Identification of Higenamine nad its Metabolites in Rat by Gas Chromatography/Mass Spectrometry

  • ($\pm$)-Higenamine is known as a cardiotonic principle of aconite root (root of Aconitum spp., Ranunculaceae). A simple and sensitive detection method for higenamine was developed by using gas chromatography-mass spectrometry (GC/MS). The recovery of higenamine after extraction and concentration with XAD-2 resin column was around 95% from rat biological fluids such as bile, plasma and urine. The limits of detection of higenamine in these biological fluids were approximately 0.1 ng/ml each. It has well been suggested that tetrahydroisoquinolines possessing catechol moiety such as higenamine should be subjected to the catechol-O-methyl transferase (COMT) activity in vivo. We detected two major peaks of presumed metabolites of higenamine in the total ion chromatogram obtained from the rat urine sample after the oral adminstration of ($\pm$)-higenamine. The scan mass spectrum of one of the metabolties coincided with those obtained from coclaurine $(C_6$-O-methyl higenamine) and those of the other metabolite are suggestive of isococlaurine $(C_7$-O-methyl higenamine).

Sorption Behavior of 1-Methylcyclopropene on Adsorbing Agents for Use in Extending the Freshness of Postharvest Food Products

  • The physiochemical interactions of 1-methylcyclopropene (1-MCP) and adsorbing agents can be described using a very powerful tool, inverse gas chromatography (IGC). Sorption behavior of 1-MCP on various adsorbing agents was assessed using the profile peaks of 1-MCP at an infinite dilution concentration using the IGC technique. Chromatogram peaks of 1-MCP adsorption were not observed for the adsorbing agent activated carbon. The forms of sorption isotherms followed Henry's law, and behaved according to the binding site theory. Specific retention volume and distribution coefficients for 1-MCP on the adsorbing agents were determined at 50, 60, 70, and $80^{\circ}C$, respectively. Silica gel had a much higher number of binding sites for 1-MCP compared to Tenax-TA and activated clay agents. Meanwhile, activated carbon proved to be a very strong binding agent for 1-MCP based on 1-MCP efficiency experiments on the selected adsorbing agents. However, as a proper means of delivering 1-MCP molecules to fresh food products, activated carbon is not fit for the binding and release of 1-MCP gas under dry or high humidity conditions because activated carbon has a strong affinity for 1-MCP, even when treated with distilled water.

생약복방제의 조성 변화가 부패성 효모 Zygosaccharomyces sp. 의 성장에 미치는 영향 (Effects of Changes in Composition of Herb Extract Product on Growth of Spoilage Yeast, Zygosaccharomyces sp.)

  • The aim of the present study was to investigate effects of food preservative addition and changes in composition of herb extract product on the growth of spoilage yeast, Zygosaccharomyces sp. Herbs such as Panax ginseng, Cinnamomum cassia, Lycium chinense, Zyzyphus juiuba and Jingiber officinale were altogether put into water and essence was extracted at 80oC, and then the extract was concentrated at 75oC. The herb extract product was made by adding vitamins, amino acids and honey to the concentrated herb extract. The amount of gas produced from the herb extract product was increased as inoculated cell number increased but decreased as Brix concentration increased. Gases were produced in small amount when incubation was made at 4oC but large amounts of gases were produced at 25 or 40oC of incubation. The gas production and growth of Zygosaccharomyces sp. were measured after browning reaction was induced by heating at 85oC for 12 hours. It appeared that heating treatment did not induce any significant change in the gas production and growth of the cell. The effects of addition of various sugar to the herb extract produce were also invesigated. Amounts of gas production were in the order of glucose>sucrose>oligosaccharide>stevioside. The viable cell count was measured as 6.0$\times$107 CFU/g when glucose was added to the herb extract product. The viable cell counts were 5.0$\times$106, 3.0$\times$103, and 3.0$\times$102 CFU/g in sucrose, oligosaccharide and stevioside added herb extract product, respectively. The amount of gas production from the herb extract product was remarkably reduced by addition of such food preservatives as sodium benzoate and DF 100. TLC(thin layer chromatography) chromatogram of the herb extract showed stability of the herb extract in the above treatments.

Development and validation of a portable gas chromatograph method for quantitative determination of oxygen and toxic gas impurities in medical oxygen using planar microchromatographic columns and detectors

  • This study examines portable Gas Chromatography (GC) for the quantitative analysis of oxygen and impurities, focusing on the development and validation of a method to determine oxygen, carbon monoxide, carbon dioxide, methane, and nitrogen in medical compressed oxygen gas. The goal is to ensure the quality of medicalgrade oxygen. The method's validation assessed its metrological characteristics, demonstrating specificity through clear chromatographic separation of the target gases and the absence of these peaks in the carrier gas chromatogram. It exhibited linearity within the designated concentration ranges, while precision met permissible standards, with the relative standard deviation for intermediate precision being less than 4% for carbon monoxide (0.00025 - 0.00099 %), less than 3 % for methane (0.0005 - 0.00246%) and carbon dioxide (0.0050 - 0.0150 %), less than 2% for nitrogen (0.1 - 0.7 %), and less than 0.01% for oxygen (99.27 - 99.98%). Overall, the validation results confirm the suitability of this analytical method for the quantitative determination of the aforementioned gases in medical compressed oxygen using portable GC with microchromatographic columns and detectors.

