Pituitary Adenylate Cyclase-activating Polypeptide Inhibits Pacemaker Activity of Colonic Interstitial Cells of Cajal

  • This study aimed to investigate the effect of pituitary adenylate cyclase-activating peptide (PACAP) on the pacemaker activity of interstitial cells of Cajal (ICC) in mouse colon and to identify the underlying mechanisms of PACAP action. Spontaneous pacemaker activity of colonic ICC and the effects of PACAP were studied using electrophysiological recordings. Exogenously applied PACAP induced hyperpolarization of the cell membrane and inhibited pacemaker frequency in a dose-dependent manner (from 0.1 nM to 100 nM). To investigate cyclic AMP (cAMP) involvement in the effects of PACAP on ICC, SQ-22536 (an inhibitor of adenylate cyclase) and cell-permeable 8-bromo-cAMP were used. SQ-22536 decreased the frequency of pacemaker potentials, and cell-permeable 8-bromo-cAMP increased the frequency of pacemaker potentials. The effects of SQ-22536 on pacemaker potential frequency and membrane hyperpolarization were rescued by co-treatment with glibenclamide (an ATP-sensitive $K^+$ channel blocker). However, neither $N^G$-nitro-L-arginine methyl ester (L-NAME, a competitive inhibitor of NO synthase) nor 1H-[1,2,4]oxadiazolo[4,3-${\alpha}$]quinoxalin-1-one (ODQ, an inhibitor of guanylate cyclase) had any effect on PACAP-induced activity. In conclusion, this study describes the effects of PACAP on ICC in the mouse colon. PACAP inhibited the pacemaker activity of ICC by acting through ATP-sensitive $K^+$ channels. These results provide evidence of a physiological role for PACAP in regulating gastrointestinal (GI) motility through the modulation of ICC activity.

Evaluation of Bioethanol Productivity from Sorghum × Sudangrass Hybrid for Cellulosic Feedstocks (셀룰로오스계 원료작물로서 수수-수단그래스 교잡종의 바이오에탄올 생산량 평가)

  • The world demand of renewable bioenergy as an alternative transportation fuel is greatly increasing. Research for bioethanol production is currently being progressed intensively throughout the world. Therefore it will be necessary to develop bioethanol production with cellulosic materials. In this study, the yield of ethanol production was evaluated by simultaneous saccharification and fermentation (SSF) using sodium hydroxide pretreated sorghum ${\times}$ sudangrass hybrids. Composition analysis of 11 varieties of sorghum ${\times}$ sudangrass hybrids was performed for selection of excellent variety to efficiently produce bioethanol. The content of cellulose, hemicellulose, lignin and ash of these varieties were 32~39%, 19~24%, 17~22% and 6~11%, respectively. Among these varieties, 4 varieties of sorghum ${\times}$ sudangrass hybrids were selected for the evaluation of ethanol yield and those were pretreated with 1 M NaOH solution at $150^{\circ}C$ for 30 min using high temperature explosion system. After pretreatment, samples were neutralized with tap water. It contained 52~57% of cellulose. Simultaneous saccharification and fermentation (SSF) was carried out for 48 h at $33^{\circ}C$ by Saccharomyces cerevisiae CHY1011 using Green star variety. The yield of ethanol was 92.4% and the amount of ethanol production was estimated at 6206 L/ha.

  • As considering a comparison study of the structure of mind and psychologic type of C.G Jung to Ji-In-Lon and Sa-Sang constituition of Lee Je Ma, I've got the following conclusion. 1. The study of the structure of mind of Jung and Ji-In-Lon of Lee Je ma have a important meaning to each theory of type of men. They developed their own peculiar theory of type men by the analysis of the mind of men. 2. Jung explained the organization of recognition by four phase theorical structure; consciousness, personal unconsciousness, collective unconsciousness, self. Lee Je Ma explained the organization of recognition by four phase theorical structure;Sa, Sim, Sin, Mul. And theory of type of men, Jung established four type; Tae-Yang-In, Tae-Um-In, So-Yang-In, So-Um-In, They all established the four phase structure, which have things in common. 3. Jung studied the consciousness and unconsciousness and Lee Je Ma studied the Sung-In, Eui, Ye, Ji, and Sim-Yock-Bi, Bak, Tam, Na. I think they have a close relationship. And I think there is relationship between consciousness, unconsciousness to Gun-Ja-Ji-Sim, So-In-Ji-Sim. 4. I think there is thought of relationship between Lee Je Ma's Dae-Dong, Gak-lip, Hang-gi-ji, Hang-gi-hang and Ho-sun, Oh-ak, Sa-sim, Tae-hang and relationship of internal organ's size to Jung's the first function, second function, third function, fourth function. 5. I think the relationship between the type of Jung;thinking type, feeling, type, sensation type, intuition type to the type of Lee Je Ma; Tae-Yang-In, Tae-Um-In, So-Yang-In, So-Yang-In, So-Um-In needs more study.

