건조방법에 따른 분류의 리올로지 특성 (Effect of drying method on rheological properties of milk powders)

  • 건조방법 및 원료성분을 달리하여 분유를 제조하고 이들분유의 몇 가지 리올로지 특성을 Instron Universal Testing Machine으로 측정하여 얻어진 결과는 다음과 같다. 드럼건조된 전지분유의 부피밀도는 $0.382\;g/cm^3$으로 탈지 및 조제 분유보다 높았고, 분무 건조된 탈지 분유의 Hausner ratio, 평정각 및 compressibility 는 각각 1.127, $32.5^{\circ}$, 0.029로 탈지 분유는 자유롭게 흐르는 것을 보여주었으며, 드럼건조된 전지분유의 compressibility는 수분 함량이 4.96%에서 3.89%로 감소함에 따라 0.079에서 0.052로 낮아졌다. 전지분유 그래뉼의 compressibility는 0.056으로서 분유의 compressibility보다 낮았다. 분무 건조된 분유는 구형으로서, 동결건조나 드럼건조된 분유보다 더 견고함을 나타냈다.

Genomic DNA Extracted from Ancient Antarctic Glacier Ice for Molecular Analyses on the Indigenous Microbial Communities

  • From ancient Antarctic glacier ice, we extracted total genomic DNA that was suitable for prokaryotic 16S rDNA gene cloning and sequencing, and bacterial artificial chromosome (BAC) library and end-sequencing. The ice samples were from the Dry Valley region. Age dating by $^{40}Ar/^{39}Ar$ analysis on the volcanic ashes deposited in situ indicated the ice samples are minimum 100,000-300,000 yr (sample DLE) and 8 million years (sample EME) old. Further assay proved the ice survived freeze-thaw cycles or other re-working processes. EME, which was from a small lobe of the basal Taylor glacier, is the oldest known ice on Earth. Microorganisms, preserved frozen in glacier ice and isolated from the rest of the world over a geological time scale, can provide valuable data or insight for the diversity, distribution, survival strategy, and evolutionary relationships to the extant relatives. From the 16S gene cloning study, we detected no PCR amplicons with Archaea-specific primers, however we found many phylotypes belonging to Bacteria divisions, such as Actinobacteria, Acidobacteria, Proteobacteria $({\alpha},\;{\beta},\;and\;{\gamma})$, Firmicutes, and Cytophaga-Flavobacterium-Bacteroid$. BAC cloning and sequencing revealed protein codings highly identical to phenylacetic acid degradation protein paaA, chromosome segregation ATPases, or cold shock protein B of present day bacteria. Throughput sequencing of the BAC clones is underway. Viable and culturable cells were recovered from the DLE sample, and characterized by their 16S rDNA sequences. Further investigation on the survivorship and functional genes from the past should help unveil the evolution of life on Earth, or elsewhere, if any.

선원사지(禪源寺地)출토 지류 유물의 분석 및 보존 처리 방법 (Analysis and the Conservation Treatment of the Paper Relics of the Seonwon Temple Area)

  • 사적 259호 선원사지(禪源寺地) 4차 발굴에서 출토된 지류(紙類) 유물들을 분석을 하고 이를 통해 보존상황에 맞는 보존처리방법을 연구하였다. 지류(紙類) 유물이 펼쳐볼 수 없는 상황에서 가장 중요한 보존처리 과정은 유물의 분리이고, 분리는 지류(紙類) 유물의 상태에 따라 분리방법을 달리하여 연구하였다. 건조한 상태의 유물 중에서 경화정도가 약한 것은 Gore-Tex chamber를 제작하여 분리하고, 경화정도가 강한 것은 여과수를 사용하여 분리하였다. 젖은 상태의 유물은 진공동결건조방법을 통해서 분리하였다. 지류유물의 분석은 재료의 특징을 알기 위한 종이의 조사, 전자현미경과 SEM을 통한 섬유의 분석, X선 형광분석법(XRF)에 의한 글씨 재료를 분석하였다. 유물의 제작 연대를 추정하기 위하여 가속기 질량 분석기에 의한 탄소연대측정을 하였다. 분석과 보존 처리 과정을 통하여 출토 유물의 분류가 되었고, 유물의 분류는 원형을 상실한 유물들의 원형보존을 가능하게 하였다. 본 논문에서는 가장 열악한 상태의 지류유물의 보존처리 방법의 적용을 제시해 보았고, 탄소연대측정을 통하여 자료도 제시할 수 있었다.

