본 연구에서는 식이단백질 수준이 성장기 흰쥐의 신장 발달과 기능에 미치는 영향을 조사하기 위해 평균체중이 97.5$\pm$1.9g인 숫컷 흰쥐를 5%, 15% 그리고 50% casein 식이로 6주간 사육하였다. 체중증가량은 단백질 수준이 높을수록 높았으며 특히 단백질수준이 신장무게에 미친영향이 간, 비장, 흉선 등이 다른 장기에 비해 컸다. 신장내 총 DNA와 RNA 함량에서는 유의적인 차이가 없었다. 총 protein과 단위 g당 protein 함량은 식이내 단백질수준이 높을 수록 증가했다. 사구체여과율(GFR)은 식이단백질 수준이 높을수록 높았으나 신혈류(RBF)는 각 군 사이에 유의적인 차이를 보이지 않았다. 뇨질소배설량은 단백질 수준에 비례하여 유의적인 차이를 보였고 노칼슘배설량은 5%군이 낮은 경향을 보였다. 본 연구 결과로 성장기의 흰쥐에서 고단백식이는 신장의 무게증가를 촉진하며 사구체여과율을 증가시킴을 알 수 있다.
To investigate the fidelity of transgene transmission and expression, we produced transgenic mice carrying bovine $\beta$-casein/bovine grwoth hormone(bGH) fusion gene and examined transmission efficiency and expression level of the transgene in the founders and their progeny. The transgene was composed of 1.8 kb bovine $\beta$-casein promoter and 2.1 kb bGH gene. Ten transgenic mice were produced. Milk and mammary gland were collected from eight transgenic lines at 10-day lactation and a, pp.ied to Western and Northern blot analyses. The bGH expression was detected in four of them. The concentrations of bGH in milk were highly variable from 4$\mu\textrm{g}$/ml to 600$\mu\textrm{g}$/ml depending on the lines. The bGH mRNA level in mammary gland was closely correlated with the bGH concentration in milk in each transgenic line. These results indicated that bGH transgene expression was a, pp.opriately regulated in the mammary gland and secreted into milk in transgenic mice. By using two transgenic lines(#2, #7) secreting a considerable amoung of bGH into their milk, the inferitance and maintenance of transgenic phenotype were assessed in successive four generations. The mean transmission frequencies of transgene in lines #2 and #7 were 34% and 40%, respectively. The bGH concentration in milk were 80, 240, 120, 60$\mu\textrm{g}$/ml in each G0(generation 0), G2, G3, G4 generation of line #2 and 600, 1600, 860, 900$\mu\textrm{g}$/ml in each G1. G2, G3, G4 generation of line #7. These results demonstrated that bovine $\beta$-casein/bGH gene was stably transmitted from generation to generation in a Menelian fashion in trasgenic mice and consistenly expressed in their milk throughout the generations, although there was a little variation in the transmission frequency and expression level of the transgene between generations.
우리나라에서 유통되고 있는 자주복의 독성을 조사하기 위하여 1995년 1월, 4월 및 9월에 부산시내 자갈치 어시장에서 총 27개체를 구입하여 부위별 독성을 조사하였다. 간장의 경우 독성이 10MU/g 이상인 유독개체 출현율은 14.8%, 최고 독성치는 160 MU/g, 평균 독성치는 7$\pm$6 MU/g(평균치$\pm$표준오차)이었다. 생식소의 경우는 각각 16.7%, 600 MU/g 및 50$\pm$35 MU/g 이었고, 껍질에서는 각각 14.8%, 26 MU/g 및 5$\pm$1MU/g이었다. 그러나 근육에서는 독이 검출되지 않았다. 계절에 따른 독성은 1월 및 4월의 일부시료에서는 유독부위가 있었으나 9월에는 시료어의 전부위에서 독성이 검출되지 않았다. 자주복의 독성은 부위나 계절에 따른 차이가 있지만 주 가식부인 근육과 껍질에서는 거의 독이 검출되지 않거나 약한 독성이 검출되어 식품 위생상 비교적 안전한 종으로 판단되었다.
In order to observe the effect of tannin on plasma lipid status in human, tannin was supplemented with increasing amounts from 0 to 0.5, 1.0, 2.5g/d early week until 4th week. The results obtained from this research are a follows. Concentration of total lipid in plasma was significantly lower compared to the control group when the level of tannin supplementation was 0.5g/day while the level of tannin 1.0g/day and 2.5g/day did not affect to change the total lipid level. There seems to be no obvious improvement of the plasm lipid status respect to different tannin levels. Despite the result, the excretion of total bile acids in feces was increased and total lipid in plasma was reduced significantly during first time supplementation compared the control.
