The Effects of SosihohapYukmijihwang-tang Administration on biochemical abnormality of Liver (소시호합육미지황탕(小柴胡合六味地黃湯)이 간의 생화학적 이상에 미치는 영향에 대한 임상보고)

  • Objectives:The purpose of this study was to examine the efficacy of SosihohapYukmijihwang-tang on seven patients with biochemical abnormality of the liver. Methods: SosihohapYukmijihwang-tangwas administered to patients for over 10 days continuously. Tests of their AST, ALT, r-GTP, ALP were taken before and after taking medicine. Results: Through this method, we found a positive effect of taking this herbal medicine on biochemical abnormality of the liver. Conclusions: The results suggest that taking this herbal medicine not only reduces hepatotoxicity, but also improves liver function. We need more study about this.

Mutations within the Putative Active Site of Heterodimeric Deoxyguanosine Kinase Block the Allosteric Activation of the Deoxyadenosine Kinase Subunit

  • Replacement of the Asp-84 residue of the deoxyguanosine kinase subunit of the tandem deoxyadenosine kinase/deoxyguanosine kinase (dAK/dGK) from Lactobacillus acidophilus R-26 by Ala, Asn, or Glu produced increased $K_m$ values for deoxyguanosine on dGK. However, it did not seem to affect the binding of Mg-ATP. The Asp-84 dGK replacements bad no apparent effect on the binding of deoxyadenosine by dAK. However, the mutant dGKs were no longer inhibited by dGTP, normally a potent distal end-product inhibitor of dGK. Moreover, the allosteric activation of dAK activity by dGTP or dGuo was lost in the modified heterodimeric dAK/dGK enzyme. Therefore, it seems very likely that Asp-84 participates in dGuo binding at the active site of the dGK subunit of dAK/dGK from Lactobacillus acidophilus R-26.

A study on design of spherical hob for internal gear hobbing (내접치차 가공용 구형 호브의 설계에 관한 연구)

  • Internal gear is the important part of the planetary reduction gear system. This study aims at developing the numerical analysis method for design of spherical hob to manufacture internal gear. Methods of calculation of hob tooth profile, generating tooth profile and tooth profile errors such as the errors of pressure angle, tooth thickness and pitch of spherical hob must be made smaller than those of standard tooth profile. And, the smaller the pressure angle and the number of equivalent pinion tooth become, the larger the tooth profile errors are. One design example of spherical hob is proposed.

  • Cosmetic products including toner and essence were manufactured to evaluate the effect of green tea polyphenols. In addition, irradiation was applied to remove an undesirable color of green tea polyphenol(GTP), which may cause a problem in marketing. The growth inhibition rates of GTP, PT, and PE on all cell lines were shown to be over 80% at 500 ppm concentration. Especially the growth inhibition rates of GTP, PT, and PE on human melanoma(G361) cells were shown to be over 80% at only 100 ppm concentration. Results indicate that the addition of irradiated green tea polyphenol may be effective in the manufacturing of functional cosmetics including toner and essence with various anti-cancer activities.

Internal Perfusion of ADP in Mouse Oocytes Increases Outward $K^+$ Currents (ADP에 의한 생쥐 난자의 외향전류 증가 효과)

  • To find the mechanism underlying the ADP-induced increase in the outward current in ovulated mouse oocytes, we examined changes in voltage-dependent currents using the whole cell voltage clamp technique and the internal perfusion technique. Eggs were collected from the oviduct of superovulated mice with PMSG and hCG. Membrane potential was held at -60 mV (or -80 mV in the case of recording $Ca^{2+}$ currents) and step depolarizations or hyperpolarizations were applied for 300 ms. By step depolarizations, outward currents comprising steady-state and time-dependent components were elicited. They were generated in response to the positive potential more than 20 mV with severe outward rectification and were blocked by external TEA, a specific $K^{+}$ channel blocker, suggesting that they be carried via $K^{+}$ channels. Internally-perused 5 mM ADP gradually increased outward $K^{+}$ currents (IK) 1 min after perfusion of ADP and reached slowly to maximum (150~170%) 5 min later over the positive potential range, implying that ADP might not be acted directly to the $K^{+}$ channels. IK were decreased by 5 mM ATP without affecting the steady-state component of outward current. In contrast to the effect of ADP and ATP on IK, both effect of ATP and ADP on inward $Ca^{2+}$ currents (ICa) could not be detected due to the continuous decrease in current amplitudes with time-lapse ("run-down" phenomena). To check if there is a G protein-involved regulation in the ionic current of mouse oocytes, 1 mM GTP was applied to the cytoplasmic side, and the outward current and inward currents were recorded. ICa was promptly increased in the presence of GTP whereas IK was not changed. from these results, it is concluded that the ATP-dependent regulation is likely linked in the ADP-induced increase in the outward $K^{+}$ current, and G protein-involved cellular signalling might affect ion channels carrying $Ca^{2+}$ and $K^{+}$ in mouse oocytes.

