The discrimination of the agricultural origin, especially locally produced or imported from the products such as Ganoderma lucidum, sesame and arrowroot were investigated by using the electronic nose. Volatile components from these products were discriminated by twelve of conducting polymer sensors without any pretreatment. Pattern recognition was carried out. Multiple discriminant analysis showed the difference between imported agricultural product and locally produced ones such as Ganoderma lucidum, sesame and arrowroot. Unknown habitat of sesame and arrowroot could be identified by multiple discriminant analysis whether the imported or the locally produced one.
This study was attempted to investigate the effect of Ganoderma lucidum (Young-Jii) extract on the activities of GPT GOT Al. P LDH and the level of total bilirubin and total cholesterol in serum of $CCl_4$-intoxicated rats, and on the level of total lipids triglyceride phospholipids and total cholesterol in the serum of experimentally induced hyperlipemic rats, and on the effect of body and liver weight in rats. The results were shown as follows; In $CCl_4$-intoxicated rats, the extract showed a significant decrease in the activities of GPT and Al.P, a slight decrease in the activity of GOT and LDH; The level of total bilirubin was slightly affected, but significantly decreased at a dose as high as 500 mg/kg; the level of total cholesterol was increased dose dependently. In hyperlipemic rats, the extract caused a significant decrease in the level of total lipids and triglyceride and the rate of decrease was more pronounced with repeated treatments for 10 days; the level of phospholipids and total cholesterol were slightly decreased with repeated treatment of the extract at a dose of 300 mg/kg for 10 days; A significant body weight gain was shown with the treatment of the extract.
The highest antitumor activity was observed in water soluble AS fraction of the Ganoderma lucidum IY 009. AS fraction did not show any cytotoxicity on sarcoma 180 cell but stimulated antibody production, opsonization of macrophage in ICR mouse and superoxide ion production from isolated macrophage. AS fraction activated complement C3 in human serum, and their antitumor activity was inhibited by EDTA, a chelator of cation related complementary activation. AS fraction exerted om prolong of life span and ingibition of tumor growth in the leukemia P388 or L1210 transplanted inbreed mouse,k BDF1 but krestin did not. AS fraction did not show any serious and lethal effects through oral administration on ICR mouse, and LD$_{50}$ of those was above 2,230 mg/kg.
To obtain a new strain of Ganoderma lucidum by protoplast fusion technique, its protoplast formation and regeneration were studied. Several factors affecting the protoplast formation and regeneration were investigated to find their optimum conditions. The mycelium was grown for four days on the cellophane membrane placed on G. Incidum complete medium (GCM). When various commercial lytic enzymes were examined for protoplast isolation, the combination of Novozym 234 and $\beta$glucuronidase was found to be effective. An osmotic stabilizer, 0.6 M sucrose in 20 mM phosphate buffer pH 5.8, gave the highest yield of protoplasts. Three-hour incubation in shaking incubator was most suitable for releasing protoplasts. To increase the protoplast yield, pretreatment with 2-mercaptoethanol was carried out. The regeneration frequency in GCM containing 0.6M MgSO$_4$ 7$H_2O$ was shown to be 0.66%.
To study antioxidative activities of Ganoderma lucidum, its extracts were fractionated by various organic solvents with different polarity the extracts were purified by thin chromatography, silicagel column chromatography and preparative liquid chromatography. In antioxidative activity tests using thiocyanate method, TBA method and weighing method, fraction 5 from the hexane extract and fraction II from the methanol extract showed antioxidative activity. When the antioxidative activities were expressed as TBA value using a homogeneous liver extracte of rats, the relative antioxidative activities of fraction 5 and fraction II were increased by 13.0% and 54.6%, respectively.
