Mitochondrial heat shock protein 75 (mtHSP75) is a member of the HSP90 family and plays essential roles in refolding proteins of the mitochondrial matrix. Mitochondria provide energy in the form of ATP and generate reactive oxygen species (ROS). Heat shock proteins (HSPs) are activated in response to stress, and protect cells. In this study, we characterized the mtHSP75 of the big-belly seahorse Hippocampus abdominalis. The protein (BsmtHSP75) is encoded by an open reading frame (ORF) of 2,157 nucleotides, has 719 amino acids (aa), and is of molecular mass 82 kDa. BsmtHSP75 has two functional domains, a histidine kinase-like ATPase (HATPase_c) domain (123-276 aa) and an HSP90 family domain (302-718 aa). BsmtHSP75 was expressed in all tested tissues of healthy seahorses. The ovary contained the highest transcription level, followed (in order) by the blood, brain, and muscle. Pouch tissue showed the lowest expression level. The expression of BsmtHSP75 was significantly (P<0.05) up-regulated on viral or bacterial challenge, suggesting that BsmtHSP75 plays a role in the immune defense against bacterial and viral pathogens.
Cinnamoyl-CoA reductase (CCR, EC 1.2.1.44) catalyses the reduction of cinnamic acid CoA esters into their corresponding aldehydes, the first step of the phenylpropanoid pathway specially dedicated to monolignol biosynthesis. A cDNA clones encoding CCR have been isolated from Panax ginseng C.A. Meyer and its expression was investigated in response to abiotic stresses. The cDNA, designated PgCCR which is 865 nucleotides long and has an open reading frame of 590 bp with a deduced amino acid sequence of 176 residues. The PgCCR encoded protein possesses substantial homology with CCRs isolated and cloned from other sources; the highest identity (51.8%) was observed with CCR from Tomato (Lycopersicon esculentum). Under various stress conditions, expression patterns of the PgCCR were highly induced in adventitious and hairy roots by several abiotic stresses. These results indicated that PgCCR plays protective role against diverse environmental stresses.

Small heat shock proteins (sHSPs) are widely distributed, and their function and diversity of structure have been much studied in the field of molecular chaperones. In plants, which frequently have to cope with hostile environments, sHSPs are much more abundant and diverse than in other forms of life. In response to high temperature stress, sHSPs of more than twenty kinds can make up more than 1% of soluble plant proteins. We isolated a genomic clone, NtHSP18.3, from Nicotiana tabacum that encodes the complete open reading frame of a cytosolic class I small heat shock protein. To investigate the function of NtHSP18.3 in vitro, it was overproduced in Escherichia coli and purified. The purified NtHSP18.3 had typical molecular chaperone activity as it protected citrate synthase and luciferase from high temperature-induced aggregation. When E. coli celluar proteins were incubated with NtHSP18.3, a large proportion of the proteins remained soluble at temperatures as high as $70^{\circ}C$. Native gel analysis suggested that NtHSP18.3 is a dodecameric oligomer as the form present and showing molecular chaperone activity at the condition tested. Binding of bis-ANS to the oligomers of NtHSP18.3 indicated that exposure of their hydrophobic surfaces increased as the temperature was raised. Taken together, our data suggested that NtHSP18.3 is a molecular chaperone that functions as a dodecameric complex and possibly in a temperature-induced manner.

