The interest in mini watermelon (Citrullus lanatus) with small fruits weighing 2-3 kg has increased by the increasing trend in one-person households and consequent tendency to consume small meals. Watermelon grafting onto cucurbit rootstocks is a very effective way to control soil-borne diseases, such as Fusarium wilt; however, this practice negatively impacts the fruit quality. This study was conducted to investigate the growth, fruit set, and fruit quality of mini watermelon grafted onto wild watermelon accessions (Citrullus spp.) resistant to Fusarium wilt. Five watermelon accessions (Galactica, IT 208441, PI 482322, PI 500303, and PI 593358) were evaluated as rootstocks for the mini watermelon "Ministar". Non-grafted "Ministar" and "Ministar" grafted onto "Shintozwa" (Cucurbita maxima D. C. moschata D.) or "Bullojangsaeng" (Lagenaria leucantha) were used as controls. The roots of the transplants grafted onto "PI 593358" and "Shintozwa" weighed more than those on other rootstocks. Additionally, the transplants on "PI 593358" showed better growth and fruit set in the field than the other transplants. However, the total soluble solid contents and fruit quality indices of the transplants on "PI 593358" and "Shintozwa" were lower, whereas the total fruit quality index of those on "PI 482322" was higher. Thus, the wild watermelon accessions tested can potentially be used as basic germplasm for developing watermelon rootstocks instead of cucurbit rootstocks. The most promising accession for this purpose was found to be "PI 482322".
Bacillus spp. isolated from mushroom medium wastes were evaluated for their biocontrol potentials on control of Sclerotinia rot of lettuce. The Bacillus isolates were more effectively obtained from waste suspension when directly added into nutrient agar(NA) medium than plating on the agar medium. Totally 42 isolates obtained from the wastes B23 and B42 showed highest antifungal activity against eight fungal pathogens such as Sclerotinia sclerotiorum, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, Fusarium oxysporum, Colletotrichum gloeosporioides, Cladosporium cucumerinum, and Botrytis cinerea and B23 and B42 were finally selected for further studies. Optimal concentration of the isolates was $10ml(10^7cfu/ml)$ to suppress the Sclerotinia rot of lettuce. Supplements such as starch, glycerol, and egg-yolk successfully maintained the bacterial population for 30 days in vitro and increased bio-control potentials against the disease. The bacterial isolate B23 alone showed 72% control value, furthermore it presented 95% control value when supplemented with 0.2% of starch, glycerol, and egg-yolk. The promising Bacillus isolates B23 and B42 were identified as Brevibacillus brevis and Bacillus stearothermophillus, respectively, based on morphological and physiological characteristics according to API database.
As part of our search for less toxic antimicrobial agents from natural resources, the antimicrobial activity of Elfvingia applanata $(P_{ers.})\;K_{arst.}$ extract was examined alone and in combination with naringenin. EA, the aqueous extract from the carpophores of E. applanata, was lyophilized and a dark brownish powder was obtained. Antimicrobial activity of EA was tested in vitro against nineteen strains of bacteria and eleven strains of fungi by serial broth dilution method, and expressed by minimal inhibitory concentration (MIC). Among nineteen strains of bacteria tested, the antimicrobial activity of EA was the most potent against Proteus vulgaris showing MIC of 1.125 mg/ml. EA also inhibited the growth of the selected fungi at higher concentrations ranging from 7.5 mg/ml to 15.0 mg/ml. To investigate the effect of antimicrobial combinations of EA with naringenin, the fractional inhibitory concentration index (FICI) was determined by checkerboard assay for each strain. The antimicrobial combinations of EA with naringenin resulted in partial synergism against Staphylococcus aureus only, and showed additive effect in two strains including Klebsiella pneumoniae and Salmonella typhi. Antagonism was not found.