  • Objectives: Fabry disease (FD) is a lysosomal storage disease caused by the inappropriate accumulation of globotriaosylceramide (Gb3) in tissues due to a deficiency in the enzyme ${\alpha}$-galactosidase A. Hypertrophic cardiomyopathy is one of the chronic complications of FD. We tried to evaluate the prevalence of Fabry disease in the Korean patients with left ventricular hypertrophy (LVH). Methods: A total of 257 patients with LVH were recruited and they were 172 males (mean 56 years, range 30-81 years) and 84 females (mean 66 years, range 45-85 years). Urinary Gb3 was used to screen FD by high performance liquid chromatography-tandem mass spectrometry. Confirmatory tests were done by alpha-galactosidaseA activity using fluorometric assay and by GLA mutation analysis using sequencing. Results: Four patients were screening positive by urinary Gb3 analysis (cutoff, 25 ug/mmol creatinine). But, one female patient was diagnosed with FD confirmed by enzyme analysis in leukocytes as well as by genetic analysis (1/257 patients, 0.4%). She showed 54.3 ug/mmoL creatinine of Gb3 and 15.5 nmole/hr/mg protein (reference range, $55.2{\pm}12.7nmole/hr/mg$ protein) of alphagalactosidase A activity. And she had a heterozygous GLA mutation of c.796G>A (p.D266N). Her daughter was found to be a carrier for FD confirmed by GLA mutation analysis. Asymptomatic carrier showed 25.5ug/mmol creatinine of Gb3 and 42.5 nmole/hr/mg protein (reference range, $55.2{\pm}12.7nmole/hr/mg$ protein) of alpha-galactosidase A activity. Conclusions: The prevalence of FD in Koran patients with LVH was detected as 0.4%. Although the prevalence seems to be low, screening studies are of great importance for detecting hidden cases as well as for identifying other effected family members.

Changes of Bacterial Diversity Depend on the Spoilage of Fresh Vegetables (신선 채소류의 부패에 따른 세균의 다양성 변화 및 세균에 의한 채소 부패 조사)

  • Almost 10~30% of vegetables were discarded by the spoilage from farms to tables. After harvest, vegetables are often spoiled by a wide variety of microorganisms including many bacterial and fungal species. This investigation was conducted to extent the knowledge of relationship the spoilage of vegetables and the diversity of microbes. The total aerobic bacterial numbers in fresh lettuce, perilla leaf, and chicory were $2.6{\sim}2.7{\times}10^6$, $4.6{\times}10^5$, $1.2{\times}10^6\;CFU/g$ of fresh weight, respectively. The most common bacterial species were Pseudomonas spp., Alysiella spp., and Burkholderia spp., and other 18 more genera were involved in. After one week of incubation of those vegetables at $28^{\circ}C$, the microbial diversity had been changed. The total aerobic bacterial numbers increased to $1.1{\sim}4.6{\times}10^8$, $4.9{\times}10^7$, and $7.6{\times}10^8\;CFU/g$ of fresh weight for lettuce, perilla leaf, and chicory that is about $10^2$ times increased bacterial numbers than that before spoilage. However, the diversity of microbes isolated had been simplified and fewer bacterial species had been isolated. The most bacterial population (~48%) was taken up by Pseudomonas spp., and followed by Arthrobacter spp. and Bacillus spp. The spoilage activity of individual bacterial isolates had been tested using axenic lettuce plants. Among tested isolates, Pseudomonas fluorescence and Pantoea agglomerans caused severe spoilage on lettuce.

Acute Oral Toxicity and Anti-obesity Effect of Diglyceride Preparation Containing Conjugated Linoleic Acid in Rat (공액리놀레산 함유 디글리세라이드 식용유지 조성물의 rat에 대한 단회 경구투여독성 및 항비만 효과)

  • The present study was carried out to investigate the acute oral toxicity and anti-obesity effects of a diglyceride preparation containing conjugated linoleic acid (DG+CLA). To test its acute oral toxicity, the DG+CLA was injected into 30 rats (15 males and 15 females) at dosage of 2,000 mg/kg and 5,000 mg/kg. Mortality rates, clinical signs, and body weight changes were monitored for 14 days following administration. According to the results, the lethal dose ($LD_50$) of DG+CLA was determined as >5,000 mg/kg in both sexes. There were no significant changes in general conditions, clinical signs, body weight, and gross lesions between the vehicle control and DG+CLA groups. For the anti-obesity studies, obese Zucker rats were randomly divided into 4 groups and fed saline, soybean oil, diglyceride, and DG+CLA, respectively, for 8 weeks. The DG+CLA groups presented significant differences in body weight, food efficiency ratio, serum lipid levels, and fat weight. Overall, the results showed that the DG+CLA did not have acute oral toxicity and reduced body weight, serum lipid levels, and fat gain.

