Isolation of Anaerobic Bacteria from Clinical Specimens in Veterinary Medical Teaching Hospital (대학 동물병원 임상 검체로부터 분리된 혐기성 세균과 항생제 감수성 양상)

  • The emergence of bacterial resistance to antibiotics during therapy is a matter of great problem in clinical medicine. This may be because many veterinarians have used inappropriate antibiotics without bacteriological culture. Therefore, the purpose of this study is to determine isolation of anaerobic bacteria as pathogens from veterinary clinical specimens as well as susceptibility pattern for choosing antibiotics. Various anaerobic bacteria were isolated from clinical specimens of dogs, cats, rabbits at Veterinary Medical Teaching Hospital of Seoul National University from May 2001 to October 2002. The total number of isolated anaerobic bacteria was 13 isolates; Bacteroides spp. (3 isolates), Fusobacterium spp. (2 isolates), Peptostreptococcus spp. (2 isolates), Porphyromonas gingivalis (2 isolates), Prevotella spp. (3 isolates), and Propionibacterium acnes (1 isolate). For evaluating the antibiotic susceptibility patterns of the isolates disk diffusion method was used. All isolates were susceptable to all tested antibiotics except only one Fusobacterium varium was resistant to norfloxacin.

  • This study was carried cut to determine the immunological response of uterus-induced by Lipopolysaccharides (LPS) in Holstein cows. The LPS isolated from Bacteroids helcogenes and Fusobacterium varium was injected at the rate of 100 ${\mu}g$ with 30 ml of phospahte buffer saline(PBS) in each cow(n=5). Three cows were acted as control. There was no difference in total polymorphonuclear leukocytes(PMNL) concentration in uterine fluid between control and LPS groups at 24, 48 and 72 hrs after LPS treatment. There was significant difference in rate of PMNL between control and LPS groups at 24(41.7% vs 72.1%), 48(41.0% vs 81.6%) and 72 hrs(44.3% vs 79.0%) after LPS treatment. There was no difference in PMNL viability between control and LPS groups at 24, 48 and 72 hrs after LPS treatment. There was significant difference in rate of phagocytic PMNL between control and LPS groups at 48 hr after LPS treatment(1.1% vs 7.7%).

분만후 젖소의 자궁내 미생물상 조사 및 Lipopolysaccharide의 분리

  • 분만후 젖소의 자궁내 미생물상을 조사하고 미생물로부터 분리한 Lipopolysaccharide를 적용하여 소의 번식효율 증진에 기여하고자 분만후 젖소의 도축장 유래 자궁을 채취하여 혐기적 상태에서 균분리 동정을 실시하였다. 균분리 동정을 위하여, 시료를 1cm$\times$1cm로 채취하여 혐기상태에서 거품이 생길 때까지 vortexing한 후 균액 300$\mu l$를 뽑아 혐기배지에 도말하였고 도말한 plate는 $37{\circ}C$ 혐기chamber에서 24시간 배양하였다. 혐기배지에서 자란 균의 colony를 따서 Mac, BHI+B, BHI 배지에 배양한 후 Gram stain을 실시하였다. BHI 배지에서 자란 균의 colony를 따서 BUA+B 배지에 계대배양하였고 BUA+B 배지에서 자란 균중에 가장 마지막으로 자란 균을 따서 An-IF에 넣고 탁도를 63%T로 맞춘 후 An micro plate에 100$\mu l$씩 분주하였다. 분주한 plate를 $37{\circ}C$ 혐기 chamber에서 20~24시간 동안 배양한 후 Biolog를 실시하였다. 시료의 UV측정을 위하여 Sonic Processor로 세포를 분쇄하였고 분쇄한 세포를 $4{\circ}C$에서 10,000rpm으로 10분간 원심분리한 후 상층액을 분리하여 0.45$\mu m$ 필터로 여과한 다음 여과액을 취하여 UV로 standard(E.coli O26 B6 LPS)와 sample(10배 및 20배 희석액)을 측정하였다.   • This study was carried out to investigate the effect of intra-uterine infusion of lipopolysaccharide (LPS) on reproductive performance in lactating dairy cows. Conception rate at first service was higher (56.0%) in dairy cows treated with LPS compared to non-treated cows (20.0 %). Cumulative conception rate by overall services (more than 2 services) after LPS treatment was also higher (64.0%) in dairy cows treated with LPS compared to non-treated cows (40.0%). Services per conception was significantly higher in control ($2.0{\pm}0.1$) compared with LPS ($1.2{\pm}0.4$) treated group (p<0.05).

  • This study was carried out to investigate the effect intra-uterine infusion of lipopolysaccharides(LPS) on the reproductive performance of dairy cows with retained placenta, The LPS isolated from Bacteroids helcogenes and Fusobacterium varium was injected at the rate of 100 ${\mu}g$ with 30 ml of phospahte buffer saline in each cow(n=33) at day 20 postpartum. Twelve cows with retained placenta were not heated with LPS and served as a control. Conception rate after first service was 0% and 33,3% for control and LPS groups, respectively. Service period was $149.6{\pm}34.3\;and\;53.0{\pm}12.5$ days for control and LPS groups, respectively(p<0.05). Services per conception was $3.6{\pm}0.8\;and\;2.1{\pm}0.3$ for control and LPS groups, respectively(p<0.05). In conclusion, intra-uterine infusion of LPS in cows with retained placenta can reduce the service period and services per conception. Reduced service period can improve the dairy farm profitability by decreasing cost un feeding and management of cows.