Postharvest decay of onion bulbs was examined by inspecting the commercial packages in the market or in storage. Bulb rot incidence was unexpectedly high, and onion bulbs with 1st quality grade were rotten most severely by 51%, followed by 32% for 2nd and 21% for 3rd grades. This indicates that larger bulbs had higher incidences of bulb rots. Major pathogens associated with basal and neck rots were Fusarium oxysporum and Aspergillus sp. or Botrytis allii, respectively, of which basal rot was most prevalent and damaging during storage. Among the epiphytic microorgani는 from onion plants, several Bacillus and Paenibacillus spp. and previously selected Pseudomonas putida and Trichoderma harzianum had inhibitory efficacy against bulb rot pathogens. Among these B. amyloliquefaciens BL-3, Paenibacillus polymyxa BL-4, and P. putida Cha 94 were highly inhibitory to conidial germination of F. oxysporum and B. allii. P. putida Cha 94, B. amyloliquefaciens BL-3, P. polymyxa BL-4, and T. harzianum TM were applied in the rhizoplane of onion at transplanting. Initially antagonist populations decreased rapidly during the first one month. However, among these antagonists, rhizoplane population densities of BL-3, Cha 94, and TM were consistently high thereafter, maintaining about 10$^4$-10$^{5}$ cells or spores per gram of onion root up to harvest time. The other bacterial antagonist BL-4 survived only for two months. TM was the most effective biocontrol agent against basal rot, with the number of rotten bulbs recorded at 4%, while that of the control was 16%. Cha 94 was effective for the first 20 days, but basal rot increased thereafter and had about the same control efficacy as that of BL-3 and BL-4. When the antagonists were applied to the topping areas of onion bulbs at harvest, TM was the most effective in protecting the stored onion bulbs from neck rotting. The second effective antagonist was BL-3. TM and BL-3 completely suppressed the neck rot in another test, suggesting that biocontrol of postharvest decay of onion using these microorganisms either at the time of transplanting or at harvesting may be promising.
Five nematophagous fungi, Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata and Fusarium oxysporum were evaluated for nematicidal effect on Meloidogyne hapla in greenhouse. Treatment of nematophagous fungi reduced the root galling by M. hapla and increased red-pepper growth in naturally infested pot soil. Number of galling were significantly less inall pots in 4 different inoculum densities of 5 nematophagous fungi compared to untreated plots. Especially, treatment of F. oxysprum resutled significant reduction of root gall of red-pepper. The increased shoot growth was significantly higher in pretreated plots by A. arthrobotryoides, A. conoides, A. oligospora, D. lobata and F. oxysporum at inoculum concentration of 1:100 but other treatments were not significantly increased shoot growth. Two promising fungi, D. lobata and F. oxysporum were selected in greenhouse test and in vitro results of previously experiment and applied to field plot naturally infested by M. hapla serverely. Number of galls were remarkably fewer in plots treated with D. lobata and F. oxysporum at either 1:70 or 1:100 concentration compared to the untreated plots. The shoot growth of red-pepper was increased strikingly in the plots following the red-pepper was increased strikingly in the plots following the treatment of both fungus than greenhouse test.

Background: Airborne fungi are ubiquitous in the air and exposure to an airborne fungus can be a significant risk factor. The composition of fungi has been potentially important for human health, especially for respiratory diseases like asthma and atopic dermatitis. Therefore, we attempted to ascertain what kind of airborne fungi affect human health at a nationwide level. Objectives: This study was carried out to provide information on indoor fungi distribution at multi-use facilities throughout South Korea. Methods: We classified our data by region and public facility after collection, cultivation, and identification via the sequencing of the ITS (internal transcribed spacer) region. We investigated whether or not the proliferation of HaCaT cells was affected by the identified airborne fungi. Results: In our data, the most isolated airborne fungi by region were Penicillium spp (Seoul, Daegu), Periconia sp (Gyeonggi-do), Iprex sp (Gangwon-do), Phanerochaete sp (Busan), Bjerkandera sp (Gwangju), and Aspergillus sp (Jeju-do). In the public facilities, the most detected fungi were Cladosporium sp (public transport), Penicillium sp (apartment house, retail market, financial institution, karaoke room), Bjerokandera sp (underground parking lot, public toilet, medical institution), Periconia sp (retail store), and Fusarium sp (general restaurant). Next, we selected twenty airborne fungi to examine their cytotoxicity and proliferation of human skin cells. In this experiment, the proliferation of the cells was influenced by most of the identified fungi. In case of the cytotoxicity test, most genera except for Rhodotorula sp and Moesziomyces sp showed cytotoxicity in HaCaT cells. Conclusions: The distribution of mold in the indoor air in multi-use facilities in South Korea differs from region to region, and this is an indicator that should be considered in future health impact studies. In addition, as a result of culturing about 20 types of bacteria dominant in indoor air, it was found that most (90%) inhibit the growth of skin cells, which can be harmful to health. An in-depth study of the health effects of floating fungi is needed.

