Bioactive Carotenoid, Fucoxanthin as Chemotaxonomic Marker and Antioxidative Agent from the Marine Bacillariophycean Microalga Hantzschia marina

  • Allenic and epoxy carotenoid, fucoxanthin (1) was isolated from the marine bacillariophycean microalga Hantzschia marina and the structure was assigned on the basis of comprehensive spectroscopic analyses. Fucoxanthin was detected only from diatom among three families (green algae, diatom and blue-green algae) of the marine microalgae tested. Fucoxanthin showed free radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and peroxynitrite $(ONOO^-)$ with $IC_{50}$ values of $32\;{\mu}M\;and\;60\;{\mu}M$, respectively.

Bio-functions of Marine Carotenoids

  • Carotenoids being most important pigments among those occurring in nature, have received increased interest owing to their beneficial effects on human health. An effort is made to review marine carotenoids as important bioactive compounds with reference to their presence, chemical, and biofunctional benefits they afford. The potential beneficial effects of marine carotenoids were particularly focused on astaxanthin and fucoxanthin, major marine carotenoids found in marine animals and aquatic plants, respectively. Both carotenoids show strong antioxidant activity which is attributed to quenching singlet oxygen and scavenging free radicals. The potential role of the carotenoids as dietary antioxidants has been suggested as being one of the main mechanism by which they afford their beneficial health effects such as anticancer activity and anti-inflammatory effect. Only recently, antiobesity effect and antidiabetic effect have been noted as specific and novel bio-functions of fucoxanthin. Nutrigenomic study reveals that fucoxanthin induces uncoupling protein 1 (UCP1) expression in white adipose tissue (WAT) mitochondria to lead to oxidation of fatty acids and heat production in WAT. Fucoxanthin improves insulin resistance and decreases blood glucose level, at least in part, through the down-regulation of tumor necrosis factor $\alpha$ ($TNF{\alpha}$) in WAT of animals.

Anti-Cancer Mechanism and Possibility of Nano-Suspension Formulation for a Marine Algae Product Fucoxanthin

  • Recently, use of natural products available from marine sources, and especially algae products, are receiving more attention. Scientific evidence for claimed nutraceutical and therapeutical effects of one such marine algae product, fucoxanthin, is discussed in this paper with a summary of the currently available literature regarding its antioxidant, anti-obesity and anticancer activities. It is safe for use in humans, but as it has poor solubility a nano-suspension mode of delivery may be adopted to improve efficacy of supplments. We conclude from ourliterature review that the marine algae product fucoxanthin has significant antioxidant, anti-obesity and anticancer activity with established mechanisms of action.

The effect of fucoxanthin rich power on the lipid metabolism in rats with a high fat diet

  • This study determined the effects of fucoxanthin on gene expressions related to lipid metabolism in rats with a high-fat diet. Rats were fed with normal fat diet (NF, 7% fat) group, high fat diet group (HF, 20% fat), and high fat with 0.2% fucoxanthin diet group (HF+Fxn) for 4 weeks. Body weight changes and lipid profiles in plasma, liver, and feces were determined. The mRNA expressions of transcriptional factors such as sterol regulatory element binding protein (SREBP)-1c, Carnitine palmitoyltransferase-1 (CPT1), Cholesterol $7{\alpha}$-hydroxylase1 (CYP7A1) as well as mRNA expression of several lipogenic enzymes were determined. Fucoxanthin supplements significantly increased plasma high density lipoprotein (HDL) concentration (P < 0.05). The hepatic total lipids, total cholesterols, and triglycerides were significantly decreased while the fecal excretions of total lipids, cholesterol, and triglycerides were significantly increased in HF+Fxn group (P < 0.05). The mRNA expression of hepatic Acetyl-CoA carboxylase (ACC), Fatty acid synthase (FAS), and Glucose-6-phosphate dehydrogenase (G6PDH) as well as SREBP-1C were significantly lower in HF+Fxn group compared to the HF group (P < 0.05). The hepatic mRNA expression of Hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) and Acyl-CoA cholesterol acyltransferase (ACAT) were significantly low while lecithin-cholesterol acyltransferase (LCAT) was significantly high in the HF+Fxn group (P < 0.05). There was significant increase in mRNA expression of CPT1 and CYP7A1 in the HF+Fxn group, compared to the HF group (P < 0.05). In conclusion, consumption of fucoxanthin is thought to be effective in improving lipid and cholesterol metabolism in rats with a high fat diet.

