Effect of the Polymorphisms of Keratin Associated Protein 8.2 Gene on Fibre Traits in Inner Mongolia Cashmere Goats

  • The aim of the experiment was to detect polymorphisms in the keratin-associated protein 8.2 (KAP8.2) gene to determine associations between the genotype and fibre traits in Chinese Inner Mongolia cashmere goats. The fibre traits data investigated were cashmere fibre diameter, combed cashmere weight, cashmere fibre length and guard hair length. Five hundred and forty-two animals were used to detect polymorphisms in the complete coding sequence of the hircine KAP8.2 gene by means of PCR-SSCP. The results identified six genotypes, AA, BB, DD, AB, AD and BD, coded for by three different alleles A, B and D. Two SNPs in the coding region were confirmed by sequencing, which were A214G and T218C respectively. The relationships between the genotypes and cashmere fibre diameter, combed cashmere weight, cashmere fibre length and guard hair length were analyzed. There were significant differences (p<0.01) between the associations of the different genotypes with cashmere fibre diameter, cashmere weight and hair length. Cashmere length was the only trait that was not associated with the genotypes. The genotype AA (0.73) was found to be predominant in Inner Mongolia cashmere goats and the animals with this genotype had the thinnest cashmere fibre diameter compared with the other genotypes. These results suggested that polymorphisms in the hircine KAP8.2 gene may be a potential molecular marker for cashmere fibre diameter in cashmere goats.

Molecular Characterization of Echovirus 30-Associated Outbreak of Aseptic Meningitis in Korea in 2008

  • Evaluation of the primary etiologic agents that cause aseptic meningitis outbreaks may provide valuable information regarding the prevention and management of aseptic meningitis. In Korea, an outbreak of aseptic meningitis caused by echovirus type 30 (E30) occurred from May to October in 2008. In order to determine the etiologic agent, CSF and/or stool specimens from 140 children hospitalized for aseptic meningitis at Soonchunhyang University Cheonan Hospital between June and October of 2008 were tested for virus isolation and identification. E30 accounted for 61.7% (37 cases) and echovirus 6 accounted for 21.7% (13 cases) of all the human enteroviruses (HEVs) isolates (60 cases in total). For the molecular characterization of the isolates, the VP1 gene sequence of 18 Korean E30 isolates was compared pairwise using the MegAlign with 34 reference strains from the GenBank database. The pairwise comparison of the nucleotide sequences of the VP1 genes demonstrated that the sequences of the Korean strains differed from those of lineage groups A, B, C, D, E, F, and G. Reconstruction of the phylogenetic tree based on the complete VP1 nucleotide sequences resulted in a monophyletic tree, with eight clustered lineage groups. All Korean isolates were segregated from other lineage groups, thus suggesting that the Korean strains were a distinct lineage of E30, and a probable cause of this outbreak. This manuscript is the first report, to the best of our knowledge, of the molecular characteristics of E30 strains associated with an aseptic meningitis outbreak in Korea, and their respective phylogenetic relationships.

  • PIGK(phosphatidylinositol glycan, class K) is a subunit of GPI transamidase that cleaves the signal peptide in proproteins and replaces it with GPI. In addition, the structure and synthesis of GPI are critically involved in some of the cellular actions of insulin. Therefore, PIGK would be essential for mammalian development and many specific cellular functions as well as for metabolic activity of insulin associated with GPI. Two types of" full-length cDNAs of porcine PIGK were cloned through RT-PCR and RACE experiments. One is thought to be a normal form(consist of 395 amino acids) and the other is considered as an alternative spliced form(consist of 371 amino acids) which contains additional 63 bps in intron 7. Since a stop codon was contained within the insertion, the spliced form has a shorter coding sequence than that of normal form. A missense mutation (T314I) in exon 6 was detected and used for genotyping to estimate association with the growth and fat deposition traits for 545 $F_2$ animals(Korean native boars ${\times}$ Landrace). From the PCR-RFLP analysis using HpyCH4III, CT genotype showed highly significant relationship(P< 0.01) with carcass fat contents.

Function of rax2p in the Polarized Growth of Fission Yeast

  • Cell polarity is critical for the division, differentiation, migration, and signaling of eukaryotic cells. RAX2 of budding yeast encodes a membrane protein localized at the cell cortex that helps maintain the polarity of the bipolar pattern. Here, we designate SPAC6f6.06c as $rax2^+$ of Schizosaccharomyces pombe, based on its sequence homology with RAX2, and examine its function in cell polarity. S. pombe $rax2^+$ is not essential, but ${\Delta}rax2$ cells are slightly smaller and grow slower than wild type cells. During vegetative growth or arrest at G1 by mutation of cdc10, deletion of $rax2^+$ increases the number of cells failing old end growth just after division. In addition, this failure of old end growth is dramatically increased in ${\Delta}tea1{\Delta}rax2$, pointing to genetic interaction of $rax2^+$ with $tea1^+$. ${\Delta}rax2$ cells contain normal actin and microtubule cytoskeletons, but lack actin cables, and the polarity factor for3p is not properly localized at the growing tip. In ${\Delta}rax2$ cells, and endogenous rax2p is localized at the cell cortex of growing cell tips in an actin- and microtubule-dependent manner. However, ${\Delta}rax2$ cells show no defects in cell polarity during shmoo formation and conjugation. Taken together, these observations suggest that rax2p controls the cell polarity of fission yeast during vegetative growth by regulating for3p localization.