The purpose of this study was to investigate damages of garlics occurred under cold storage and farmhouse storage condition, influence of cultural and environmental factors on storage spoilage of garlics, and to establish control strategies to reduce damages occurred under storage of garlics. Decays of garlics were highly related with cultural condition (paddy field or upland soil), ventilation, storage temperature and relative humidity, continuous cropping years, and harvesting stage. Early-harvested garlics were more decayed than late-harvested garlics. Garlics cultivated on paddy field were less decayed than ones cultivated on upland soil under farmhouse storage condition. The densities of Penicillium spp. and Fusarium spp. were higher on plot with long term continuous cropping cultivation history than on plot with short term continuous cropping cultivation history. However there is no relation between continuous cropping years and percent of decay of garlics. As a result of investigating influence of environmental factors on decay of garlics, P. hirsutum caused severe spoilage under high relative humidity condition, while F. oxysporum and Stemphyllium botryosum were not related with relative humidity. The three postharvest pathogens grew well above woe. In addition when P. hirsutum and S. botryosum were cultured for two months, they grew even at $-1^{\circ}C$. Except for environmental factors, wounds occurred through farming works. had an effect on storage spoilage of garlics. Garlics only hurt with a toothpick without inoculation of pathogens were decayed more severe than those inoculated with pathogens without wounds. Seven agro-chemicals showed highly suppressive effect were selected by measuring mycelial growth of three major pathogens of garlics on potato dextrose agar amended with 0.1% (v/v) of each fungicide. When they were foliar-sprayed on garlics 30 days before harvesting, it was confirmed that they suppressed storage spoilage of garlics. Also when garlics were sprayed with and drenched into the suspension of Benoram WP very after harvesting garlics, garlic damages by postharvest pathogens were reduced remarkably.

An antagonistic bacteria was isolated from rhiaosphere of pepper and corn and identified as Bacillus (B.) subtilis. These B. subtilis B-5 was transformed and marked with the plasmid pCPP4 which possess neomycine resistan. gene. The marked stranins showed growth inhibition to Rhizoctonia (R.) solani, Fusarium (F.) solani, and F. oxysporum in vitro, and were used in studying growth promoting effects on sesame and cabbage. All the identified strains utilize glucose, sucrose, fructose, lactose, mannitol and sorbitol as carbon source, but not rhamnose, and the marked strains also showed characteristics similar to wild-type strains. Germination rate of chinese cabbage and sesame seeds was increased by about 10% or more in the plot to which these strains were inoculated and the effect was higher in soil than in petri dish. The early growth promoting effects of these strains appeared higher, as compared with control plot, in the plots to which B. subtilis B-5 and pathogenic fungi was inoculated together. When the marked strains, B. subtilis B-5NEOr, were inoculated in the rhizosphere of chinese cabbage and sesame with $1.1{\times}10^8CFU/g$ dry soil, the number of inoculated strain was decreased slowly to the level of $10^5{\sim}10^6CFU/g$ dry soil after 4 weeks and the number of Pseudomonas spp. maintanied the level of $10^5CFU/g$ dry soil throught total period, but the number of fungi was decreased rapidly from the early level of $10^8CFU/g$ dry soil to $10^3CFU/g$ dry soil after 4 weeks.

A bacterial strain isolated from a Malva verticillata leaf was identified as Bacillus velezensis MV2 based on the 16S rRNA sequencing results. Complete genome sequencing revealed that B. velezensis MV2 possessed a single 4,191,702-bp contig with 45.57% GC content. Generally, Bacillus spp. are known to produce diverse antimicrobial compounds including bacteriocins, polyketides, and non-ribosomal peptides. Antimicrobial compounds in the B. velezensis MV2 were extracted from culture supernatants using hydrophobic interaction chromatography. The crude extracts showed antimicrobial activity against both gram-positive bacteria and gram-negative bacteria; however, they were more effective against gram-positive bacteria. The extracts also showed antifungal activity against phytopathogenic fungi such as Fusarium fujikuroi and F. graminearum. In time-kill assays, these antimicrobial compounds showed bactericidal activity against Bacillus cereus, used as indicator strain. To predict the type of antimicrobial compounds produced by this strain, we used the antiSMASH algorithm. Forty-seven secondary metabolites were predicted to be synthesized in MV2, and among them, fourteen were identified with a similarity of 80% or more with those previously identified. Based on the antimicrobial properties, the antimicrobial compounds may be nonribosomal peptides or polyketides. These compounds possess the potential to be used as biopesticides in the food and agricultural industry as an alternative to antibiotics.