HPLC-PDA를 이용한 기능성 화장품 중 푸코잔틴의 정량 (Determination of fucoxanthin in cosmeceutical products by HPLC-PDA)

  • 푸코잔틴을 함유한 원료 및 이를 유효성분으로 처방된 기능성 화장품의 품질관리를 위하여 photodiode array (PDA) 검출기를 갖춘 HPLC를 이용한 분석법을 확립하고 원료 및 제품 함량 분석에 응용하고자 본 연구를 수행하였다. 컬럼은 옥타데실화한 실리카 겔을 충진한 것을 사용하였으며 PDA의 측정파장은 499 nm로 설정하였다. 분석법을 밸리데이션하기 위하여 검량선의 직선성, 검출 및 정량 한계, 재현성, 회수율을 조사한 결과 양호한 결과를 얻었다. 검량선의 상관계수는 1.000로 0.5 ~ 100 ppm의 농도에서 직선성이 양호하였다. 또한 검출 한계는 0.1 ppm이었으며 정량 한계는 0.5 ppm이었다. 시스템의 적합성을 확인하기 위하여 피크의 재현성을 평가한 결과 피크 면적 값의 RSD (n=5) 값은 2.0%이었으며 머무름 시간의 RSD는 0.09% 이었다. 표준물질 첨가법의 실험 결과 회수율은 $101.6{\pm}0.77%$이었다. 끝으로 확립된 분석법을 원료 및 제품 분석에 적용하였다. 두 종류의 푸코잔틴 함유 원료의 푸코잔틴 함량은 각각 $49.6{\pm}3.3%$$1.03{\pm}0.016%$이었다. 또한 푸코잔틴이 150 ppm이 되도록 처방한 제품의 푸코잔틴의 함량은 $156.7{\pm}4.7ppm$으로 확인되었다. 위 실험결과로 볼 때 본 실험방법은 기능성 화장품에서 푸코잔틴을 정량분석하는 것에 적용될 수 있을 것으로 판단되었다.

Fucoxanthin derivatives from Sargassum siliquastrum inhibit matrix metalloproteinases by suppressing NF-κB and MAPKs in human fibrosarcoma cells

  • Fucoxanthin is known to be an effective cell proliferation inhibitor with anti-tumor and anti-angiogenic activities. However, there is a lack of data regarding the biological effects of cis isomers of fucoxanthin. To assess the potential therapeutic properties of 9'-cis-(6'R) fucoxanthin (FcA), and 13-cis and 13'-cis-(6'R) fucoxanthin complex (FcB) isolated from Sarggassum siliquastrum, we investigated their inhibitory effects on matrix metalloproteinases (MMPs) in phorbol 12-myristate 13-acetate (PMA)-induced human fibrosarcoma (HT1080) cells. FcA and FcB reduced MMP-2 and MMP-9 protein and mRNA levels, as well as the migration of these cells, in a dose-dependent manner. Additionally, FcA and FcB increased levels of MMPs inhibition factors such as tissue inhibitor of metalloproteinase-1. FcA and FcB significantly inhibited the transcriptional activity of nuclear factor ${\kappa}B$ (NF-${\kappa}B$) and by inhibiting c-Jun N-terminal kinase (JNK) and p38 mitogen-activated protein kinases. Our results demonstrate that suppression of the NF-${\kappa}B$, JNK, and p38 signaling pathways may inhibit PMA-induced MMP-2 and MMP-9 activity. Therefore, FcA and FcB may be useful in noninvasive therapeutic strategies against fibrosarcoma metastasis.

