Effects of Enzymatic Hydrolysates from Hamcho (Salicornia herbacea L.) on Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (함초(Salicornia herbacea)의 효소적 가수분해물이 스트렙토조토신-유발 당뇨쥐의 혈당 강하 및 혈청 지질 개선효과)

  • This study was designed to investigate the effects of enzymatic hydrolysates (EH) from Hamcho (Salicornia herbacea L.) on blood glucose and serum lipid status in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into normal and 5 diabetic groups. The diabetic groups were fed enzymatic hydrolysate-free control (DM) diets or diets supplemented with 0.02% (DM-2), 0.04% (DM-4), 0.08% (DM-8), and 0.16% (DM-16) of enzymatic hydrolysate for 4 weeks. Body weight gains were lower in five diabetic groups than that of the normal group. Blood glucose was decreased in EH-supplemented groups as compared to the normal group, and especially the lowest blood glucose levels were found in DM-4 and DM-8 groups. Activities of three disaccharidase in the middle part of the intestine, such as maltase, sucrase and lactase, in EH-supplemented groups were significantly lower than those of DM group. There was no significant differences in the activities of glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) among all experimental groups. Serum triglyceride in DM group was significantly increased as compared to the normal group, but those of EH-supplemented groups were decreased to the normal level. Total cholesterol level in DM group was higher than EH-supplemented groups and normal group, but that of DM-16 group was significantly decreased to the normal level. HDL cholesterol level in DM group was significantly decreased compared to the normal group, but that of EH-supplemented groups was increased to the normal level. These results suggest that enzymatic hydrolysate from Hamcho has hypoglycemic and hypolipidemic effects on STZ-induced diabetic rats and may be useful as a dietary supplement for the treatment of diabetes.

  • Heterocyclic amines (HCAs) are potent mutagens generated during the cooking of meat and fish, and several of these compounds produce tumors in conventional experimental animals. During the past 5 years or so, HCAs have been tested in a number of novel in vivo murine models, including the following: lacZ, lacI, cII, c-myc/lacZ, rpsL, and $gpt{\Delta}$ transgenics, $XPA^{-/-}$, $XPC^{-/-}$, $Msh2^{+/-}$, $Msh2^{-/-}$ and $p53^{+/-}$ knock-outs, Apc mutant mice ($Apc^{{\Delta}716}$, $Apc^{1638N}$, $Apc^{min}$), and $A33^{{\Delta}N{\beta}-cat}$ knock-in mice. Several of these models have provided insights into the mutation spectra induced in vivo by HCAs in target and non-target organs for tumorigenesis, as well as demonstrating enhanced susceptibility to HCA-induced tumors and preneoplastic lesions. This review describes several of the more recent reports in which novel animal models were used to examine HCA-induced mutagenesis and carcinogenesis in vivo, including a number of studies which assessed the inhibitory activities of chemopreventive agents such as 1,2-dithiole-3-thione, conjugated linoleic acids, tea, curcumin, chlorophyllin-chitosan, and sulindac.

Antioxidative Effects of Electro-acupuncture at $KI_6$ on Rats Treated with AAPH (조해(照海) 전침의 항산화 효과에 대한 실험적 연구)

  • Objectives : This study investigated the antioxidant effect of electro-acupuncture at $KI_6$ on rats treated with 2,2'-azobis(2-amidinopropane) dihydrochloride(AAPH). Methods : After electro-acupuncture at $KI_6$ on 48 rats that were given AAPH(50mg/kg) every day, the author observed several changes in weight, liver index, albumin, total bilirubin, LDL-cholesterol, LDH, glucose, GOT, GPT, SOD and catalase activity, and glutathione, NO, and MDA concentrations. Results : 1. In the $KI_6-EA$ group, the albumin level increased and LDL-cholesterol decreased compared with the control and holder groups. 2. In the $KI_6-EA$ group, SOD activity increased compared with the holder group, glutathione concentration increased compared with the control and holder groups, and catalase activity increased compared with the control group. 3. In the $KI_6-EA$ group, NO concentration decreased compared with the control and holder groups. Conclusions : This case shows that electro-acupuncture at $KI_6$ plays a significant antioxidant role by elevating defenses against cell toxicity caused by oxidative stress and suppressing oxidative injury.

