Expression of Arabidiopsis CAX4 in tomato fruits increases calcium level with no accumulation of other metallic cations

  • We generated transgenic tomato plants with Arabidopsis thaliana $H^+$/cation exchanger gene (C4X4) by Agrobactrium-mediated transformation. We confirmed transgene copy number and transcription by Southern and Northern blot analyses. The intact CAX4-expressing tomato (Lycopersicon esculentum) fruits contained 63-71% more calcium ($Ca^{2+}$) than wild-type fruits. Moreover, ectopic expression of C4X4 in tomato fruits did not show any significant increase of the four kinds of metallic cations analyzed ($Mg^{2+}$, $Fe^{2+}$, $Mn^{2+}$, and $Cu^{2+})$. The C4X4-expressing tomato plants including their fruits did not show any morphological alternations during whole growth period. These results suggest the enhanced Ca-substrate specificity of CAX4 exchanger in tomato. Therefore, intact CAX4 exchanger can be a useful tool for $Ca^{2+}$ nutrient enrichment of tomato fruits with reduced accumulation of undesirable cations.

The Anthelmintic Principle of "O-Mae", the Roasted Fruits of Prunus mume, Against Clonorchis sinensis (오매의 간디스토마 살충성 물질에 관한 연구)

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

  • Jujube is one of korean most favorite fruits. Harvesting operations of the jujube farming, however, completely depend on hand labor. Especially, collecting operation requires about three times of hand labor compared with separate operation by impacting or shaking of twigs in the hand-harvesting system of jujube fruits. Consequently, jujube farmers sincerely demand the mechanization of the collecting operation of jujube fruits in the harvesting operation. A new collecting system was designed and constructed five parts-collecting pad, guide ring system, sorting system, frame with driving system, and handle with operation levers, which performed collecting operation of jujube fruits without missing fruits in five part functions, however, an extra future more deliberate study would need how to handle conveniently the collecting pad of the prototype in the narrow space of row of the jujube farm.

Antimicrobial Activity of Thinned Strawberry Fruits at Different Maturation Stages

  • Among the phenolic compounds that is generally present in strawberry fruits, five simple phenolics, three flavonoids, and a stilbene were tested for their antimicrobial activity against seven fungi and one oomycete. trans-Cinnamic acid showed strong antimicrobial activity, and the antimicrobial effect of the simple phenolics decreased with an increase in the number of hydroxyl groups. Phytophthora capsici was the most susceptible to the phenolic compounds tested in this study. trans-Cinnamic acid, p-hydroxybenzoic acid, and kaempferol were mainly detected in 'Seolhyang' strawberry fruits, and the total phenolic contents of the fruits decreased during their development. Extracts of the green (1-10% red color) and red (above 90% red color) strawberry fruits reduced the mycelial growth and zoospore germination rate of P. capsici, and the extract of red strawberry fruit showed strong antimicrobial activity against the zoospore germination of P. capsici. These results indicate that strawberry fruits contain antimicrobial phenolic compounds and that strawberry fruit extract can be used as a natural fungistat.

Studies on the Changes of Vitamin C content in the Hot Green Pepper Fruits during the Circulation Periods (풋고추의 유통과정중 Vitamin C 함량의 변화)

  • This study was carried out to observe the changes of Vitamin C content and to preserve the quality for the circulation periods of hot green pepper fruits wrapped with polyethylene film under the indoor temperature, as well as to find the proper time for harvest of hot green pepper fruits as a hot taste food and vegetable. the results obstained were as follows: 1. The consignment rate of hot green pepper fruits was highest, 60.2% on the 25th day after flowering, the nest 25.4% on the 20 th day, 10.3% on the 30th day and 4.1% on the 35th day. 2. the content of Vitamin C I each pepper fruits was highest o the 30th day, considerably high on the 25th day and 20th day. 3. The total content of Vitamin C in each part of hot green pepper fruits; the content in pericarp was much higher than in placenta and seed, and that of hydro type Vitamin C was almost the same. 4. The reduction rate of vitamin C during circulation periods was highest on the 3rd day; 42% in pericarp, 50% in placenta, 65% in seed. But in the package wrapped up with polyethylene film, the reduction rate in the same period was 5% in pericarp, which was very low in reduction, and those in placenta and seed were almost the same as in the unwrapped state.

Brown Rot of Apricot and Mume Caused by Phomopsis vexans

  • Brown rot symptoms severely occurred on fruits of apricot and mume grown in Changnyeong, Suwon and Yeongi in Korea during a disease survey in June 2002. A total of 32 isolates of Phomopsis sp. was obtained from the fruit rot symptoms. All isolates were identified as Phomopsis vexans based on their morphological and cultural characteristics. Four isolates of the fungus were tested for pathogenicity to fruits of apricot and mume by artificial inoculation. All isolates induced brown rot symptoms on the fruits of apricot and mume by wound inoculation. Unwounded inoculation also induced symptoms on mume fruits but not on apricot fruits. The symptoms on the fruits induced by artificial inoculation were similar to those observed in the orchard. The pathogenicity tests revealed that mume was more susceptible to the pathogen than apricot. This is the first report of brown rot of apricot and mume caused by P. vexans.

