  • Recently, In Korea region becoming subtropical climate, freezing index values have been increasing in winter. The climate condition in Korean became polarized. Since freezing index is increased recently, track heaving from frost is often occurred, which is ignored in past couple of years. Methods to prevent frost-heaving are circumfusion of calcium chloride and replacement of gravel ballast in usual. The effects of those methods are minimal, incontinuous, and not eco-friendly. Therefore, maintenances are performed repeatedly every year. This study discussed utilizing the waste-tire-chips and insulators in the small structures(bridge and box etc) for prevention of frost-heaving. In order to prove effect of waste-tire-chips, laboratory and field tests were conducted with CBR and measurement of frost-heaving amount to prove and verify effect of insulators.

Effects of Aging Period Prior to Freezing on Meat Quality of Hanwoo Muscle (Longissimus dorsi)

  • This study was conducted to evaluate the effect of the aging period prior to freezing on the meat quality of Hanwoo longissimus dorsi (LD) muscle. Three different combinations of aging and freezing periods (0/90, 20/70, and 40/50) were examined using LD muscle at 24 h postmortem under an identical storage time of 90 d. The pH and lightness slightly increased with increasing aging period. However, there were no significant (p>0.05) differences in redness and yellowness. The solitary freezing treatment (0/90) had the significantly (p<0.05) lowest moisture content. The un-aged treatment had a significantly (p<0.05) higher total loss than the aged treatments due to an increase in thaw drip loss. The aging significantly improved the myofibrillar fragmentation index and shear force of Hanwoo LD muscle (p<0.05). In addition, the aged treatments produced a higher flavor, tenderness, juiciness, and overall acceptability relative to un-aged treatment. However, there was no significant (p>0.05) difference in shear force and sensorial properties between 20 and 40 d aging prior to freezing. Therefore, 20 d aging prior to freezing may be a sufficiently effective strategy to improve the tenderness and sensorial properties of Hanwoo LD muscle.

굴절률을 이용한 수용액의 빙충전율 측정 (Measurement of the Ice Packing Factor of an Aqueous Solution Using the Index of Refraction)

  • In the present paper, a new method is proposed for the measurement of the ice packing factor (IPF) of an ice slurry using the index of refraction. The purpose of the new method is to improve the resolution of the measurement and to increase its resistance to electric noise as compared to the standard IPF measurement technique that relies on measurement of the freezing temperature. These two methods are similar in that they both obtain a concentration of aqueous solution from measured physical quantities and calculate the IPF using a relation between concentration and IPF. We experimented and compared the two methods, whose results were also compared with results from the calorimeter method obtaining the IPF directly They are in good agreement (within $5\%$), which demonstrates the validity of the newly proposed method.

돼지난포란의 동결과 체외수정에 관한 연구 (Freezing and In Vitro Fertilization of Porcine Oocytes)

  • This study was undertaken in an effort to product embryos through in vitro maturation(IVM), in vitro fertilization(IVF) and in vitro culture(IVC) after cryopreservation of immature and mature porcine oocytes. The experiments were conducted to investigate IVM rate of oocytes frozen with 3 different cryoprotectants and to examine IVF and IVC of frozen-thawed oocytes. The CEI(cumulus cells expansion index) after IVM of frozen-thawed immature oocytes was higher in oocytes frozen with PG+PEG(propylene glycol plus polyethylene glycol) than those frozen with single cryoprotectant and this index was almost 90% of unfrozen oocyte's index(2.39 vs. 2.66). The IVF rate of all frozen oocytes was very low(68% of unfrozen oocytes) and the IVF rate of frozen immature oocytes was slightly higher than that of frozen mature oocytes(39.0% vs. 34.4%), but polyspermic penetration was higher in frozen immature oocytes(21.9% vs. 19.1%). The cleavage rate after IVF of frozen-thawed oocytes was 9.3% for frozen mature oocytes and 11.3% for frozen immature oocytes and this rate was significantly lower(P<0.05) than that of control(60.7%). The development to 8-cell stage was greatly lower in frozen mature oocytes than in frozen immature oocytes. The results indicate that the use of PG plus PEG as cryoprotectant may be very effective for vitrification of porcine oocytes and the frozen-thawed immature porcine oocytes can be used fro in vitro embryo production based on IVM, IVF and IVC system.

Impacts of post-mortem ageing prior to freezing on technological and oxidative properties of coarse ground lamb sausage in a model system

  • Objective: The objective of this study was to evaluate the effects of ageing time of lamb loins prior to freezing on technological characteristics and oxidation stability of coarse ground lamb loin sausage using in a model system. Methods: Lamb loins (M. longissimus lumborum, n = 25) were aged at $-1.5^{\circ}C$ for 0, 1, 2, 3, and 8 wk and then frozen for the remaining days (a total of 30 wk). The aged/frozen/thawed lamb loins were ground, and model sausages were formulated with 75% aged/frozen/thawed lamb loin, 25% water, 1.5% sodium chloride (NaCl) and 0.3% sodium tripolyphosphate. The pH and thaw/purge loss of aged/frozen/thawed lamb loins were evaluated, and protein functionality (protein solubility and emulsifying capacity), water-holding capacity and textural properties of model sausages were determined. Cooked model sausages were vacuum-packaged in a plastic bag and displayed under continuous fluorescent natural white light ($3^{\circ}C{\pm}1^{\circ}C$). Colour and lipid oxidation of the cooked model sausages were evaluated on 0 and 21 d of display storage. Results: Ageing prior to freezing had no impact on pH and purge/thaw loss of lamb loins and the colour of cooked sausages (p>0.05) made from the loins. Lamb loins aged for at least 3 wk prior to freezing numerically improved total and myofibrillar protein solubilities (p>0.05) and emulsion activity index (p = 0.009) of meat batter, but decreased cooking loss (p = 0.003) and lipid oxidation (p<0.05) of model sausages. Conclusion: This study suggests that post-mortem ageing of raw meat prior to freezing could improve water-holding capacity and lipid oxidative stability of sausage made from the meat.

