볶음 치커리의 생리활성 (Biological Activities of Roasted Chicory Root)

  • 국내산 치커리의 생리적 기능성을 알아보기 위해 실험한 결과는 다음과 같다. 복음치커리의 물추출물은 간세포의 독성을 유발시키지 않았으며, 5 mM의 galactosamine을 5시간 배양하였을 때 LDH의 활성이 가장 높게 나타났다. 5 mM의 galactosamine으로 독성을 유발시킨 흰쥐의 간세포에 볶음치커리 물추출물을 투여한 결과 간세포로 부터 유리되는 LDH의 활성을 유의성 있게 감소시켜 간세포 보호 활성을 나타내었다. 볶음치커리 물추출물 800 mg을 당뇨 유발 흰쥐에 10일간 투여한 결과 혈당치는 297 mg/dl로 유의적으로 감소하는 경향을 나타냈었고, 체중은 대조구에 비해 20% 높게 나타났다. Salmonella thyphimurium YG 1024의 복귀돌연변이 콜로니수를 조사한 결과 복음치커리 물추출물 ($10{\sim}5,000\;{\mu}/g$)의 처리구에서는 돌연변이를 유발시키지 않았으며, 2-aminofluorene을 처리한 양성 대조군에 물추출물을 plate당 $1,000{\sim}5,000\;{\mu}/g$ 농도로 처리한 결과 돌연변이 억제 활성은 없었다.

  • Objectives : This study was done to investigate the protective effects of Gagaminjakdowha-Tang on liver injury of rats induced by $CCl_4$ and d-galactosamine. Methods : All animals were divided into 5 groups, those were normal group(untreated), control group(treated with 0.9% Saline solution), sample I group(200mg/kg administrated), sample II group(400mg/kg administrated), Silymarin(200mg/kg administrated) group. Liver injury of rats were induced by $CCl_4$ and d-galactosamine, and then the serumtransaminase(ALT & AST) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities, Liver cytosol malondialdehyde(MDA), catalase, superoxide dismutase(SOD), glutathione S-transferase(GST) and glutathione-peroxidase(GPX) for enzyme activities were measured. Results : The inhibitory effects on the serum ALT activities were noted in both sample I and sample II group. The inhibitory effects on the serum AST activities were noted in only sample II group. The inhibitory effects on the serum ALP activities were noted in both sample I and sample II group. The inhibitory effects on the serum LDH activities were noted in only sample II group. The inhibitory effects on the liver cytosol malondialdehyde were noted in only sample II group. The decresed effects on the liver cytosol catalase activities were inhibited in only sample II group. The decresed effects on the liver cytosol superoxide dismutase activities were inhibited in only sample II group. The decresed effects on the liver cytosol GST activities were inhibited in only sample II group. The decresed effects on the liver cytosol GPX activities were inhibited in only sample II group. The inhibitory effects of the serum ALT activities were noted in both sample I and sample II. The inhibitory effects of the serum AST activities were noted in only sample II group. The inhibitory effects of the serum ALP activities were noted in only sample II group. The inhibitory effects of the serum LDH activities were noted in both sample I and sample II group. Conclusions : Gagaminjakdowha-Tang has protective effects against liver injury in rats induced by $CCl_4$ and d-galactosamine.

흰쥐의 간손상(肝損傷)에 대한 가미유간탕(加味愈肝湯)의 간(肝) 보호효과(保護效果) (Protective Effects of Gami Yugan-tang on Liver Damage in Rats)

  • In order to investigate the protective effects of Gami Yugan-tang on liver damage in rats induced by $CCl_{4}$ and d-galactosamine, the serum transaminase(ALT & AST), alkaline phosphatase(ALP), lactic dehydrogenase(LDH), superoxide dismutase(SOD), catalase, glutathione S-transferase(GST), glutathione peroxidase(GPX) for enzyme activities, and lipid peroxidation were measured. All animals were divided into 4 groups: normal group (untreated), control group (treated with 0.9% saline solution), sample I group (treated with 740mg/kg Gami Yugan-tang), and sample II group (treated with 1,480mg/kg Gami Yugan-tang). The results were as follows : 1. The results of liver damage in rats induced by $CCl_4$ : The protective effects of ALT were displayed in sample I and sample II, and AST, ALP, LDH, SOD, catalase, GST, GPX, and lipid peroxidation were noted in sample II group. It showed slight necrosis of hepatic cell and pathologic changes, for example, inflammatory cells infiltration were improved in sample II group compared to the control group. 2. The results of liver damage in rats induced by d-galactosamine : The inhibitory effects of AST, ALT, LDH, and ALP activities were noted in both sample I and sample II groups. The findings from this experiment suggests that Gami Yugan-tang has protective effects against liver damage in rats induced by $CCl_{4}$ and d-galactosamine.