전복(Haliotis discus hannai) 내장(內臟)의 Carotenoid에 관(關)한 연구(硏究) (Studies on the carotenoids in the viscera of abalone (Haliotis discus hannai))

  • 전복(Haliotis discus hannai) 내장(內臟)에 함유(含有)되어있는 carotenoid에 관(關)하여 시험(試驗)하였다. 우리나라 근해단(近海岸)에서 채취(探取)된 전복의 내장(內臟)을 acetone-methanol (4 : 1) 혼합용매(混合溶媒)로 추출(抽出)하고 이 추출액중(抽出液中)의 carotenoid를 silica gel TLC, $Mg(OH)_2$ 여과지(濾過紙) chromatography 및 $Mg_2(OH)_2CO_3$ TLC에 의하여 분리정제(分離精製)하였다. 분리정제(分離精製)한 11종(種)의 carotenoid에 대(對)하여 epoxide test, partition ratio 측정(測定), $NaBH_4$에 의한 환원(還元), 알카리처리(處理), 표준(標準) carotene와 비교시험(比較試驗), electronic absorption spectrophotometry, 및 infrared spectroscopy 등(等) 여러가지 시험(試驗)은 하였다. 확인(確認)된 carotenoid는 ${\alpha}$-carotene, ${\beta}$-carotene, lutein, zeaxanthin, siphonaxanthin, siphonein fucoxanthin, loroxanthin 유사물(類似物) 및 fucoxanthinol 류이물등(類似物等) 9종(種)이며 이 밖에 2종(種)의 미량(微量) carotenoid는 확인(確認)되지 못했다. fucoxanthin은 특(特)히 KOH처리(處理)에 의하여 fucochrome이나 semifucoxanthol과 동일(同一)한 chromophore를 갖는 산물(産物)로 변환(變換)됨을 발견(發見)하였다. 전복 내장(內臟)에서 확인(確認)된 색소(色素)들 가운데siphon-axanthin, siphonein, fucoxanthin, loroxanthin 유사물(類似物) 및 fucoxanthinol 유사물(類似物) 등은 지금까지 패류(見類)에서 발견(發見)되었다는 보고(報告)가 없다.

Effects of Fucoxanthin Addition to Ground Chicken Breast Meat on Lipid and Colour Stability during Chilled Storage, before and after Cooking

  • Effects of fucoxanthin (FX), a major carotenoid in algae, on lipid peroxidation and meat colour in ground chicken breast meat were investigated. FX and/or ${\alpha}$-tocopherol (Toc) were added to ground samples at a content level of 200 mg/kg. The samples were subjected to a chilling storage test before and after cooking. In the storage test before cooking, neither FX nor Toc affected the thiobarbituric acid reactive substances (TBARS) on days 1 and 6, and FX decreased the L* value and increased the a* and b* values. In the storage test after cooking, both FX and Toc decreased TBARS values on days 1 and 6. FX decreased the L* value and increased the a* and b* values, similar to what occurred in the storage test before cooking. Based on these results, we concluded that FX is a potent ingredient for improvement of the appearance and shelf life of chicken meat and its products.

LED (Light-Emitting Diode)를 이용한 미세조류 (Chaetoceros calcitrans)의 성장 및 생화학적 조성 변화 (Changes in the growth and biochemical composition of Chaetoceros calcitrans cultures using light-emitting diodes)

  • The marine microalgae Chaetoceros calcitrans was cultured under a fluorescent lamp (CON) and light-emitting diodes (LEDs) of various wavelengths (blue, LB; red, LR; green, LG; white, LW); changes in growth, fucoxanthin, chlorophyll-a, amino acid and fatty acid profiles were investigated. LR-exposed cultures exhibited the highest specific growth rate (SGR) (0.34), whereas LG-exposed cultures showed the lowest SGR (0.26). After cultivation for 10 days, the maximum dry cell weight (g/L) of LR-exposed cultures was significantly higher than that of those exposed to other light conditions (LR${\geq_-}$CON>LB${\geq_-}$LW${\geq_-}$LG). Eicosapentaenoic acid (EPA) levels were significantly higher in CON-exposed cultures compared to those exposed to LW (P<0.05), with no marked difference compared to those exposed to LB, LR and LG (P>0.05). The fucoxanthin content was highest in LB-exposed cultures ($6.3{\mu}g/mL$), whereas LW showed the lowest ($3.6{\mu}g/mL$; P<0.05). Chlorophyll-a content was highest in cultures exposed to LB compared to other light sources. These results suggest consistent differences in growth and biochemical composition after exposure to light of different wavelengths.
