대두 단백 효소 가수분해물의 항균활성

  • 대두 단백질을 효소로 가수분해 하였을 때 생성되는 peptide의 항균활성을 조사하고 천연항균제로서 이용 가능성을 검토하기 위하여 분리 대두 단백질에 5종의 단백질 가수분해 효소를 작용시켜 생성된 가수분해물의 항균력을 측정하고 한외여과하여 분자량별로 분리된 각 fraction의 항균활성과 HPLC로 정제하여 항균성 peptide 의 아미노산 결합순서를 분석하여 다음과 같은 결론을 얻었다. 분리대두 단백질에 5종의 단백질 분해효소를 작용시켜 제조한 가수분해물 중 Asp.saitoi protease로 작용시킨 것이 항균활성이 높았다. Asp. saitoi protease로 작용시킨 대두 단백질의 가수분해물을 membrane filter로 여과한 결과 분자량 1000-3000 fraction에서 항균활성이 가장 높았다. 분자량 1000-3000 범위을 가진 가수분해물의 MIC는 0.5-0.8mg/$m\ell$ 이었으며 그람 양성균과 음성균 모두의 증식을 억제하는 경향을 보였다. 분리 대두 단백으로부터 얻어진 항균성 peptide는 121$^{\circ}C$, 10분간 열처리하여도 안정하였으며 한외여과에 의하여 분자량 1000-3000범위의 가수분해물을 동결건조하여 gel filteration하였을 때 2개의 fraction에서 항균 활성을 나타내었다. HPLC결과 RT 16.02 의 peak에서 항균활성이 확인되었고 질량은 1,633이었으며 아미노산 결합순서는 H$_2$N-G-P-P-G-V-V-A-T-V-V-A-A-R-COOH 이었다.

벼의 공기 밀폐저장 특성 (Airtight Storage Characteristics of Rough Rice)

  • This study was performed to investigate airtight storage characteristics of rough rice using airtight flexible pve container. A storage test of rough rice of 4 tonnes was carried out to determine the changes of gas composition of air in the container, grain moisture content, air temperature and relative humidity , the presence of insects ,germination rate, crack ratio , fat acidity , 1000-kernel weight, and brown rice recovery over storage period of 5 months in Suwon. Concentration of oxygen was decreased from 20% to 16% and carbon dioxide was increased of from 0.03% to 1.6%. The grain moisture content was decreased from 14.4%(w.b) to 14.1%(w.b) for 5 months storage period . Insect population levels were low but these increased after 5 months storage. Most of insects were dead, Fat acidity increased from 7.5(mg KOH/100g) to 10.2(mg KOH/100g). Other storage factors such as germination rate, brown rice recovery and 1000-kernel , and 1000-kernel weight slightly decreased and crack ratio was slightly increased. Qualities of rough rice during 5 months storage period under hemetic air conditions were maintained fairly good considering the above changes of quality factors during storage.

Effect of Activation Temperature on CO2 Capture Behaviors of Resorcinol-based Carbon Aerogels

  • In this study, carbon aerogel (CA) was synthesized using a soft-template method, and the optimum conditions for the adsorption of carbon dioxide ($CO_2$) by the carbon aerogel were evaluated by controlling the activation temperature. KOH was used as the activation agent at a KOH/CA activation ratio of 4:1. Three types of activated CAs were synthesized at activation temperatures of $800^{\circ}C$(CA-K-800), $900^{\circ}C$(CA-K-900), and $1000^{\circ}C$(CA-K-1000), and their surface and pore characteristics along with the $CO_2$ adsorption characteristics were examined. The results showed that with the increase in activation temperature from 800 to $900^{\circ}C$, the total pore volume and specific surface area sharply increased from 1.2165 to $1.2500cm^3/g$ and 1281 to $1526m^2/g$, respectively. However, the values for both these parameters decreased at temperatures above $1000^{\circ}C$. The best $CO_2$ adsorption capacity of 10.9 wt % was obtained for the CA-K-900 sample at 298 K and 1 bar. This result highlights the importance of the structural and textural characteristics of the carbon aerogel, prepared at different activation temperatures on $CO_2$ adsorption behaviors.

