Occurrence Pattern of an Unidentified Moth Captured by Sex Pheromone Trap of the Oriental Fruit Moth, Grapholita molesta, and Its Discriminating Molecular Markers (복숭아순나방(Grapholita molesta) 성페로몬 트랩에 포획된 미동정 나방의 발생패턴과 판별 분자지표)

  • An unidentified moth was captured in sex pheromone traps of the oriental fruit moth, Grapholita molesta, especially at spring season in apple orchards and their vicinity. Though the captured males were similar in appearance to G. molesta males, they were easily distinguished by a matted difference in body size. Their occurrence pattern was also similar to that of overwintering G. molesta population from April to May, at which more males were captured in the pheromone traps installed in the vicinity of apple orchards than within apple orchards. After May, they were no longer captured in the pheromone traps. To investigate any larval damage due to this unidentified moth, molecular markers needed to be developed. Four PCR-RFLP markers originated from cytochrome b region of mitochondrial DNA could distinguish this unidentified moth from G. molesta.

Effect of Particle Pre-Treatment on Properties of Jatropha Fruit Hulls Particleboard

  • The objective of the research was to evaluate the effect of particle pre-treatment on physical, mechanical, and durability of jatropha fruit hulls (JFH) particleboard. The pre-treatments included were immersing in cold water, hot water, and acetic acid solution. After each treatment, the particles were dried up to 3% moisture content. Urea-formaldehyde (UF) resin was used to fabricate particleboards with board size, thickness and density target of 25 cm by 25 cm, 0.80 cm, and $0.70g/cm^3$, respectively. Board pressed at $130^{\circ}C$ for 10 minutes, and $25kg/cm^2$ pressure. The evaluation of particleboard followed the JIS A 5908-2003. Whilist their resistance to subterranean termite test (mass loss, mortality, antifeedant value and feeding rate) refers to the Indonesian standard (SNI 01.7207-2006). The physical and mechanical properties of particleboards showed that all pre-treatments decreased the pH of particles. Overall, all particle immersing treatments resulted of better physical and mechanical properties of particleboard than those of untreated ones. The acetic acid treatment resulted the best physical and mechanical properties of particleboard. Based on the mass loss of JFH particleboard, hot water and acetic acid treated particleboards were classified into weak resistance to subterranean attack. The other two treatments were classified into very weak resistance. Hot water treated particleboard provided the highest mortality and antifeedant as much as 87.40% and 34.20%, respectively. Based on antifeedant classification, hot water treated particleboards were classified into moderately strong resistance, while other treatments were categorized into weak resistance. The lowest feeding rate value ($45.30{\mu}g/termite/day$) was attained by hot water treatment.

Cudrania tricuspidata Root Extract as Whitening and Antiwrinkle Cosmetic Agent (꾸지뽕나무 뿌리 열수추출물로부터 미백 및 주름개선 화장품 소재의 개발)

  • Water extracts of root, leaf, branch and fruit of Cudrania tricuspidata were tested to use as natural antiwrinkle and whitening cosmetic ingredient. Root extract indicated 86.5 mg/g polyphenol concentration and 55.5 mg/g flavonoid concentration. From MTT test, root, branch and fruit extract did not show cell toxicity up to $1,000{\mu}g/mL$, however, leaf extract displayed significant cell toxicity in concentrations higher than $200{\mu}g/mL$. Cudrania tricuspidata root extracts displayed excellent 81% tyrosinase inhibition (whitening effect), 58% elastase inhibition (antiwrinkle effect) at $500{\mu}g/mL$ and showed minor UVB absorbance. When solution and lotion formulation were made including Cudrania tricuspidata root extract, they are stable at $25^{\circ}C$ for 21 days, but showed significant deviation in viscosity and particle size at $47^{\circ}C$. From the study, Cudrania tricuspidata root extract demonstrated good potential for natural whitening and antiwrinkle cosmetic agent.