Exo-polysaccharide (BWS) obtained from submerged cultivation of Ganoderma lucidum mycelium was fractionated. Antitumor activity of their fractions was investigated in comparison with the mycelial polysaccharide fractions. Eight kinds (BWS-DN, BWS-DA, BWS-DN-GI, BWS- DA-GI, MWS-DN, MWS-DA, MWS-DN-GI and MWS-DA-GI) of polysaccharide fractions were obtained by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel chromatography from BWS and MWS, which were isolated from culture fiuid and mycelial cell, respectively. The anticomplementary activities (ITCH$_{50}$%) of the exo-polysaccharide fractions showing 15% to 30% were lower than those of mycelial polysaccharide fractions showing 15% to 70%. The acidic fractions of BWS-DA and BWS-DA-GI fractionated from BWS, showed the highest activity of 30%. In the MTT assay, BWS-DN and MWS against mouse leukemia L1210 exhibited high inhibition ratio of 86 and 89%, respectively at the concentration of 600 $\mu$g/ml. High inhibition ratio of 50% (IC$_{50}$) was achieved for BWS, BWS-DA and MWS-DA fractions against human colon adenocarcinoma COLO-205 and for BWS-DA, BWS-DN and MWS-DN fractions against human leukemia HL-60 at the concentra- tion of 300 $\mu$g/ml among the six polysaccharide fractions, respectively.
In the previous study we described the antitumor effect of GLB, a protein-polysaccharide fraction separated from the growing tips of Ganoderma lucidum, against sarcoma 18 0 solid tumor in ICR mice. In this study, we separated an acidic protein-polysaccharide fraction, GLB-A, and a basic protein-polyaccharide fraction, GLB-B, from GLB by differential precipitation, and elucidated their antitumor and immunomodulatory activities. When ip injected at the dose of 50mg/kg/day into the ICR mice, GLB-A and GLB-B inhibited the growth of ip implantated sarcoma 180 cells by 32.4% and 21.0%, respectively. Of these, GLB-A increased the % lymphoblast in the spleen of the tumor-bearing and the normal mice by 20.9% and 123.0%, and the CD4/CD8 ratio by 73.3% and 22.4%, respectively. GLB-A also increased the expression of CD25 (IL-2 receptor alpha ch0ain) in normal mice by 82.0%. These results strongly suggest that GLB-A is a promising candidate for antitumor immunomodulatory medicine.
This study was carried out to investigate the acute toxicity and safety of Ssanghwa-tang extract fermented with Ganoderma lucidum. To evaluate the acute toxity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of Ssanghwa-tang and fermented Ssanghwa-tang extracts were orally administered to 20 male and 20 female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight, and autopsy. Compared with the control group, we could not find any toxic alteration in all treated groups (1250, 2500 and 5000 mg/kg). $LD_{50}$ of Ssanghwa-tang and fermented Ssanghwa-tang extracts might be over 5000 mg/kg and it is very safe to ICR mice.
In this study, the effects of Ganoderma lucidum (GL) on fat metabolism were performed in 3T3-L1 adipocytes. The effects of GL on 3T3-L1 preadipocytes differentiation were also examined. Our results showed that GL decreased the TG content by ORO staining. To elucidate the mechanism of the effects of GL on lowering TG content in 3T3-L1 adipocytes, we examined whether GL modulate the expressions of transcription factors and adipokines related to control of energy expenditure process because adipokines regulate adipocyte mass and increased expenditure may consume much TG in adipocytes. As a result, the expression of C/$EBP{\beta}$, C/$EBP{\delta}$, C/$EBP{\alpha}$, and $PPAR{\gamma}$, genes, which induce the adipose differentiation and adipose-specific FAS, aP2, and adipsin genes, which compose fat formation were decreased. In addition, GL increased the expression of leptin, UCP2, adiponectin in 3T3-L1 adipocytes, resulting in energy homeostasis. In conclusion, GL could regulate transcript factor related to induction of adipose differentiation and control TG content by up-regulation of adipokines related to fat burn.
The effect of exo-polymer from Ganoderma lucidum on the swimming endurance capacity of mice was investigated. The administration of the exo-polymer (100 mg/kg body weight) increased the swimming endurance capacity of mice by about 10 min and reduced the muscle and liver glycogen exhaustion by $18.5\%\;and\;67.2\%$, respectively. A substantial decrease in serum lactate accumulation ($50.7\%$) was also achieved under the influence of the exo-polymer. The exo-polymer was determined to be a glycoprotein with a molecular weight of 780 kDa and found to contain $82.8\%$ carbohydrate and $17.2\%$ protein.
