먹물버섯의 생성.자가소화 과정에서 laccase 및 chitinase의 발현 (Chitinase and Laccase Expression during the Fruit Body Development in Coprinellus Congergatus)

  • 먹물버섯은 버섯 시원체로부터 버섯이 성숙되는 과정에서 자가소화가 일어나 먹물이라 불리는 검은 액체를 생성한다. 이 과정에서 멜라닌을 생성하는 laccase, 균류 세포벽 성분의 하나인 키틴을 분해하는 chitinase의 관련을 분석하고자 Northern hybridization 방법을 이용하여 유전자의 발현을 분석하였다. 시원체가 생성되고 버섯이 성숙되어 먹물을 생성하는 시기에 따라 멜라닌색소 생성 효소인 laccase와 킨틴분해효소인 chitinase의 발현이 증가하는 것이 확인되었다.

Effects of a Health Drink Containing the Extract of the Hovenia Dulcis Fruit Stalk and Theracurmin, on Ethanol-Induced Hangover

  • The fruit stalk of Hovenia dulcis (H.dulcis) is traditionally used to relieve hangovers in Korea. Theracurmin is a highly absorbable curcumin preparation which increases the bioavailability of curcumin. Curcumin is known for its antioxidant and anti-inflammatory effects. However, the role of this combination in lowering alcohol levels in the body, thereby alleviating the severity of alcohol-induced hangover has not been investigated. Therefore, we conducted a study to investigate the eliminatory effects of a health drink containing the extract of the H. dulcis fruit stalk and theracurmin (theracurmin drink) on ethanol-induced hangover in rats. The theracurmin drink delivered orally to rats 30 mins before the administration of 40% ethanol (5 g/kg body weight), lowered the concentration of ethanol and acetaldehyde in the blood samples collected 1, 3, and 5 h after ethanol administration. Furthermore, the theracurmin drink increased the activities of alcohol dehydrogenase and aldehyde dehydrogenase enzymes. The effectiveness of the theracurmin drink was thus superior to that of other health drink products, suggesting that its consumption may alleviate or prevent an alcohol-induced hangover.

RGB LED 광원이 느타리류의 자실체 특성에 미치는 효과 (The effect of RGB LED lights on oyster mushroom (Pleurotus spp.) fruit-body characteristics)

  • 생육 시 사용하는 LED 조명색이 느타리류의 갓색과 수량, 형태에 어떤 영향을 미치는지 알고자 하였다. 주요결과는 다음과 같다. 자실체의 전체적인 형태는 LED 조명색별로 차이를 보였는데, 청색과 자색이 일반적으로 생산하는 버섯 형태와 유사하였고 녹색은 조명이 없거나 약한 곳에서 자란 버섯형태를 보였다. 전체 품종의 청색, 녹색, 자색 LED에서 키운 버섯의 갓명도의 평균은 각각 57.0, 57.4, 59.4였다. 원형1호와 황금산타리를 제외하고는 세가지 LED 색에 따라 갓 명도의 평균이 통계적으로 유의한 차이가 있었다. 갓의 적색도는 LED 조명과 품종에 따라 많은 차이가 관찰되었다. 청색, 녹색, 자색에서 생육한 자실체의 갓색이 모두 음의 값을 나타낸 품종은 곤지7호의 갓색돌연변이인 곤지7호M이 유일하였다. 곤지7호를 제외한 8개 품종에서 청색에서 갓의 적색도가 가장 높았다. 전체 품종의 LED 조명색별 평균 수확량은 자색이 68.0 g, 청색 58.3 g, 녹색 50.1 g 이었다. 청색에서 가장 수확량이 많은 품종은 곤지7호로 92.8 g이었고 같은 품종의 녹색 처리구에서는, 77.1 g, 자색에서는 98.6 g이 수확되었다. 발이소요일수는 자색에서 생육한 버섯의 평균값이 5.3일로 가장 짧고, 청색에서 5.8일, 자색에서 5.8일 순이었다. 자색에서 가장 짧은 발이소요일수를 보인 품종은 6개, 녹색에서는 3개, 청색에서는 2개였다. 자실체의 길이는 생육 시 조사한 LED 조명색에 따라 녹색(66.4 mm) > 자색(51.8 mm) > 청색 (46.8 mm)의 순으로 나타났다. 본 연구결과가 시장에서 요구하는 형질을 갖춘 버섯을 생산하는 기초가 될 것으로 사료한다.