Antioxidant activity of aqua-acupuncture of Polygoni Radix rubra in rats treated with AAPH (적하수오(赤何首烏) 약침(藥針)의 AAPH처리된 흰쥐에 대한 항산화작용)

  • This study was done to elucidate the antioxidant activities of Polygoni Radix rubra(赤何首烏) by way of aqua-acupuncture methods. After 10% & 20% concentrations of liquid extract of Polygoni Radix rubra(PRR) were acupunctured on Joksamri(足三里) of rats with acute and chronic experimental oxidation by AAPH(2, 2' -azobis (aminoidinopropane), hydrochloride), various kinds of experiments were performed. The results were obtained as follows: 1. In acute oxidant experiment, the amount of TBARS was significantly increased from 0.5 hr to 1.5 hr after AAPH treatment 2. In acute oxidant experiment by AAPH, uric acid as significantly increased while the others didn't show any difference such as total protein, albumin, bilirubin. 3. In chronic oxidant experiment by AAPH, PRR significantly increased the body weight in comparison with control group, whereas liver and spleen tended to increase, lung and kidneys tended to decrease. 4. In chronic oxidant experiment by AAPH, PRR significantly decreased the GPT, hemolysis of erythrocytes, TBA and BUN, where GOT was significantly reduced in 20% PRR treated group, and TG was significantly in 10% PRR treated group. These results suggest that PRR has potent antioxidant activity and needs more studies on microsomal fraction of liver and kidney.

Dietary Effects of Lactic Acid Bacteria on Growth, Hematological and Immune Responses of Grey Mullet, Mugil cephalus (유산균 배양액 첨가 사료에 의한 숭어, Mugil cephalus의 성장, 혈액 및 비특이적 면역학적 반응 효과)

  • This study was conducted to investigate the dietary effects of lactic acid bacteria (LAB) supplementation on growth, hematological and immune responses of grey mullet, Mugil cephalus. Three replicate groups of grey mullet (body length, $62.8{\pm}1.04\;mm$ ; body weight, $3.2{\pm}0.13\;mg$) were fed the experimental diets with 0 (control), 1, 2.5 and 5,0 % of LAB for 3 months. Total body length growth rate was increased in 2.5 % supplementation group compared to control (P<0.05). No differences were observed in hematological parameters (hematocrit, hemoglobin, RBCs) and serum chemistry (calcium, magnesium, total protein, glucose, GOT and GPT). The antioxidant activity of grey mullet fed the 5.0 % LAB diets was significantly higher than that of other groups (P<0.05). Both intracellular superoxide anion production and lysozyme activity of kidney were higher in the 2.5 % LAB diet than in the control (P<0.05).

Effect of Medium and Long Chain Triglyceride Diets on the Serum Enzyme Activity in the Rats (MCT와 LCT가 혈청(血淸)의 효소활성(酵素活性)에 미치는 영향(影響))

  • To investigate the effect of feeding rats medium-chain triglyceride(MCT), triglyceride containing primarily $C_8$ and $C_10$ fatty acids, it were compared to the effects of feeding triglycerides composed of long-chain triglyceride (LCT), corn oil and lard, on the serum enzyme activity. For 4 weeks rats fed a diet containing 20% MCT or LCT ${\cdot}$ MCT, as compared with LCT, had the following effects: 1) The total lactic dehydrogenase(LDH) activities in the serum of all experimental group were significantly decreased then those of control group. 2) The activities of glutamic oxaloacetic transaminase (GOT) in the serum of all experimental group were decreased than those of control group. 3) The activities of glutamic pyruvic transaminase (GPT) in the serum of all experimental group were decreased, MCT and LCT group were singinficantly decreased than of control group. 4) The activities of ${\alpha}-amylase$ in the serum of all groups were significantly increased than those of control group. 5) According to electrophoresis, LDH of LDH isoezyme activities in MCT and Lard group were increased with those of $LDH_5$ in corn oil group were increased than those of control gourp. It is suggested that the MCT and LCT fed to rats influence on the activity of various serum enzymes.

