Investigation of Nanofiber and Thermosensitive Scaffold for Intervertebral Disc through Organ Culture (기관배양을 통한 추간판 재생용 나노파이버 및 온도 감응성 지지체에 대한 검증)

  • The purpose of this study is to investigate the potential of a novel tissue engineering approach to regenerate intervertebral disc. In this study, thermosensitive scaffold (chitosan-Pluronic hydrogel) and nanofiber were used to replace the nucleus pulposus (NP) and annulus fibrosus of a degenerated intervertebral disc, leading to an eventual regeneration of the disc using the minimally invasive surgical procedure and organ culture. In preliminary study, disc cells were seeded into the scaffolds and cellular responses were assessed by MTT assay and scanning electron microscopy (SEM). Based on these results, we could know that tissue engineered scaffolds might provide favorable environments for the regeneration of tissues. Organ culture was performed in fresh porcine spinal motion segments with endplates on both sides. These spinal motion segments were classified into three groups: control (Intact), injured NP (Defect), and inserting tissue engineered scaffolds (Insert). The specimens were cultivated for 7 days, subsequently structural stability, cell proliferation and morphological changes were evaluated by the relaxation time, quantity of DNA, GAG and histological examination. In these results, inserting group showed higher relaxation time, reduced decrement of DNA contents, and accumulated GAG amount. Consequently, the tissue engineered scaffolds used in this study seen to be a promising base scaffolds for regenerative intervertebral disc due to its capacity to absorb external dynamic loading and the possible ideal environment provided for disc cell growing.

A study on carbonization on the surface of steel sheet during the BAF annealing process (BAF 소둔공정에서 탄화 현상에 관한 연구)

  • In steel industries, cold-rolled sheet manufacturing is one of the links between the front and rear important industries for national competitiveness and high value-added production. In particular, in small and medium-sized enterprises, one of the major problems is the carbonization phenomenon of the steel sheet during the annealing process. Carbonization occurring on the surface of the coil help reduce steel production. After conducting various experiments to identify the cause of carbonization on the surface of a cold-rolled steel, the following results were obtained: (1) An analysis of the rolling oil, which is used in the field, revealed it to contain approximately 40 ppm carbonized material. (2) A comparison of the thermal denaturation characteristics of the fresh rolling oil and using rolling oil by FT-IR analysis showed that thermal denaturation had occurred, as shown by the significant decrease in the relative intensity of the 2900 and $1750cm^{-1}$ peaks. (3) The thermal decomposition of the rolling oil took place for the rolling oil at approximately $220^{\circ}C$. Furthermore, annealing experiments at $200^{\circ}C$ showed that the carbonization phenomenon of the sample was not observed. On the other hand, carbonization was observed at temperatures higher than $240^{\circ}C$.

A Study for Use of Wild Rhododendron mucronulatum for. albiflorum as Landscape plant (야생 흰진달래의 조경식생화를 위한 연구)