Isolation and Charaterization of Bioactive Peptides from Hwangtae (yellowish dried Alaska pollack) Protein Hydrolysate

  • Hwangtae, dried Alaska pollack, is a major storage product in the fish processing industry. Hwangtae is prepared by removing the internal organs and drying outdoors during the cold witner months by allowing it to thaw during the daytime and re-freeze at night under sub-zero ($-10^{\circ}C$) conditions and gradually dry from December until the next April for around 5 months from Myungtae. In this study, ground Hwangtae was hydrolyzed using two proteolytic enzymes (pepsin and alcalase) which produced five soluble active peptides from Hwangtae (yellowish dried Pollack, Theragra chalcogramma) protein. Two different peptides with strong antioxidative activity were isolated from the hydrolysate using consecutive chromatographic methods of Sephadex G-25 gel, ion-exchange chromatography on a Sepharose-Sephadex C-25 gel, and high-performance liquid chromatography. The isolated peptides, APO1 and APO2, were composed of 16 and 13 amino acid residues, respectively. Both peptides contained a Gly residue at the C-terminus and the repeating motif Gly-Pro-Hyp. The peptide with a molecular weight less than 1,000 Daltons (APACE) obtained from enzymatic hydrolysates of Hwangtae exhibited the highest ACE inhibitory activity. The APACE peptides was composed of 4 amino acid residues (Gly-Leu-Leu-Pro). These results suggest that Hwangtae hydrolysates could be a good source of peptides with ACE inhibitory activity. Biochemical analysis indicated that two 70 kDa peptides (APG1 and APG2) isolated from the hydrolysate had gelatinoytic activity, which was shown to be a calcium dependent protease type as showed by gelatin SDS PAGE.

Development of ginseng powder using high hydrostatic pressure treatment combined with UV-TiO2 photocatalysis

  • Background: Korean ginseng (Panax ginseng Meyer) powder is in rising demand because powder forms of foods are convenient to handle and are highly preservable. However, ginseng powder (GP) manufactured using the conventional process of air drying and dry milling suffers nutrient destruction and a lack of microbiological safety. The objective of this study was to prepare GP using a novel process comprised of UV-TiO2 photocatalysis (UVTP) as a prewashing step, wet grinding, high hydrostatic pressure (HHP), and freeze-drying treatments. Methods: The effects of UVTP and HHP treatments on the microbial population, ginsenoside concentration, and physiological characteristics of GP were evaluated. Results: When UVTP for 10 min and HHP at 600 MPa for 5 min were combined, initial 4.95 log CFU/g-fw counts of total aerobes in fresh ginseng were reduced to lower than the detection limit. The levels of 7 major ginsenosides in UVTP-HHP-treated GP were significantly higher than in untreated control samples. Stronger inhibitory effects against inflammatory mediator production and antioxidant activity were observed in UVTP-HHP-treated GP than in untreated samples. There were also no significant differences in CIELAB color values of UVTP-HHP-treated GP compared with untreated control samples. Conclusion: Combined processing of UVTP and HHP increased ginsenoside levels and enhanced the microbiological safety and physiological activity of GP.

소회향이 흰쥐의 국소뇌혈류량 및 평균혈압에 미치는 영향 (Effects of FOENICULI FRUCTUS on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats)

  • This experimental study was designed to investigate the effects of FOENICULI FRUCTUS freeze dry powder (FF) on the change of cerebral hemodynamics [regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP)] in normal and further to determine the mechanism of action of FF. The results in normal rats were as follows ; FF significantly increased rCBF in a dose-dependent, but decreased MABP, This results were suggested that FF significantly increased rCBF by dilating PAD. The FF-induced increase in rCBF was significantly inhibited by pretreatment with 1H[1,2,4]oxadizolo[4,3-a]quinoxalin-1-one (ODQ, 10 ${\mu}$g/kg, i.p.), an inhibitor of guanylate cyclase and indomethacin (IDN, 1 mg/kg, i.p.), an inhibitor of cyclooxygenase and propranolol (PPN, 3 mg/kg, i.p.), a blocker of adrenalic f receptor and Lu-Nitro-L-Arginine (L-NNA, 1 m9/kB, i.p.), an inhibitor of nitric oxide synthase. The FFE-induced decrease in MABP was significantly increased by pretreatment with L-NNA and was increased by pretreatment with PPN, Dut was inhibited by pretreatment with ODQ and IDN, This results were suggested that the mechanism of FF was mediated by nitric oxide synthase and adrenalic ${\beta}$ receptor.

Effects of Dietary n-3/n-6 Fatty Acid Ratio on In Vitro Fermentation Characteristics and Fatty Acid Profiles

  • This study was conducted to examine the effect of dietary n-3/n-6 fatty acid (FA) ratio on in vitro dry matter digestibility (IVDMD), fermentation indices and FA profile. Rice bran was mixed with oil sources (cotton seed oil and linseed oil) to make the diets at 0.02, 0.29 and 0.61 of dietary n-3/n-6 FA ratio. These diets (0.5g) were placed into the incubation bottles with 40 ml of anaerobic culture medium, which contained rumen fluid and Van Soest medium at 1:2 ratio. Five replicates of each diet and two blanks were incubated at $39^{\circ}C$ for 48 hours. After incubation, the incubated contents were centrifuged. The residues were freeze-dried for DMD and FA analyses. The supernatant was used for pH, $NH_3-N$ and volatile fatty acid analyses. The concentrations of lactate (p<0.001) and iso-valerate (p<0.001) decreased linearly with increasing dietary n-3/n-6 FA ratio, but acetate concentration (p=0.056) and the ratio of acetate to propionate (p=0.005) was increased linearly. The concentrations of n-3, n-6 FA and the ratio of n-3/n-6 FA in residues increased (p<0.001) linearly with increasing dietary n-3/n-6 FA ratio, but C18:1n-9 FA concentration was decreased (p<0.001) linearly. With these results, it could affect fermentation characteristics and FA profile of rumen content by dietary n-3/n-6 FA ratio.