D-Galactosamine으로 유발된 간손상에 대한 민들레 열수추출물의 예방효과 (Hepatoprotective Activity of Dandelion (Taraxacum officinale) Water Extract against D-Galactosamine-Induced Hepatitis in Rats)

  • 본 연구에서는 실험동물에 민들레 열수추출물 식이를 급여한 후, GalN으로 간손상을 유발함으로써 그 예방효과를 혈액중의 생화학적 변화 및 간조직의 효소적인 변동을 통해서 규명하고자 하였다. GalN의 투여로 현저히 증가하였던 AST, ALT의 활성은 민들레 열수추출물 투여로 억제되었으나 군간의 차이는 보이지 않았고 ALP의 활성과 TBARS 함량은 3%의 추출물을 급여한 군에서 유의적인 감소를 보였다. GalN의 투여로 현격하게 높아졌던 혈중 $TNF-{\alpha}$의 농도 또한 감소하는 경향을 확인하였으나 유의적인 차이는 보이지 않았다. GalN의 투여로 억제되었던 catalase, GSH-reductase, Mn-SOD의 활성은 민들레 추출물 투여로 유의적인 회복이 관찰되었으나 GSH-px의 활성은 그 경향만을 확인할 수 있었다. 조직 검경을 통해 민들레 열수추출물의 간염 예방효과를 확인한 결과 GalN으로 인해 유발된 광범위한 간세포의 괴사와 변성, 지방변성 등이 민들레 열수추출물식이로 다소 감소하는 것을 관찰할 수 있었다. 이상의 결과로 민들레 열수추출물은 AST, ALT와 ALP의 활성 및 산화적 스트레스를 감소시키고 활성산소 해독계에 관여하는 효소의 활성을 증가시킴으로써 GalN으로 인한 간 손상을 예방하는 것으로 사료된다.

홍삼의 전처리에 의한 사염화탄소 및 갈락토사민 유발 간독성에 대한 보호효과 (Effects of Pretreated Korean Red Ginseng on Carbon Tetrachloride and Galactosamine-induced Hepatotoxicity in Rats)

  • In present study, we examined whether or not the pretreatment of Korean Red Ginseng (KRG) could protect hepatotoxicity induced by carbon tetrachloride (CCl$_4$) and D-galatosamine (GalN). For this study, we not only tested activity of various plasma enzymes (AST, ALT, SDH, LDH), which are used as indicators of liver disease, but also checked the change of liver components such as lipid, glutathione and cytochromes content, and several liver enzyme activity. Pretreatment of KRG for two weeks significantly reduced the elevated plasma enzyme activities induced by CCl$_4$ and GalN. Pretreatment of KRG also restored the hepatic enzymes, malonedialdehyde formation, and depletion of reduced glutathione content induced by CCl$_4$ and GalN to near normal level. However, ${\gamma}$-glutamylcysteine synthetase activity was lot affected by KRG. These results suggest that KRG shows the hepatoprotective effect by reducing lipid peroxidation, by reducing the activity of free radical generating enzymes, and by preserving the hepatic glutathione.

Purification and Characterization of a New Galactoside Specific Lectin from Trichosanthes kirilowii Root

  • A new lectin, named TRA, was purified from Trichosanthes kirilowii root by acid-treated Sepharose 6B, Mono-Q, and TSK-gel 3000SW column sequential chromatography. The lectin appeared homogeneous by native gel electrophoresis at pH 4.3 and gave two protein bands of Mr=31 and 28 kDa by SDS-PAGE. The N-terminal amino acid sequences of the polypeptides of TRA have not been reported in amino acid sequences of the lectins. TRA lectin formed a precipitate with asialofetuin, neuraminidase-treated fetuin. A sugar inhibition assay indicated that N-acetyl-D-galactosamine, among the monosaccharides tested, was the most potent inhibitor of TRA-induced hemagglutination. Asialofetuin showed a 260-times stronger inhibitory activity than N-acetyl-D-galactosamine. TRA lectin also showed agglutination with normal leukocytes and lymphoma cells, but not with premature hemopoietic cells. These results suggest that TRA is a novel plant lectin.

