Isoprostane Characteristics in Sick House Syndrome, Atopy and Asthma Patient

  • An imbalance between oxidants and antioxidants, in favor of oxidants leading to oxidative stress, is known to play an important role in the pathogenesis of various diseases. Isoprostanes are structurally stable isomers of the conventional enzymatically derived prostaglandins, which are produced in vivo primarily by a free radical catalyzed peroxidation of polyunsaturated fatty acids. In asthmatics, disease severity can occur from environmental exposure to air pollution. Some surveys suggested that air pollutants, especially diesel-exhaust particulates, could trigger allergic sensitization and development of atopic diseases. Sick house syndrome (SHS) presents healthy damage owing to the indoor environment of a building. The aim of this study was to examine isoprostane as a parameter fur oxidative stress in environments related diseases such as sick house syndrome, atopy and asthma. We measured plasma and urinary levels of isoprostane from health volunteers, sick house syndrome, atopy and asthma patients. Plasma isoprostane concentrations in asthma and sick house syndrome group were significantly higher than in control. Urinary isoprostane levels were significantly higher in volunteers with sick house syndrome and asthma compared with health volunteers. These findings suggest that plasma and urinary isoprostane measurement may have useful clinical implications for investigating sick house syndrome and asthma. The interventions that decrease exposure to environmental reactive oxygen species might be beneficial in these diseases.

Comparison of Mineral Contents and Antioxidant Activities of Domestic and Chinese Wolfiporia extensa for Origin Identification (국내산과 중국산 복령의 원산지 구별을 위한 성분 분석 및 항산화 활성)

  • This study aimed to discriminate the geographical origins of domestic and chinese Wolfiporia extensa. They were subjected to oxygen (45.32 to 48.07%), carbon (38.09 to 40.12%), hydrogen (6.05 to 6.78%), and nitrogen (0.16 to 0.23%). Antioxidant activity was examined by DPPH free radical scavenging activity. According to the results of the experiment, no significant differences were found between domestic ($IC_{50}$, 7.25 mg/ml) and Chinese ($IC_{50}$, 8.35 mg/ml) W. extensa. However, as determined by the inorganic mineral contents were significantly different between domestic and Chinese W. extensa. The amount of potassium in domestic and Chinese W. extensa was and $33.14{\pm}17.27%$, $47.60{\pm}8.78%$, respectively. The results of this study suggested that the analysis of inorganic components by ED-XRF should be useful for origin identification of W. extensa.

  • Green Tea consists of 15-30% catechins (a type of polyphenol), which act as super antioxidants, inhibitory action against aging process. Antioxidants fight radical-free oxygen, an agent which can begin the process of cancer by damaging essential body chemicals and harming DNA. This study was peformed to develop the elderly diet using Green Tea as an inhibitory action against aging process. Nokchaborijook (Baley gruel added rice powder and Green Tea) was manufactured by various levels of Green Tea(1, 4, 7%) and the grinding period(15, 30, 45 sec). The optimum levels of added Green Tea and grinding periods on Nokchaborijook were determined with the results of sensory evaluation by response surface methodology and analysis of composition. The Anti-oxidant Vitamin A, C, E and Flavonoid were increased with increased levels of Green Tea and grinding periods be decreased. As the levels of Green Tea and grinding periods were increased, the green color and penetrated force became stronger. Among the sensory attributes, Bitterness, Green Tea Flavor and Hashness were greater depending on increasing Green Tea. Jujube flavor was greater relying on decreasing Green Tea. Greenness and spreadability of particles were increased as both the amounts of Green Tea and the grinding period were increased. This result was used to determine the optimum conditions of adding levels of Green Tea and grinding periods. The optimum conditions of Nokchaborijook was established as adding of 5.8% Green Tea grinded for 15 seconds.

The Antifibrotic and Antioxidant Activities of Hot Water Extract of Adventitious Root Culture of Panax ginseng (ARCP)

  • The anti-fibrotic effects of hot water extract of adventitious root culture of Panax ginseng (ARCP) and the possible mechanisms were investigated on $CCl_4-induced$ hepatotoxicity model mice. Fibrosis was induced by a mild treatment of $CCl_4$. Then silymarin as a positive control drug and ARCP or carrier alone as a negative control were treated. Serum GPT, GOT and ALP activity levels were lowered by 25, 21 and 11% for silymarin treated group and by 48, 39 and 14% for ARCP treated group compared to carrier treated alone. Hepatic collagen for ARCP treatment group was reduced by 18% and MDA contents decreased a little more. Pro-fibrotic gene ($TGF-{\beta}1$, TIMP-1 and ${\alpha}-SMA$) expression increased following the $CCl_4$ treatment, but both the silymarin and the ARCP treatments decreased the expressions of these genes by 20% to 50%. The antioxidant effect of ARCP was studied by DPPH free radical scavenging activity. In addition, a generation of reactive oxygen species (ROS) was also reduced in $H_2O_2-treated$ HepG2 cells upon the ARCP treatment. In summary, ARCP has antioxidant property, and can have some protection against oxidative stress; more importantly, ARCP can efficiently protect mice against $CCl_4-induced$ fibrosis.