The Heavy Metal Tolerant Soil Bacterium Achromobacter sp. AO22 Contains a Unique Copper Homeostasis Locus and Two mer Operons

  • Copper-containing compounds are introduced into the environment through agricultural chemicals, mining, and metal industries and cause severe detrimental effects on ecosystems. Certain microorganisms exposed to these stressors exhibit molecular mechanisms to maintain intracellular copper homeostasis and avoid toxicity. We have previously reported that the soil bacterial isolate Achromobacter sp. AO22 is multi-heavy metal tolerant and exhibits a mer operon associated with a Tn21 type transposon. The present study reports that AO22 also hosts a unique cop locus encoding copper homeostasis determinants. The putative cop genes were amplified from the strain AO22 using degenerate primers based on reported cop and pco sequences, and a constructed 10,552 base pair contig (GenBank Accession No. GU929214). BLAST analyses of the sequence revealed a unique cop locus of 10 complete open reading frames, designated copSRABGOFCDK, with unusual separation of copCD from copAB. The promoter areas exhibit two putative cop boxes, and copRS appear to be transcribed divergently from other genes. The putative protein CopA may be a copper oxidase involved in export to the periplasm, CopB is likely extracytoplasmic, CopC may be periplasmic, CopD is cytoplasmic/inner membrane, CopF is a P-type ATPase, and CopG, CopO, and CopK are likely copper chaperones. CopA, B, C, and D exhibit several potential copper ligands and CopS and CopR exhibit features of two-component regulatory systems. Sequences flanking indicate the AO22 cop locus may be present within a genomic island. Achromobacter sp. strain AO22 is thus an ideal candidate for understanding copper homeostasis mechanisms and exploiting them for copper biosensor or biosorption systems.

Comparative Study of Minimum Ripple Switching Loss PWM Hybrid Sequences for Two-level VSI Drives

  • Voltage source inverters (VSIs) are widely used to drive induction motors in industry applications. The quality of output waveforms depends on the switching sequences used in pulse width modulation (PWM). In this work, all existing optimal space vector pulse width modulation (SVPWM) switching strategies are studied. The performance of existing SVPWM switching strategies is optimized to realize a tradeoff between quality of output waveforms and switching losses. This study generalizes the existing optimal switching sequences for total harmonic distortions (THDs) and switching losses for different modulation indexes and reference angles with a parameter called quality factor. This factor provides a common platform in which the THDs and switching losses of different SVPWM techniques can be compared. The optimal spatial distribution of each sequence is derived on the basis of the quality factor to minimize harmonic current distortions and switching losses in a sector; the result is the minimum ripple loss SVPWM (MRSLPWM). By employing the sequences from optimized switching maps, the proposed method can simultaneously reduce THDs and switching losses. Two hybrid SVPWM techniques are proposed to reduce line current distortions and switching losses in motor drives. The proposed hybrid SVPWM strategies are MRSLPWM 30 and MRSLPWM 90. With a low-cost PIC microcontroller (PIC18F452), the proposed hybrid SVPWM techniques and the quality of output waveforms are experimentally validated on a 2 kVA VSI based on a three-phase two-level insulated gate bipolar transistor.

Bacillus subtilis BC-P1 균주를 이용하여 제조한 청국장의 발효 및 품질 특성분석 (Fermentation and Quality Characteristics of Cheonggukjang Fermented with Bacillus subtilis BC-P1)

  • 본 연구진은 청국장에서 수종의 미생물을 분리하였으며, 이중 소화 효소능이 뛰어난 균주를 선별하였으며, 유전학적 분석결과를 통해 본 균주가 Bacillus 속 균주임을 확인하고 Bacillus subtilis BC-P1이라 명명하였다. Bacillus subtilis BC-P1을 starter로 사용하여 청국장을 제조하였으며, protease, xylanase, chitinase activity, 혈전 분해능, 영양성분 분석 및 아미노산 분석을 통하여 B. subtilis BC-P1를 starter로 활용한 청국장 제품의 개선 가능성을 타진하였다. Bacillus subtilis BC-P1를 starter로 제조한 청국장은 기존 제품에 비해 우수한 소화효소 생성능을 보였으며, 특히 혈전분해효소 생성능이 우수함을 확인하였고, 대조군에 비해 증가된 환원당 및 총 아미노산 함량을 확인 할 수 있었다. 따라서 본 균주를 starter로 하여 청국장을 제조할 경우 기능성분, 생리활성의 증가, 품질 향상을 기대할 수 있을 것으로 판단되며, 향후 대규모 생산시 균주, 제조 조건에 관한 연구가 필요할 것으로 사료된다.

