Genetic diversity of forty-four strains of Xanthomonas campestris pv. vesicatoria from diverse geographic origins was investigated using random amplified polymorphic DNA (RAPD) of genomic DNA. One hundred and thirty-seven amplified fragments were produced by polymerase chain reaction with a set of 14 random primers, and the sizes of amplified DNA fragments ranged approximately from 0.3 to 3.2 kb. Cluster analysis of genetic similarity among the strains generated the dendrogram that clearly separated all strains from each other. The 44 strains of X. campestris pv. vesicatoria were classified into 4 major genomic DNA RAPD groups and 15 subgroups at the genetic similarity of 0.60 and 0.92, respectively. The strains from foreign countries formed discrete subgroups, but the United States strain 87-77 clustered closely with some of Korean strains together. Thirty-nine Korean strains were classified into 11 subgroups, and especially Masan strain Ms93-1 clustered distinctly far from the other Korean strains. RAPD polymorphism suggests strongly the occurrence of genetic differentiation of X. campestris pv. vesicatoria and the existence of genetically distinctive subgroups among the populations in Korea.
The genetic variations in three major river populations viz. the Halda, the Jamuna and the Padma of the Indian major carp, Catla catla were analyzed by Random Amplified Polymorphic DNA (RAPD) markers. Four decamer primers were used for amplifying DNA of 10 individuals from each population. The proportion of polymorphic loci and the gene diversity estimates were 59.4 and 0.20 for the Halda, 37.5 and 0.14 for the Jamuna and 46.9 and 0.16 for the Padma populations respectively indicating the existence of a relatively high level of genetic variation in the Halda river population. The inter-population similarity indices, gene flow and genetic distance values indicated that the Jamuna-Padma population pair of catla was genetically closer than the Halda-Jamuna and the Halda-Padma population pairs in compliance with the geographical distances among them. The coefficient of gene differentiation ($G_{ST}$=0.13) reflects some degree of genetic differentiation among three populations of catla studied. The data suggest that the RAPD technique could be used to discriminate different river populations of catla.

Genetic variation was surveyed at four microsatellite loci and 1416 base pairs (bp) of the mitochondrial DNA (mtDNA) cytochrome c oxidase I gene (COI) to clarify the genetic structure of the small yellow croaker, Larimichthys polyactis, in the Yellow and East China Seas, especially regarding four provisional populations, (one Korean and three Chinese populations). Based on microsatellite DNA variations, the estimated expected heterozygosity ($H_E$) in each population ranged from 0.776 to 0.947. The microsatellite pairwise $F_{ST}$ estimates showed no significant genetic differentiation between the populations. MtDNA variations also indicated no genetic structure in L. polyactis, but very high variability. The absence of genetic differentiation among and within populations of L. polyactis may either result from the random migration of the adult or the passive dispersal of the eggs and larvae.
Using the method of 'random sampling in typical colonies of the central area of the habitat', 60 Small-tailed Han sheep were obtained in Jining city, Shangdong province. The variations of Small-tailed Han sheep at 12 structural loci encoding blood proteins were detected by several electrophoresis techniques and their gene frequencies were then estimated. The same data of four other sheep populations from Near-sea Mainland in East Asia were cited for the analysis of genetic differentiation. The average heterozygosities of five populations, namely Kharkhorin sheep, Ulaanbaatar sheep, Small-tailed Han sheep, Hu sheep and Cham Tribe sheep were 0.3447, 0.3285, 0.3157, 0.3884 and 0.2300, respectively. The coefficient of gene differentiation among four populations, Kharkhorin sheep, Ulaanbaatar sheep, Small-tailed Han sheep and Hu sheep, was 0.045557, and that between these four breeds and Cham Tribe sheep was 0.088005, indicating that the level of gene differentiation among the former four sheep populations of Mongolian group was comparatively lower than that between Cham Tribe sheep and other four sheep populations. The origin of Cham Tribe sheep deserve further research. The documentary research on the evolution of Small-tailed Han sheep and Hu sheep from Mongolian sheep was further verified by the biochemical experiments in the study. It was reasonably deduced that Hu sheep, Small Tailed Han sheep and Cham Tribe sheep were decreasingly influenced by the bloodline of Mongolian sheep.