Thirty-day-old rice seedlings, IR-20 variety were transplanted into the experimental plots and were sprayed with endosulfan (35% EC) after 52 days at the rate of 0.64 kg a.i/ha. Residues of endosulfan in the plant, soil, and water were found to decrease steadily upto 15 days. A second application of the pesticide was made on the 31st day and the plant was harvested on 56th day after the first application. The residue level on hay and grains was 0.7 ${\mu}g/g$. This level is seven times higher than FAO/WHO-prescribed tolerance level of 0.1 ${\mu}g/g$. Metabolites of endosulfan were traced out in plants, soil, and water during the pre- and post-harvest period.
Objectives: This survey was designed to conduct the first nationwide dietary exposure assessment on hazardous substances including the intakes of functional food and herbal medicine. In this paper, we introduced the survey design and the results of the dietary exposure status and internal exposure levels of lead (Pb), cadmium (Cd), and mercury (Hg). Methods: We selected 4867 subjects of all ages throughout Korea. We conducted a food survey, dietary survey, biomonitoring, and health survey. Results: Pb and Cd were the highest (median value) in the seaweed ($94.2{\mu}g/kg$ for Pb; $594{\mu}g/kg$ for Cd), and Hg was the highest in the fish ($46.4{\mu}g/kg$). The dietary exposure level (median value) of Pb was $0.14{\mu}g/kg$ body weight (bw)/d, $0.18{\mu}g/kg$ bw/d for Cd, and $0.07{\mu}g/kg$ bw/d for Hg. Those with a blood Pb level of less than $5.00{\mu}g/dL$ (US Centers for Disease Control and Prevention, reference value for those 1 to 5 years of age) were 99.0% of all the subjects. Those with a blood Cd level with less than $0.30{\mu}g/L$ (German Federal Environmental Agency, reference value for non-smoking children) were 24.5%. For those with a blood Hg level with less than $5.00{\mu}g/L$ (human biomonitoring I, references value for children and adults, German Federal Environmental Agency) was 81.0 % of all the subjects. Conclusions: The main dietary exposure of heavy metals occurs through food consumed in a large quantity and high frequency. The blood Hg level and dietary exposure level of Hg were both higher than those in the European Union.
This study was investigated to prove the relation between the ingredient of Samul Chol-Pyon and its microorganism pollution level during the storage. As a result, the pollution degree in total aerobic bacteria, yeast, mold, and colitis germs of rice power turned out to have a lower one than the oriental medicine materials do. In case of preserved write Chol-Pyon, the total aerobic bacteria pollution level was 8.8 $\times$ 10$^3$CFU/g the highest degree among other ones in their among other ones in their early pollution levels and in the oriental medicine materials, the pollution level was degreased as its annex increased. Moreover, yeast propagated fast in its first day of storage, but mold grew somewhat slowly than yeast and total aerobic bacteria did. The present study has evaluated the $CO_2$ adsorption amount of activated carbon pellets (AC). Coconut shell based test AC were modified with surface impregnation of glycine, glycine metal salts and monoethanolamine for low level $CO_2$ (3000 ppm) adsorption. Physical and chemical properties of prepared adsorbents were analyzed and the adsorbed amount of $CO_2$ was investigated by using pure and 3,000 ppm $CO_2$ levels. The impregnation of nitrogen functionalities was verified by XPS analysis. The adsorption capacity for pure $CO_2$ gas was found to reach upto 3.08 mmol/g by AC-LiG (Activated carbon-Lithium glycinate), which has the largest specific surface area ($1026.9m^2/g$). As for low level $CO_2$ flow the primary amine impregnated adsorbent showed 0.26 mmol/g of adsorption amount, indicating the highest selectivity. An adsorbent with potassium-glycine salts (AC-KG, Activated carbon-Potassium glycinate) instead of amine presented with 0.12 mmol/g of adsorption capacity, which was higher than that of raw activated carbon granules (0.016 mmol/g).
