine how hypoxia could cause changes in the cellular phenomena and microenvironment in a non-small cell culture system and to examine the effects of EGCG on a HIF-1$\alpha$ and VEGF in A549 cell line. Methods: A549 cells, a non-small cell lung cancer cell line, were cultured with DMEM and 10% fetal bovine serum. A decrease in oxygen tension was induced using a hypoxia microchamber and a $CO_2-N_2$ gas mixture. Gas analysis and a MTT assay were performed. The A549 cells were treated with EGCG (0, 12.5, 25, 50 ${\mu}mol/L$), and then examined by real-time-PCR analysis of HIF-1$\alpha$, VEGF, and $\beta$-actin mRNA. Results: Hypoxia reduced the proliferation of A549 cells from normoxic conditions. EGCG inhibited HIF-1$\alpha$ transcription in A549 cells in a dose-dependent manner. Compared to HIF-1$\alpha$, VEGF was not inhibited by EGCG. Conclusion: HIF-1$\alpha$ can be inhibited by EGCG. This suggests that targeting HIF-1$\alpha$ with a EGCG treatment may have therapeutic potential in non-small cell lung cancers.

  • The sympathoadrenal system plays an important role in homeostasis in widely varing external environments. Conflicting findings, however, have been reported on its response to hypoxia. We investigated the effect of hypoxia on the sympathoadrenal system in dogs under halothane anesthesia by measuring levels of circulating catecholamines in response to graded hypoxia. Ten healthy mongreal dogs were mechanically ventilated with different hypoxic gas mixtures. Graded hypoxia and reoxygenation were induced by progressively decreasing the oxygen fraction in the inhalation gas mixture from 21%(control) to 15%, 10% and 5% at every 5 minutes, and then reoxygenated with 60% oxygen. Mean arterial pressure, central venous pressure and mean pulmonary arterial pressure were measured directly using pressure transducers. Cardiac output was measured by the thermodilutional method. For analysis of blood gas, saturation and content, arterial and mixed venous blood were sampled via the femoral and pulmonary artery at the end of each hypoxic condition. The concentration of plasma catecholamines was determined by radioenzymatic assay. According to the exposure of graded hypoxia, not only did arterial and mixed venous oxygen tension decreased markedly at 10% and 5% oxygen, but also arterial and mixed venous oxygen saturation decreased significantly. An increased trend of the oxygen extraction ratio was seen during graded hypoxia. Cardiac output, mean arterial pressure and systemic vascular resistance were unchanged or increased slightly. Pulmonary arterial pressure(PAP) and pulmonary vascular resistance(PVR) were increased by 55%, 76% in 10% oxygen and by 82%, 95% in 5% oxygen, respectively(p<0.01). The concentrations of plasma norepinephrine, epinephrine and dopamine increased by 75%, 29%, 24% in 15% oxygen and by 382%, 350%, 49% in 5% oxygen. These data suggest that the sympathetic nervous system was activated to maintain homeostasis by modifying blood flow distribution to improve oxygen delivery to tissues by hypoxia, but hemodynamic changes might be blunted by high concentration of nitrous oxide except PAP and PVR. It would be suggested that hemodynamic changes might not be sensitive index during hypoxia induced by high concentration of nitrous oxide exposure.

  • The feasibility for the employment of manganese nodule as an adsorbent for $SO_{2}$ gas has been investigated. The specific surface area of manganese nodule particle, which used in the experiments, was ca. $221.5m^{2}/g$ and the content of sulfur in manganese nodule was observed to significantly increase after $SO_{2}$ was adsorbed on it. The EPMA for the distilled water-washed and methanol-washed manganese nodule particle after $SO_{2}$ adsorption showed that its sulfur content was slightly decreased to 14.7% and 13.1% respectively, from 15.4% before washing. The XRD analysis of manganese nodule showed that todorokite and birnessite, which are manganese oxides, and quartz and anorthite were the major mineralogical components and weak $MnSO_{4}$ peaks were detected after $SO_{2}$ was adsorbed on manganese nodule. For an comparative investigation, limestone was also tested as an adsorbent for $SO_{2}$, however, no peaks for $CaSO_{4}$ were found by XRD analysis after the adsorption of $SO_{2}$. As the size of adsorbent increased, time for breakthrough was decreased and the adsorbed amount of $SO_{2}$ was also diminished. The $SO_{2}$ adsorption was hindered when its flow rate became high and the adsorption capacity of manganese nodule was observed to be superior to that of limestone. In addition, the mixture of manganese nodule and limestone did not show an increase in the adsorption of $SO_{2}$. Finally, as the temperature was raised, the adsorbed amount of adsorbate on manganese nodule was found to be decreased.

