The Design and Fabrication of μCCA-μGI Device for Toxicity Evaluation of Acetaminophen (아세트아미노펜 독성평가를 위한 μCCA-μGI 디바이스의 개발)

  • Deficiencies in the early ADMET(absorption, distribution, metabolism, elimination and toxicity) information on drug candidate extract a significant economic penalty on pharmaceutical firms. Microscale cell culture analogue-microscale gastrointestinal(${\mu}CCA-{\mu}GI$) device using Caco 2, L2 and HEp G2/C3A cells, which mimic metabolic process after absorption occurring in humans was used to investigate the toxicity of the model chemical, acetaminophen(AAP). The toxicity of acetaminophen determined after induction of CYP 1A1/2 in Caco 2 cells was not significant. In a coculture system, although no significant reduction in viability of HEp G2/C3A and L2 cells was found, approximately 5 fold increase in the CYP 1A1/2 activity was observed. These results appear to be related to organ-organ interaction. The oral administration of a drug requires addition of the absorption process through small intestine to the current ${\mu}CCA$ device. Therefore, a perfusion coculture system was employed for the evaluation of the absolution across the small intestine and resulting toxicity in the liver and lung. This system give comprehensive and physiologic information on oral uptake and resulting toxicity as in the body. The current ${\mu}CCA$ device can be used to demonstrate the toxic effect due to organ to organ interaction after oral administration,

Discovery and Evaluation of Polymorphisms in the AKT2 and AKT3 Promoter Regions for Risk of Korean Lung Cancer

  • AKT is a signal transduction protein that plays a central role in the tumorigenesis. There are 3 mammalian isoforms of this serine/threonine protein kinase-AKT1, AKT2, and AKT3-showing a broad tissue distribution. We first discovered 2 novel polymorphisms (AKT2 -9826 C/G and AKT3 -811 A/G), and we confirmed 6 known polymorphisms (AKT2 -9473 C/T, AKT2 -9151 C/T, AKT2 -9025 C/T, AKT2 -8618G/A, AKT3 -675 A/-, and AKT3 -244 C/T) of the AKT2 and AKT3 promoter region in 24 blood samples of Korean lung cancer patients using direct sequencing. To evaluate the role of AKT2 and AKT3 polymorphisms in the risk of Korean lung cancer, genotypes of the AKT2 and AKT3 polymorphisms (AKT2 -9826 C/G, AKT2 -9473 C/T, AKT2 -9151 C/T, AKT2 -9025 C/T, AKT2 -8618G/A, and AKT3 -675 A/-) were determined in 360 lung cancer patients and 360 normal controls. Statistical analyses revealed that the genotypes and haplotypes in the AKT2 and AKT3 promoter regions were not significantly associated with the risk of lung cancer in the Korean population. These results suggest that polymorphisms of the AKT2 and AKT3 promoter regions do not contribute to the genetic susceptibility to lung cancer in the Korean population.

Optimization of Headspace Sampling Using Solid Phase Microextraction For Volatile Organic Acids in Different Tobacco Types

  • A Solid-phase micro extraction(SPME) was evaluated as a tool for headspace sampling of tobacco samples. Several experimental parameters (sampling temperature, pH, and type of SPME fibers) were optimized to improve sampling efficiency in two aspects ; maximum adsorption and selective adsorption of volatile organic acids onto SPME fibers. Among four types of SPME fibers such as PDMS(Polydimethylsiloxane), PA(Polyacrylate), Car/PDMS (Carboxen/Polydimethylsiioxane) and PDMS/DVB(Polydimethylsiioxane/Divinylbenzene) which were investigated to determine the selectivity and adsorption efficiency. A variety of tobacco samples such as flue cured, burley and oriental were used in this study. The effect of these parameters was often dominated by the physical and chemical nature (volatility, polarity) of target compounds. This method allowed us to make important improvements in selectivity and sensitivity. The Car/PDMS fiber was shown to be the most efficient at extracting the 10 selected volatile organic acids. The parameters were optimized: $80^{\circ}C$ adsorption temperature, 30 min of adsorption time, $240^{\circ}C$ desorption temperature, 1 min of adsorption time.