Potable handheld gas chromatograph(PHGC)를 이용한 인삼속(Panax species) 식물들의 향기패턴 분석 (Analysis of Aroma Pattern of Panax Species by Potable Handheld Gas Chromatograph)

  • 분말상태의 인삼속 식물들의 품종 및 원산지를 판별하기 위하여 SAW 센서가 내장된 PHGC를 이용하여 향기 패턴을 분석하였다. 한국 백삼을 1로 기준할 때 전체적인 Rt에 대한 frequency pattern의 면적비는 화기삼 $0.248{\sim}0.871$, 전칠삼 $0.030{\sim}0.674$, 중국산 백삼 $0.005{\sim}0.212$ 범위로 나타났다. 분명한 차이를 나타내는 몇 개의 특정 향기성분의 면적비를 보면 $Rt_{20.02}$에서 한국산 백삼 1일 때, 중국산 백삼 0.212, 화기삼 0.343, 전칠삼 0.065이었다. 또한 $Rt_{21.70}$$Rt_{24.90}$에서 검출되는 향기성분의 면적비도 품종간의 차이가 뚜렷하였다. 한국산 백삼과 중국산 백삼에서 검출된 Rt_26.15 향기성분의 면적비는 각각 1과 0.185로 나타나 원산지간의 차이도 분명히 나타났다. $Rt_{26.15}$의 향기성분은 화기삼과 전칠삼에서는 검출되지 않았다. Frequency pattern, derivative pattern을 Vapor $print^{TM}$을 사용하여 도형화하여 비교한 결과 한국 백삼(Korean Panax ginseng C.A. Meyer), 화기삼(미국삼, 서양삼, Panax quinquefolium L.), 전칠삼(Panax notoginseng F.H. Chen), 중국산 백삼(Chinese Panax ginseng)은 서로 다른 패턴을 보여주어 품종간의 차이는 물론 원산지의 차이도 뚜렷하게 나타났다.

인삼 육성계통 캘러스부터 Polyacetylene의 분석 및 생합성에 미치는 배양조건 (Analysis and Culture Conditions for Biosynthesis of Polyacetylene from Callus of Ginseng Superior lines)

  • 인삼 callus의 기내배양에 의해서 항암물질을 대량생산하기 위한 연구의 일환으로 인삼 우수계통으로부터 기내에서 callus를 유기하여 polyacetylene 고함유 세포주를 선발하고, 인삼 callus의 polyacetylene 생산에 미치는 식물호르몬 및 배지의 영향을 조사하였다. 식물호르몬 CPA가 첨가된 MS배지에서 유도된 인삼 우수계통 callus에서는 polyacetylene 중에서 panaxynol은 TLC와 GC에 의해서 전혀 검출할 수 없었으나, callus에 따라서는 panaxydol이 형성됨을 확인할 수 있었다. 특히, 강력한 항암효과를 가지고 있는 panaxydol은 우수계통 callus 18종 중에서 5종이 형성하였으며, 30201계통 callus에서 가장 많이 검출되었다. 또한 인삼 10301계통 callus는 CPA가 단독으로 첨가된 배지에서 panaxydol을 생성하지 못하였으나, CPA 2 mg/L와 BA 0.05 mg/L 첨가구에서는 callus의 생장도 양호하였으며, panaxydol도 생성하였다. 인삼 callus의 생장과 panaxydol의 합성에는 MS배지보다 SH 배지가 더 양호하였으며, NAA와 sucrose는 전혀 영향을 미치지 않았다.

디젤기관에서 산소성분 첨가에 의한 배기가스 배출특성의 실험적 연구 (The Experimental Study on Exhaust Emission Characteristics with Oxygen Component Addition in Diesel Engine)

  • Recently, our world is faced with very serious and hard problems related to the air pollution due to the exhaust emissions of the diesel engine. So, lots of researchers have studied to reduce the exhaust emissions which influenced the environment strong. In this paper, the effect of oxygen component in fuel on the exhaust emissions has been investigated for diesel engine. And, we tried to analysis the quantities of the low and high hydrocarbon among the exhaust emissions in diesel engine. It have been investigated by the quantitative analysis of the hydrocarbon $C_1\simC_6$ using the gas chromatography. This study carried out by comparing the chromatogram with diesel fuel and diesel fuel blended DGM(diethylene glycol dimethyl ether) 5%. The results of this study show that the hydrocarbon $C_1\simC_6$ among the exhaust emissions of the mixed fuels are exhausted lower than those of the diesel fuel at the all load and speed. In particular, high boiling point hydrocarbons such as $C_5$ and $C_6$ were reduced remarkably in high speed and load region.