  • Rhododendron mucronulatum for. albiflorum, native species is a shrub that has white flowers on May to June, and rare species endangered by people's rash digging or cutting. But its physiological ecological characteristics and propagation method are not being known at all. Therefore, this study was executed to utilize this species as the planting material for landscaping by analysing its habitat environment and growth form, and also experimenting its seed and vegetative propagation, and it field culture and utilization. The results are as follows; 1. The elevation, gradient and direction of this species were 295-1,350m, 10-36$^{\circ}$, northwest respectively. It was found that the species is shade-liking plant that grows under forest cover of average 51.33%. 2. The soil pH and water content of its habitat were 5.4, 25.41% respectively. The organic matter content was 6.29% that was higher than 3.2%, the average organic matter content of forest soil in Korea. 3. Representative plant community within which this species was living was Quercus mongolica community, and its main neighboring species were Lindea obtusiloba, Fraxinus sieboldiana, Rhus trichocarpa, Rhododendron Schlippenbachii, Rododendron mucronulatum. 4. The leaf length and width of this species were 39.18mm, 12.60mm respectively. This result showed that generally its leaf size was larger than that of R. micranthum, R. yedoense var. poukhanense and R. mucronulatum var. ciliatum and smaller than that of R. mucronulatum and R. schlippenbachii. 5. The whole size of its pollen was, as 59${\times}$61$\mu\textrm{m}$, the largest of plants of Rhododendron family including R. mucronulatum and R. mucronulatum var. ciliatum. 6. The result of seed germination experiment at intervals of 5$^{\circ}C$ from 15$^{\circ}C$ to 30$^{\circ}C$ presented the highest germination rate of 94.7% at 20$^{\circ}C$ numerically, but high percent germination at all temperature levels without significant difference. And the seed of this plant proved to be sun-liking seed at requiring dormancy in germination. 7. Through seed germination experiment by treatment of growth regulators such as GA. Thiourea and Kinetin under dark condition, it was found that the effect of GA treatment on germination increase and acceleration was the highest. 8. In greenwood cutting, rooted rate by treatment of various concentration of IBA and NAA on clay and vermiculite bed was not wholly high, but 100ppm plots of both IBA and NAA of clay bed showed relatively good rooted rate. 9. As result of field culture experiment for finding out optimum growth temperature and light intensity, growth conditions such as height, number of leaves, fresh weight and chlorophyll contents were the best at night/day temperature of 20/25$^{\circ}C$ and under 1/2sun. Also, the photosynthetic rate was the highest at 25$^{\circ}C$. Accordingly, it was found that optimum temperature and light intensity for growth of this plant are 25$^{\circ}C$ (day temperature), 50% of natural light respectively.

Characterizing a full spectrum of physico-chemical properties of (20S)-and (20R)-ginsenoside Rg3 to be proposed as standard reference materials

  • The authentication of the physico-chemical properties of ginsenosides reference materials as well as qualitative and quantitative batch analytical data based on validated analytical procedures is a prerequisite for certifying good manufacturing practice (GMP). Ginsenoside Rb1 and Rg1, representing protopanaxadiol and protopanaxatriol ginsenosides, respectively, are accepted as marker substances in quality control standards worldwide. However, the current analytical methods for these two compounds recommended by Korean, Chinese, European, and Japanese pharmacopoeia do not apply to red ginseng preparations, particularly the extract, because of the relatively low content of the two agents in red ginseng compared to white ginseng. In manufacturing fresh ginseng into red ginseng products, ginseng roots are exposed to a high temperature for many hours, and the naturally occurring ginsenoside Rb1 and Rg1 are converted to artifact ginsenosides such as Rg3, Rg5, Rh1, and Rh2 during the heating process. The analysis of ginsenosides in commercially available ginseng products in Korea led us to propose the inclusion of the (20S)- and (20R)-ginsenoside Rg3, including ginsenoside Rb1 and Rg1, as additional reference materials for ginseng preparations. (20S)- and (20R)-ginsenoside Rg3 were isolated by Diaion HP-20 adsorption chromatography, silica gel flash chromatography, recrystallization, and preparative HPLC. HPLC fractions corresponding to those two ginsenosides were recrystallized in appropriate solvents for the analysis of physico-chemical properties. Documentation of those isolated ginsenosides was achieved according to the method proposed by Gaedcke and Steinhoff. The ginsenosides were subjected to analyses of their general characteristics, identification, purity, content quantification, and mass balance tests. The isolated ginsenosides showed 100% purity when determined by the three HPLC systems. Also, the water content was found to be 0.534% for (20S)-Rg3 and 0.920% for (20R)-Rg3, meaning that the net mass balances for (20S)-Rg3 and (20R)-Rg3 were 99.466% and 99.080%, respectively. From these results, we could assess and propose a full spectrum of physico-chemical properties of (20S)- and (20R)-ginsenoside Rg3 as standard reference materials for GMP-based quality control.