  • In this paper, we analyze the Transition of Isc by natural solar spectrum of c-Si and a-Si PV module. Commonly, performance of photovoltaic (PV) module is estimated under the standard test condition (STC). That is, solar irradiance $1kW/m^2$, solar spectrum distribution: AM1 5G, module temperature $25^{\circ}C$ This means it rarely meets actual outdoor conditions. The solar spectrum always changes. So it is rare to fit the standard solar spectrum AM1 5G defined in ASTM G173-03 or IEC 60904-3. Thus spectral response of PV module is different depending on the material. so we estimated the variation of Isc at every minutes by comparing c-Si PV module with a-si PV module for outdoor conditions.

In vitro anticancer and antioxidant effects of acetone extract of Eucommia ulmoides oliver leaves (두충잎 아세톤 추출물의 in vitro 항암 및 항산화 효과)

  • In vitro anticancer and antioxidant effects of acetone extract from leaves of Eucommia ulmoides Oliver were investigated. The extraction yield and total phenolic content of the acetone extract were $1.13{\pm}0.033%$ (w/w) and $36.7{\pm}1.96mg$ gallic acid equivalents/g-extract, respectively. $GI_{50}$ values of the acetone extract for human non-small cell lung cancer cells (A549), human colon cancer cells (SNU-C4), human cervical cancer cells (HeLa), and human embryonic lung epithelial cell (L132) were 53.4, 53.8, 88.3, and $153.9{\mu}g/mL$, respectively. The acetone extract effectively inhibited the proliferation of human non-small cell lung cancer (A549) and colon cancer (SNU-C4) cells in a concentration-dependent manner, but was less cytotoxic with human normal cells (L132). $EC_{50}$ values of the acetone extract for free radical scavenging, reducing power, and lipid peroxidation inhibition were about 2,000, 275.8, and $257.9{\mu}g/mL$, respectively. The acetone extract showed a potent reducing power and lipid peroxidation inhibitory activity in a concentration-dependent manner.

CpG Islands Detector: a Window-based CpG Island Search Tool

  • CpG is the pair of nucleotides C and G, appearing successively, in this order, along one DNA strand. It is known that due to biochemical considerations CpG is relatively rare in most DNA sequences. However, in particular subsequences, which are a few hundred to a few thousand nucleotides long, the couple CpG is more frequent. These subsequences, called CpG islands, are known to appear in biologically more significant parts of the genome. The ability to identify CpG islands along a chromosome will therefore help us spot its more significant regions of interest, such as the promoters or 'start' regions of many genes. In this respect, I developed the CpG islands search tool, CpG Islands Detector, which was implemented in JAVA to be run on any platform. The window-based graphical user interface of CpG Islands Detector may facilitate the end user to employ this tool to pinpoint CpG islands in a genomic DNA sequence. In addition, this tool can be used to highlight potential genes in genomic sequences since CpG islands are very often found in the 5' regions of vertebrate genes.

Characteristics of a new button mushroom variety 『Dahyang』 (갈색 양송이 신품종 '다향'의 형태적 특성)

  • Dahyang, a CNARES's newly bred cultivar, is a CM061202 strain isolated and selected from 161 single-spores of brown button mushroom. It was selected as a new cultivar after comparison of yield, mycelial growth and quality with No. 705. Mycelial growth of Dahyang on compost extract mushroom complete medium at $25^{\circ}C$ for 15 days was 63mm when it was compared with that of No. 705 with 38mm. The cultivation temperature of Dahyang was 15 to $19^{\circ}C$ which was $2^{\circ}C$ higher than that of No. 705 with 15 to $19^{\circ}C$ cultivated on rice straw bed. The color of pileus of Dahyang was brown. Pinhead producing period was 30 days which is 1 day late than that of No. 705 with 29 days. Dahyang yielded 37% more with $49.1kg/3.3m^2$ than that of No. 705 with $35.8kg/3.3m^2$, respectively. The weight was 16.0g which was heavier than 14.0g of No. 705. The thickness and diameter were also more than those of No. 705. The stipe was shorter and thicker than that of No. 705. Also physical characteristics such as hardness and elasticity were better than that of No. 705.