  • This study was conducted to investigate effects of terpenoids-rich plant extracts (TRPE) on the in vitro ruminal fermentation characteristics and methane production. The ruminal fluid was collected from a cannulated Hanwoo cow fed concentrate and timothy in the ratio of 6 to 4. The TRPE as Mint (Mentha arvensis var. piperascens), Pine (Pinus densiflora), Japan cedar (Cryptomeria japonica), Sichuan pepper (Zanthoxylum piperitum), Hinoki cypress (Chamaecyparis obtuse) and Japanese black pine (Pinus thunbergii) were used in this study. The 15 mL of mixture, contains McDougall buffer and rumen fluid in the ratio of 2 to 1. The mixture was dispensed anaerobically 50 mL serum bottles and it is contained 0.3 g timothy substrate and 5% TRPE. The bottles were incubated at $39^{\circ}C$ for 3, 6, 9, 12, 24, 48 and 72 hours. The pH value decrease by increased incubation times and the pH values at all times were significantly (p<0.05) higher in treatments than in control. The digestibility of dry matter at 3 hours was significantly (p<0.05) higher in mint treatment than in control. Productions of total gas and carbon dioxide at before 12 hours was significantly lower (p<0.05) in treatments than in control. The methane production at 24 hours was significantly (p<0.05) lower in treatments than in control. The concentrations of acetic acid and propionic acid at 24 hours were significantly higher (p<0.05) in mint and pine treatments than in control. In conclusion, the terpenoid-rich plant extracts were shown to decreased methane emission and without adversely affected ruminal fermentation. Therefore, the terpenoid-rich plant extracts as mint and pine were shown to decreased methane production and it has potential possibility for ruminal fermentations.

  • This study was carried out to investigate the in vivo and in vitro inhibitory effect of a traditional herbal complex (HC) extract prepared from a mixture of four oriental herbs (Dioscorea Rhizoma, Glycine soja Sieb. et Zucc, Bombycis corpus, Fermented Glycine soja) that have been widely used for the treatment and prevention of diabetes mellitus on hyperglycemia. The water extract of HC showed potent inhibitory effect on $\alpha$-glucosidase with $IC_{50}$ value of 1.24 mg/mL. Additionally, the ethanol extract of HC was also found to exhibit significant inhibitory effect against protein tyrosine phosphatase $1{\beta}$ ($PTP1{\beta}$), which is known as a major regulator of both insulin and leptin signaling. In the $PTP1{\beta}$ inhibitory assay, the most active n-hexane fraction obtained from the ethanol extract of HC, was identified as a mixture of fatty acid derivatives by gas chromatography-mass spectrometry (GC-MS). In high-fat diet-low dose streptozotocin (STZ)-induced diabetic rat, the water extract of HC improved the oral glucose intolerance as compared with rosiglitazone. HC also caused a marked decrease of body weight and fasting blood glucose and a significant improvement on glucose tolerance in metabolic syndrome mice model. These findings support that this traditional HC may be useful in the control of blood glucose in diabetes mellitus and metabolic syndrome.

  • The triglyceride composition of perilla oil was investigated by high performance liquid chromatography (HPLC) in combination with gas liquid chromatography (GLC). The triglycerides were separated from perilla oil by thin layer chromatography (TLC), and fractionated into five groups on the basis of their partition numbers by reverse phase HPLC on a column packed with ${\mu}-Bondapak\;C_{18}$ using methanol-chloroform mixture as a solvent. Each of these collected fractions gave one to three peaks in the GLC chromatograms according to the acyl carbon number of the triglyceride, and fatty acid composition of the triglyceride was also analyzed by GLC. The results indicate that the perilla oil consists of fifteen kinds of triglycerides, and the major triglycerides in perilla oil were as follows: 68.0% of $(C_{18:3},\;C_{18:3},\;C_{18:3})$, 6.7% of $(C_{18:2},\;C_{18:3},\;C_{18:3})$, 5.9% of $(C_{18:1},\;C_{18:3},\;C_{18:3})$, 4.3% of $(C_{16:0},\;C_{18:3},\;C_{18:3})$, 3.8% of $(C_{18:1},\;C_{18:2},\;C_{18:3})$, 3.2% of $(C_{18:1},\;C_{18:1},\;C_{18:3})$, 2.0% of $(C_{16:0},\;C_{18:2},\;C_{18:3})$, 1.5% of ($C_{18:2},\;C_{18:2},\;C_{18:3})$, 1.0% of $(C_{16:0},\;C_{18:1},\;C_{18:3})$.

  • High performance liquid chromatography (HPLC) was applied to the analysis of triglycerides of rice bran oil. The triglycerides were clearly separated in five peaks by HPLC on a column packed with ${\mu}-Bondapack$ C18 using methanol-chloroform mixture as a solvent. Compositions of the triglyceride and fatty acid of the fraction was also analyzed by gas liquid chromatography (GLC). Each of these collected fractions gave three to four peaks in the GLC chromatograms according to the carbon number of the triglyceride. The fitty acid compositions of these triglycerides were mainly composed of C16:0, C18:1 and C18:2 fatty acids. The major triglycerides of the rice bran oil were found to be those of (C16:0, C18:1, C18:2;16.64%), $(2{\times}C18:1,\;C18:2;16.18%)$, $(3{\times}C18:1;13.7%)$, $(C16:0,\;2{\times}C18:1;12.77%)$,$(C18:1,\;2{\times}C18:2;9.16%)$ and $(C16:0,\;2{\times}C18:2;6.42%)$