표고버섯과 느타리 버섯의 항암효과 (Anti-cancer Activity of Lentinus edoeds and Pleurotus astreatus)

  • 연구는 버섯을 이용한 기능성식품 개발 및 버섯가공 산업 육성에 기초자료를 제공하고자 표고버섯 및 느타리버섯 자실체, 균사체 및 폐상의 조단백다당체 분말을 0, 5, 25 mg/kg 농도로 생리식염수에 용해시켜 마우스(DBA/2와 ICR)에 주사하여 i) 백혈병$(L_{1210})$, 간암$(H_{22}),\;mouser{\;}sarcoma180\;(S_{180})$에 대한 항암효과, ii) 면역기전에 관련된 장기중량변화, 용혈반 형성 세포수 변화 등 조단백다당이 면역에 미치는 영향, 그리고 in vitro 세포독성실험을 수행하였다. 본 실험에서 사용한 모든 조단백다당류는 백혈병에 대해 항암효과를 보였는데 그 중 표고버섯자실체 25 mg/kg 처리구가 가장 높은 저지율(86%)을 보였다. 간암$(H_{22})$에 대한 항암효과는 표고버섯균사체 5 mg/kg 처리구를 제외하고 모든 처리구에서 저지효과를 보였는데, 그 중 느타리버섯 폐상 25 mg/kg 처리구가 가장 높은 저지율(87.6%)을 보였으며, 다음은 표고버섯자실체 25 mg/kg 처리구(71%)였다. Sarcoma180에 대한 항암효과는 표고버섯균사체와 자실체 25 mg/kg 처리구에서 각각 30.9,와 33.9%의 저지율을 보였다. In vitro 세포독성검사에서 각 시료의 최종농도 $50,\;100,\;200,\;400\;{\mu}g/{\mu}L$에서 $L_{1210}$에 대한 유의적 세포사망률은 보이지 않았다. 면역효과 실험에서 간장과 비장중량은 농도증가에 따라 증가하는 추세이나 현저한 차이는 없었다. 표고버섯균사체와 자실체 처리시 항체생성능력을 지닌 용혈반 형성 세포수는 대조군에 비해 현저히 높게 나타났다.

Inhibitory Effect of Aqueous Extracts from the Fruit Body of Lentinus edodes on Rat Intestinal Mucosa $\alpha$-glucosidase Activity and Reducing the Increase of Blood Glucose after Streptozotocin-induced Diabetic Rats

  • The aqueous extract from the fruit body of Lentinus edodes was evaluated for inhibitory activities against $\alpha$-glucosidase isolated from Spargue-Dawley male rats. A aqueous extract of Lentinus edodes exhibited 13.8% inhibitory activity on using 2 mM p-nitrophenyl $\alpha$-D-glucopyranoside as a substrate ($IC_{50}$ 75.3 mg/ml). The aqueous extract of Lentinus edodes inhibition type on $\alpha$-glucosidase was determined to be competitive inhibition. When it was oral administered to increase of blood glucose levels after STZ-induced in a dose dependent dietary. These results suggest that aqueous extract of Lentinus edodes effect a metabolism of intestine, and thereby reducing the increase of blood glucose after STZ-induced.

흑목이 버섯 자실체의 흑갈색 색소 동정 및 특성 (Isolation and Characterization of Dark Brownish Pigments from Fruit Body of Auricularia auricula)

  • A dark brownish pigment from fruit body of Auricularia auricula was isolated and characterized in this report. The pigment was obtained with a yield of 0.61%(w/w) by alkaline extraction and subsequent purification steps. It showed the positive $FeCl_3$ test which was the indication of phenolic compounds. A synthetic melanin showed a similar spectrometric characteristics to the pigment extract regarding a characteristic UV absorption between 200-250 nm and infrared absorptions profiles in the finger print region including absorption peaks at 1701 and 1624 $cm^{-1}$. Its element analysis indicated that its atomic copmposition is close to that of DOPA melanin (eumelanin). With the result of its antioxidant activity in the TNBT (5-thio-2-nitrobenzoic acid) assay, we concluded that the dark brownish pigment from A. auricula is a melanin-like compound having a powerful antioxidative activity.