Rabbit's Cervical Tracheal Replacement with Cryopreserved Homograft - Effects on the Viability and Rejection - (토끼 경부기관의 초냉동보관 동종이식편 기관 이식술 - 생육성 및 거부반응에 미치는 영향 -)

  • Background: There are no ideal substitutes for tracheal replacement. Therefore we investigated the possibility of clinical use of cryopreserved tracheal homograft with special interest in the viability and rejection of the epithelial cell and cartilage. Material and Method: Rabbit's trachea was sected and stored in liquid nitrogen tank for 1 month. Tracheal replacement was done in 45 rabbits with autograft(n=15, Group 1), fresh allograft(n=15, Group 2) and cryopreserved homograft(n=15, Group 3). After 7, 14, and 30 days, 5 rabbits in each group were sacrificed and the regeneration of epithelium and cartilage and the degree of rejection were assessed by counting the monocellular infiltration. Result: Investigation at day 7, showed no difference in epithelial regeneration, however, at days 14 and 30, Group 1 showed better regeneration of epithelium than groups 2 and 3. There was no difference of epithelial regeneration between group 2 and 3. There was little rejection at day 7, but at days 14 and 30, there was significant rejection in group 2 and group 3.(P<0.05). Group 3 showed lesser rejection than group 2 at days 14 and 30, but it was not statistically significant. Cartilage showed no rejection and maintained its viability in groups 2 and 3. Conclusion: Cryopreserved tracheal homograft can maintain its viability, therefore it may represent a possibility of clinical application for tracheal replacement. However, cryopreservation can not eliminate the antigenicity of the trachea completely. Furthere studies for lowering the antigenicity and rejection should be performed for an ideal substitute for tracheal replacement.

Effect of Environmental Conditions and Chemical Treatments on Seed Germination of Astilbe koreana (Kom.) Nakai (자생 숙은노루오줌의 종자발아에 미치는 환경조건과 화학적 처리의 영향)

  • Factors affecting germination of seeds in the forms of various environment and chemical compounds. The present study was aim to produced effective seed propagation method of Astilbe koreana (Kom.) Nakai which had expected high value for the cut flower, ornamental and pharmaceutical material. Seed width and length ranged 0.62, 2.22 ㎜, respectively, and weight of thousand seeds was 40.5 ㎎. As result of imbibition test, showed moisture content of fresh seed (2.57%) increased rapidly by water-soaking treatment under 24 hours, recording to maximum value of 29.8%, and expansion of the seed coat was observed. Seed germination was the best at 15℃ and light conditions (40.8%) among temperature and light conditions treated. Percent germination of seeds was improved under the low (15, 20℃) than high temperature (25, 30℃). In addition, the seed was not germinated at dark condition regardless of temperature. Seeds of A. koreana thus seemed that it had low temperature germinability conditions. To improve germination rate, seeds were submerged in various concentrations of growth regulators such as GA3 and kinetin, and minerals as KNO3 and KCl. As a results, KNO3 treatment, regardless of concentrations, promoted germination compared to control. Especially, percent of germination (77.8%), germination energy (96.1%), mean germination time (11.3 days) and T50 (6.5 days) were effectively improved by treatment of KNO3 20 mM.

Sodium Chloride Decomposing Method in Food Waste Compost using Triple Salt (삼중염을 이용한 음식물 쓰레기 퇴비의 염분(NaCl) 분해방법)

  • The characteristics of food waste in our country is high contents of water and sodium chloride(NaCl). Average water contents of household garbage was 80.0%, and those of wastes of restaurants and of wholesale market of agricultural products was 76.9% and 90.0%, respectively. The NaCl contents were high in household garbage and restaurant's waste as 3.36% and 4.84%, respectively. The NaCl contents of food waste composts made by various techniques known upto now were under the level of 1% by fresh weight basis. But these techniques has some problem that is environment pollution from treated water and high equipment cost. The application to agricultural land of food waste compost that is not sufficiently removed NaCl was considered to be improper due to salt accumulation in soils and plant growth inhibition by salt stress. The purpose of this study is to decompose NaCl in food waste compost using triple salt and this method is differ from existing chemical method. Also, reaction of NaCl with triple salt produced KCl that is basic material of potassium fertilizer. The experiment results of growing lettuce produced difference between food waste compost and treated food waste compost with triple salt. The latter got more high sprouting ratio and a growth rate.