Background: Juniperus chinensis L. populations are distributed locally on several areas including coastal cliffs which are difficult to access in the central eastern Korea. Wild populations inhabit relatively barren environments such as rocky areas and cliffs, which are very sensitive to even minor environmental disturbances including artificial interventions and natural disturbances, and thus demonstrate great fluctuations in the population size and density. This study aims to analyze the genetic diversity, differentiation, and genetic structure of each population in order to provide useful data required to establish a substantial conservation strategy of J. chinensis. Results: The genetic diversity of J. chinensis at the population level (P = 78.7%, h = 0.282, S.I. = 0.420) was somewhat higher compared with those measured in the same genus, Juniperus. The genetic differentiation degree among nine populations established naturally in central eastern Korea was 11.50% and that among sub-populations within the same area was 5.52%. On the other hand, genetic variation of individuals within the populations was 82.93%. But frequency of the main allele was different among loci. In particular, fixation of allele frequency and occurrence of rare allele in the highly isolated population suggest a likelihood that genetic drift would occur in populations of this plant. As the result of analysis on the genetic structure of nine populations, nearby populations and isolated populations tended to form separate clusters from each other as the hypothetical number of clusters (K) increase. Conclusions: This result implies that if the population size of J. chinensis is reduced due to environmental change and artificial and/or natural disturbances in the future, it could affect negatively on the genetic diversity of the plant species. In order to maintain and conserve genetic diversity of J. chinensis, ecological network, which can help genetic exchange among the local populations, should be prepared, and conservation strategies in situ as well as ex situ are also required with continuous monitoring.

The 7 sheep microsatellite markersOarFCB48, OarAE101, MAF33, OarFCB11, MAF70, OarFCB304 and OarFCB128, which were located on chromosomes 2, 4, 6, 9, 17 and 19, were selected to PCR in Hu sheep, Tong sheep and their closely related species,the goat. They were studied with the amplifying result of 7 microsatellite sites of Hu Sheep, Tong Sheep and goats, the data of allele number and range of allele' size of amplifying were analyzed with ANOVA. The results showed that there were no significant differences (p<0.05) in microsatellite DNA sites among 3 populations. Concerning the conservation of microsatellites in closely related species, selecting microsatellite sites located on the chromosome where the Robertsonian fusion was caused between sheep and goat, may be used in research into genetic differentiation and evolutionary relationships between sheep and goats.
To identify sand crab Ovalipes punctatus populations and establish management units for each population, mtDNA COI regions were analyzed. As a result, the clade of O. punctatus in Korea were separated by two with a genetic distance of 0.17-2.08%, and there was no significant difference in the result of pairwise FST values representing genetic differentiation by sampling areas (p > 0.05). Also, no geographical separation found in the distribution of haplotypes and the results of the haplotype network. This result suggests that O. punctatus larvae were dispersed for a long time by the ocean current by suffering meroplanktonic period for 1 month, and increased the gene flow due to the development of the swimming legs for the increase in mobility. Therefore, in the results of mtDNA COI region analysis of O. punctatus in the East Sea, Yellow Sea, South Sea and East China Sea (Ieodo) of Korea, no clear intra-species differentiation was found.
