Effect of oral administration of Ginsenoside-Rb2 on rotavirus infection

  • Glycosaminoglycans(PT -Gag) were isolated from the porcine testis. From the PT -Gag. we obtained two different types of Gag fractions using Dowex macroporous Resin MSA-1 column. PT-Gag-1.5% NaCl and PT -Gag-16% NaCl. Various biological activities of the GAGs were examined in aspect of anticoagulant and immunomodulating activity. The anticoagulant activity of the GAGs was evaluated by activated partial thromboplastin time (aPTT ) assay and thrombin time (TT) assay. The GAGs of porcine testis markedly increased the clotting times of both of aPTT and TT. showing that PT-Gag-16% NaCl was more effective than PT-Gag-1.5% NaCl. The immunomodulating activity of the GAGs was examined in relation to regulation of cytokine production of mutine peritoneal macrophages. Treatment with the GAGs promonently enhanced the prodution of cytokines. IFN-${\gamma}$, from macrophages. Taken together. GAGs isolated from porcine testis possess biological functions such as anticoagulant and immunomodulating activity.

Immunomodulating and Anticoagulant Activity of Glycosaminoglycans Derived from Porcine Testis

  • Glycosaminoglycans (GAGs) were isolated from the porcine testis, and their immuno-modulating and anticoagulant activity was investigated. From anion exchange chromatography (Dowex Macropolous Resin) used for further isolation of porcine testis GAGs (PT-GAGs), two fractions (PT-GAG-1.5 and PT-GAG-16) eluted by different salt concentration were obtained. In immunomodulating activity test, PT-GAG-1.5, but not PT-GAG-16, significantly enhanced the growth of murine peritoneal macrophages. In addition, treatment with PT-GAG-1.5 induced the production of cytokines, interleukin-1$\beta$ (IL-1$\beta$), interferon-${\gamma}$ (IFN-${\gamma}$) and tumor necrosis factor-$\alpha$ (TNF-$\alpha$), from murine microphages. Unexpectedly, both of PT-GAGs had no effect on the growth of murine splenocytes. The anticoagulant activity of PT-GAG-1.5 and PT-GAG-16 was examined by activated partial thromboplastin time (aPTT) assay and thrombin time (TT) assay. Both of PT-kGAGs significantly increased the clotting times of aPTT and TT in a dose-dependent manner. The anticoagulant activity of PT-GAG-16 was found to be higher than that of PT-GAG-1.5. These results suggest that PT-GAGs possess biological activities such as immunomodulating activity and anticoagulant activity.

미더덕 껍질로부터 Glycosaminoglycans의 추출 (Extraction of Glycosaminoglycans from Styela clava Tunic)

  • GAGs의 추출 방법 중 효소의 사용으로 추출 효율의 상승과 원료수급의 문제점을 해결할 수 있었다. 그러나 효소 사용에 따른 생산단가의 상승과 단백질 함량의 증가에 따른 화장품 규격을 만족시킬 수 없음으로, 1/60 M sodium phosphate buffer로 105$^{\circ}C$로 열수 추출하는 것이 화장품 규격을 만족시키는 방법으로 나타났다. 이렇게 추출한 GAGs의 SO$_4$ 함량은 31.2%이고 회분의 함량이 22.2%로 GAGs 추출 향상을 위해 사용되어지는 sodium phosphate의 사용으로 회분 함량이 높아졌음을 알 수 있고 무기질 분석에서 Na의 함량이 3.0 mg%로 총 무기질의 47.6%를 차지하고 있었다. 시료를 HPLC로 분석하였을 때 chondroitin sulfate A, C와 동일한 분석시간을 나타내어 chondroitin sulfate의 함유 가능성을 나타내고, glucosamine과 galactose가 78.0% 존재하는 것을 당조성 분석을 통해 확인하였다. 일반 성분, HPLC분석, 당 조성분석과 아미노산 분석결과로 glucosamine과 galacturonic acid가 주가 되어 threonine으로 연결된 구조를 하는 GAGs라는 것을 알 수 있다. 화장품 원료 기준에 맞추기 위해서는 sulfate 함량이 35.0~45.0%, 단백질 함량은 14.0~22.0%로 유지해야하므로 제단백결과 5.0%, 10.0%, 20.0% TCA (w/v) 처리, 10.0%, 20.0%, 40.0% HCI (v/v) 처리, UF(ultra filtration)를 포함한 10.0% TCA (w/v), 20.0% SSA (w/v), 25.0% HCI (v/v) 처리 결과 5.0% TCA (w/v) 및 10.0% HCI (v/v)이 화장품 기준에 적합하며 경제적이며 효율적이라는 것을 알 수 있었다.

