가압조건이 정지상태 과냉각 수용액의 해소에 미치는 영향 (Effect of pressurization on dissolution of a supercooled aqueous solution with a stationary state)

  • Supercooled type ice slurry system is hard to keep a proper supercooling degree when solution becomes supercooling state. This is the reason of the ice blockage in pipe or cooling part due to an unstable cooling state. In this study, a cooling experiment was performed to pressurized solution in a stationary state. The behaviors during the supercooled aqueous solution were investigated at fixed flow rate of brine and aqueous solution of ethylene glycol 7 mass%. Also the effect to the freezing point of supercooled aqueous solution was investigated to the different pressure 101, 202, 303, and 404 kPa. At results, the pressure of the aqueous solution in the cylinder increased the supercooling degree increased and dissolution of supercooled point decreased.

점안용 소염약침액의 안점막자극실험 (Eye Irritation Test of Soyeom Pharmacopuncture Solution for Eye Drop)

  • Objectives This study was done to investigate the safety of Soyeom pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration(2005. 10. 21, KFDA 2005-60). After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, the auther observed eye irritation of the cornea, iris, conjunctiva at 1, 2, 3, 4 & 7day. Results 1. After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, there wasn't physical problem at 9 rabbits. 2. After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, there wasn't eye irritation of the cornea, iris, conjunctiva at 1, 2, 3, 4 & 7day. Conclusions I suggested that Soyeom pharmacopuncture solution didn't induced eye irritation in rabbits.

The Effect of Solution Pressure to the Release in a Supercooled Aqueous Solution

  • Supercooled type ice storage system with aqueous solution (or water) may have trouble with non-uniform release of supercooling even though it contributes to the simplicity of system and ecological improvement. The non-uniform release increases the instability of the system because it may cause an ice blockage in pipe or cooling part. In order to suppress the release of the supercooling, a cooling experiment was tried to an ethylene glycol(EG) 3 mass% solution corresponding with pressurization. Also, the frequency ratio of the release of the supercooling was measured to the pressurization from 101 to 505 kPa. At results, the frequency ratio of supercooling release tends to decrease as the pressure of the aqueous solution increased in each cooling rate. Moreover, it tends to decrease as the cooling rate of the solution decreased in each pressure.

  • In previous studies, we reported that sow which was transferred OPS-freezing embryos not able to deliver a piglet (Kim et al, 2004). This study was conducted to investigate a possibility of gilt as recipients which produce piglets after transfer of OPS-freezing embryos. All transferred embryos were prepared by in vitro production (IVP) system. In vitro culture (IVC) medium used glucose-free NCSU23 supplemented with 5mM sodium pyruvate, 0.5 mM sodium lactate and 4 mg/ml bovine serum albumin for 2 days at $39^{\circ}C$. From day 3 of IVC, 10% fetal bovine serum albumin was added to the culture medium. In preparing of freezing embryos, embryos were treated with 7.5 $\mu g/ml$ cytochalasin-B for 30 min and centrifuged at $13,000{\times}g$ for 13 min. And then, embryos were exposed sequentially to an ethylene glycol (EG) solution, aspirated into open pulled straw (OPS), and plunged or thawed into the liquid nitrogen. In embryo transfer (ET), we used two kinds of type (surgical method vs. non-surgical method). In surgical method of embryo transfer, $55\sim65$ embryo were transferred in both uterine horn of two recipient gilts by plastic straw. Non-surgical method which is like artificial insemination was performed on three gilts. Each 140 frozen embryos were transferred to two gilts and 40 fresh embryos to one gilt. Pregnancy establishment was shown one recipient at 45 days after ET. However, the one recipient was also aborted at 58 days after ET. These results suggest that gilts can be considered as a candidate of recipients for OPS-freezing embryo transfer.