Growth Period Effects on the Protective Properties of Aloe vera Against t-BHP-Induced Oxidative Stress in Chang Cells

  • Aloe vera has been used in traditional medicine for the therapy of a variety of disorders, such as wounds and burns. However, few studies have examined the antioxidant capacities of A. vera plants during different growth periods. In order to investigate the effects of growth on antioxidant activity, A. vera was prepared from 2-, 4-, 6-, 8-, and 12-month-old aloe. The extracts from 6-month-old A. vera showed the highest contents of flavonoids (9.750 mg catechin equivalent/g extract) and polyphenols (23.375 mg gallic acid equivalent/g extract) and the highest ferric reducing antioxidant power (0.047 mM ferrous sulfate equivalent/mg extract). The extract from 6-month-old A. vera exhibited the highest free radical scavenging potential, and the lowest IC50 values were found for 2,2-diphenyl-1-picrylhydrazyl (0.26 mg/ml) and alkyl radicals (0.50 mg/ml). In addition, the extract from 6-month-old A. vera showed the greatest effects on cell viability in normal liver cells. Based on these findings, the extract from 6-month-old A. vera was examined further in order to determine its protective potential against tert-butyl hydroperoxide (t-BHP)-induced oxidative stress. The extract from 6-month-old A. vera at a concentration of 0.25 mg/ml showed the highest protective activity against t-BHP-induced reactive oxygen species production. These findings suggested that harvesting regimens were critical in the regulation of effects of the bioactive potential of A. vera on antioxidant activity.

Effects of the Sciripi rhizoma on Antioxidation and Melanogenesis (삼릉추출물이 항산화와 멜라노제네시스에 미치는 영향)

  • Whitening effect, which decreases the skin pigmentation, is the one of important targets in cosmetics. This study was investigated the effects of Scirpi rhizoma on ant ioxidation and melanogenesis. S.rhizoma is a rhizome of Scirpus fluviatilis G. a perennial Cyperaceae species of wide occurrence in Asia, Europe, Africa and North America. S.rhizoma shown scavenging activities of free radicals and reactive oxygen species (ROS) with the IC50 of 638${\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 21.7${\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. S.rhizoma treatment (48 h) suppressed the biosynthesis of melanin up to 27% and reduced tyrosinase activity up to 31% at 100${\mu}g/ml$ in B16 melanoma cells. S.rhizoma was also able to significantly inhibit tyrosinase and TRP-1 expres- sion in protein level. These results suggest that S.rhizoma inhibited melanin biosynthesis by regulating tyrosinase activity and expression in B16 melanoma cells. Therefore S.rhizoma may be useful as new whitening agent due to the antioxidant effect and the inhibitory effect against melanogenesis.

Structural, Electrochemical, DNA Binding and Cleavage Properties of Nickel(II) Complex [Ni(H2biim)2(H2O)2]2+ of 2,2'-Biimidazole

  • A nickel(II) complex $[Ni(H_2biim)_2(H_2O)_2](ClO_4)_2{\cdot}H_2O$ (1) of biimidazole ligand has been synthesized and characterized (Where $H_2biim$ = 2,2'-biimidazole). The single crystal X-ray diffraction of the complex shows a dimeric structure with six coordinated psudo-octahedral geometry. The cyclic voltammograms of complex exhibited one quasireversible reduction wave ($E_{pc}=-0.61V$) and an irreversible oxidation wave ($E_{pa}=1.28V$) in DMF solution. The interaction of the complex with Calf-Thymus DNA (CT-DNA) has been investigated by absorption and fluorescence spectroscopy. The complex is an avid DNA binder with a binding constant value of $1.03{\times}10^5M^{-1}$. The results suggest that the nickel(II) complex bind to CT-DNA via intercalative mode and can quench the fluorescence intensity of EB bind to CT-DNA with $K_{app}$ value of $3.2{\times}10^5M^{-1}$. The complex also shown efficient oxidative cleavage of supercoiled pBR322 DNA in the presence of hydrogen peroxide as oxidizing agent. The DNA cleavage by complex in presence of quenchers; viz. DMSO, KI, $NaN_3$ and EDTA reveals that hydroxyl radical or singlet oxygen mechanism is involved. The complex showed invitro antimicrobial activity against four bacteria and two fungi. The antimicrobial activity was nearer to that of standard drugs and greater than that of the free ligand.

