Attitudes toward Genetically Modified Foods and Willingness to Purchase Them among Housewives (주부의 유전자재조합식품의 태도와 구매의사)

  • In this study, housewives' attitudes toward genetically modified foods (GM foods) and their willingness to purchase GM foods were examined. The findings of this study could provide useful information for consumer education and consumer policy development regarding GM foods. The specific purposes of this study were: (1) to examine consumers' attitudes toward GM foods, (2) to analyze the effect of the perception of GM foods and demographic variables on consumers' attitudes toward GM foods, and (3) to analyze the effect of the perception of GM foods and demographic variables on consumers' willingness to purchase GM foods. The questionnaire used in the survey was constructed by the author, based on existing literature. The survey was conducted with 1,100 housewives, and 723 of the completed survey forms were used in the final analysis. Frequencies, percentages, means, standard deviation, t-tests, ANOVA, Duncan-test, Pearson's Correlation, factor analysis, and discriminant analysis were employed for data analysis methods. Major findings are: (1) Consumers' attitudes toward GM foods consist of three factors, that are, attitude regarding potential danger, attitude regarding the use of GM technique on plants, and attitude regarding the use of GM technique on animals. (2) Consumers with a higher level of education tend to perceive GM food as more dangerous, whereas consumers with a lower level of education tend to accept more the use of GM technique on plants. (3) Consumers who tend not to consider GM foods as dangerous, and those who acknowledged benefits in using GM technique on plants are more willing to buy GM foods.

Multiplex PCR Detection of the GT73, MS8xRF3, and T45 Varieties of GM Canola

  • A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect three varieties of genetically modified (GM) canola. The construct-specific primers were used to distinguish the following three varieties of GM canola; GT73, MS8xRF3, and T45, using multiplex PCR. The FatA (fatty acyl-ACP thioesterase) gene was used as an endogenous canola reference gene in the PCR detection. The primer pair Canendo-FIR containing a 105 bp amplicon was used to amplify the FatA gene and no amplified product was observed in any of the 15 different plants used as templates. The GT73-KHUF1/R1 primer recognized the 3'-flanking region of GT73, resulting in an amplicon of 125 bp. The Barstar-F1/MS8xRF3-R primer recognized the junction region of bars tar and the NOS terminator introduced into MS8xRF3, resulting in a 162 bp amplicon, and the T45-F2/R2 primer recognized the junction region of PAT and the 35S terminator introduced into T45, resulting in an amplicon of 186 bp. This multiplex PCR allowed for the detection of construct-specific targets in a genomic DNA mixture of up to 1% GM canola containing GT73, MS8xRF3, and T45.

Effects of Hydrocolloids on Quality Characteristics of Bread with Rice Flour (쌀을 첨가하여 제조한 식빵의 품질 특성에 미치는 Hydrocolloids의 영향)

  • The effects of different hydrocolloids, including hydroxypropyl methylcellulose (HPMC), xanthan gum (XG), guar gum (GG), and glucomannan (GM), on bread quality characteristics were investigated. The composite flour used for bread production consisted of 80% bread flour and 20% rice flour with 1% different hydrocolloids based on baker's%. Loaf volume, specific loaf volume, baking and cooling loss rate, moisture content, crumb texture and color, and sensory evaluation were determined. Breads containing HPMC and GM showed the highest loaf volumes, but the difference was not significant. Bread containing GM showed the lowest baking and cooling loss rate (7.03 and 7.78%, respectively), and the highest moisture content. Breads containing HPMC and GM showed increased springiness and decreased hardness based on texture profile analysis. Cohesiveness, chewiness, and gumminess of breads containing GM and HPMC showed their lowest values, whereas breads containing XG revealed their highest values. Bread with HPMC showed lightest coloured crumbs. In the sensory evaluation, bread containing GM and HPMC presented their highest scores, whereas bread containing XG showed the lowest scores. Overall, HPMC and GM significantly and positively affected quality characteristics of bread.