Molecular characterization of a lectin, BPL-4, from the marine green alga Bryopsis plumosa (Chlorophyta)

  • A novel lectin specific to N-acetyl-D-galactosamine as well as N-acetyl-D-glucosamine was isolated from Bryopsis plumosa and named as BPL-4. Sodium dodecyl sulfate polyacrylamide gel electrophorese (SDS-PAGE) and matrix-assisted laser desorption / ionization-time of flight (MALDI-TOF) mass spectrometry data showed that this lectin was a monomeric protein with molecular weight 12.9 kDa. The N-terminal amino acid sequences of the lectin were determined by Edman degradation and the full cDNA sequence encoding this lectin was obtained using the degenerate primers designed from the amino acid sequence. The size of the cDNA was 414 bp containing single open reading frame (ORF) encoding the lectin precursor. The homology analysis showed that this lectin might belong to H lectin group. BPL-4 showed high sequence similarity (60.6%) to BPL-3, which is a previously reported lectin from the same species. The comparative analysis on the lectin's primary structure showed two conserved domains including one possible active domain of H lectin group.

돈육 Oligopeptide를 이용한 간질환식의 효과 (Effect of Pork Meat Oligopeptides as a Foodstuff for Experimental Hepatitic Rats)

  • A formula diet based on pork meat oligopeptides(pork meat protein hydrolysates) was designed for experimental hepatitic rats. The rats were given D-galactosamine for 6 days. During this period, the rats were provided with a 12% casein diet or the formula diet which was low in aromatic amino acids and rich in branched chain amino acids. The formula diet was prepared using pork meat oligopeptides as the principal nitrogen source. The hepatitic rats given the formula diet had lower plasma GOT and GPT concentrations. Additionally, the fischer ratio of the plasma was significantly lower in those rats. However, there was no significant difference in the plasma insulin-like growth factor-I concentration before and after acid-ethanol extraction among groups. These results suggest that the formula diet was better for the animals than the casein diet. Furthermore, these findings suggest that pork meat oligopeptides are an excellent material for preparation of formula diets for patients suffering from hepatitis.

한국산 치자(梔子) 엑스 및 Geniposide의 약물성(藥物性) 간장해(肝障害)에 대한 보호효과(保護效果) (Protective Effects of Geniposide and Extract of Korean Gardeniae Fructus -On Hepatic Injury Induced by Toxic Drugs in Rats-)

  • This study was attempted to investigate the effect of Gardeniae Fructus on GOT, GPT, Al.p, LDH activities and level of total cholesterol in serum of $CCl_{4}$ and $_{D}-galactosamine$ intoxicated rats, and bile excretion. The geniposide and extract caused a remarkabel decrease of GPT activities, level of total cholesterol in serum of $CCl_{4}$ intoxicated rats at EtOH Ex. 300, 500 mg/kg p.o., MeOH Ex. and geniposide 100 mg/kg p.o., and GOT, Al.p, LDH activities were significantly decreased compared with control group. It caused a remarkable decrese of GPT, Al.p, LDH activities in serum of $_{D}-galactosamine$ intoxicated rats, and GOT activities was significantly decreased compared with control group. The geniposide and extract caused a remarkable increase of bile excretion, when administration of EtOH extract 500 mg/kg p.o., MeOH extract 100 mg/kg i.d., MeOH extract 50 mg and geniposide 50 mg/kg i. v. compared with normal-control group.

시호의 메탄올 추출물이 일차배양한 뇌, DRG 및 간세포에 미치는 영향 (Effects of the Methanol Extract of Bupleuri Radix on Primary Cultured Brain Cells, DRG and Hepatocytes)

  • Effects of the methanol extract of Bupleuri Radix on primary cultured chicken embryonic brain cells, dorsal root ganglia (DRG) and rat hepatocytes were studied. The methanol extract of Bupleuri Radix at the concentration ranging from $10{\;}{\mu}g/ml\;to\;100{\;}{\mu}g/ml$ significantly recovered the cytotoxicity of rat hepatocytes induced by the treatment of galactosamine; at the concentration of $100\;{\mu}g/ml$, values of GOT and GPT in the culture medium were reduced by the 60% and 75%, respectively of those in the absence of the methanol extract of Bupleuri Radix. The addition of the methanol extract of Bupleuri Radix. into chicken embryonic brain cells which were cultured with a deficient medium significantly increased the number of cells promoting the neurite outgrowth. However, the methanol extract of Bupleuri Radix showed no effect on the activities of PDHC and acetylcholinesterase in primary cultured brain cells.
