Changes in free amino acids and hardness in round of Okinawan delivered cow beef during dry- and wet-aging processes

  • Background: Aging trials are conducted to determine characteristics associated with dry- and wet-aging processes of beef from delivered cows grown in Okinawa, i.e., dams that have finished giving birth (Okinawan delivered cow beef). Changes in free amino acids, hardness, and other factors were analyzed in round of Okinawan delivered cow beef during dry- and wet-aging processes along with a comparison with characteristics of beef imported from Australia. Results: Functional amino acids did not increase during both dry- and wet-aging processes. However, proteinogenic amino acids increased significantly (P < 0.05) and hardness tended to decrease during both dry- and wet-aging processes. On comparison between dry- and wet-aging processes by analysis of variance, drip and cooking losses were significantly lower during the dry-aging process than during the wet-aging process. However, there was no significant difference in free amino acids or hardness in this comparison. Conclusion: There was no significant difference between dry- and wet-aging methods for all studied variables related to free amino acids or hardness in this study.

  • BACKGROUND: Effect of various light wavelength and plant defense molecules were evaluated on the hardness and the contents of free amino acid including ${\gamma}$-aminobutyric acid (GABA) in soybean sprouts. METHODS AND RESULTS: Germinating soybean seeds were treated with various single wavelength of light (380, 440, 470, and 660 nm) or without light at $25^{\circ}C$ for six days. Soybean seeds were also treated with stress-signaling molecule ethephon or $H_2O_2$ at the same time. Soybean sprouts treated with 470 nm substantially raised the hardness almost two times than the control. The free amino acid contents were higher in 470 nm and $H_2O_2$ treated soybean seeds than the control. Nutritionally beneficial GABA contents were increased by the treatments of 470 nm, 440 nm, ethephon, and $H_2O_2$. CONCLUSION: These results suggest that the hardness and the contents of amino acids can be regulated by stimuli, which stimuli could be composed of various wavelength and plant defense molecules. Especially, single wavelength 470 nm illumination has the effect of increasing GABA contents with increased hardness.

Chemical and Textural Properties in Commercial Fermented Soybean Curds of Sufu

  • A survey aiming to find out the chemical and textural properties of commercial fermented soy bean curd called sufu was conducted. Sixteen brands of plain sufu produced in the Northern or the Southern part of China were collected and examined for their crude protein, crude fat, texture profiles, free amino acids, and free fatty acid contents. Twenty-one free amino acids were extracted and derivatized using a commercial kit followed by separation and analyzed by the gas chromatography-mass spectrometry (GC-MS). Similarly, ten free fatty acids were extracted using alumina, eluted, separated and analyzed. The content ranges of crude fat and protein were 22~36% and 31~38%, respectively. In texture profile analysis, ranges of the texture parameters were 131~493 g (hardness), 0.4~0.5 (cohesiveness), -137 to -50 gs (adhesiveness), 0.6~1 (springiness), 47~220 g (gumminess) and 32~177 g (chewiness). Twenty-one different free amino acids, especially alanine, glycine, $\alpha$-aminobutyric acid, valine, leucine, allo-isoleucine, aspartic acid, glutamic acid and lysine in large amount, as well as ten fatty acids in total, notably linoleic acid (9-octadecanoic acid), oleic acid (9,12-octadecadienoic acid), linolenic acid (9,12,15-octadecadienoic acid), hexadecanoic acid and octadecanoic acid were found. This information provides important quality reference ranges for product developers and manufacturers to optimize and produce the plain sufu.

Changes in free amino acid content and hardness of beef while dry-aging with Mucor flavus

  • Background: A mold strain thought to be suitable for dry-aging process was isolated. The information about the scientific aspects of molds related to dry-aging beef is scarce. We, therefore, conducted aging trials to determine the characteristics of the isolated mold strain associated with dry-aging process. Specifically, during the dry-aging of beef with the mold strain, the changes in the free amino acid content, hardness, productive loss, drip and cooking loss were analyzed. These characteristics were compared with those obtained while dry-aging in the absence of a mold. Results: The isolated mold strain was identified as Mucor flavus. The free amino acid content in the mold-aging beef decreased or remained constant during the aging process. However, that in the trimming sections of the beef dramatically increased in the presence of mold. In addition, hardness of mold-aging beef gradually decreased during the aging process and finally decreased significantly. Conclusion: Amino acids such as GABA (gamma-aminobutyric acid), proline, and aspartic acid were produced by our mold strain, M. flavus during its growth on beef meat, and the mold conferred savory odors to the dry-aged beef.