Physical Activity- and Alcohol-dependent Association Between Air Pollution Exposure and Elevated Liver Enzyme Levels: An Elderly Panel Study

  • Objectives: The deleterious effects of air pollution on various health outcomes have been demonstrated. However, few studies have examined the effects of air pollution on liver enzyme levels. Methods: Blood samples were drawn up to three times between 2008 and 2010 from 545 elderly individuals who regularly visited a community welfare center in Seoul, Korea. Data regarding ambient air pollutants (particulate matter ${\leq}2.5{\mu}m$ [$PM_{2.5}$], nitrogen dioxide [$NO_2$], ozone [$O_3$], carbon monoxide, and sulfur dioxide) from monitoring stations were used to estimate air pollution exposure. The effects of the air pollutants on the concentrations of three liver enzymes (aspartate aminotransferase [AST], alanine aminotransferase [ALT], and ${\gamma}$-glutamyltranspeptidase [${\gamma}$-GTP)]) were evaluated using generalized additive and linear mixed models. Results: Interquartile range increases in the concentrations of the pollutants showed significant associations of $PM_{2.5}$ with AST (3.0% increase, p=0.0052), ALT (3.2% increase, p=0.0313), and ${\gamma}$-GTP (5.0% increase, p=0.0051) levels; $NO_2$ with AST (3.5% increase, p=0.0060) and ALT (3.8% increase, p=0.0179) levels; and $O_3$ with ${\gamma}$-GTP (5.3% increase, p=0.0324) levels. Significant modification of these effects by exercise and alcohol consumption was found (p for interaction <0.05). The effects of air pollutants were greater in non-exercisers and heavy drinkers. Conclusions: Short-term exposure to air pollutants such as $PM_{2.5}$, $NO_2$, and $O_3$ is associated with increased liver enzyme levels in the elderly. These adverse effects can be reduced by exercising regularly and abstinence from alcohol.

Members of the ran family of stress-inducible small GTP-binding proteins are differentially regulated in sweetpotato plants

  • Ran is a small GTP-binding protein that binds and subsequently hydrolyzes GTP. The functions of Ran in nuclear transport and mitotic progression are well conserved in plants and animals. In animal cells, stress treatments cause Ran relocalization and slowing of nuclear transport, but the role of Ran proteins in plant cells exposed to stress is still unclear. We have therefore compared Ran genes from three EST libraries construed from different cell types of sweetpotato and the distribution pattern of Ran ESTs differed according to cell type. We further characterized two IbRan genes. IbRan1 is a specific EST to the suspension cells and leaf libraries, and IbRan2 is specific EST to the root library. IbRan1 showed 94.6 % identity with IbRan2 at the amino acid level, but the C-terminal region of IbRan1 differed from that of IbRan2. These two genes showed tissue-specific differential regulation in wounded tissues. Chilling stress induced a similar expression pattern in both IbRan genes in the leaves and petioles, but they were differently regulated in the roots. Hydrogen peroxide treatment highly stimulated IbRan2 mRNA expression in the leaves and petioles, but had no significant effect on IbRan1 gene expression. These results showed that the transcription of these two IbRan genes responds differentially to abiotic stresses and that they are subjected to tissue-specific regulation. Plant Ran-type small G-proteins are a multigenic family, and the characterization of each Ran genes under various environmental stresses will contribute toward our understanding of the distinctive function of each plant Ran isoform.

An Investigation on the Metabolic Syndromes and Health-Related Risk Factors among Male Workers (근로자들의 대사증후군과 건강위험 요인과의 관련성 연구)

  • This study is performed to estimate the prevalence of metabolic syndrome among male workers and to identify the relationships with many related factors including anthropometry, hematological index, serum lipid level, dietary-related behaviors and health-related behaviors. According to the age groups, the 20s are significantly higher in normal and risk groups than in the metabolic syndrome (MS) group, the 30s are significantly higher in MS group than the other groups. The levels of AST and ${\gamma}$-GTP both show significant differences in the order of MS group ($30.3{\pm}8.8U/l$, $91.1{\pm}40.2U/l$) > risk group ($25.7{\pm}8.1U/l$, $41.8{\pm}20.2U/l$) > normal group ($22.8{\pm}6.0U/l$, $26.6{\pm}10.7U/l$). For the frequency of breakfast consumption, the response of 'Every day' is significantly higher in MS group than normal and risk groups, but the response of 'Not at all' is significantly higher in normal group than MS and risk groups. The drinking amount is positively correlated with ${\gamma}$-GTP in normal group, and it is negatively correlated with the hematocrit level, but it is positively correlated with the systolic blood pressure in MS group. AST is positively correlated with glucose concentrations of the MS group. The ALT is positively correlated with waist circumferences and systolic blood pressure in the risk group. The results of this study show that breakfast frequency, education level, drinking amount, drinking frequency, exercise frequency, AST, ALT and ${\gamma}$-GTP levels are all important risk factors of MS. Therefore, it is very important to maintain a healthy life style for the prevention of MS incidence.