This study intended to obtain an useful information on the prevalence of subjective symptoms, and to clarify the interrelationships between blood lead and lead related symptoms in low level lead exposure. The 93 male workers exposed to lead and 56 male nonexposed workers were examined for their blood lead(PBB), Zinc-protoporphy(ZPP), hemoglobin(HB) and personnal history, and completed 15 questionnaires related to symptoms of lead absorption : also measured lead concentration in air (PBA) in the workplace. The results obtained were as follows ; 1. The means of blood lead (PBB), blood ZPP and hemoglobin (HB) among workers exposed to lead were $26.1{\pm}8.8{\mu}g/dl,\;28.3{\pm}26.0{\mu}g/dl$ and $16.2{\pm}1.2g/dl$ : whereas those of nonexposed workers were $18.7{\pm}5.1{\mu}g/dl,\;20.6{\pm}8.7{\mu}g/dl$ and $17.3{\pm}1.1g/dl$. The means of above three indicies between two groups showed significant difference statistically (p<0.05). 2. The means of blood lead (PBB), blood ZPP and hemoglobin of workers exposed .to different lead concentration in air were as follows : When it was below $25{\mu}g/m^3$, the indices were $24.7{\pm}79,\;26.1{\pm}26.8{\mu}g/dl\;and\;16.4{\pm}1.1g/dl$ respectively : These indices were $27.1{\pm}8.5,\;23.9{\pm}10.92{\mu}g/dl\;and\;16.2{\pm}1.3g/dl$ when the lead concentration in air was $25{\sim}50{\mu}g/m^3$ : and they were $3.4{\pm}9.3,\;42.3{\pm}31.3{\mu}g/dl\;and\;15.5{\pm}1.2g/dl$ when the concentration of lead was above $50{\mu}g/m^3$. Although there were statistical difference in blood lead and hemoglobin among three different lead concentration in air, there was no statistical difference of blood ZPP among the three groups with different exposure levels (p>0.05). 3. The most frequent by complained symptom was 'Generalized weakness and fatigue', and fewest symptom was 'Intermittent pains in abdomen' 4. Only two symptoms out of fifteen symptoms checked by themselves revealed significant difference between exposed and nonexposed groups. These were 'Intermittent pains of abdomen' and 'Joint pain or arthralgia' (p<0.05), No positive correlation was found between the levels of blood lead and symptom groups categorized as gastrointestinal, neuromuscular and constitutional symptoms, 5. Blood lead (r=0.3995) and ZPP (r=0.2837) showed statistically significant correlation with mean lead concentration in air, whereas correlations were not demonstrated between blood lead and lead related symptoms or blood ZPP and lead related symptoms. 6. Blood lead (PBB) and ZPP showed association (r=0.2466) and the equation PBB=23.75+0.0842 ZPP was derived. 7. On stepwise multiple regression, using blood lead level as a dependent variable and ZPP, hemoglobin (HB), age, work duration (WD) and symptom prevalence as a independent variables, only ZPP significantly contributed a lot to blood lead level. 8. While the ZPP measurement was found to be a good indicator in evaluating health effect of lead absorption in low level lead exposure, lead related symptoms were not sensitive enough to evaluate of lead absorption in low level exposure.
Objectives: Blood mercury levels among adults living in certain areas of the Gyeongsang Provinces have been shown to be very high (Kunwee County $29.6{\mu}g/L$, Yeongcheon-city $26.7{\mu}g/L$). The purpose of this project was to determine mercury exposure levels in schoolchildren and factors related with their mercury levels in high mercury exposure areas identifyed by the 2007 Korea National Environmental Health Survey. Methods: From June to September 2010, 1,097 students from grades 3 to 6 at 19 elementary schools participated in this study, including 294 students from 10 elementary schools in Kunwee County, 529 students from Yeongcheon City, 122 students from two elementary schools in Pohang City, North Gyeongsang Province, and 152 students from two elementary schools in Ulsan Metropolitan City. Biological samples from schoolchildren, including whole blood, urine and hair, were collected to measure total mercury at the time of a health check up. Information about children was collected by questionnaire. Total mercury concentrations in blood were measured using the Direct Mercury Analyzer 80 with the gold-amalgam collection method. Results: The mean mercury levels were $2.70{\mu}g/L$ in 1,091 blood samples, $2.25{\mu}g/g-creat.$ in 820 urine samples and $1.03{\mu}g/g$ in 1,064 hair samples. Blood mercury levels in the schoolchildren was slightly higher than the result of $2.4{\mu}g/L$ from a 2006 survey of elementary school children on exposure and health effects of mercury by the National Institute of Environmental Research. However, 0.3% and 4.5% of participants exceeded the reference level of blood mercury by CHBMII ($15{\mu}g/L$) and the US EPA ($5.8{\mu}g/L$), respectively. The reference level of urine by CHBMII ($20{\mu}g/L$) was exceeded by 0.4% of participants. As factors, residence period in the study areas, residence type, father's education level and income all showed significant associations with mercury level in the biological samples. The number of dental amalgam sides showed an association with urine mercury. Fish intake preference and fish intake frequency were important factors in mercury levels. In particular, intake of shark meat and recent intake of shark meat were associated with higher mercury levels. In this regard, participation in the performance of an ancestral rite showed a relation with higher mercury levels. Conclusion: The intake of shark meat was very important factor to high mercury exposure level. It is recommended to monitor and manage students with high mercury exposures who exceeded CHBM II and EPA guidelines, and include blood mercury testing in the Children's Health check up for this province.