ISSR 표지를 이용하여 전나무 6개 집단의 유전다양성과 유전구조를 분석하였다. 6개 ISSR primer로 유전다양성을 추정한 결과, 집단별 다형성 유전자좌의 비율은 평균 85.6%, 이형접합도 기대치($H_e$)는 0.288로 나타났다. AMOVA 결과, 전체 유전변이에서 5.6%는 집단간, 94.4%는 집단내 개체간 차이에 기인하는 것으로 나타났다. 베이즈 추론에 근거한 유전분화는 ${\theta}^{II}$와 $G_{ST}$가 각각 0.045, 0.038로, 근연교배 정도는 0.509로 추정되었다. Mental 검증에서 집단간 지리적 거리가 멀수록 유전적으로 상이한 것으로 판명되었다(r = 0.74, P < 0.05). UPGMA 방법과 PCA 결과에 따라서 남원, 청도, 문경 집단을 한 군집으로, 인제, 홍천, 평창 집단을 다른 군집으로 나눌 수 있었다. 베이즈 군집분석에서는 유전변이 분포에 따라서 남원, 문경 집단이 한 군집으로 인제, 홍천, 평창, 청도 집단이 다른 한 군집으로 묶여서 2개의 상위 군집으로 나뉘었다. 빈도주의 분석에 따른 '군집'을 반영한 AMOVA 결과에서 전체 유전변이의 3.9%를 군집의 영향으로 설명할 수 있었으며, 전나무 집단의 지리적 분포는 베이즈 분석보다는 UPGMA 방법에 의한 구분과 일치하는 것으로 나타났다.
Objective: The growth differentiation factor 8 (GDF8) gene plays a key role in bone formation, resorption, and skeletal muscle development in mammals. Here, we studied the genetic variants of GDF8 and their contribution to body conformation traits in Chinese Dabieshan cattle. Methods: Single nucleotide polymorphisms (SNPs) were identified in the bovine GDF8 gene by DNA sequencing. Phylogenetic analysis, motif analysis, and genetic diversity analysis were conducted using bioinformatics software. Association analysis between five SNPs, haplotype combinations, and body conformation traits was conducted in 380 individuals. Results: The GDF8 was highly conserved in seven species, and the GDF8 sequence of cattle was most similar to the sequences of sheep and goat based on the phylogenetic analysis. The motif analysis showed that there were 12 significant motifs in GDF8. Genetic diversity analysis indicated that the polymorphism information content of the five studied SNPs was within 0.25 to 0.5. Haplotype analysis revealed a total of 12 different haplotypes and those with a frequency of <0.05 were excluded. Linkage disequilibrium analysis showed a strong linkage (r2>0.330) between the following SNPs: g.5070C>A, g.5076T>C, and g.5148A>C. Association analysis indicated these five SNPs were associated with some of the body conformation traits (p<0.05), and the animals with haplotype combination H1H1 (-GGGG CCTTAA-) had greater wither height, hip height, heart girth, abdominal girth, and pin bone width than the other (p<0.05) Dabieshan cattle. Conclusion: Overall, our results indicate that the genetic variants of GDF8 affected the body conformation traits of Chinese Dabieshan cattle, and the GDF8 gene could make a strong candidate gene in Dabieshan cattle breeding programs.
As a part of the on-going effort to conserve endangered Scrophularia takesimensis Nakai in Korea, its genetic structure and diversity from 3 population, consisted of 14 subpopulations in Ulleung Island were analyzed using RAPD band patterns. Out of 60 primers tested, 33 generated amplified bands with its genome, including 149 polymorphic and 67 monomorphic bands. The highest number (146) was found in northern population, especially, 64 in HY subpopulation; the smallest (40) in eastern population. An examination of its genetic structure with AMOVA revealed that about 60% of all variations could be assigned to among subpopulations within populations. Population differentiation among populations and subpopulations is seriously going now because of habitat fragmentation due to human activities, such as road and small port construction. Although the habitats of S. takesimensis in Ulleung Island, Korea are disappeared at an alarming rate, significant levels of genetic variation still exist at species level, and population level, especially northern population. Therefore, three conservation strategies should be needed urgently; 1) preservation of populations as it stands, 2) establishment of recovery plan to connect population and subpopulations genetically, and 3) long-term monitoring.