Biodegradation of Aniline by Pseudomonas Rhodesiae isolated from River Water (강물에서 분리한 Pseudomonas rhodesiae의 아닐린 분해)

  • Two Bacterial strains 1-C and 51-C capable of utilizing aniline as a sole source of carbon and energy were isolated from river waters. Both strains were identified as Pseudomonas rhodesiae based on their physiological and biochemical characteristics and 16S rRNA gene sequence. The strains were able to grow on the mineral salt media containing aniline at concentrations up to 6,000 ${\mu}g/mL$. Pseudomonas rhodesiae 51-C completely degraded aniline in a mineral salt medium containing 300 ${\mu}g/mL$ of aniline as a sole carbon and energy source within 16 hours. The optimum pH and temperature for its growth and aniline degradation were 7.0 and $30^{\circ}C{\sim}35^{\circ}C$, respectively. This is the first report of aniline degradation by P. rhodesiae strains.

Antioxidative and Anti-diabetic Activity of C3GHi, Novel Black Rice Breed (Cyanidine-3-glucoside (C3G) 색소의 함유량이 증대된 쌀 신품종(C3GHi)의 항산화 및 항당뇨 활성)

  • C3GHi, a novel black rice variety developed by conventional breeding has more high contents of cyanidine-3-glucoside (C3G) and a more strong antioxidant than normal black rice. In this study, we investigate the antioxidative potential and anti-diabetic activity of C3GHi and extract of it for the purpose of development of functional materials or replacement meal for diabetic patients. Using in vitro antioxidative activity assay system such as DPPH radical quenching assay, superoxide anion radical scavenging assay, inhibition of lipid peroxidation and DNA breakage assay, C3GHi extract was observed to have more high antioxidative activity than normal black rice breed. The glycemic index of a freeze dried powder of C3GHi brown rice showed $43.7\pm18.8$, which belonged to low GI food (GI of 55 or less). Using in vivo diabetic model such as db/db mice model and streptozotocininduced diabetic model, C3GHi rice extract decrease blood glucose level and inhibit oxidative stress in blood. From the results, we think that the C3GHi varieties have a potential for the functional materials or ingredient of meal replacement for diabetic patients.

Phylogenetic Relationship among Several Korean Coastal Red Tide Dinoflagellates Based on their rDNA Internal Transcribed Spacer Sequences

  • The nucleotide sequences of the internal transcribed spacer regions (ITS1 and ITS2) of ribosomal DNA (rDNA), and the 5.85 rRNA gene, have been determined for 13 strains of dinoflagellates in order to analyze the phylo-genetic relationship. The DNA sequences contained considerable variation in the ITS regions, but little in the 5.85 rDNA. In addition, the ITS1 was more variable than the ITS2 in all species examined. The nucleotide length of this region varied from 519 bp to 596 bp depending on the taxa. The investigated taxa were divided into three large groups based on the ITS length, i. e., a group with short ITS region (A. fraterculus and Alexandrium sp.), a with ITS region group (P. micans, P. minimum and P. triestinum) and a with ITS region group (G. impudicum, C. polykrikoides, G. sanguineum, G. catenatum and H. triquetra). The relationship between nucleotide length of ITS1 and that of ITS2 was negative, whereas G+C content and nucleotide length showed positive correlation. In phylogenetic analyses producing NJ trees, the topology was similar cluster and clearly divided the taxa into three groups based on 5.8S rDNA that were similar to those based on morphological characteristics. In particular, G. impudicum was more closely related to G. catenatum than to C. polykrikoides using phylogenetic analysis. From this study, we chew that the length of ITS region contributes to discriminate Korean harmful algal species and ITS analysis is a useful method for resolving the systematic relationships of dinoflagellates.