양송이의 균사생장(菌絲生長) 및 자실체(子寶體) 수량(收量)에 미치는 복토재료(覆土材料)의 이화학적(理化學的) 성질(性質)에 관(關)한 연구(硏究) (Studies on the Physico-Chemical Characteristics of Different Casing Materials Affecting Mycelial Growth and Yield of Cultivated Mushroom, Agaricus bisporus (Lange) Sing.)

  • Since the importance of casing in fruit body formation of Agaricus bisporus has been emphasized, physico-chemical characteristics of casing materials were discussed by many workers and a mixture of peat and mineral soil as proper casing material has been adopted in many of mushroom growing countries. Because of limited resources of peat in Korea, it is necessary to find practical performance and substitutional materials for casing. The effect of casing on mycelial growth and mushroom yield of A. bisporus varied with materials, its combination and practices etc. The experiments to be discussed in this paper are concerned with pH and Ca of casing material which influence A bisporus, and changes of physico-chemical characteristics with mixing ratio of casing materials and its effect on A. bisporus. The optimum range of moisture content of each material, management of watering and application of physico-chemical characteristics casing materials was also investigated and re-use of weathered spent compost for casing material was described. 1. The effect of calcium on mycelial growth of A. bisporus at various pH in Halbschalentest showed different results with calcium sources. Best results were obtained around neutrality and fresh weight of fruit bodies grown in the range of pH 7 to 8 was highest among the tested levels. 2. Available moisture, pore space, organic matter, cation exchangeable capacity and exchangeable cation was increased by an increase of mixing ratio of peat in casing materials, while an adverse effect was obtained by addition of sand. 3. Mycelial growth on clay loam was more rapid at a lower bulk density of 0.75g/cc and at 20% moisture content on a dry weight basis at the same bulk density. 4. Mixing ratio of casing materials, 60 to 80 per cent by volume of peat mixed with 20 to 40 per cent of clay loam produced the highest yield of fresh fruit bodies and sand the lowest. However, per cent of open cap was highest in peat and lowest in sand. 5. Days required for fruit body initiation was shortened in mixtures of peat and clay loam by one to three days compared with other materials and the formation of flushes was clear. 6. The effect of some physico-chemical characteristics of casing materials on the fresh weight of fruit bodies were estimated by a multiple regression equation; Y=-923.86+$8.18X_1+8.04X_2+7.90X_3+0.12X_4+2.03X_5-0.82X_6-0.54X_7$ where $X_1,X_2,X_3,X_4,X_5,X_6,X_7$ are sand, silt, clay, available moistuer, porosity, organic matter and exchangeable cation respectively. The productivity of certain casing material could be predicted from this equation. 7. Fresh weight of fruit bodies was positively correlated with porosity exchangeable cation, organic matter, available moisture, silt and clay of materials; while sand was negatively correlated. On the contrary, sand was the unique factor reducing per cent of open cap. 8. Distribution of three phases of high productive casing material was concentrated in the range of 10 to 30 per cent solids, 15 to 30 per cent liquids, and 50 to 60 per cent in air volume. 9. Fresh weight of fruit bodies from peat was not affected with heavy watering but in clay loam and sandy loam severe crop losses occurred. Fresh weight of individual fruit was increased and open caps were decreased with heavy watering but light watering resulted in adverse effects: its effect was especially great in peat. 10. Optimum range of moisture content by weight on a dry basis was different with each casing material. To maintain optimum moisture content concerned with yield of fruit bodies and open cap, sandy loam and peat mixtures required daily watering of 0.6, 0.6 to 1. 2 and 1.2 to 2.4 liters per $3.3m^2$ of bed area, respectively. 11. Maximum yield of fruit body was recorded in the range of pF 2. 0 to 2. 5 of casing materials if organic matter content was below 4.2 per cent and in pF 1. 3 to 1.8 if above 7.1%. 12. pF curve of a certain casing material could be draws from moisture content at various pF values by multiple regression equations provided texture, organic matter and calcium of the casing material are given. Optimum moisture range of the casing materials also could be estimated by the equation. 13. It was possible to improve the phyico-chemical characteristics of clay loam and sandy loam by addition of weathered spent compost although the effect was less than in the case of peat. Fresh weight of fruit bodies wsa increased by addition of weathered spent compost but its effect was not as remarkable as peat. Accordingly, further studies will be required.

