Evaluation of the body weight and laying performance of diallel crossed Korean native chicken layers from hatch to 40 weeks of age

  • The current experiment was conducted to evaluate the effect of diallel crossbreeding on the body weight and laying performance of Korean native chicken from hatch to week 40. A total of 1,000 one-day-old chicks were allotted to 10 cages per crossbreeding treatment with 10 birds per cage on a random basis and then raised until 16 weeks and subsequently moved to layer battery cages and raised until 40 weeks. Ten crossbred treatments (YC, YD, YF, YK, CD, CF, CK, DF, DK, FK) that were obtained from the diallel crossbred of five pure lines were used in the current experiment. The body weight and mortality were measured biweekly from hatching to week 20 and every four weeks from week 20 to week 40. The number of eggs was measured daily. YC and YD crossbreeds showed a higher body weight (p < 0.05) and FK crossbreed showed a lower body weight (p < 0.05) during the whole xperimental period. The week 20 weight range was 1,501 to 1,729 g and the week 40 weight range was from 1,829 to 2,179 g. Earlier onset of egg-laying was noted in the YC and YD groups whereas late onset was observed for the DF and DK groups. YK reached its peak earliest at week 25 with 89.15% while the DK crossbreed attained its peak at week 36 with 89.69%. The YC and YD crossbreeds showed the improved body and egg-laying performance. Conclusively, there are variations in the body weight and laying performance of Korean native chickens with diallel crossing.

Effects of Iron Supplementation on Iron Status and Immunity Status of Elite Female Soccer Players (엘리트 여자 축구선수의 철분보충이 체내 철분상태와 면역 및 항산화에 미치는 영향)

  • This study was performed to evaluate the effect of iron supplement for 4 weeks on iron status, immunity, and antioxidant status of national female soccer players (n = 25). This study was performed at summer hard training period right before competition. A single blind design was used to divide the subjects into iron-supplement (IS) or placebo group (P). Iron-supplement group was supplemented with iron (40 mg/d) for 4 weeks. The mean age of the subjects was 23.3 $\pm$ 2.5 years old. Mean height and body weight of the subjects were 164.4 $\pm$ 5.7 em and 57.4 $\pm$ 4.6 kg, respectively. The mean carrier as soccer player was 11.0 $\pm$ 2.6 years and mean training time was 7.0 $\pm$ 1.3 hr/day. The mean hemoglobin, hematocrit, total iron binding capacity and ferritin concentrations before iron supplementation were not different between two groups. After 4 weeks of summer training and iron supplementation, serum ferritin level was significantly increased only in IS group after supplementation. Mean corpuscular volume and total iron binding capacity were significantly decreased in both groups. Meanwhile, hemoglobin and red blood cell count were significantly lowered only in placebo group. The IgM concentration increased significantly in both groups, but IgG concentration had increasing tendency only in IS group (p < 0.064). Therefore, iron supplementation during hard training period may be helpful to improve work capacity of the athletes by improving ferritin status and humoral immune responses.

Growth of Larval Sweetfish Plecoglossus altivelis Fed Moina macrocopa (물벼룩 Moina macrocopa 급이가 은어 Plecoglossus altivelis 자어의 성장에 미치는 영향)

  • The effect of live food and commercial diet on sweetfish Plecoglossus altivelis culture was assessed by determining the daily growth rate and feed conversion ratio. After feeding for 8 weeks, the Moina macrocopa diet resulted in a higher growth rate of 5.101 compared to commercial (3.441) and Artemia (2.729) diets. In addition fish fed the M. macrocopa diet had the weight gain, averaging 41.29 g at the end of the experiment. The content of n-3 highly unsaturated fatty acids (HUFA) was highest in the commercial diet, while free fatty acids were not detected in the M. macrocopa diet. Triglycerides, the energy source in fishes, constituted 40.0% of the M. macrocopa diet, which was significantly higher than in the commercial and Artemia diets at 20.0 and 12.0%, respectively These results indicate that freshwater M. macrocopa can be used as feed for sweetfish larvae, even without docosahexaenoic acid (DHA) enrichment from Artemia.

  • Information seeking behaviour is an activity of an individual in the process of identifying information that suits his/her knowledge pursuit. It is observed from the review of literature that there is no single study on the information seeking behaviour of faculties of pharmacy either at state level or national levels in India. Therefore this research has been conducted to bridge the gap on information seeking behaviour of pharmacy faculty in Tamil Nadu in view of the recent developments in information seeking behaviour, with objectives such as: to identify the information needs and seeking behaviour of faculty of the pharmacy educational institutions in Tamil Nadu (India); to examine the motivating factors for information seeking behaviour of the pharmacy faculty; to examine faculty opinions about the comprehensiveness or otherwise of respective institutions' library collections; and to analyse the extent of use and dependence on various sources of information for teaching and research. A total of 729 questionnaires have been distributed among 41 pharmacy educational institutions in Tamil Nadu, out of which 601 have responded, and the response rate is 82.44%. Based on the findings of the study certain implications have been derived as measures to enhance the quality of the pharmacy libraries in Tamil Nadu.