인간 정자의 완만.급속 동결보존 방법이 융해 후 정자 운동성 지수와 생존율 및 정자 형태에 미치는 영향 (Effect of Cryopreservation by Slow and Rapid Freezing on the Sperm Motility Index, Viability and Morphology of Post-thaw Human Spermatozoa)

  • 정자의 동결보존을 위한 새로운 기술개발 목적은 동결과정에서 최소한의 손상으로, 응해 후 최대한 높은 활력도의 정자를 얻는 것이다 정자가 난자와 수정하기 위해서는 적당한 생존성과 운동성을 유지해야 하는데, 가장 일반적인 방법으로는 정자의 진진 운동성과 첨체의 정상 여부 및 형태 검사방법 등이 있다 본 연구는 사람 정액을 동결보존 할 때 semi-programmable freezer를 이용한 완만동결 방법과, 액체질소의 vapor를 이용한 급속동결 방법이 응해 후 정자의 운동양상과 생존율 및 형태에 미치는 영향을 알아보기 위해 실시하였다. 동결-응해 후 정자의 MOT, VCL, VSL, VAP는 각각 급속 동결방법에서 47.40$\pm$20.06%, 38.12$\pm$15.58 $\mu$m/s, 28.19$\pm$14.10 $\mu$m/s, 33.64$\pm$15.15 $\mu$m/s로 완만 동결방법인 43.39$\pm$18.79%, 33.91$\pm$13.50 $\mu$m/s, 19.98$\pm$10.88 $\mu$m/s, 24.60$\pm$11.72 $\mu$m/s보다 유의적으로 높았으나(p<0.05), LIN은 완만동결 방법이 34.64$\pm$11.36으로 급속동결 방법인 28.83$\pm$10.35보다 더 좋은 성적을 나타내었다(p<0.05). 고 활력정자를 나타내는 HYP 역시 급속동결 방법에서 2.77$\pm$2.71%로 완만동결 방법의 1.33$\pm$1.57%보다 유의적으로 높게 나타났다(p<0.05). 그러나 정자의 운동양태를 나타내는 MAD, WOB, DNC, DNM에 있어서는 완만동결 방법이 급속동결 방법에 비해 조금 더 좋은 성적을 보였으나 유의적인 차이는 없었다. 동결-응해 후 정자의 생존율 및 정상형태의 정자는 완만동결 방법이 각각 62$\pm$2.1%, 44$\pm$8.3%, 급속동결 방법은 60$\pm$2.2%, 46$\pm$7.7%로 유의적인 차이가 없이 비슷한 결과를 보였다. 따라서 사람 정액의 동결 보존 시 많은 시간과 고가의 장비가 필요한 완만동결 방법보다는 짧을시간동안 액체 질소만으로 간단히 시행할 수 있는 급속동결 방법이 더 효과적이라고 사료된다.

제설제 사용으로 인한 노면 미끄럼저항 특성 연구 (A Study of Skid Resistance Characteristics by Deicing Chemicals)

  • 미끄럼 저항은 타이어와 도로노면의 마찰저항을 나타내는 지수로 차량의 주행 안전성에 지대한 영향을 미친다. 타이어의 상태, 노면의 마모도, 주행속도, 노면의 건조, 습윤, 결빙 등에 따라 미끄럼저항의 크기는 달라진다. 특히 겨울철 도로노면의 경우 강설 후 기온강하에 도로포장표면이 결빙이 발생할 경우 미끄럼저항이 급격히 저하되며, 미끄럼저항을 회복하고자 제설 작업을 수행하게 된다. 제설작업 후에는 습윤상태로 노면조건이 바뀌게 되는데, 제설제가 노면에 잔류하게 되면 일반적인 습윤상태에서의 미끄럼저항을 확보치 못할 수 있다. 본 연구에서는 국내 포장도로의 대다수를 차지하는 밀입도 아스팔트와 타이닝 콘크리트 포장노면에 대하여 실험실 조건에서 노면 결빙시, 제설중 제설후 잔류물이 노면에 있을 경우의 조건에서의 미끄럼저항을 정량적으로 평가하였다. 연구 결과 노면 결빙시 미끄럼저항 회복시간은 수용액 살포방법이 고체염 살포방법 보다 빠르며 제설제 중에는 소금이 가장 빠르게 나타났다. 노면에 잔류한 제설제는 콘크리트 시편이 아스팔트 시편보다 미끄럼저항 값이 높게 나타났으며 습윤 노면 상태와 비교시 미끄럼저항 확보율은 54~80% 이다.

양태, Platycephalus indicus의 정자의 냉동보존 (Practical Procedure of Sperm Cryopreservation of the Bar-tailed Flathead Platycephalus indicus)

  • 양태, Platycephalus indicus 정자의 동결보존을 위해 적정 동해방지제와 동결높이에 따른 예비동결(0, 2, 4, 6cm), 그리고 해동온도에 따른 정자의 운동성과 생존율로 비교하였다. 적정 동해방지제로 dimethylsulphoxide(DMSO), glycerol, methanol에 대해 실험하였으며, DMSO와 glycerol에서 methanol에 비하여 높은 생존율과 운동성을 나타내었다. 또한, 동결보존에 있어 예비동결은 액체질소표면으로부터 2 cm 높이에서 높은 생존율과 운동성을 보였고, 해동온도로는 $20^{\circ}C$가 가장 적합하였다.