생쥐 1-세포기배의 초급속 동결에 있어서 평형 온도와 노출시간의 영향 (The Effect of Equilibration Temperature and Exposure Time on the Ultrarapid Freezing of 1-cell Mouse Zygote)

  • The present study was to assess the effect of ultrarapid freezing on the development of 1-cell mouse zygote using cryoprotectants, DMSO (dimethyl sulfoxide) or PROH (1,2-propanediol). We investigated the effect of the type and concentration of cryoprotectant, and of the temperature and time of prefreezing equilibration on their capacity to develop to the blastocyst stage in vitro. The concenration, the equilibration temperature, and the exposure time seemed to serve as an important factor in ultrarapid freezing of 1-cell mouse zygotes. In addition to the exposure time and the concentration of cryoprotectant appeared to playa key role in the development of the embryo. In general, the development of the embryo was more effective at $3^{\circ}C$ than $23^{\circ}C$ and 4.5 M than 3 M for 3 to 5 minutes. At $23^{\circ}C$ the development of the embryo was stimulated by DMSO while at $3^{\circ}C$ it was stimulated by PROH. Thus it has been suggested that there exists a correlation between the concentration of cryoprotectants and exposure time in the development of the embryo. In conclusion, we found that for ultrarapid freezing of mouse 1-cell embryos in DMSO, or PROH-based solution, viability shown optimum depending on the cryoprotectant, the concentration of the cryoprotectant and on the temperature and the duration of equilibration.

냉각법 및 영상 처리기법을 이용한 서브마이크론 정전분무 액적의 크기분포 측정 (Measurement of Size Distributions of Submicron Electrosprays Using a Freezing Method and an Image Processing Technique)

  • The size distributions of electrospray droplets from the Taylor cone in cone-jet mode are directly measured by using a freezing method and a transmission electron microscope (TEM) image processing technique. These results are compared with the data obtained by an aerodynamic size spectrometer (TSI Aerosizer DSP). The use of glycerol seeded with NaI and a freezing method make it possible to sample droplets with their original sizes preserved. Since pictures of droplets are taken with TEM with very low vapor pressure of the solution, evaporation is suppressed by freezing. For liquid flow rates below 1 nl/sec, the measured droplet diameters by the TEM image processing technique and the aerosizer are in the range of 0.25 to 0.32 m add 0.3B to 0.40m, respectively. Comparing the TEM data with the aerosizer measurements, it has been revealed that the TEM image processing technique can afford more accurate values of droplet size distributions in the submicron range of 0.1 to 0.4m.

  • In vitro survival of the mouse morulae frozen by vitrification method(Kasai et al., 1990) was investigated in the present study. The embryos were plunged into LN2 directly after exposure to the vitrification solutions(EFS, GFS and DFS). The results were obtained as follows. The viability of morulae after freezing and thawing was high in EFS(96.7∼100.0%) and GFS vitrification solution(93.3∼96.7%), and the lowest in DFS vitrification solution(0.00∼0.03%).

냉동 딸기의 비가열 전처리 기술 개발 및 최적 냉동조건 수립 (Non-thermal Treatment of Postharvest Strawberry and Establishment of Its Optimal Freezing Condition)

  • 수출용 냉동딸기 제조 시 미생물학적 안전성 확보를 위한 수단으로 이산화염소수와 acetic acid 병합처리를 최적 비가열 전처리 기술로써 적용하여 -20, -70, $-196^{\circ}C$로 냉동한 후 미생물 수, 품질변화 및 관능평가를 조사하였다. 냉동방법 및 세척 처리에 따른 색도 차이는 나타나지 않았고, 비타민 C 함량은 $-70^{\circ}C$ 냉동에서 35.33 mg/100 g FW로 가장 대조구와 유사하였으며, drip loss도 $-70^{\circ}C$ 냉동이 14.39%로 가장 낮게 나타났다. 관능평가 역시 $-70^{\circ}C$ 냉동이 -20, $-196^{\circ}C$ 냉동보다 높은 점수를 받았으며, 세척처리는 비타민 C 함량, drip loss 및 관능평가에 큰 영향을 끼치지 않는 것으로 나타났다. 또한, 냉동딸기에 50 ppm 이산화염소수와 1% acetic acid를 병합 처리하여 냉동 후 미생물 수 변화를 측정한 결과, 냉동 전과 같이 병합처리 된 딸기 시료에서 미생물이 검출되지 않았다. 따라서 저장성이 높은 수출용 냉동딸기 생산을 위해서는 gas nitrogen convection chamber를 이용한 $-70^{\circ}C$에서의 급속냉동 처리가 보다 효과적인 냉동방법이며, 냉동 처리만으로는 미생물 제어가 어렵기 때문에 냉동 전 비가열 전처리를 통해서 냉동딸기의 미생물학적 안전성을 확보해야 한다고 판단된다.