엽록체 DNA를 이용한 섬괴불나무(Lonicera insularis Nakai)의 종내변이 및 지리학적 연구 (Intraspecific variation and geographic study of Lonicera insularis (Caprifoliaceae) based on chloroplast DNA sequences)

  • 섬괴불나무(Lonicera insularis Nakai)는 인동과(Caprifoliaceae) 인동속(Lonicera L.) 에 속하는 관목으로 울릉도와 독도의 해안가 지역에 분포하는 한국특산식물이다. 섬괴불나무와 인동속의 근연종인 괴불나무, 청괴불나무, 각시괴불나무, 길마가지나무, L. morrowii, 병꽃나무 7분류군과의 유연관계를 밝히고, 섬괴불나무의 종내 변이를 확인하기 위해 엽록체 DNA 5개 영역의 염기서열을 분석하였다. 분석결과 전체 3.2kb의 염기서열이 결정되었고, 섬괴불나무와 L. morrowii 에서 2개의 엽록체 DNA haplotype(CP01-02)이 결정되었다. 섬괴불나무는 한 개의 염기치환으로 CP01 type과 CP02 type으로 구별되었으며, 일본의 L. morrowii와는 CP02 type을 공유하였다. 섬괴불나무에서 확인 된 두 개의 CP type은 1) 진화적으로 뚜렷한 두 개 이상의 계통을 가지며, 이는 하나 이상의 유입경로를 통해 울릉도로 유입되었을 가능성을 높게 지지한다. 그리고, 2) 형태학적으로 구별되는 섬괴불나무와 L. morrowii의 CP type의 공유는 두 종의 지리적인 장벽에 의한 이소적종분화의 결과로 추론할 수 있다. 본 연구에서 확인된 울릉도와 독도의 섬괴불나무의 종내 변이 및 다양성에 관한 결과는 울릉도 및 독도 식물의 분자생물지리학적 연구로 대양섬의 생물학적 진화양상과 종 분화 과정에 중요한 기초 자료로 활용될 것이다.

Isolation, Expression Pattern, Polymorphism and Association Analysis of Porcine TIAF1 Gene

  • TIAF1 is a TGF-${\beta}$1-induced anti-apoptotic factor that plays a critical role in blocking TNF (tumor necrosis factor) cytotoxicity in mouse fibroblasts and participates in TGF-${\beta}$-mediated growth regulation. In this study, we obtained the full-length cDNA sequence of the porcine TIAF1 gene. Real-time PCR further revealed that the TIAF1 gene was expressed at the highest level in liver and kidney with prominent expressions detected in uterus, and lower levels detected in heart, spleen, lung, stomach, small intestine, skeletal muscle and fat of Large White pigs. Sequence analysis indicated that a 6 base-pair deletion mutation existed in the exon of the TIAF1 gene between Meishan and Large White pigs. This mutation induced deletion of Gln and Val amino acids. PCR-RFLP was used to detect the polymorphism in 394 pigs of a "Large White${\times}$Meishan" $F_{2}$ resource population and four purebred pig populations. The frequencies of the A allele (with a 6 bp deletion) were dominant in Chinese Meishan and Bamei pigs, and the frequencies of the B allele (no 6 bp deletion) were dominant in Large White and Landrace pigs. Association analyses revealed that the deletion mutation had highly significant associations (p<0.01) with meat marbling score of the thorax-waist longissimus dorsi (LD) muscle (MM1) and intramuscular fat percentage (IMF), and significant associations (p<0.05) with carcass length (CL). The results presented here supply evidence that the 6 bp deletion mutation in the TIAF1 gene affects porcine meat quality and provides useful information for further porcine breeding.

돼지 MC1R 유전자변이의 양돈산업 적용 (Commercial Application of Porcine MC1R Gene Polymorphisms to Korean Pork Industry)

  • 본 연구는 한국재래돼지의 MC1R 유전자형을 결정하고 MC1R 유전자의 변이와 육질과의 연관성을 규명하기 위하여 실시하였다. 재래돼지의 MC1R 유전자형을 분석한 결과 중국재래돼지품종이 속하는 $E^{D1}$ 내에서도 0201형에 속하는 것으로 나타났으며 몇몇의 재래돼지에서는 $E^P$ 유전자형이 출현하였는데 이는 Berkshire 품종의 혼입으로 인한 것으로 사료된다. 흑색의 모색과 관련되어 있다고 보고된 Leu102Pro (T1132C) 변이를 재래돼지와 Yorkshire의 양방향 교배로 생산된 F2 집단의 171 두를 대상으로 분석하고 육질형질과의 연관성을 분석하였다. 모든 육질형질에 대하여 유의적인 효과를 나타내지 않았으며 이로써 돼지의 흑색모색과 육질과는 연관성이 없으며 다른 육질관련 유전자에 의해 영향을 받는다는 근거를 제공하였다. 또한 Duroc 품종에서 AA으로 고정되어 있어 다른 품종과의 구분 기준이 되는 Ala243Thr (G1554A) 변이를 이용하여 종돈검정소에서 출하된 Landrace, Yorkshire, Duroc, Berkshire 순종집단에 대한 유전자형 빈도를 조사하였으며 Berkshire를 제외한 나머지 품종에서 이형접합체가 일부 출현하였다.