Patho-Histologic Study of Mouse which Fed with Heavy Metal Containing Shell Fish (중금속 오염 패류를 먹이로 한 마우스 장기의 조직학적 변화에 관한 연구)

  • Clams were collected from mouth part of Mangeong Gang(river) of which heavy metals were heavily contaminated. Scapharca subcrenata, one of the clams, were fed to mice for 1-2 months with regular food stuffs. Eventhough difference of body weight was recognized between the experimental group and normal diat group, accumulation of heavy metals examined, cadmium was found 1.16 $\mu\textrm{g}$/gm in liver, 3.31 $\mu\textrm{g}$/gm in kidney and 0.21 $\mu\textrm{g}$/gm in blood of experimental group at 60th day, whereas 0.14 $\mu\textrm{g}$/gm, 0.35$\mu\textrm{g}$/gm and 0$\mu\textrm{g}$/gm respectively in normal diet group ate tje same period. No specific histo pathological finding was found in brain and kidney, although slighr fatty change and focal necrosis were found in liver tissues of the experimental group in second month.

Monitoring of Genetically Modified Soybean and Processed Foods in Korean Market using PCR (PCR을 이용한 국내시장에 유통중인 유전자재조합 콩 및 가공식품의 모니터링)

  • A method using PCR was developed for the monitoring of genetically modified soybean (GMS) and GMS derived foods utilized in the market. We designed 3 pairs of specific oligonucleotide primers based on epsps and pat inserted in GMS and ferritin gene as internal standards. Template DNAs isolated from soybean and processed foods were used for multiplex PCR with 3 primer sets. PCR, used with specific primer sets for GMS detection, showed the amplified DNA fragments with GMS template DNA. In this study, GMS containing epsps was detected from soy processed foods manufactured before GM food labeling system, however, GMS containing epsps or pat was not detected from soy processed foods manufactured after GM food labeling system.

  • The different social reflections about the benefits and the potential risks of genetically modified (GM) crops have evolved with .different reactions in different countries. Many countries including Korea are working toward setting down new guidelines. Korea requires companies to label all food that contains more than 3% GM ingredients. One of the rapid and convenient detection methods of GM ingredients is amplification of the introduced DNAs by polymerase chain reaction (PCR). Many PCR kits for this purpose are commercially available. The objective of this study was to evaluate performance of commercialized GM crop detection kits. The results showed that 6 out of 15 kits tested did not meet the requirements even purposed by the manufacturers themselves in terms of stability, reproducibility, and detection limits, suggesting a potential quality control problem in their design stage or production line. The evaluation also suggests that, although the duplex and triplex detection kits allowed unambiguous detection in a single PCR reaction, the monoplex detection kits were the most sensitive to the detection of GM ingredients. The detection limits also differ between soybean and corn. Results from this study will be useful in the development of sound qualitative tracking systems of GM ingredients for monitoring throughout the cultivation of GM crops, their trans-boundary movement, and food production using GM grains as well as for complying with government guidelines associated with GM crops.

Analysis of Levels of Risk Perception using Psychometric Paradigm and Factors Affecting Concerns about Food Risk Elements of Housewives in Daegu (심리측정 패러다임을 이용한 대구 지역 주부들의 식품 위해요소에 대한 위험 지각 수준 및 우려도에 영향을 미치는 요인 분석)

  • This study examined the levels of risk perception of food risk elements by adopting a psychometric paradigm and analyzed factors affecting concerns about food risk elements to obtain basic materials for food safety policy. The data were collected from 296 housewives in Daegu, Korea, by a self-administered questionnaire. Frequency distributions, Pearson's correlation coefficient analysis, factor analysis, Cronbach's ${alpha}$, and multiple regression analyses were conducted by SPSS 21.0. The mean level of concern for food safety was 3.75/5.00 points, and the mean percentage of correct answers about heavy metal contamination was highest among food risk elements. The respondents perceived radioactive contaminated foods, GM foods, and endocrine disruptors as a new, delayed, scientifically unknown, involuntary, serious, and uncontrollable risk in risk perception. According to the result of factor analysis for risk perception, two factors such as non-controllability and dread were categorized. In the risk perception map, radioactive contaminated foods and GM foods were considered as an uncontrollable and dreaded risk, heavy metal contamination, endocrine disruptors, and pesticide residues as a controllable and dreaded risk, and foodborne illness and food additives as a controllable and less dreaded risk. On the other hand, the levels of concerns about food risk elements were higher in order of radioactive contaminated foods, GM foods, and endocrine disruptors. The results of multiple regression analysis showed that age, concern for food safety, percentage of correct answers about food risk elements, non-controllability, and dread influenced the concerns about food risk elements. These results imply that food safety policies should consider differences in consumer's risk perception of food risk elements.

