Gas Chromatographic Determination of Benfuresate through On-column Injection (On-column Injector를 이용한 benfuresate의 분석)

  • Benfuresate의 잔류분석을 위해 GC-FPD(S-mode)를 이용하여 분석조건을 설정 중 열적 불안정으로 인해 열분해 산물과 모 화합물이 혼재됨이 확인되었다. 가스크로마토그라프를 이용한 열분해의 주된 요인은 주입구의 온도와 주입구내의 기화 정도인 것으로 판단되었으며, 주입구를 Chauhan과 Debre 가 고안한 on-column으로 교체 후 분석한 결과 분해산물이 없는 단일 봉우리의 크로마토그램을 얻을 수 있었고, 절대량으로 0.6-4ng의 정량 범위 내에서 유의성 높은 검량선을 얻을 수 있는 주입구 온도는 20$0^{\circ}C$였다.

  • Instrumental determination was performed for the analysis of residues of ethylene oxide(EO), ethylene chlorohydrin(ECH) and ethylene glycol(EG) in white ginseng powders which were deaerated for 30 min after EO fumigation. Gas chromatographic data showed that EO residues were 570 ppm in the immediate samples after deaeration and 170 ppm in the stored ones for 8 days at 30$\pm$1$^{\circ}C$, respectively, showing a decreasing tendency with elapsed time. On the other hand, ECH and EG residues in samples as the secondary products of EO were 17, 650 ppm and 727 ppm stored for one month, and 9, 595 ppm and 221 ppm stored for 3 months, respectively.

  • Mixed photocatalyst containing cadmium sulfide and zinc sulfide was prepared on silica gel powder and Nafion film. Photo-irradiation of aqueous mixture containing the photocatalysis generated hydrogen by water cleavage reaction. Use of sodium sulfide as sacrificial reagent help the photo-reaction. Evolution of the hydrogen was measured by gas chromatographic analysis. Composition of the catalyst was determined by atomic absorption spectrophotometer. 0.2 mL of of hydrogen was generated per hour. The maximun catalytic activity was obtained after 8-12 hours later. Hydrogen generation efficiency by the two different catalytic system was compared and showed that the Nafion-based catalyst is more efficient than the silicagel-based catalyst for the photoreaction.

  • A solid-phase microextraction and gas chromatography-mass spectrometry for the extraction and analysis of methyl-tert-butyl ether has been described. methyl-tert-butyl ether was extracted from aqueous solution using SPME fiber coated polydimethylsiloxane and analysed by GC-MS with capillary column. Extraction parameters and chromatographic separation conditions were optimized. The applied method represented good analytical performance in terms of precision (3-8%, RSD) and accuracy(93-102%, mean recovery) with a method detection limit of 0.03 ppb.

Development of Analytical Method for Polychlorinated Biphenyls and Organochlorine Pesticides in Air (대기 중 PCBs와 유기염소계 살충제의 정량을 위한 분석 방법 개발)

  • An analytical method was investigated for the meaiiurement of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) concentrations in air samples. Procedures required for column chromatographic clean up. silicagel (stage I) and gel permeation chromatography (stage II), were discussed. Identification and quantification of PCBs and OCPs were performed using a combination of gas chromatography/mass spectrometry/ selected ion monitoring. Recovery tests calculated from six samples are 68∼137% for PCBs and 58∼130% for OCPs except for endrin aldehyde. Instrumental detection limits determined for the PCBs and OCPs varied from 0.05 to 0.18 pg/m3 and from 0.71 to 16.82 pg/㎥, respectively. The method has been applied to the analysis of air samples collected at Ansung city, Kyonggi province, Korea. This method may serve as a screening protocol for the simultaneous determination of PCBs and OCPs in air.

  • acs gene cloning was constructed by subcloning the 2.2-kb MunI-MunI restriction fragment of 638 and 639 which include acs gene from the kohara phage into the unique EcoRI site of pUC18 and pJM9131 containing the PHA biosynthesis genes. Then recombinant E. coli fadRatoC(Con) mutants containing the polyhydroxyalkanoate(PHA) biosynthesis genes are able to incoporate s significant levels of 3-hydroxyvalerate (3HV) into the copolymer [P(3HB-co-3HV)]. Quantitative determination of PHB and P(3HB-co-3HV) was performed by gas-chromatographic analysis of extracts obtained from methanolysis of lyophilized cells.

  • "Ha-Neul-Ta-Ri" (Trichosanthes kirilowii Max., Cucurbitaceae) cultivated in Korea and used in popular medicine, was investigated for sterol composition of the seed oil. This seed oil contains unusual sterols having ${\delta}^{7, 22}_-$, ${\delta}^{7, 24}_-$ and ${\delta}^{7, 22, 25}_-$ di and trienols. ${\delta}^{7, 22}_-$ Stigmastadienol and ${\delta}^{7, 22, 25}_-$ Stigmastatrienol were identified as the major sterols in the seed oil of Trichosanthes kirilowii by gas loiquid chromatographic analysis. The results showed that contents of the seed sterols were campesterol 1.0-1.2%, stigmasterol 1.5-1.7%, ${\delta}^{7}_-$ campstenol 0.6-0.8%, sitosterol 10.6-11.4%, ${\delta}^{7, 22}_-$ stigmastadienol 39.1-41.8%, ${\delta}^{7, 22, 25}_-$ stigmastatrienol 28.9-29.3%, ${\delta}^{7, 25}_-$ stigmastadienol 14.9-15.5% and ${\delta}^{7, 24}_-$ stigmastadienol 0.7-1.0%.

  • A simple and reproducible pretreatment method was developed for the determination of dioxins in milk sample. Liquid-liquid extraction (LLE) was used for the initial extraction of the analyte from milk. For the elimination of interferences coextracted from milk, acid treatment followed by multilayer silica gel, and then alumina column clean-up were performed. The clean extract could be obtained without carbon column or high performance liquid chromatographic (HPLC) clean-up procedure. Polychlorinated biphenyles (PCBS) and dioxins were separated on neutral alumina activated at 180 ℃ for 12 hours. The final extract was analyzed by HPLC and high resolution gas chromatography/high resolution mass spectrometry (HRGC/HRMS). The recovery of dioxins spiked in milk at 75-300 ppt level was 83.3-98.9% and their relative standard deviation was 4.1-14%.

  • By saponification and extraction of corn oil foots abandoned as waste during oil refining, a mixture of phytosterols was obtained, and its major components were determined as .betha.-sitosterol, campesterol and stigmasterol by gas chromatographic analysis. The mixture is very cheap and regarded as an excellent substrate for direct fermentation of C-17 keto steroid intermediate for various steroid pharmaceuticals.

  • Propofol(2, 6-diisopropylphenol) is rapid, short-acting intravenous anaesthetic agent. It is used for the induction and maintenance of general anaesthesia or sedation. The recommended doses are 2-2.5mg/kg given as a titration infusion over about 30min to achieve anaesthesia. Recently, we encountered 4 fatalities related to propofol. One death is a suicide by self-administered of propofol and the others are therapeutic misadventures during surgical care. The propofol level in the blood and tissues were determined by gas chromatographic analysis with mass spectral detection. (omitted)