무화과 콘서트 처리에 의한 품종별 쇠고기의 이화학적 특성 변화 (The Effect of Fig Conserve Additive on the Physicochemical Characteristics of Beef Obtained from Various Breeds)

  • To prevent the denaturation of ficin(EC 3.4.22.3) that is a proteolytic enzyme in fig(Ficus carica L.), fig conserve was heated to 55oC. The fig conserve was added as a tenderizer to native Korean cattle beef(KCB), dairy cattle beef(DCB), castrated dairy cattle beef(CDCB), and imported beef(IB). The composition of free amino acids, hydroxyproline content, shear force, cooking loss, morphological changes and sensory evaluation were then investigated to observe the effect of tenderizing beef with fig conserve. Free amino acids and cooking loss of treated beef were higher than those of control, whereas hydroxyproline and shear force were lower. Glutamine in treated beef decreased relatively but asparagine increased. Hydroxyproline was found, in increasing order of abundance, in DCB, CDCB, IB and KCB. By portion, loin was higher than tenderloin in free amino acids, hydroxyproline and shear force but was lower in cooking loss. Observation with a light microgram revealed a surprising loss of muscle fiber in treated beef. In sensory evaluation of uncooked beef, the control was redder than the treated beef(p<0.01~p<0.05). Treatment with fig conserve increased the juiciness of both cooked CDCB and IB(p<0.001) and decreased their hardness(p<0.01~p<0.001).

인삼 및 청국장 물추출-혼합액으로 제조한 코팅미의 이화학적 특성 (Physicochemical Characteristics of Coated Rice Manufactured by the Mixture of Ginseng and Chungkukjang Water Extract)

  • New application of rice was carried out by coating the rice with the mixture solution of ginseng and chungkukjang water extract The physicochemical characteristics of the coated rice were investigated. Moisture content of uncoated (15.26%) and coated rice$(15.57\sim15.66%)$ was not different significantly. The contents of crude protein, crude fat, and crude ash of the coated rice were higher as much as $4.28\sim11.82%\;8.47\sim47.46%$, and $11.54\sim42.31%$ than those of control, respectively. As total free amino acids in coated rice were increased by augmenting the amount of the extract, total free amino acids according to rate was increased to $3.1\sim7.8$ times. The major amino acids in the coated rice was alanine$(19.56\sim39.88\;mg%)$, leucine$(5.14\sim17.66\;mg%)$, and proline$(9.98\sim16.82\;mg%)$. Of those amino acids, alanine and $\gamma-aminobutyric$ acid in only coated rice were detected to the level of $19.56\sim39.88\;mg%$ and $7.78\sim12.36\;mg%$ respectively. The calcium amount of coated rice was increased to 15% to 20%. As increasing the coating rate, hardness, cohesiveness, chewiness, and brittleness of coated rice were decreased, but springiness was increased. Before cooking, the color of coated rice appeared yellow and after cooking turned to the light yellow. The color intensity was increased feasibly as increasing the coating rate. The sensory characteristic of rice coated manufactured by adding 15% of the extract was best and found to be similar to that of the control.

새우의 맛성분과 미세구조에 미치는 가열 및 건조방법의 영향 (Effects of Cooking and Drying Methods on the Taste Component and Microstructure of Shrimp)

  • 가열처리 및 건조방법이 새우의 맛성분과 미세구조에 미치는 영향을 밝히고자 하였다. 생새우의 핵산관련물질은 ATP,ADP,AMP,IMP, inosine 및 hypoxanthine이 검출되었는데 이 중 AMP는 흔적량 검출되었으나, 가열 및 건조방법에 따라서 AMP 함량은 100g당 $23.5{\sim}45.7{\mu}mole$로 증가하였는데 이는 열처리 및 건조에 의하여 ATP 및 ADP가 AMP로 분해된 것으로 판단되었다. 생새우 중의 유리아미노산은 arginine의 함량이 가장 많았고 다음으로 glycine, lysine, proline, alanine등으로 이들 주요 아미노산 함량이 전체 유리아미노산 함량의 70%를 차지하였다. 총유리아미노산 함량은 생새우에서 건물 100g당 1,198mg이던 것이 끓인후 열풍 및 동결건조한 것은 각각 342mg 및 503mg으로 감소하였는데, 이는 가열처리시 수용성 아미노산이 용출된 것으로 판단되었다. 생새우를 가열처리 없이 열풍 및 동결건조한 것은 가열처리나 마이크로파 처리후 열풍 및 동결건조한 것보다 견고성 및 부서짐성이 높았으나 응집성과 검성은 낮았다. 동결건조한 것은 열풍건조한 것에 비하여 근육섬유질의 조직이 부드러웠고 마이크로파 처리후 열풍 및 동결건조한 것은 각각 조밀한 망상조직과 다공질의 망상조직을 형성하였다.