Genetic Diversity and Spatial Structure in Populations of Abelia tyaihyoni (줄댕강나무 (Abelia tyaihyoni) 집단의 유전다양성 및 공간구조)

  • The genetic diversity and the spatial structure in two populations of Abelia tyaihyoni in Yeongwol region were studied by employing I-SSR markers. In spite of the limited distribution and small population sizes of Abelia tyaihyoni, the amount of genetic diversity estimated at the individual level was comparable to other shrub species (S.I.=0.336, h=0.217). Genetic diversity at the genet level was very similar to that at individual level. (S.l.=0.339, h=0.219). About 18.7 percent of total variation was allocated between two populations, which was slightly higher or similar level as compared with other shrub species. Genotypic diversity estimated by the ratio of the number of genets ($N_G$) over the total number of individuals (N) and a modified Simpson's index ($D_G$) were also higher than those of other shrubs. The maximum diameter of a genet did not exceed 5.5 m. The high level of gene and genotypic diversity, and the relatively limited maximum diameter of a genet suggested that the clonal propagation is not the most dominant factor in determining the population structure of Abelia tyaihyoni. Spatial autocorrelation analysis revealed significant spatial genetic structure within 12 m and 18 m distances in two populations A and B, respectively. Autocorrelations among individuals at the both individual and genet levels in each population didn't show any considerable differences. As a sampling strategy for ex-situ conservation of populations showing continuous distribution, a minimum distance of 18 m between individuals was recommended. For the populations with many segments, it was considered very crucial to sample materials from as many segments as possible.

ⳑ-Methionine inhibits 4-hydroxy-2-nonenal accumulation and suppresses inflammation in growing rats

  • BACKGROUND/OBJECTIVES: 4-Hydroxy-2-nonenal (HNE) is a biomarker for oxidative stress to induce inflammation. Methionine is an essential sulfur-containing amino acid with antioxidative activity. On the other hand, the evidence on whether and how methionine can depress HNE-derived inflammation is lacking. In particular, the link between the regulation of the nuclear factor-κB (NF-κB) signaling pathway and methionine intake is unclear. This study examined the link between depression from HNE accumulation and the anti-inflammatory function of ⳑ-methionine in rats. MATERIALS/METHODS: Male Wistar rats (3-week-old, weighing 70-80 g) were administered different levels of ⳑ-methionine orally at 215.0, 268.8, 322.5, and 430.0 mg/kg body weight for two weeks. The control group was fed commercial pellets. The hepatic HNE contents and the protein expression and mRNA levels of the inflammatory mediators were measured. The interleukin-10 (IL-10) and glutathione S-transferase (GST) levels were also estimated. RESULTS: Compared to the control group, hepatic HNE levels were reduced significantly in all groups fed ⳑ-methionine, which were attributed to the stimulation of GST by ⳑ-methionine. With decreasing HNE levels, ⳑ-methionine inhibited the activation of NF-κB by up-regulating inhibitory κBα and depressing phosphoinositide 3 kinase/protein kinase B. The mRNA levels of the inflammatory mediators (cyclooxygenase-2, interleukin-1β, interleukin-6, inducible nitric oxide synthase, tumor necrotic factor alpha) were decreased significantly by ⳑ-methionine. In contrast, the protein expression of these inflammatory mediators was effectively down regulated by ⳑ-methionine. The anti-inflammatory action of ⳑ-methionine was also reflected by the up-regulation of IL-10. CONCLUSIONS: This study revealed a link between the inhibition of HNE accumulation and the depression of inflammation in growing rats, which was attributed to ⳑ-methionine availability. The anti-inflammatory mechanism exerted by ⳑ-methionine was to inhibit NF-κB activation and to up-regulate GST.

  • Laminaria japonica roots have not been used practically in Korea. In this study, in order to promote the use of these by-products, the anti-inflammatory activity of an ethanol extract of Laminaria japonica root (LJREE) was investigated using a lipopolysaccharide (LPS) to induce an inflammatory response in RAW 264.7 cells. To examine the potential anti-inflammatory effects of LJREE, levels of nitric oxide (NO) and pro-inflammatory cytokines, such as interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interleukin-$1{\beta}$ (IL-$1{\beta}$), and cell proliferation were measured. We found that NO levels decreased in a dose-dependent manner, the production of IL-6, TNF-${\alpha}$, and IL-$1{\beta}$ was suppressed, and the production of IL-$1{\beta}$ was inhibited over 30% after treatment with $100{\mu}g/mL$ LJREE. In conclusion, the proliferation of RAW 264.7 cells, measured by MTT assay, confirmed that LJREE may have significant effects on inflammatory factors without any cytotoxicity, making it a potential anti-inflammatory agent.

