오배자의 항혈전 효과 (Antithrombotic Effect of Galla Rhois)

  • The possibility of Galla Rhois(GR) water extract as an antithrombotic agent was investigated. The effect of GR on platelet aggregation in human platelet-rich plasma(PRP) induced by collagen and ADP in vitro and coagulation parameters in a pathological model induced by endotoxin and hydrocortisone acetate(HA) in vivo were examined. In platelet aggregation assay, GR extract significantly inhibited platelet aggregation induced by collagen and ADP in a dose-dependent manner. GR extract significantly increased the number of platelet and shortened prothrombin time(PT) and activated thromboplastin time(APTT) as compared with the control in pathological model induced by endotoxin and HA. Also, GR extract significantly increased fibrinogen level as compared with the control in a pathological model induced HA. These results suggest that GR may be a promising antithrombotic agent.

  • We have succeeded in designing a PHUND (portable head up navigation display), which has a compact system with a MD panel and full color display device using RGB LED sources. The PHUND has been developed as an alternative to conventional built-in type HUD system targeting the high volume aftermarket with an affordable price.

冠岳山에 있어서 空中에 飛散하는 花紛層에 關한 硏究 (A Study on the Airborne Pollen Calender in Mt. Kwanak)

  • The airborne pollens were investigated by Durhan's pollen trap in Mt. Kwanak from March 1, 1984 to February 28, 1985. The observed airborn pollens were identified 22 families and 49 genera, which were divided into 27 tree pollens. Most airborne pollens were found from late-March to late-September and a few from early-November to late-February, 1985. The first observed pollen of the year was Alnus on March 5 and the last was Artemisia on Decomber 19, 1984. The maximum amount of daily airborne pollens was 823 grains/cm2 on May 16. Peak day and the maximum amount of major airborne pollens were as follows; 166 grains/cm2/day of Alnus pollen on April 4, 38 gr. of Populus on April. 13, 22 gr. of Quercus on May 10, 791 gr. of Pinus on May 26, 139 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia on Sep. 10 and 236 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia airborne pollen for one year was 12,733 grains/cm2 which were composed of 61% tree pollens and 39% herb ones. Decreasing order of pollen counts was Pinus, Alnus, Quercus and Populus in trees, and Graiminae, Artemisia and Ambrosia in herbs. Most of the airborn pollens were deposited in the day-time and a few at night, the maximum at 2∼4p.m.and the mininum at 8∼10 p.m.

A new function of glucocorticoid receptor: regulation of mRNA stability

  • It has long been thought that glucocorticoid receptor (GR) functions as a DNA-binding transcription factor in response to its ligand (a glucocorticoid) and thus regulates various cellular and physiological processes. It is also known that GR can bind not only to DNA but also to mRNA; this observation points to the possible role of GR in mRNA metabolism. Recent data revealed a molecular mechanism by which binding of GR to target mRNA elicits rapid mRNA degradation. GR binds to specific RNA sequences regardless of the presence of a ligand. In the presence of a ligand, however, the mRNA-associated GR can recruit PNRC2 and UPF1, both of which are specific factors involved in nonsense-mediated mRNA decay (NMD). PNRC2 then recruits the decapping complex, consequently promoting mRNA degradation. This mode of mRNA decay is termed "GR-mediated mRNA decay" (GMD). Further research demonstrated that GMD plays a critical role in chemotaxis of immune cells by targeting CCL2 mRNA. All these observations provide molecular insights into a previously unappreciated function of GR in posttranscriptional regulation of gene expression. [BMB Reports 2015; 48(7): 367-368]

Expression of a Glutathione Reductase from Brassica rapa subsp. pekinensis Enhanced Cellular Redox Homeostasis by Modulating Antioxidant Proteins in Escherichia coli

  • Glutathione reductase (GR) is an enzyme that recycles a key cellular antioxidant molecule glutathione (GSH) from its oxidized form (GSSG) thus maintaining cellular redox homeostasis. A recombinant plasmid to overexpress a GR of Brassica rapa subsp. pekinensis (BrGR) in E. coli BL21 (DE3) was constructed using an expression vector pKM260. Expression of the introduced gene was confirmed by semi-quantitative RT-PCR, immunoblotting and enzyme assays. Purification of the BrGR protein was performed by IMAC method and indicated that the BrGR was a dimmer. The BrGR required NADPH as a cofactor and specific activity was approximately 458 U. The BrGR-expressing E. coli cells showed increased GR activity and tolerance to $H_2O_2$, menadione, and heavy metal ($CdCl_2$, $ZnCl_2$ and $AlCl_2$)-mediated growth inhibition. The ectopic expression of BrGR provoked the co-regulation of a variety of antioxidant enzymes including catalase, superoxide dismutase, glutathione peroxidase, and glucose-6-phosphate dehydrogenase. Consequently, the transformed cells showed decreased hydroperoxide levels when exposed to stressful conditions. A proteomic analysis demonstrated the higher level of induction of proteins involved in glycolysis, detoxification/oxidative stress response, protein folding, transport/binding proteins, cell envelope/porins, and protein translation and modification when exposed to $H_2O_2$ stress. Taken together, these results indicate that the plant GR protein is functional in a cooperative way in the E. coli system to protect cells against oxidative stress.