서울 및 근교에 거주하는 한국인의 연령별 식생활 비교 및 평가 : (3) 만성퇴행성질환의 위험요인 (Comparative Analysis and Evaluation of Dietary Intakes of Koreans by Age Groups : (3) Risk Factors for Chromic Degenerative Diseases)

  • This study was performed to analyse some rick ffactors for chronic degenerative diseases of the Korean dict. The study subjects consisted of preschool children(PC), elementary school children(EC), high school students(HS), college students(CS), and adults over 30 years old living in Seoul and the surrounding areas. The subjects over 30 years old were classfied to 30-49 years, 50-69 years, and over 70 years. A dietary survey was conducted using 24-hour recall method and data were collected from 2392 subjects. The dietary risks for chronic degenerative disease such as food sources of fat consumption, high cholesterol intake, low dietary fiber intake, and low calcium intake and high protein intake were analyzed. Younger subjects had more animal fat proportion than older subjects did. HS and CS had the least fat proportion from fishes and shellfishes. Each mean cholesterol intake of PC, EC, HS, and CS was over 300mg/day, and HS load the highest level of 415mg/day. Cholesterol density of the diet of PC was the highest of 175mg/1000kcal and the level decreased when the age increased. The subjects consuming cholesterol over 100 mg/1000kca1 were about 70% of PC and HS. More than 20% of PC, EC, and HS consumed calcium under 75% of RDA with protein consumption over 125% of RDA. Dietary fiber intakes were 18g for HS, CS, 30-49 years, and 50 ∼ 69 years, 14g for EC and over 70 years, and 12g for PC. Ninety percent of PC and EC, 80% of HS and CS, and 70% of tole subjccts over 30years consumed dietary fiber under the level of 10g/1000kca1. The major food sources of dietary fiber were vegetables and grains. From these results, children and adolescents had ricks for chronic degenerative diseases on all the analyzed items. The results of this study would provide the basic information for development of dietary interventions to improve diet find health.

Immunogenicity Study of Recombinant Human Basic Fibroblast Growth Factor

  • The immunogenicity of the recombinant human basic fibroblast growth factor (rh-bFGF) was investigated by tests for active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA), passive hemagglutination (PHA) and guinea pig maximization test (GPMT) in mice or guinea pigs. Guinea pigs were sensitized with rh-bFGF ($100-1000\;\mu\textrm{g}/kg$) or rh-bFGF-CFA mixture ($1000\;\mu\textrm{g}/kg$). All animals sensitized with rh-bFGF alone or mixture with CFA showed symptoms of anaphylactic shock. IgE antibodies to rh-bFGF were detected in sera obtained from rh-bFGF and rh-bFGF-Alum ($1000\;\mu\textrm{g}/kg$) sensitized mice, indicating that rh-bFGF has immunogenicity eliciting potential. IgG and/or IgM antibodies to rh-bFGF were also detected in all the sera obtained from sensitized mice by PHA. In the GPMT for delayed type skin reaction, no skin reaction was observed in sensitized guinea pigs after intradermal injection and dermal application of 0.01% rh- bFGF. However, these positive reactions were consistent with the results of another rh-bFGF, showing that rh- bFGF is a heterogenous protein to rodents. Considering the fact that rh-bFGF is a genuine human protein of which structure is identical to the endogenous human bFGF, it is thought that rh-bFGF is rarely associated with immunological problems in clinical use.

  • This study was undertaken to assess overall effects of bisphenol A, a monomer widely used in manufacturing polycarbonate plastics or epoxy resin, exposure on immune system of mice. For in vitro evaluation, serial concentration of SPA was added into culture of various immune cells from normal female ICR mice, and for in vivo or ex vivo assessment, mice were orally exposed to BPA dissolved in olive oil as doses of 500, 1000, 2000 mg/g b.w. for acute expose or 100, 500, 1000 mg/kg/day b.w. 5days a week for subacute exposure.   • 본 연구에서는 팔의 굽힘(flexion)과 폄(extension)운동 시 물체의 무게에 따른 저크의 변화를 통해 부드러움을 정량화하고, 물체무게와 움직임의 형태(type)에 따른 동작의 부드러움을 비교하고자 한다. 10 명의 오른손잡이 남자 대학생(평균 $24.8{\pm}1.1$ 세)을 실험 참여자로 선정하였다. 물체무게(0g, 1000g)와 움직임의 형태(손등 위/아래)를 달리하여 팔의 굽힘과 폄 운동을 실시하였다. 또한 3 축 가속도 센서를 이용하여 x 와 z 축의 가속도를 측정하였고, 측정된 신호로 부터 저크값을 추출 하였다. 실험은 총 8 분으로 4 개의 block 으로 구성되며 각 block 은 resting 구간 1 분과 lifting 구간 1 분으로 구성된다. 실험 결과 물체무게 0g 에 비해 1000g 에서 가속도와 정규저크 값이 유의미하게 감소하였다.