Establishment and Characterization of Carboplatin-Resistant Retinoblastoma Cell Line

  • Carboplatin-based chemotherapy is the primary treatment option for the management of retinoblastoma, an intraocular malignant tumor observed in children. The aim of the present study was to establish carboplatin-resistant retinoblastoma cell lines to facilitate future research into the treatment of chemoresistant retinoblastoma. In total, two retinoblastoma cell lines, Y79 and SNUOT-Rb1, were treated with increasing concentrations of carboplatin to develop the carboplatin-resistant retinoblastoma cell lines (termed Y79/CBP and SNUOT-Rb1/CBP, respectively). To verify resistance to carboplatin, the degree of DNA fragmentation and the expression level of cleaved caspase-3 were evaluated in the cells, following carboplatin treatment. In addition, the newly developed carboplatin-resistant retinoblastoma cells formed in vivo intraocular tumors more effectively than their parental cells, even after the intravitreal injection of carboplatin. Interestingly, the proportion of cells in the G0/G1 phase was higher in Y79/CBP and SNUOT-Rb1/CBP cells than in their respective parental cells. In line with these data, the expression levels of cyclin D1 and cyclin D3 were decreased, whereas p18 and p27 expression was increased in the carboplatin-resistant cells. In addition, the expression levels of genes associated with multidrug resistance were increased. Thus, these carboplatin-resistant cell lines may serve as a useful tool in the study of chemoresistance in retinoblastoma and for the development potential therapeutics.

Prospective Study of the Immunologic Factors Affecting the Prognosis of Severe Community-Acquired Pneumonia (중증 지역사회획득 폐렴환자의 예후에 영향을 미치는 면역지표에 대한 연구)

  • Background : In the severe community-acquired pneumonia, it has been known that the immune status is occasionally suppressed. This study was performed to identify the immunologic markers related with the prognostic factors in severe community-acquired pneumonia. Methods : 23 patients with severe community-acquired pneumonia were involved in this study, and divided into survivor (16) and nonsurvivor (7) groups. In this study, the medical history, laboratory tests(complete blood counts, routine chemistry profile, immunoglobulins, complements, lymphocyte subsets, cytokines, sputum and blood culture, urine analysis), and chest radiographs were scrutinized. Results : 1) Both groups had lymphopenia(total lymphocyte count $995.6{\pm}505.7/mm^3$ in the survivor and $624.0{\pm}287.6/mm^3$ in the nonsurvivor group). 2) The T-lymphocyte count of the nonsurvivor group($295.9{\pm}203.0/mm^3$) was lower than the survivor group($723.6{\pm}406.5/mm^3$) (p<0.05). 3) The total serum protein(albumin) was $6.0{\pm}1.0(2.7{\pm}0.7)\;g/d{\ell}$ in the survivor and $5.2{\pm}1.5(2.3{\pm}0.8)g/d{\ell}$ in the nonsurvivor group. The BUN of the nonsurvivor group($41.7{\pm}30.0mg/d{\ell}$) was higher than that of the survivor group($18.9{\pm}9.8mg/d{\ell}$)(p<0.05). The creatinine concentration was higher in the nonsurvivor group($1.8{\pm}1.0mg/d{\ell}$) than that in the survivor group($1.0{\pm}0.3mg/d{\ell}$)(p<0.05). 4) The immunoglobulin G level was higher in the survivor group($1433.0{\pm}729.5mg/d{\ell}$) than in the nonsurvivor group($849.1{\pm}373.1mg/d{\ell}$) (p<0.05). 5) The complement $C_3$ level was $108.0{\pm}37.9mg/d{\ell}$ in the survivor group and $88.0{\pm}32.1mg/d{\ell}$ in the nonsurvivor group. 6) A cytokine study showed an insignificant difference in both groups. 7) Chronic liver disease, DM, and COPD were major underlying diseases in both groups. Conclusion : These results suggest that decreased a T-lymphocyte count and immunoglobulin G level, and an increased BUN and creatinine level may be associated with the poor prognosis of severe community-acquired pneumonia.

Optimization of Preparation Conditions and Quality Characteristics of Sweet Pumpkin Stock (단호박 스톡 제조조건의 최적화 및 품질 특성)

  • The stock that is the first step for preparation of soups and purees links to the taste of food. Many types of vegetable have been used in stocks, but this study focused on stocks prepared with sweet pumpkin. The stock preparation conditions including the weight of sweet pumpkin, the water volume, and the boiling time at $97^{\circ}C$ were optimized by response surface methodology. The quality characteristics of the resulting stock were investigated. The color, flavor, taste and overall acceptability were dependent parameters. A model equation was proposed with regard to the sweet pumpkin weight, water volume, and boiling time at $97^{\circ}C$. A sweet pumpkin weight of 357.9 to 403.0 g, a water volume of 689.8 to 768.5 mL, and a boiling time of 9.9 to 10.3 min at $97^{\circ}C$ were found to be the optimal stock preparation conditions. The quality characteristics of the sweet pumpkin stock prepared under the optimized conditions were pH 6.64, total acidity 0.18%, soluble solids $2.39\;^{\circ}Brix$, color value (L, 99.07 a, -2.43 b, 11.82), total polyphenol 280.75 mg/L, and electron donating ability 21.32%.
