Characteristics of virus-like particles in color mutants of oyster mushrooms (원형느타리백색변이체에 존재하는 바이러스의 특성)

  • This study was carried out to characterize virus-like particles (VLPs) in Pleurotus ostreatus. The white and the dark gray mutants frequently observed in mushroom farms of Pleurotus ostreatus (Wonhyeong-neutari). A 5.8kb segments of dsRNA was detected only in the white mutants but not in the dark gray mutants. The VLPs were purified from the fruit bodies by Polyethylene Glycol (PEG) and ultracentrifugation. Electron microscopy analysis showed that VLPs were isometric about 14, 20~45nm in diameter. Further study is needed to reveal the morphological and yield variations of mushroom strains including VLPs observed in the mushroom farms. Also it is needed to maintain fundamental research for taxonomy, diagnosis, and physiology of VLPs in the mushroom strains.

Study on ICT convergence in Lentinula edodes (Shiitake) cultivation system using Automated container (컨테이너형 수출용 버섯식물공장시스템설계 및 표고버섯 생산 연구)

  • In the 21st century, information and communication technology (ICT) worldwide presents a new vision for agriculture. Time and place, as well as the high-tech industry, to overcome barriers to the fusion of the so-called "smart agriculture," are changing the agricultural landscape. Core container production in precision agriculture for mushroom cultivation, optimal temperature, humidity, irradiation, self-regulation of factors such as carbon dioxide, and environment for mushroom cultivation were adopted. Lentinula edodes (shiitake) is an edible mushroom native to East Asia, cultivated and consumed in many Asian countries. It is considered to be medicinal in certain practices of traditional medicine. We used different controlled light sources (Blue-Red-White-combined LED, blue LED, red LED, and fluorescent light) with different LED radiation intensities (1.5, 10.5, and $20.5{\mu}mol/m^2s$ for LEDs) to compare growth and development. Mushrooms were treated with light in a 12-hour-on/12-hour-off cycle, and maintained in a controlled room at $19{\sim}21^{\circ}C$, with 80~90% humidity, and an atmospheric $CO_2$ concentration of 1,000 ppm for 30 days. Growth and development differed with the LED source color and LED radiation intensity. Growth and development were the highest at $10.5{\mu}mol/m^2s$ of blue LED light. After harvesting the fruit bodies, we measured their weight and length, thickness of pileus and stipe, chromaticity, and hardness. The $10.5{\mu}mol/m^2s$ blue-LED-irradiated group showed the best harvest results with an average individual weight of 39.82 g and length of 64.03 mm, pileus thickness of 30.85 mm and pileus length of 43.22 mm, and stipe thickness of 16.96 mm with fine chromaticity and hardness. These results showed that blue LED light at $10.5{\mu}mol/m^2s$ s exerted the best effect on the growth and development of L. edodes (shiitake) mushroom in the ICT-system container-type environment.

Interspecific Hybridization between Pleurotus cornucopiae and Pleurotus florida Following Protoplast Fusion (원형질체(原形質體) 융합(融合)에 의한 노랑느타리버섯과 사철느타리버섯의 종간(種間) 교잡(交雜))

  • Interspecific fusion products were obtained by protoplast fusion induced by polyethylene glycol, from auxotrophic mutants, Pleurotus cornucopiae and P. florida. The fusants were classified into allodiploidy, stable heterokaryon, and spontaneously segregated heterokaryons. Fruiting body of the clamped or clampless fusants was produced by light-dark cycle on the sawdust medium in glass bottles. Most of these clampless fusants produced mature basidiocarps. The pilei showed various mixed colors resembling the parents. All fruit bodies presented clamp connections except two fusants. When small tissues of stipe from basidiocarps were cultured on a complete medium, mycelial colonies grew more vigorously than that of the original clampless fusants. Five fusants in three crosses were analysed with the distribution of progenies and segregation of genetic characters by random spore analyses. The genetic markers were shown to segregate and recombine in the first generation of monospores isolated from basidiocarps. The analysis indicated the heter-okar-yosis and strong evidence for haploidy of vegetative nuclei, a sexual cycle consisting of nuclear fusion and meiosis. Genotypes of a large number of auxotiophic progenies were not detected. The aberration ratio of segregants indicated the gene interaction resulting from different genome structure between vegetatively incompatible species.

Tolerance of Agaricus bisporus to Bacterial Brown Blotch by Pseudomonads (세균성갈색무늬병에 대한 양송이 내성 검증)

  • Pseudomonads cause bacterial brown blotch disease, which causes great damage to the common mushroom Agaricus bisporus. The tolerance of A. bisporus to pseudomonads was tested and found to not be correlated with mycelium growth ability. The offsprings of the tolerant strain (ASI1085) to pseudomonads were not as tolerant as their parents in the mycelium stage. But, tolerance decreased compared to mycelium in the fruiting body. The offsprings of the weakly tolerant strain (ASI1321) were even more weak in the mycelium stage. It is presumed that the tolerance of the parents is transferred to later generations. The tolerance in the mycelium was not correlated in the fruiting body. Therefore, the browning of the fruiting body is thought to be induced by other factors. Pseudomonas tolaasii caused higher browning than Pseudomonas agarici. Pseudomonas reactans did not have a significant effect on the mycelium, but affected the browning of the fruit bodies. P. agarici had higher ability to inhibit mycelium growth than fruiting body growth.