느타리(1100㎖병)의 균사 배양 중 배양기 내부 통기성 개선 (Studies on the aeration improvement of inner bottle(1100ml) culture system during the mycelial culture of Pleurotus ostreatus)

  • 본 연구는 느타리버섯 $1,100m{\ell}$ 배양용기의 뚜껑종류 및 크기에 따른 배양중 이산화탄소농도변화, 수분함량, 유리당, 및 수량을 조사하였다. 이산화탄소 농도는 기존의 비통기성 무스폰지 뚜껑구가, 상하 천공과 하 천공 모두에서 천공 구멍이 작을수록, 또한 상대비교는 하 천공구에서 높게 유지되었다. 배양중 이산화탄소 변화는 배양 뚜껑 종류와는 관계없이 배양 경과 6~11일경 이산화탄소발생량이 최고치에 도달하였다. 뚜껑종류별 느타리버섯($1100m{\ell}$병)의 수량은 상하 뚜껑 천공의 19~38mm천공에서 11.4~23.8%로 증수되어 폭넓게 수량이 좋았으며, 하 천공에서 26~47mm천공 뚜껑에서 대조구에 비하여 6.5~17.9%가 수량이 증수되었다. 또한 빠른 균사배양속도를 고려하면 상하 33mm천공 뚜껑에서 기존 무스폰지 뚜껑보다도 23.8%의 높은 수량으로 제일 양호하였다.

느타리버섯 재배를 위한 적정 LED혼합광 및 광량 (The suitable mixed LED and light intensity for cultivation of oyster mushroom)

  • LED혼합광에 따른 자실체의 생육특성을 조사한 결과 갓직경은 녹색광+적색광을 제외한 모든 처리구가 형광등 처리구와 대등하였으며, 갓직경/대길이는 청색광+백색광에서 가장 컸다. 대의 길이는 무광과 녹색광+적색광에서 가장 길었고, 수량은 녹색광+적색광을 제외한 혼합광에서 대조구와 대등하였으며, 상품수량은 청색광+백색광에서 가장 많았다. 또한 버섯류의 대표적인 항산화물질인 ergothioneine은 청색광+백색광에서 다른 처리구에 비해 가장 높았다. 광량에 따른 생육특성 중 갓직경은 광량이 작을수록 작아지는 경향이었고, 대길이는 $1{\mu}mol/m^2/s$에서 가장 길었으며, 갓직경/대길이는 $1{\mu}mol/m^2/s$에서 0.34로 다른 처리구에 비해 가장 작았다. 수량은 모든 처리구에서 대등하였으나 광량이 작을수록 유효경수와 상품수량이 많았다.

떫은감 진피 복합추출물의 급성 역류성 식도염 개선 효과 (Improving Effects on Rats with Reflux Esophagitis Treated with Combined Extract of Young persimmon fruit and Citrus peel)

  • Objectives : The present study was conducted to evaluate protective effects of Combined Extract of young persimmon fruit and citrus peel (PCM) in Reflux Esophagitis(RE) rats.Methods : Twenty-four Sprague-Dawley (SD) rats were divided four groups and each group had six rats ; Normal group, RE control group, RE group treated PCM 50 ,100 mg/kg body weight group. Reflux esophagitis was induced that tied the pylorus and fundus in SD rats stomach. PCM was administered at 50, 100 mg/kg body weight 2 hrs prior to induction of RE. After 6 hrs, the effects of PCM treated rats were compared with those of normal and control rats. We have performed an analysis such as pH of stomach secretion, oxidative stress biomarkers in serum, and western blot.Results : The increased esophageal mucosa damage by RE was markedly improved by PCM treatment in a dose-dependent manner. Also, the administration of PCM decreased the elevated serum reactive oxygen species (ROS) and peroxynitrite (ONOO-) in serum. The protein expressions of anti oxidant such as SOD, catalase, GPx exhibited down-regulation by PCM treatment in tissues. And, PCM effectively reduce inflammatory cytokines such as inflammation-related proteins cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), Tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) in RE rats. In addition, NFκB and p-IκBɑ were decreased in PCM-adiministrated RE rats. But there was no difference on stomach secretion pH between reflux esophagitis rats and PCM administration rat group.Conclusions : In conclusion, administration of PCM (50, 100 mg/kg body weight) made esophagus have less inflammation and injury by decreased NFκB path way. These findings suggest that PCM could have Improving effects on reflux esophagitis.