Study on Optimum Water Supply by Solar Radiation in Cut Rose(Rosa hybrida cv Cardinal) (일사비례제어에 의한 절화장미(Rosa hybrida cv Cardinal)의 급액량 구명)

  • This study was carried out to find optimum accumulative solar radiation in 'Carnidal' of Perlite Media. The pH was stable from 6.0 to 6.7 during cultivation. Electric conductivity by drainage was higher water than by supply water and electric conductivity was increased at later. Inorganic compound, such as phosphoric, kalium and magnesium were accumulated in crossed system. Especially, kalium and magnesium were highly accumulated. When solar radiation high, consumption of the amount of nutrient solution were increased. Sap flew was $273g{\cdot}hr^{-1}$ per hour from 10 : 30 to 11 : 00 AM. However there was no relation-ship between solar radiation and the mount of sap flew. When amount of solar radiation was $250W{\cdot}hr^{-1}$, cut rose 'Cardinal' nutrient consumption was 212.8 mL at nutrient supply of 50 mL. The yield of cut rose 'Cardinal' was 154.6 ea/10a in perlite media. In the cut rose 'Cardinal', nutrient solution was 50 mL as supplied at solar radiation of $200W{\cdot}hr^{-1}$ and nutrient solution was 30mL as supplied at solar radiation of $250W{\cdot}hr^{-1}$ at low solar radiation in perlite.

The Effect of 4 Weeks of Treadmill Exercise and Protein Diet on Immunoglobulin and Antioxidant Enzyme in Rats (4주간의 트레드밀 운동과 단백질 식이가 흰쥐의 면역글로불린 및 항산화효소에 미치는 영향)

  • The purpose of this study was to investigate the effects of treadmill exercise and a protein diet on immunoglobulin and antioxidant enzymes in rats. Forty-four male Sprague-Dawley rats, 5 weeks old, were used. Experimental groups were divided into exercise with protein diet group (A, n=11), exercise group (B, n=11), protein diet group (C, n=11), and the control group (D, n=11). Exercise was administered through a treadmill running program (14~18 m/min, $0^{\circ}$ grade, 20 min/day, 5 day/wk) and these rats were given a 40% protein diet for 4 wk. The results of this study are as follows: the protein diet group showed a significant increase in IgG of immunoglobulin compared to the exercise group and control group; the exercise with protein diet group and protein diet group showed a significant increase in SOD activity of antioxidant enzymes compared to the control group; the exercise with protein diet group, exercise group and protein diet group showed a significant increase in GPx activity of antioxidant enzymes compared to the control group; the exercise with protein diet group showed a significant increase in CAT activity of antioxidant enzymes compared to the protein diet group and control group. In conclusion, treadmill exercise and a protein diet were found to help with immunoglobulin and antioxidant enzymes. Further research regarding the effects of exercise and protein diets is required.

  • The increased concern for the security of the oil supply and the negative impact of fossil fuels on the environment, particularly greenhouse gas emissions, has put pressure on society to find renewable fuel alternatives. Compared to the traditional biofuel, ethanol, higher alcohols offer advantage as gasoline substitutes because of their higher energy density and lower hygroscopicity. For this reason, microbial fermentation is known as potential producers for sustainable energy carriers. In this study, bacterial responses including cellular and molecular toxicity were studied in three different microorganisms, such as Escherichia coli, Clostridium acetobutylicum, and Saccharomyces cerevisiae. In this study, it was analyzed specific stress responses caused by ethanol and buthanol using four different stress responsive genes, i.e. fabA, grpE, katG and recA. The expression levels of these genes were quantified by semi-quantitative reverse transcription-PCR. It was found that four genes have shown different responsive patterns when E. coli cultures were under stressful conditions caused by ethanol and buthanol, respectively. Therefore, in this study, the stress responsive effects caused by these alcohols and the extent of each stress response can be analyzed using the expression levels and patterns of different stress responsive genes.

Lithium Battery Anode Properties of Ball-Milled Graphite-Silicon Composites (볼밀링법으로 제조된 흑연-실리콘 복합체의 리튬전지 음전극 특성)

  • To use as an anode material of lithium secondary battery, graphite-silicon composite powders are prepared by ball-milling with silicon nanoparticles (average diameter 100 nm, 0~50 wt%) and graphite powder (average diameter $15{\mu}m$) and their electrochemical properties are examined. As the silicon content increases, the graphite becomes smaller by the ball-milling and amorphous phase appears whereas the silicon do not suffer the change of nanocrystalline phases and embeds within the amorphous phase of graphite. Cyclic voltammetry at low scan rate reveals that typical oxidation peaks of graphite and silicon appear at 0.2~0.35 and 0.55~0.6 V, respectively, with higher reversibility for repeated cycles. In contrast, the high-scan-rate redox behavior is very irreversible for repeated cycles. High irreversible capacity is exhibited in the initial charging-discharging cycles, but it diminishes as the cycle number increases. The saturated discharge capacity achieves about 485 mAh $g^{-1}$ at 50th cycle for the composite of Si 20 wt%. This is due to the formation of amorphous graphite morphology by the adequate composition (C:Si=8:2 w/w), which efficiently buffers the volume change during alloying/dealloying between silicon and lithium.