Anti-Oxidant Activities of Paeoniflorin Derivatives

  • We previously showed that the root extract of Paeonia lactiflora Red Charm might have anti-oxidant activities, however it was not clear which components might be involved in this activity. Our boinformatics analysis indicated the root extracts of Paeonia lactiflora Red Charm has a couple of potential anti-oxidant materials. One of them is paeoniflorin. We hypothesized that one of components present in Paeonia lactiflora Red Charm, paeoniflorin and its derivatives might be related to anti-oxidant activity. In this study, we compared paeoniflorin and its derivatives with the root extract of Paeonia lactiflora Red Charm using DPPH assays to measure its antioxidant activities. Paeoniflorin showed the highest radical scavenging activity(%) in $1000{\mu}g/m{\ell}$. its derivative showed the high levels radical scavenging activity(%) in $600{\mu}g/m{\ell}$, $800{\mu}g/m{\ell}$, $1000{\mu}g/m{\ell}$ similar to ascorbic acid. Taken together, these results suggest that Paeoniflorin and its derivatives may play a role in anti-oxidant acitivity in the root extracts of Paeonia lactiflora. Much of future studies may be needed to develop a potential new anti-oxidant candidates with anti-aging and anti-cancer effects.

  • IPDI, PTAd 및 DMPA(음이온 중심)을 사용하여 폴리우레탄 아이오노머를 합성하고 여기에 물을 첨가하여 수분산 폴리우레탄을 제조하였다. 폴리올 분자량(Mn)이 분산상태 및 에멀젼 주사 필름의 열적, 기계적, 점탄성적 성질 및 팽윤에 미치는 영향을 측정하였다. 폴리올 분자량이 증가할수록 에멀젼 입경과 soft segment $T_{g}$는 감소하였으며, 용매팽윤, 에멀젼 점도, hard segment $T_{g}$는 증가하였다. 필름의 인장강도는 Mn = 1000에서 최소치를 나타냈으며, 파단신율은 폴리올 분자량 증가와 더불어 증가하였는데 이러한 결과는 soft-hard segment 상분리와 고분자량 PTAd의 결정화로 적절히 설명할 수 있었다.

식품유해균에 대한 차류 추출물의 항균효과 (Antimicrobial Activities of Commercially Available Tea on the Harmful Foodborne Organisms)

  • Use of chemical preservative for controlling harmful microorganisms in food products has been debated due to public concerns about food quality because of perceived toxic and carcinogenic potential. Thus, use of non toxic natural antimicrobial agents has become essential. This study was investigated to determine the antimicrobial activity of water or ethanol extract of commercially available tea, and of solvent fractionated ethanol extracts obtained from steamed green tea. Both of water and ethanol extracts of green tea(steamed or roasted), oolong tea and black tea exhibited strong antimicrobial activity against gram positive and negative bacteria, but not effective against yeast and mold. Also, antimicrobial activity of ethanol extract of 4 different kinds of tea was stronger than that of water extract. Among 4 different tea, ethanol extract of steamed green tea was further fractionated. One thousand g/disk buthanol extract had the strongest antimicrobial activity against bacteria and mold. The concentration of the antimicrobial activity of buthanol extract in tested microorganisms ranged from 125~1000 g/disk except for Rhizopus javanicus. Antimicrobial activity of buthanol extract of steamed green tea was not destroyed by heating at 100oC for 60 min and at 121oC for 15 min, which is very stable over heat treatment. The inhibitory effect of the buthanol extract on the growth of Listeria monocytogenes and Staphylococcus aureus was investigated. Growth of both strains was started in the presence of 250 and 500 g/ml after 12 and 24 hour respectively, whereas complete inactivation of both strains was occurred in the presence of 1000 g/ml.