다목적 농산물건조기를 이용한 오디 건조특성 구명 (Studies on the drying characteristics of mulberry fruits with multipurpose agricultural products dryers)

  • 본 연구는 장기 냉동저장 오디에 대해 현재 사용하고 있는 방법이 보관 및 유통비용 등이 커 농가에 부담이 되므로, 이에 고가 건조기를 대체할 수 있는 값이 저렴한 상용화된 다목적농산물건조기를 통한 건조기 개량 및 반건조기술을 개발하였다. 연료와 히터의 건조온도를 $60^{\circ}C$로 설정하고, 상온통풍은 상온보다 온도를 낮추어 건조시험 한 결과 연료 3시간, 히터 1시간, 상온통풍 51시간인 조건에서 다른 조건과 비교해서 연료소모 및 전기사용량 면에서 가장 양호하였고, 품질에 있어서도 다른 조건에 비해 현저히 끈기가 있는 반 건조 특성을 나타내었으며, 완료된 건조시료의 상태도 상품성이 뚜렷한 암갈색을 나타냈다. 경도(압축력)은 건조 전보다 건조 후에, 항온보다는 변온에서 높게 나타났다.

Characterization of starch and gum arabic-maltodextrin microparticles encapsulating acacia tannin extract and evaluation of their potential use in ruminant nutrition

  • Objective: The use of tannin extract and other phytochemicals as dietary additives in ruminants is becoming more popular due to their wide biological actions such as in methane mitigation, bypass of dietary protein, intestinal nematode control, among other uses. Unfortunately, some have strong astringency, low stability and bioavailability, and negatively affecting dry matter intake and digestibility. To circumvent these drawbacks, an effective delivery system may offer a promising approach to administer these extracts to the site where they are required. The objectives of this study were to encapsulate acacia tannin extract (ATE) with native starch and maltodextrin-gum arabic and to test the effect of encapsulation parameters on encapsulation efficiency, yield and morphology of the microparticles obtained as well as the effect on rumen in vitro gas production. Methods: The ATE was encapsulated with the wall materials, and the morphological features of freeze-dried microparticles were evaluated by scanning electron microscopy. The in vitro release pattern of microparticles in acetate buffer, simulating the rumen, and its effect on in vitro gas production was evaluated. Results: The morphological features revealed that maltodextrin/gum-arabic microparticles were irregular shaped, glossy and smaller, compared with those encapsulated with native starch, which were bigger, and more homogenous. Maltodextrin-gum arabic could be used up to 30% loading concentration compared with starch, which could not hold the core material beyond 15% loading capacity. Encapsulation efficiency ranged from $27.7%{\pm}6.4%$ to $48.8%{\pm}5.5%$ in starch and $56.1%{\pm}4.9%$ to $64.8%{\pm}2.8%$ in maltodextrin-gum arabic microparticles. Only a slight reduction in methane emission was recorded in encapsulated microparticles when compared with the samples containing only wall materials. Conclusion: Both encapsulated products exhibited the burst release pattern under the pH conditions and methane reduction associated with tannin was marginal. This is attributable to small loading percentages and therefore, other wall materials or encapsulation methods should be investigated.

건조방법을 달리한 보리 잎의 생리활성 (Physiology Activity of Barley Leaf Using Different Drying Methods)

  • 다양한 방법으로 건조한 보리 잎의 생리활성을 분석한 결과, 보리 잎의 추출수율은 건조방법 간에 유의적인 차이를 보이지 않았다. 총 페놀함량은 건조방법별 함량에 유의적인 차이가 없었으나, 총 플라보노이드 함량은 열처리 후 건조한 보리 잎에서 생잎 다음으로 높게 나타났고, 음건, 진공 동결 건조, 전자레인지 후 건조한 보리 잎에서는 유의성을 보이지 않았다. SOD 유사활성은 생잎 $95.69{\pm}0.33%$, 열처리 후 건조한 보리 잎 $94.96{\pm}0.77%$, 음건 $92.04{\pm}1.35%$, 진공 동결건조 $91.53{\pm}0.40%$, 전자레인지 건조 $92.58{\pm}0.27%$로 90% 이상의 높은 활성을 보였으며, 대조군인 항산화제 sesamol $88.42{\pm}0.80%$, tocopherol $88.86{\pm}0.61%$, BHT $86.62{\pm}0.89%$에 비해 유의적으로 높은 활성을 나타냈다. 또한 생잎과 전자레인지 건조한 보리 잎을 제외하고 80% 이상의 peroxyl radical 소거능 및 90% 이상의 높은 xanthine oxidase 저해능을 보였다. 본 연구결과, 건조방법별 보리 잎의 총 페놀, 총 플라보노이드 함량은 건조방법에 따라 차이를 보였으나, 항산화 능력에는 크게 영향을 미치지 않는 것으로 사료된다.