Establishment and application of a qualitative real-time polymerase chain reaction method for detecting genetically modified papaya line 55-1 in papaya products (RT-PCR을 이용한 유전자변형파파야(55-1)검사법 확립 및 파파야가공식품의 적용 연구)

  • Genetically modified (GM) papaya line 55-1, which is resistant to PRSV infection, has been marketed globally. Prompt and sensitive protocols for specific detections are essential for the traceability of this line. Here, an event- and construct-specific real-time polymerase chain reaction (RT-PCR) method was established to detect 55-1. Qualitative detection was possible for fresh papaya fruit up to dilutions of 0.005% and 0.01% for the homozygous SunUp and heterozygous Rainbow cultivars, respectively, in non-GM papaya. The method was applied in the qualitative detection of 55-1 in eight types of commercially processed papaya products. Additionally, papaya products were monitored to distinguish GM papaya using the P35S and T-nos RT-PCR detection methods. As expected, detection capacity was improved via modified sample preparation and the established RT-PCR detection method. Taking these results together, it can be suggested that a suitable method for the extraction and purification of DNA from processed papaya products was established for the detection of GM papaya.

Nutrition Survey in Younghung Island (영흥도 주민의 영양실태조사)

  • Younghung-myun is located on a small island under the jurisdiction of Gyunggi province. The island occupying a total of approximately 40sq km is populated by 5,850 people. Two areas within the island were selected for this survey; 1) Nae 4-ri in Younghung-myun (mainly engaged in farming) 2) Nae 5-ri in Younghung-myun (mainly engaged in fishery) The survey was conducted for the period of 5-12 august 1978. Intake of foods and nutrients of all members in the households surveyed by means of questionnaire was computed based on number of meals a day. On the other hand, daily intake of nutrients by an individual was calculated by sex, age and type of work based on the conversion rate of RDA (Korean Recommended Dietary Allowances for Adult). The results obtained are summarized; 1. Status of food intake Average daily food intake of an individual in the surveyed areas was 1,018gm. Out of 1,018gm above, 490gm which represent 48% of the total intake were from cereals, grains or other vegetative products. Ratio of animal-origin foods was determined as only 16% of the total intake. These findings led us to the conclusion that people in the surveyed areas live heavily on vegetative foods. 2. Status of nutrients intake 1) Overall average intake of calorie by an individual in the surveyed areas was 2026.3Kcal per day. On the other hand, average calorie intake by adult persons was 2,491.3 Kcal per day, which is lower than 2,700 Kcal recommended by RDA by approximately 7.7%. Furthermore, it was observed that most of 2,491.3 Kcal obtained by adult persons was from such vegetative foods as cereals, grains, ect. 2) Overall average intake of protein by an individual in the areas surveyed was 52.2gm per dar. On the other hand, average protein intake among adult persons was 67.7gm a day, which indicates lower intake of protein than 80gm of RDA. It was also found that 84.3% of the protein intake by an adult person was of plant origin. 3) Overall average of fat intake by an individual in the areas surveyed was 18.9gm per day, whereas average fat intake among adult persons was 23. 2gm a day. It was brought to our attention that the average fat intake by adult persons is much lower than 36gm recom mended by RDA. 4) Average calcium intake among adult persons in the areas surveyed was 457.1mg per day, that is somewhat lower than 500mg in RDA. However, average iron intake among adult persons in the areas was slightly higher with 10.5mg a day than 10mg of RDA. 5) With exception with vitamin C, intake of all other vitamin groups, such as vit. A, vit. $B_1$, vit. $B_2$ and niacin, were lower among the inhabitants than recommended by RDA. 3. Economic status Average schooling years of food handlers was 6.5 years with average monthly income of \89,000 in the areas surveyed. Petroleum was used by majority of the households for fuel.