Ecological and Physiological Characteristics of Armillaria solidipes and A. gallica in Korea (잣뽕나무버섯과 곤봉뽕나무버섯의 생태학적 및 생리학적 특성)

  • Armillaria is a white-rot fungus that decomposes organics; it is also a root pathogen that kills trees. Recently, since the forest tending practice, the incidence of Armillaria infections on tree stumps has been increasing. Herein, we have examined the morphological features of Armillaria solidipes and A. gallica collected from 10 areas in Korea, and investigated the activities of the decomposing enzymes cellulase and laccase of these two Armillaria species. A. solidipes was found in eight host tree species including Pinus densiflora, Quercus serrata, and Castanea crenata at the altitude of 107~1,396 m, while A. gallica was found in three host species including P. densiflora and Fraxinus rhynchophylla at the altitude of 119~556 m. The fruiting bodies of A. solidipes are close with lamellae, and have brown to black hairs on the stipe, while those of A. gallica are distant with lamellae and clavate-shaped, and have yellow hairs on the base. The activites of cellulase of the two species were not different, but the activity of A. solidipes laccase was higher ($12.94{\pm}0.4U/mL$) than that of A. gallica laccase ($9.14{\pm}0.3U/mL$).

  • Limonoids are abundant as bitter taste in citrus fruit and other plants. Interestingly. limonoid UDP-glucosyltransferase (LUGT) effectively ameliorates the bitterness from limonoid. The high level of LUGT expression in Escherichia coli can result in the formation of insoluble aggregates known as inclusion bodies. We isolated the soluble LUGT protein when this inclusion body was renaturated with ${\beta}$-cyclidextrin treatment after protein denaturation by urea. Our present results suggest that the isolation of LUGT from inclusion body in cells leads to shed light to characterize the enzyme for food industry purposes.

Distribution of Ectomycorrhizal Fruit Bodies According to Forest Fire Area (산불발생에 따른 외생균근성 버섯의 분포)

  • This study was conducted to investigate the diversity of ectomycorrhizal fungi by surveying sites from June 2010 to October 2011. The obtained results from investigation were as follows. The total of 2 Kingdom 3 Phylum 6 classes 15 orders 34 families 59 genera and 107 species including saprophytic and ectomycorrhizal fungi was investigated. A total of 10 families 17 genera 49 species (801 ea.) of ectomycorrhizal mushroom was investigated. The mushrooms are classified into 28 families 51 genera and 99 species in Basidiomycota, 5 families 7 genera and 7 species in Ascomycota and 1 families 1 genera and 1 species in Amoebozoa. Dorminant species were Amanitaceae (14 species) followed by Russulaceae (12 species) and Boletaceae (11 species). The populaion ectomycorrhizal mushroom was highest in sites 1 and 2, and sites 4 and 5 occurrence rarely. The mushroom occurrence of ectomycorrhizal fungi was closely related to climatic conditions such as high air temperature and lots of rainfall from July to August. The environment factors which have a favorable influence of mushroom occurrence were soil pH, organic matter content of soil and air temperature of climatic environment.

Isolation of Anticarcinogenic Isoflavone-conjugated Glycoproteins from a Submerged Liquid Culture of Agaricus blazei Mycelia by the Autolysis Process (신령버섯균사체 액체배양물의 자가분해에 의한 항암성 isoflavone-conjugated glycoprotein 분리)

  • Most beta-glucans obtained from various fruit bodies of mushrooms and mushroom mycelial cultures have high-molecular weight glycoproteins, conjugated with beta-glucans. We report that isoflavone-conjugated glycolproteins (designated as gluvone) were isolated and exhibited stronger anticarcinogenic activities. Agaricus blazei mycelia (ABM) was cultured in a liquid medium containing soybean flakes for 14 days. The liquid culture was autolyzed by incubating at $53^{\circ}C$ (pH 5.5) for 3 h. A crude glycoprotein (CGP) fraction with a cytotoxic effect on a mouse ascite cancer cell line (S-180) and a human breast cancer cell line (MCF-7) was isolated from the autolyzed ABM cultures by 80% ethanol treatment. Gluvone was isolated from the CGP with Sephadex G-75 column chromatography. It exhibited a stronger anticancer effect than CGP against the S-180 cell-induced female ICR mouse ascites carcinogenesis. Gluvone with 9,400 daltons was identified as a glycoprotein conjugated with isoflavone. According to HPLC and GC analysis, in conjunction with $^1H$-NMR spectral analysis, it contained 60% carbohydrates (glucose, fructose, and ribose), 31% protein, and 2% isoflavone (daidzein and genistein), which is a novel material. These results indicate that a strong anticarcinogenic gluvone was isolated from the autolyzed product of a submerged liquid culture of ABM, suggesting that autolysis could be a useful tool to produce antitumor agents.

Stain improvement in the white button mushroom 'Seolgang' and its varietal characteristics in Agaricus bisporus

  • The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, 'Seolgang' was developed by crossing two monokaryons 'CM020913-27' and 'SSU423-31'. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of 'Seolgang' on CDA was better at $20^{\circ}C$ and $25^{\circ}C$ when it was compared with that of '505 Ho'. The mature cap shape of new strain 'Seolgang' is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain '505 Ho', the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of '505 Ho'. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of '505 Ho'. Genetic analysis of the new strain 'Seolgang' showed different profiles compared to '505 Ho', CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.

Interspecific protoplast Fusion and Sexuality in Pleurotus (느타리의 종간(種間) 원형질체(原形質體) 융합(融合) 및 유성(有性))

  • Interspecific heterokaryons were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of P. ostreatus+P. cornucopiae, P. ostreatus+P. florida, P. ostreatus+P. sajor-caju, P. florida+P. cornucopiae, P. florida+P. sajor-caju and P. sajor-caju+P. cornucopiae protoplasts. The Fusion products of protoplasts were induced by MCM+benomyl, but segregation of sectors were not identified. Interspecific fusion products of protoplasts between incompatible strains did not form true clamp connections and did not produce fruit bodies. For induction of fertility, interspecific heterokaryons crossed with their parents by hyphal anastomosis.