  • Among the Glycosaminoglycans (GAGs), Heparin and its fractions or fragments, obtained by several different processes, are of considerable interest .In these last few years, the goal of the researchers in this field has been finding molecular species having selective action, like for instance species having only antithrombotic activity disjointed from any anticoagulant effect, and assessing the effects of these GAGs and of other GAGs, like dermatan sulphate, not only in the field of venous or arterial thrombosis but also on cell factors like smooth muscle cell proliferation and even on aspects of antiinflammatory activity.

Antibody derived from insect glycosaminoglycan

  • We prepared antibodies from insect glycosaminoglycans (GAGs) and assayed the titer. Nine polyclonal antibodies against insect GAGs were raised for development of an ELISA in biological fluids (mice serum). The 3th booster collection of antiserum of BALB/c mice as a primarily antibody was assayed for titer determination by ELISA method. In sandwich ELISA of GAGs derived from Isaria sinclairii or other insects, antiserum from insect GAGs gave satisfactory results for so potent antibody(100: 1~1000:1) raising (manufacturing) agent in range of 10 ng/ml.

오만둥이(Styela plicata)에서 글리코스아미노글리칸의 최적 추출조건 설정 및 주름개선 효능 (Establishment of Optimum Extraction Conditions and Wrinkle Improvement Evaluation of Glycosaminoglycans in Styela plicata)

  • Styela plicata are naturally-occurring marine resources easily found along the coastlines that have established their niche as functional food and nutraceuticals ingredient along with their increasing consumer demand. Ascidian contain a large amount of dietary fiber but only the meat has been utilized and consumed while the rest of its parts are discarded. Also, various studies have been conducted on the meat of ascidians while studies on the functionality of the ascidian tunics, which were mostly undervalued, were scarce. In this study, we investigated and explored the glycosaminoglycans (GAGs) contents in the tunics of S. plicata, and their potential use as functional ingredient in pharmaceutical and nutraceutical applications. Sulfated GAGs and uronic acids contents were 8.9-10.7 g/100 g and 9.4-11.3 g/100 g, respectively. Highest GAGs content was extracted with optimum Brix at 7-9. Extraction efficiency using hot water at 121℃ was 4.22% while enzyme extraction using Protamex was more efficient at 5.91%. GAGs extracted from S. plicata tunics exhibited collagenase inhibitory activity of 75.2% at 100 ㎍/mL and procollagen synthesis activity of 80.1% at 100 ㎍/mL.

  • Mucopolysaccharidosis (MPS) is a genetic disorder with deficiency of Iysomal enzymes needed for the degradation of glycosaminoglycans(GAGs). This storage disease is characterized by intra-lysosomal accumulation of GAGs. progressive mental and physical deterioration. multi-organ failure and premature death. Quality of life (QOL) is very low in MPS patients. Protein-GAGs interactions play prominent roles in cell-cell recognition and cell growth. Acharan sulfate (AS), isolated from the giant African snail Achatina fulica, is a novel member of glycosaminoglycan families. It showed antitumor activity by the inhibition of angiogenesis. Composition of Biologically Active Substances and Antioxidant Activity of New Zealand Deer Velvet Antler Extracts

  • Deer velvet antler was subjected to the extraction process using boiling water at three different temperatures (100, 110 and $120^{\circ}C$) and 70% ethanol solution. Functional components such as uronic acid, sulfated-glycosaminoglycans (sulfated-GAGs) and sialic acid in the extracts were analyzed, and their antioxidant activities were investigated using several in vitro models. Uronic acid and sulfated-GAGs content of each extract significantly decreased with increasing extraction temperature (p<0.05), while the residues obtained from the upper and middle part of the antler had a higher uronic acid content than the residues obtained from the base section. Sialic acid contents were highest in compounds extracted at $110^{\circ}C$, followed by 120 and $100^{\circ}C$. The 70% ethanol extracts also had a high levels of uronic acid content, but not for sulfated-GAGs and sialic acid. All extracts showed good antioxidant ability in a dose-dependant manner, with the $100^{\circ}C$ residue exhibiting the strongest activity compared to the 110 and $120^{\circ}C$ extracts. In relation to the hydroxyl radical scavenging activity and reduction power, the 70% ethanol extract exhibited the strongest activity. Furthermore, the velvet antler extracts inhibited apoptosis in hydrogen peroxide-induced PC-12 cells.