Component Analysis of Different Parts of Chestnut (밤의 부위별 성분 분석)

  • To obtain basic data utilizing chestnuts as a raw food material, proximate analysis was conducted. Chemical component of chestnut flesh were $63.60\%$ moisture, $118\%$ ash, $3.02\%$ crude protein, $0.615\%$ crude fat, $1.21\%$ crude fiber, and $30.37\%$ nitrogen free extract, respectively. The weight ratio of tegmen, seed coat and flesh of chestnut sample were 17.05, 14.9, and $68.05\%$, respectively. The total amino acid contents of flesh and seed coat were $2,994\;mg\%$ and $1,450\;mg\%$, respectively. The total amount of free amino acids was less than that of total amino acids. As results of mineral analysis, the content of K was higher than that of any other minerals. The contents of maltose and sucrose were higher than those of fructose and glucose. The total polyphenol contents of tegmen, seed coat, fresh, leaf and bark were $9.56\;mg\%$, $0.047\;mg\%$, $0.23\;mg\%$, $15.44\;mg\%$ and $17.85\;mg\%$, respectively.

A Study on the Flavor Constituents of the Citron (Citrus junos) (유자의 향미성분에 관한 연구)

  • To accept basic data of utilizing of citron (Citrus junos) as a raw material of industrial produce, major chemical components of citron were investigated. Weight ration of poet flesh md seed of citron were 44.7, 42.9 and 12.4%, respectively. Comparing proximate composition of peel and flesh of citron, peel showed higher in crude protein crude fat and crude ash than flesh but lower in moisture, carbohydrate and soluble solid. The major free sugars of citron were fructose, glucose and sucrose. Peel contained higher in sucrose than flesh, but lower in fructose and glucose. The content of K md P were 309 and 15.9 mg% in peel and 175 and 22.4 mg% in fresh, respectively. The main organic acids of citron were citrate, malate and oxalate. Total organic acid content of flesh (6.6%) was higher than that of peel (4.6%). Total amino acid content of peel and flesh were 671.9 and 315.7 mg%, respectively. Free amino acid content of peel and flesh were 324.3 and 280.7 mg%, respectively, and the major ones were proline, serine, glutamic acid, aspartic acid, and histidine. Total 49 volatile compounds were detected and 26 of these ones were identified in cion. The major volatile component of citron was limonene, which consists of 80% among the total volatiles in peel by all extract methods.

The Horse Manure Fertilizers and Pollutants Characteristics Analysis Research (말 분뇨의 비료성분 및 오염물질 특성 조사 연구)

  • Horse manure general composition and contaminants, heavy metals content analysis was investigated as follows. The moisture content of horse feces was 76.1% and 96% for horse urine. The average T-N, T-P, T-K concentrations of the racing horse were 0.18%, 0.10%, 0.22% respectively and 2.88%, 0.0015%, 0.84% for urine. And the average BODs, COD, SS concentrations were 26,906 $mg/{\ell}$, 36,642 $mg/{\ell}$, 89,375 $mg/{\ell}$ respectively and 14,298 $mg/{\ell}$, 7,484 $mg/{\ell}$, 6,987 $mg/{\ell}$ for urine. In addition, the Cr, Cu, Ni, Cd, Pb, Zn concentrations of feces were 2.65 mg/kg, 7.05 mg/kg, 1.24 mg/kg, 0.07 mg/kg, 0.01 mg/kg, 45.11 mg/kg respectively and 1.52 mg/kg, 1.62 mg/kg, 0.00 mg/kg, 0.03 mg/kg, 0.03 mg/kg, 4.49 mg/kg for urine on the fresh matter basis. The end of the survey was that the quality of the compost can be used as a raw material, and manure is necessary to constantly driven forward horse manure composting research.