Rapid and Specific Detection of Acidovorax avenae subsp. citrulli Using SYBR Green-Based Real-Time PCR Amplification of the YD-Repeat Protein Gene

  • The aim of this study was to develop a SYBR Green-based real-time PCR assay for the rapid, specific, and sensitive detection of Acidovorax avenae subsp. citrulli, which causes bacterial fruit blotch (BFB), a serious disease of cucurbit plants. The molecular and serological methods currently available for the detection of this pathogen are insufficiently sensitive and specific. Thus, a novel SYBR Green-based real-time PCR assay targeting the YD-repeat protein gene of A. avenae subsp. citrulli was developed. The specificity of the primer set was evaluated using DNA purified from 6 isolates of A. avenae subsp. citrulli, 7 other Acidovorax species, and 22 of non-targeted strains, including pathogens and non-pathogens. The AC158F/R primer set amplified a single band of the expected size from genomic DNA obtained from the A. avenae subsp. citrulli strains but not from the genomic DNA of other Acidovorax species, including that of other bacterial genera. Using this assay, it was possible to detect at least one genomeequivalents of the cloned amplified target DNA using 5 × 100 fg/µl of purified genomic DNA per reaction or using a calibrated cell suspension, with 6.5 colony-forming units per reaction being employed. In addition, this assay is a highly sensitive and reliable method for identifying and quantifying the target pathogen in infected samples that does not require DNA extraction. Therefore, we suggest that this approach is suitable for the rapid and efficient diagnosis of A. avenae subsp. citrulli contaminations of seed lots and plants.

Estimation of Consumers Valuation by Attributes of Green Jujube Chip (청대추칩의 속성별 소비자 가치 추정)

  • This research was launched as an attempt to help create a new value-added product called "Green Jujube Chip." To accomplish the objectives, consumers value by attributes of Green Jujube Chip should be estimated. We used Choice Experiment for analysis, which we used to arrive at pricing of specialty jujube chips based on the conditional logit model. As a result, MWTP were estimated at 1333 Won/unit for Mid-size Company Product, 1752 Won/unit for Large Company Product, 1703 Won/unit for University Affiliation, 350 Won/unit for packing unit, 548 Won/unit for rating the raw fruit, 1,322 Won/unit for HACCP, and 1,166 Won/unit for ISO22000. The consumer's WTP for Green Juju Chip, associated with the university-brand recognition, was higher than a product produced by a mid-sized company but lower than a product by a large company. Given how the consumers regard the university-brand product as a specialty item, it is necessary to create value-added products that meet such high expectation. Therefore, we recommend the producers to acquire HACCP Food Safety Certification and the value-added product has used the highest quality raw fruit.

Effect of Plastic Bagging Cultivation of Summer Squash (Cucurbita moschata) on Improvement of Quality and Extension of Storage Period (플라스틱 봉지재배가 애호박 품질 향상과 저장 연장에 미치는 효과)

  • In recent years, the bagging cultivation of summer squash (Cucurbita moschata) is applied for surface protection during harvest and extension of storage period. In this study, the effect of plastic bagging cultivation of summer squash on improvement of quality and extension of storage period was investigated. The yield and the length of fruit didn't have no difference between bagging and control treatment but the bagging treatment significantly made more regular size and light weight of fruits, and increased the number of fruits because of the limitation of their growth. The bagging treatment decreased weight loss and fruit softening during the their storage. Therefore it showed effective means for quality improvement and storage extension of summer squash.

  • In this study, two commercial mating disruptors were compared in terms of disruption of Oriental fruit moth, Grapholita molesta, male orientation, in which a new dispenser type, $SPLAT^{(R)}$, was compared with a current dispenser type, $Isomate^{(R)}$-M ROSSO. For this assessment, the last three year field monitoring data were analyzed for the efficacy of Isomate type dispenser. Then two commercial dispensers were compared in different localities with sub-locality replications during identical monitoring period from mid February to late August. There appeared to be four adult population peaks, in which the overwintering population size was positively correlated with the following reproductive population sizes. Isomate type dispenser effectively suppressed G. molesta populations during all growing seasons with some annual variation in its efficacy. Between two dispensers, SPLAT type was much effective. The difference in their efficacy may be caused by the difference in composition of major pheromone component.