Effect of Harvesting Season on the 6-Cryptoxanthin in Shiranuhi Mandarin Fruit Cultivated in Jeju Island

  • [ $\beta-Cryptoxanthin$ ] content was determined in Shiranuhi mandarin fruits harvested at monthly intervals from October to February in Jeju Island. Crude carotenoids were extracted from both peel and flesh of Shiranuhi mandarin fruits and analyzed using TLC and HPLC; $\beta-cryptoxanthin$ was indicated the Rr value of 3.2 and retention time of 23 min, respectively. $\beta-Cryptoxanthin$ contents in both peel and flesh were increased gradually as the citrus fruits ripened fully until harvesting season (February). According to the harvesting time, $\beta-cryptoxanthin$ contents in the peel were $0.15\;mg\%\;(October),\;0.28\;mg\%\;(November),\;0.38\;mg\%\;(December),\;1.23\;mg\%\;(January),\;and\;1.71\;mg\%\;(February).$In the flesh, $\beta-cryptoxanthin$ contents were lower than those of peels, having $0.06\;mg\%\;(October),\;0.08\;mg\%\;(November),\;0.19\;mg\%\;(December),\;0.26\;mg\%\;(January),\;and\;0.65\;mg\%\;(February).$ These results demonstrate that $\beta-cryptoxanthin$ in Shiranuhi mandarin fruits accumulated during ripening of the citrus fruits. In particular, the peels had much higher concentrations of $\beta-cryptoxanthin$ and have potential for use as a functional ingredient.

Analysis of Flavonoid Contents in the Fruits of Acanthopanax Species using HPLC

  • Analysis of flavonoid contents in the fruits of Acanthopanax species (A. chiisanensis, A. divaricatus, A. koreanum, A. senticosus, and A. sessiliflorus) was conducted by high performance liquid chromatography. A Discovery$^{(R)}$ C18 ($4.6{\times}250$ mm, 5 ${\mu}m$) column was used with a gradient mobile phase of water and acetonitrile (90 : 10 to 60 : 40 for 60 min) and UV detection was conducted at 350 nm. The contents of rutin, hyperin, quercetin, afzelin, and kaempferol were 0.063~0.540, 0.494~7.480, 0.584~0.704, 0.388~0.567, 0.190~0.471 mg/g, respectively, in the fruits of Acanthopanax species. Total content of flavonoids in the fruits of Acanthopanax species was highest in those of A. chiisanensis. Furthermore, hyperin was the most abundant compound in the fruits of Acanthopanax species. Consequently, our results demonstrate that the fruits of Acanthopanax species containing flavonoids have promising potential as a new income source of agriculture and industry in medicinal natural products, health supplements, and beverages.

Anti-hypertensive Effects of DHP1501, Ethanolic Extracts from Eleutherococcus sessiliflorus Fruits, via Inhibition of Angiotensin Converting Enzyme and Activation of Endothelial Nitric Oxide Synthase (오가피열매 주정추출물, DHP1501의 ACE 억제 및 eNOS 활성화를 통한 항고혈압 효능)

  • The fruits of Eleutherococcus sessiliflorus (Rupr. & Maxim.) S. Y. Hu (Araliaceae), as edible fruits, were traditionally used for ingredients of wine or tea in Eastern Asia. In addition to, the fruits of E. sessiliflorus were known for having antioxidant and anti-inflammatory effects. Recently, we investigated that the ethanolic extracts of E. sessiliflorus fruits (DHP1501) have effects on hypertension via vasorelaxation and decrease of blood pressure. In the present study, we investigated that the gene and protein expression levels of endothelial nitric oxide synthase (eNOS) was increased by treatment of DHP1501 in HUVECs. Moreover, we confirmed the angiotensin converting enzyme inhibitory activity of DHP1501 through in vitro tasks. Therefore, DHP1501 could be a candidate of functional food for alleviating hypertension.

Quantitative Analysis of Flavonoids in the Unripe and Ripe Fruits and the Leaves of Four Korean Rubus species (한국산 4종 Rubus속 숙과, 미숙과 및 잎의 플라보노이드 함량 분석)

  • The five flavonoids, isoqueritrin, juglanin, astragalin and 2-O-trans-p-coumaroyl astragalin were quantitatively analyzed in the leaves, unripe- and ripe fruits of four Korean Rubus species including Rubus crataegifolius, R. pungens var. oldhami, R. parvifolius, and R. coreanus belonging to the Rosaceae family. The HPLC analysis demonstrated that flavonoids are more abundant in the leaves than in the fruits. In the four Rubus species, ripe fruits generally contained similar or more amount of flavonoids than in unripe fruits. In particular, the quantity levels of total flavonoid (49.81${\pm}$0.50 mg/g) and isoquercitrin (35.08${\pm}$0.37 mg/g) were highest in the leaf of R. crataegifolius. In addition, the content of the flavonoids were quite low in the unripe fruits of R. coreanus that is most widely used as Rubi Fructus.