Alterations of growth performance, hematological parameters, and plasma constituents in the sablefish, Anoplopoma fimbria depending on ammonia concentrations

  • Juvenile Anoplopoma fimbria (mean length $16.8{\pm}2.2cm$, and mean weight $72.8{\pm}5.4g$) were exposed for 2 months with different levels of ammonia (0, 0.25, 0.50, 0.75, 1.00, and 1.25 mg/L). Growth performances such as daily length gain, daily weight gain, condition factor, and hepatosomatic index were significantly decreased by ammonia exposure. Hematological parameters such as red blood cell (RBC) count, hematocrit, and hemoglobin were also significantly decreased. In plasma inorganic components, calcium and magnesium were significantly decreased by ammonia exposure. In plasma organic components, there was no alteration in cholesterol and total protein. In enzyme plasma components, glutamic oxalate transaminase (GOT) and glutamic pyruvate transaminase (GPT) were significantly increased. The results of this study indicated that ammonia exposure can induce significant growth reduction and blood biochemistry alterations of A. fimbria.

Studies on the Substrances Contained in Glucomannan Lowering Liver and Serum Cholesterol Levels (구약고구마(Glucomannan)의 섬유질이 간 및 혈청지질에 미치는 영향)

  • As reported previously, it was found that glucomannan, contains a potent cholesterol-lowering material. This study was designed to characterize this material further. Both the water sluble and the alcohol soluble extracts of glucomannan were prepared, and each of these extracts were capable of reducing serum and liver cholestrol levels of rats: This finding indicates that a potent cholesterol-lowering material contained in glucomannan is an organic compound with hydrophilic groups.

  • Objective: We investigated the effects of deer antler extract (DAE) on the expression of the weight, leptin, and serum levels (lipid metabolism) of the obese SD rats induced by a $2\%$ cholesterol diet in vivo. Methods: In vivo, the weight, leptin and serum levels were measured after a period of 7 weeks for obese SD rats induced by a $2\%$ cholesterol diet and treated with DAE of three concentrations (0.938 g/kg/5ml, 1.875 g/kg/5ml, 3.75 g/kg/5ml). Results: 1. Body weight didn't change significantly. 2. Leptin levels significantly decreased (p<0.05). 3. Total cholesterol, triglyceride, LDH, sGOT & sGPT levels significantly decreased in the sample (p<0.05). 4. The HDL cholesterol levels significantly increased in the sample (p<0.05). 5. The average size of the epididymal fat cells significantly decreased as the concentration of deer antler extract increased (p<0.05). Conclusion: Deer antler effects inhibition of obese rats.

Effects of Powdered Siho(Bupleuri Radix) on Serum and Liver Lipid Composition and Antioxidative Capacity in Rat Fed High Oxidized Fat (시호(Bupleuri Radix)분말이 과산화지질을 급여한 흰쥐의 혈장 및 간장지질구성과 항산화능에 미치는 영향)

  • Effects of powdered siho(bupleuri radix) on serum and liver lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Sixty male Sprague-Dawley rats weighing 161.25$\pm$2.51g were blocked into four groups according to body weight and raised seven weeks with basal diet(normal group), basal diet and 10% oxidized fat(control group), basal diet, 10% oxidized fat and 2% powdered siho(2% powdered siho group) and basal diet, 10% oxidized fat and 3% powdered siho(3% powdered siho group). Feed intake, body weight gain and feed efficiency were not significantly different among oxidized fat diet groups. The values of total cholesterol and triglyceride in plasma and liver showed no significantly different(p>0.05) in the oxidized fat diet groups. Plasma HDL-cholesterol showed atendency to increase in powdered siho diet groups. The values of thiobarbituric acid in plasma and liver were lower in the powdered siho diet groups than control group. Plasma GOT and GPT activity showed a tendence to decrease in powdered siho groups. Liver glutathione peroxidase activity showed a tendence to increase in powdered siho diet groups. Thus, it seems that powdered siho had some antioxidative elements.