차세대 원전 대형 압력용기용 고강도 SA508 Gr.4N Ni-Cr-Mo계 저합금강 개발 (High Strength SA508 Gr.4N Ni-Cr-Mo Low Alloy Steels for Larger Pressure Vessels of the Advanced Nuclear Power Plant)

  • There is a growing need to introduce advanced pressure vessel steels with higher strength and toughness for the optimizatiooCn of the design and construction of longer life and larger capacity nuclear power plants. SA508 Gr.4N Ni-Cr-Mo low alloy steels have superior strength and fracture toughness, compared to SA508 Gr.3 Mn-Mo-Ni low alloy steel. Therefore, the application of SA508 Gr.4N low alloy steel could be considered to satisfy the strength and toughness required in advanced nuclear power plants. The purpose of this study is to characterize the microstructure and mechanical properties of SA508 Gr.4N low alloy steels. 1 ton ingot of SA508 Gr.4N model alloy was fabricated by vacuum induction melting followed by forging, quenching, and tempering. The predominant microstructure of the SA508 Gr.4N model alloy is tempered martensite having small packet and fine Cr-rich carbides. The yield strength at room temperature was 540MPa, and it was decreased with an increase of test temperature while DSA phenomenon occurred at around $288^{\circ}C$. Overall transition property of SA508 Gr.4N model alloy was much better than SA508 Gr.3 low alloy steel. The index temperature, $T_{41J}$, of SA508 Gr.4N model alloy was $-132^{\circ}C$ in Charpy impact tests, and reference nil-ductility transition temperature, $RT_{NDT}$ of $-105^{\circ}C$ was obtained from drop weight tests. From the fracture toughness tests performed in accordance with the ASTM standard E1921 Master curve method, the reference temperature, $T_0$ was $-147^{\circ}C$, which was improved more than $60^{\circ}C$ compared to SA508 Gr.3 low alloy steels.

  • The effect of improving the tensile and J-R fracture toughness properties of SA508 Gr.1a on the LBB margin for the main steam pipe is investigated. The material properties and microstructure images of the existing main steam piping material SA106 Gr.C used in domestic nuclear power plants and the newly selected material SA508 Gr.1a were compared. For each material, LBB margins were calculated and compared through finite element analysis and crack instability evaluation. The LBB margin of the improved SA508 Gr.1a is found to be greatly improved compared to that of the existing SA106 Gr.C and SA508 Gr.1a. This is because of the increased material's strength and J-R fracture toughness compared to the previous materials. In order to analyze the effect of physical property change on the LBB margin, the sensitivity of each LBB margin according to the variation of tensile strength and J-R fracture toughness was analyzed. The effect of the change in tensile strength was found to be greater than that of the change in fracture toughness. Therefore, an increase in strength significantly influenced the improvement of the LBB margin of the improved SA508 Gr.1a.

광주시민의 식이구성에 관한 조사연구 (A Study on Diets of Gwangju City Residents)

  • In order to understand the diets of Gwangju city resident, nutrition survey was carried out through four years from 1974 to 1977. 1. Average intake of nutritional elements for an adult per day was 580. 25gr in 1974, 583.62gr in 1975, 599.13gr in 1976, and 597.72gr in 1977. 2. Average calorie distribution of the nutritional elements per adult per day was 13.51% protein, 4.70% fat and 81.08% carbohydrate in 1974; 11.6% protein, 7.1% fat and 81.3% carbohydrate in 1975; 15.0% protein, 4.9% fat and 80.1 % carbohydrate in 1976; 13.79% protein, 4.3% fat, and 82% carbohydrate in 1977. 3. The ratio of the animal protein to total protein consumed was 20.01% in 1974, 25.89% in 1975, 26.01% in 1976, and 29.38% in 1977. 4. In aspect of calorie, taken-in calorie was 2456.91 cal in 1974, 2515.10 cal in 1975, 2544.58 cal in 1976, and 2519.08 cal in 1977. However most of calorie were lower than the R.D.A. (3000cal). 5. The amount of ingested protein was 78.18 gr in 1974, 87.61 gr in 1975, 89,93 gr in 1976, and 81.65gr in 1977. The amount was higher than the R.D.A. (80gr) except 1974’s protein amount. 6. As for fat consumption, it was 27.18 gr in 1974, 40.96gr in 1975, 29.61gr in 1976, and 25.64gr in 1977. But these values were much lower than the R.D.A. (40gr) except for 1975’s. 7. In aspect of the minerals (Fe, Ca), intake of Ca was 462.34mg in 1974, 400.07 mg in 1975, 488.14 mg in 1976 and 440.95mg in 1977. The consumption of Ca was lower than the R.D.A. (600mg). Fe was 10.27mg in 1974, 8.54mg in 1975, 11.36mg in 1976 and 20.84mg in 1977. Most of them were higher than the R.D.A. (10mg) except 1975. 8. It was found that thiamine and riboflavin were ingested slightly higher than the R.D.A. (1.4mg, vit. $B_1$, 1.6mg, vit. $B_2$). The amount of niacin and ascorbic acid were also higher than the R.D.A. except 15.41mg of niacin in 1975.

Gustatory Receptors Required for Avoiding the Toxic Compound Coumarin in Drosophila melanogaster

  • Coumarin is a phenolic compound that mainly affects the liver due to its metabolization into a toxic compound. The deterrent and ovicidal activities of coumarin in insect models such as Drosophila melanogaster have been reported. Here we explore the molecular mechanisms by which these insects protect themselves and their eggs from this toxic plant metabolite. Coumarin was fatal to the flies in a dosage-dependent manner. However, coumarin feeding could be inhibited through activation of the aversive gustatory receptor neurons (GRNs), but not the olfactory receptor neurons. Furthermore, three gustatory receptors, GR33a, GR66a, and GR93a, functioned together in coumarin detection by the proboscis. However, GR33a, but not GR66a and GR93a, was required to avoid coumarin during oviposition, with a choice of the same substrates provided as in binary food choice assay. Taken together, these findings suggest that anti-feeding activity and oviposition to avoid coumarin occur via separate mechanisms.