Cold Hardiness in Relation to Vine Management in 'Campbell Early' Grapevines ('캠벨얼리' 포도의 수체관리와 내한성)

  • The experiment was conducted to investigate the effects of vine management and yield on fruit quality and bud burst of 'Campbell Early' grapevines in Hwaseong and Suwon area. The extreme value of minimum temperature of Hwaseong area, frequently causing cold damage, was lower than that of Suwon area in winter. Narrow spacing, overbearing, and poor vine management were observed in Hwaseong area, where the harvest time was delayed by 7 days compared with that of Suwon area. Total carbohydrate content of bearing mother branches was lower with 1.2~1.7%, and the percentage of bud burst was extremely lower at $-15^{\circ}C$. In Suwon area, there were no differences in cluster and berry size, but soluble solids content was lower and skin coloration was poorer when yield was high. Total carbohydrate content was lower when yield was high. The percentage of bud burst was lower at cold treatments below $-20^{\circ}C$. The results suggest that appropriate yield based on fruit quality is 2.3~2.6 MT/10a.

Antioxidantive Effectiveness of Trichosanthes kirilowii Maximowicz Extracts (하늘타리(Trichosantes kirilowii Maximowicz)추출물의 항산화 효과)

  • Recently, there is a growing interest about unsaturated lecithin having excellent characteristics of skin affinity and absorbency. Accordingly, this study intended to develop a natural sulfuration material in order to enhance the stability of oxidation of unsaturated lecithin and substitute existing sulfuration materials which indicate variability and toxicity. As sulfuration components, plenol acid family, 3,5-dihydroxybenzoic acid, and flavanone were analyzed. Total polyphenol content was higher in the root extracts (133.85 mg/g) than in the fruit extracts (53.5 mg/g). Above 100 ppm polyphenol content, the free radical removal efficiency and lipid oxidation prevention of the root extracts were 20.1 and 19.2% superior compared with BHT respectively. Also, the extracts indicated high survival rate of more than 95% below 1250 ppm, showing the stability. For the stability of liposome made from an unsaturated lecithin, the root extracts were superior to the fruit extracts. Especially, 15.1 and 13.9% of sulfuration effect and zeta potential were improved with 9.3% reduced particle size compared with BHT as the control group, respectively.

The Status of Birds Consuming Fruits and Seeds of the Tree and Related Tree Species on Jeju Island, the Republic of Korea (제주도에서 나무의 열매와 종자를 섭식하는 조류와 관련 수종 현황)

  • Birds play a main role in the formation and change of forest structures as they are seed-dispersal agents. This study aims to identify birds consuming fruits and seeds of the tree and their associated fruits and seeds on Jeju Island in the context on the forest restoration in Korea. We conducted field surveys twice a month from 2013 to 2015 at nine study sites located across Jeju Island and collected available photographic and observation records. A total of 50 species of birds consuming fruits and seeds of the tree were identified and birds belonging to Bombycillidae, Pycnonotidae, Zosteropidae, Sturnidae and Fringillidae were confirmed as major birds consuming fruits and seeds of the tree. Gulping was the dominant type of feeding as well as crushing, but relationship between the size of a bird and the number of fruit and seed species used by the bird was not significant. We also documented that 118 fruit and seed species were consumed by birds and that shrubby fruits and seeds were more consumed by birds than those of other plant types. The relative consumption rate of fruits and seeds ranged from 0.02 to 0.44, but five species were the most important fruits and seeds for birds. Our finding suggest that avian frugivorous gulpers will benefit the seed dispersal, especially of five fruiting plants, providing useful baseline data for forest restoration and urban park design.