The Extract from Lysimachia foenum-graecum Induces Apoptosis in MCF-7 Breast Cancer Cells (MCF-7 유방암 세포에서 영릉향 추출물에 의한 Apoptosis 유도에 관한 연구)

  • The extract from Lysimachia foenum-graecum (LFE) has been known to possess various instructive characters including anti-oxidant, anti-obesity, fungicidal activities. However, the accurate mechanism of those effects of LFE is not well known. In that respect, we evaluated the apoptotic effect and anti-cancer efficacy of extracts of LFE in MCF-7 breast cancer cells. In this study, we hypothesized that LFE may exert cancer cell apoptosis through regulating p53 and mitochondria-mediated apoptotic proteins. And this substance can generate ROS to cause free radical-induced apoptosis. Accordingly, the generation of ROS by LFE triggers the activation of p53 which are accompanied by pro-apoptotic protein activation and suppression of pro-survival proteins. We determined with MTT assay, flow cytometry for detection of intracellular ROS and Annexin V-PI staining, Western blotting. Consequently, our researches demonstrated that the treatment of LFE to breast cancer cells resulted in an activation of p53, Puma, Bax, cleaved-PARP and an inhibition of Bcl-2 expressions.

Effects of three kinds of Radix Rehmanniae Water Extract in Cultured Rat Myocardial Cells (삼종 지황 추출물이 배양 심근세포에 미치는 영향)

  • To test the protective effect of herbal medicine on myocardial damage against oxygen free radical-induced myocardiotoxicity, cytotoxicity was examined using MTT, Beating rate and DNA synthesis assay in the presence of water extract of three kinds of Radix Rehmanniae. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. The results of these experiments were obtained as follows : Xanthine oxydase/hypoxanthine resulted in a decrease in viability, beating rate and DNA synthesis in cultured myocardial cells. Radix Rehmanniae Recens(生地黃, RRR) water extract shows effects of protection from the cardiocyte toxicity induced by xanthine oxydase/hypoxanthine treatment such as increases in beating rate. Radix Rehmanniae Preparat(熟地黃, RRP) water extract shows effects of protection from the cardiocyte toxicity induced by xanthine oxydase/hypoxanthine treatment such as increases in DNA synthesis. These results show that xanthine oxydase/hypoxanthine elicits toxic effects in cultured myocardial cells derived from neonatal rat, and suggest that water extract of three kinds of Radix Rehmanniae is very effective in the prevention of xanthine oxydase/hypoxanthine-induced cardiotoxicity.

Effect of Allopurinol Pretreatment on the Liver Damage in $CCl_4$-treated Rat (흰쥐에 있어서 사염화탄소에 의한 간손상에 allopurinol의 영향)

  • To evaluate the effect of xanthine oxidase on liver injury by $CCl_4$, liver damage was induced both in allopurinol pretreated rats (500 mg/kg. ip) and control group by twice intraperitoneal injection of $CCl_4$ (0.1 ml/100 g body wt. 50% in olive oil) at interval of one day. Increases in the levels of serum alanine aminotransferase and liver weight/body weight (%) by $CCl_4$ were significantly smaller inallopurinol pretreated rats than in control whereas the hepatic microsomal glucose-6-pholphatase activities were significantly higher in allopurinol pretreated rats than control group by $CCl_4$ treatment. These results indicates that allopurinol pretreatment may reduce the liver damage in $CCl_4$ intoxicated rats. In rats either with $CCl_4$or not, hepatic type O xanthine oxidase activities were significantly reduced by allopurinol pretreatment and the increasing rate of these enzymes to each control was remarkably lower in allopurinol pretreated rats than control. Liver cytosolic protein contents and aniline hydroxylase, aminopyrine demethylase activities were higher in allopurinol pretreated rats than coirol rats when animals were treated with $CCl_4$. On the other hand, neither allopurinol pretreated nor $CCl_4$ treatment caused any significant changes in hepatic superoxide dismutase and catalase activities. Hepatic glutathione contents were higher in $CCl_4$-treated rats than control, but no significant changes were found in both between the allopurinol treated rats and $CCl_4$-treated rats pretreated with allopurinol, and glutathione and glutathione S-transferase activities were significantly reduced in $CCl_4$-treated rats than control whereas these enzyme activities showed on significant change in both between allopurinel treated and $CCl_4$-treated rats pretreated with allopurinol. It is concluded that xanthine oxidase reaction system augment $CCl_4$ induced liver injury via even oxygen free radical system.