유향성 황색계 호접란 육성 및 전자코를 이용한 향기패턴 분석 (Breeding of Fragrant Yellow Phalaenopsis and Scent Pattern Analysis by GC/SAW Electronic Nose System)

  • 노란색 유향성 호접란을 육성하기 위해 황화계 육종모본들을 이용하여 5종류의 교배조합이 작성되었으나 $Phal.$ Brother lawrence를 종자친으로 $Phal.$ Brother saragold를 화분친으로 한 교잡에서만 정상적인 교배와 꼬투리 형성 및 종자발아가 이루어졌다. 잡종후대로 약 500개체가 실생 조직배양을 통해 육성되어 온실에 심어졌으며 개화기에 화색과 화형 위주로 60여 우수개체가 1차 선발 되었고 다시 생육속도, 병충해저항성 등을 검정한 2차 선발에서 15계통이 육성되었다. 이들을 대상으로 향기가 비교적 강하고 꽃 수명이 길어 시장에서 기호도가 높을 것으로 예상되는 6계통을 최종 선발하였다. 최종 선발된 이들 유향종들과 이들 교잡종들의 양친을 대상으로 전자코를 이용한 향기패턴분석이 이루어졌다. 전자코로 꽃 향기를 분석한 후 GC chromatogram peak와 이 피크를 이용한 Polar derivative diagram을 작성 비교한 결과 양친에서는 retention time 7s와 9s 사이, retention time 20s와 25s 사이에서 강한 peak가 나타났다. 양친과 비교하여 잡종후대의 선발계통에서는 retention time 7s와 9s 사이 즉 8s 근처에서 하나의 강한 피크가 공통적으로 나타났으며 양친에서 나타났던 retention time 20s와 25s 사이에서 강한 2개의 피크는 교잡후대 중 4개의 계통에서 이 피크가 강하게 발현되었으나 2개의 계통에서는 오히려 거의 나타나지 않았다. 그리고 $VaporPrint^{TM}$ 이미지 분석소프트웨어를 이용하여 향기패턴을 시각적으로 이미지화 한 결과 더욱 명확하게 향기패턴을 비교 분석할 수 있었는데 마찬가지로 교잡양친과 교잡후대계통 모두에서 retention time 8s 근처에서 강한 peak가 공통적으로 나타났으며 이 피크의 성분은 노란색 유향종에서 주로 강하게 발현되는 향기성분인 것으로 판단되었다. 이 피크는 향후 노란색 유향성 개체의 교잡선발 육종에 유용한 마커로서 활용할 수 있을 것으로 판단되었다.

Character Impact Compounds in Flavors of Korean Soy Sauce Manufactured with the Traditional and the Improved Meju

  • We characterized the character impact compounds of flavors of the fermented Korean soy sauce manufactured with both the traditional and the improved Meju made with different strains. The whole flavor samples were obtained by extracting each volatile flavor phase from both the traditional and the improved soy sauce. To get more detailed information, each whole volatile flavor was further fractionated into the basic, acidic, phenolic and neutral fractions. Each separated peak from the whole and fractionated flavor samples on gas chromatogram was identified by GC/MS and Kovat s retention index, and likewise the aroma of each peak was investigated by a sniffing test with the exercised panel. We were able to identify 15 groups of ingredients with the characteristic soy sauce aroma from the soy sauce made with the traditional Meju and 6 groups from the soy sauce manufactured with the improved Meju made with Aspergillus oryzae. The character impact compounds the two soy sauces were different from each other.

Analysis of Poly(Ethylene-co-Vinyl Acetate) Using Off-line Pyrolysis

  • Poly(ethylene-co-vinyl acetate) (EVA) was pyrolyzed to eliminate acetic acid of VA unit using off-line pyrolysis, and the deacetylated EVA was analyzed infrared spectroscopy (IR) and pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS). EVA film for deacetylation was prepared by solution casting on aluminum foil and it was pyrolyzed at low temperature of $300^{\circ}C$ in the off-line pyrolysis apparatus. After deacetylation, carbon-carbon double bond (C=C) was formed by 1,2-elimination of the VA unit in the EVA backbone. Most of C=C bonds were trans-1,4-unit and 1,2-unit was also observed. Presence of the 1,2-unit in deacetylated EVA indicates that terminal or branch VA units exist in the raw EVA. Py-GC/MS chromatogram of deacetylated EVA displayed much smaller acetic acid and much more abundant other pyrolysis products than that of raw EVA, which means that the pyrolysis efficiency and separation condition were improved.