Blanched 섬초(시금치) 분말을 첨가한 식빵의 품질 특성 (Quality Characteristics of White Bread Mixed with Blanched Seomcho (Spinacia oleracea L.) Powder)

  • This research is about the quality characteristics of white bread mixed with varying ratios of Seomcho powder, such as 1%, 3%, 5%, and 7% (Spinacia oleracea L.). The pH level of dough decreased as more Seomcho powder (Spinacia oleracea L.) was added, and 7% addition recorded the lowest pH. The fermentation power of dough expansion decreased as the additional ratio increased in both the first and second proofing, and it was negatively correlated to the fermentation time factor. The moisture content of bread decreased as the addition ratio increased. The bread volume became smaller as the addition ratio increased, but it correlated negatively with its specific volume. The color of the crumb, the lightness and redness decreased in comparison to the control, but the yellowness increased. The total amino acid contents increased as the addition ratio increased, and glutamic acid was positively correlated to the additional ratio. The total free amino acids increased as the additional ratio increased, but cysteine levels were high at 5% and 7% additional ratio. The textural characteristics of the bread, springiness and cohesiveness were lower than the control, but gumminess, brittleness, and hardness were higher than the control. The adhesiveness was realized at 7% additional ratio.

감마선 조사가 건조-발효소시지의 Trans 지방산, 유리아미노산 및 관능검사 특성에 미치는 영향 (Effect of Gamma-Irradiation on Trans Fatty Acid, Free Amino Acid and Sensory Evaluation of Dry-fermented Sausage)

  • 본 연구는 감마선 조사가 건조-발효햄의 조직감, 지방산, 유리아미노산 및 관능적 특성에 미치는 영향을 조사하기 위해 실시하였다. 경도(hardness) 및 부착성(adhesiveness) 의 경우 조사 처리구가 유의적으로 낮게 나타났다(p<0.01). 검성(gumminess)의 경우에는 2 및 4 kGy처리구가 대조구 및 1 kGy처리구보다 유의적으로 높은 값을 나타내었다(p<0.01). 지방산의 경우 처리구의 포화지방산(SFA)(C14:0, C16:0, C18:0)은 유의적으로 높았고(p<0.05), 불포화지방산 (UFA)(C16:1, C18:1 및 C18:3), trans 지방산, n-3 및 n-6 지방산 의 함량은 4 kGy처리에서 감소하였다(p<0.05). 감마선 조사처리는 phosposerine, aspartic acid, glutamic acid를 증가 시켰으나, lysine은 감소시켰다(p<0.05). 관능검사의 경우, 황취(sulfur odor) 및 지방산화 맛(fat pungent flavor)은 4 kGy에서 가장 높게 나타났으며(p<0.05), 곰팡이 냄새(moldy odor) 및 곰팡이 맛(moldy flavor)은 낮게 나타났다(p<0.05). 이상의 결과 감마선 조사가 조직감, 지방산조성 및 관능적 특성에 영향을 미쳤다.

홍삼 및 복분자를 첨가한 청국장의 이화학적 특성 (Physicochemical Properties of Cheonggukjang Containing Korean Red Ginseng and Rubus coreanum)

  • 본 실험은 청국장의 풍미와 품질 향상을 위한 기초자료를 제공하기 위하여 홍삼 및 복분자 추출액을 첨가하여 청국장을 제조하고 이들의 이화학적 특성을 조사하였다. 수분 및 조지방의 함량은 증자콩, 일반청국장, 홍삼청국장 및 복분자청국장에서 유의적인 차이가 없었다. 환원당 함량은 홍삼청국장이 1,992.6 mg/100 g으로 가장 높았으며, 증자콩은 138.7 mg/100 g으로 청국장에 비해 그 함량이 매우 낮았다. 유리당 함량을 살펴보면, 증자콩에서는 maltose(805 mg/100 g)의 함량이 높았고, 청국장에서는 fructose와 glucose가 주된 유리당이었으며, 특히 홍삼청국장은 일반청국장에 비해 sucrose의 함량이 높았다. 증자콩 및 청국장에서 총 17종의 유리아미노산이 검출되었으며, 홍삼청국장의 유리아미노산 함량이 증자콩이나 일반청국장에 비해 높게 나타났다. L, a 및 b값은 증자콩에서 가장 높게 나타났으며, 복분자청국장의 값이 가장 낮게 나타났고, 청국장이 증자콩에 비해 경도 및 강도가 높게 측정되었다. 이러한 결과는 홍삼을 첨가한 청국장은 일반 청국장에 비해 단맛과 감칠맛 그리고 부드러운 질감을 가질 것으로 생각되며, 청국장 제조 시 홍삼을 첨가함으로써 맛과 풍미를 향상시킬 수 있을 것으로 판단된다.