A general formulation for shape design sensitivity analysis over three dimensional beam structure is developed based on a variational formulation of the beam in linear elasticity. Sensitivity formula is derived based on variational equations in cartesian coordinates using the material derivative concept and adjoint variable method for the displacement and Von-Mises stress functionals. Shape variation is considered for the beam shape in general 3-dimensional direction as well as for the orientation angle of the beam cross section. In the sensitivity expression, the end points evaluation at each beam segment is added to the integral formula, which are summed over the entire structure. The sensitivity formula can be evaluated with generality and ease even by employing piecewise linear design velocity field despite the bending model is fourth order differential equation. For the numerical implementation, commercial software ANSYS is used as analysis tool for the primal and adjoint analysis. Once the design variable set is defined using ANSYS language, shape and orientation variation vector at each node is generated by making finite difference to the shape with respect to each design parameter, and is used for the computation of sensitivity formula. Several numerical examples are taken to show the advantage of the method, in which the accuracy of the sensitivity is evaluated. The results are found excellent even by employing a simple linear function for the design velocity evaluation. Shape optimization is carried out for the geometric design of an archgrid and tilted bridge, which is to minimize maximum stress over the structure while maintaining constant weight. In conclusion, the proposed formulation is a useful and easy tool in finding optimum shape in a variety of the spatial frame structures.
In this study, limestone powder (LS) and fly ash (FA) were used as powder materials in self-compacting concrete (SCC) in increasing quantities in addition to cement, so that the two powders commonly used in the production of SCC could be compared in the same study. Considering the reduction of the maximum aggregate size in SCC, 10 mm or 16 mm was selected as the coarse aggregate size. The properties of fresh concrete were determined by slump flow (including T500 time), V-funnel and J-ring experiments. The experimental results showed that as the amount of both LS and FA increased, the slump flow also increased. The increase in powder material had a negative effect on V-funnel flow times, causing it to increase; however, the increase in FA concretes was smaller compared to LS ones. The increase in the powder content reduced the amount of blockage in the J-ring test for both aggregate sizes. As the hardened concrete properties, the compressive and splitting strengths as well as the modulus of elasticity were determined. Longitudinal and transverse deformations were measured by attaching a special frame to the cylindrical specimens and the values of Poisson's ratio, initiation and critical stresses were obtained. Despite having a similar W/C ratio, all SCC exhibited higher compressive strength than NVC. Compressive strength increased with increasing powder content for both LS and FA; however, the increase of the FA was higher than the LS due to the pozzolanic effect. SCC with a coarse aggregate size of 16 mm showed higher strength than 10 mm for both powders. Similarly, the modulus of elasticity increased with the amount of powder material. Inelastic properties, which are rarely found in the literature for SCC, were determined by measuring the initial and critical stresses. Crack formation in SCC begins under lower stresses (corresponding to lower initial stresses) than in normal concretes, while critical stresses indicate a more brittle behavior by taking higher values.

Earthquakes can lead to substantial damage to buildings, with long-period ground motion being particularly destructive. The design of high-performance building structures has become a prominent focus of research. The double-story isolated structure is a novel type of isolated structure developed from base isolated structure. To delve deeper into the building performance of double-story isolated structures, the double-story isolated structure was constructed with the upper isolated layer located in different layers, alongside a base isolated structure for comparative analysis. Nonlinear elastoplastic analyses were conducted on these structures using different ground motion inputs, including ordinary ground motion, near-field impulsive ground motion, and far-field harmonic ground motion. The results demonstrate that the double-story isolated structure can extend the structural period further than the base isolated structure under three types of ground motions. The double-story isolated structure exhibits lower base shear, inter-story displacement, base isolated layer displacement, story shear, and maximum acceleration of the top layer, compared to the base isolated structure. In addition, the double-story isolated structure generates fewer plastic hinges in the frame, causes less damage to the core tube, and experiences smaller overturning moments, demonstrating excellent resistance to overturning and a shock-absorbing effect. As the upper isolated layer is positioned higher, the compressive stress on the isolated bearings of the upper isolated layer in the double-story isolated structure gradually decreases. Moreover, the compressive stress on the isolated bearings of the base isolated layer is lower compared to that of the base isolated structure. However, the shock-absorbing capacity of the double-story isolated structure is significantly increased when the upper isolated layer is located in the middle and lower section. Notably, in regions exposed to long-period ground motion, a double-story isolated structure can experience greater seismic response and reduced shock-absorbing capacity, which may be detrimental to the structure.
The suspended zipper bracing system is suggested to reduce the flaws of ordinary zipper braced and concentric inverted V braced frames. In the design procedure of suspended zipper bracing systems, columns and top story truss elements are strengthened. This bracing system show different performances and characteristics compared with inverted V braced and ordinary zipper frames. As a result, a different response modification factor for suspend zipper frames is needed. In this research paper, the response modification factor of suspended zipper frames was obtained using the incremental dynamic analysis. Suspended zipper braced frames with different stories and bay lengths were selected to be representations of the design space. To analyze the frames, a number of models were constructed and calibrated using experimental data. These archetype models were subjected to 44 earthquake records of the FEMA-P695 project data set. The incremental dynamic analysis and elastic dynamic analysis were carried out to determine the yield base shear value and elastic base shear value of archetype models using the OpenSEES software. The seismic response modification factor for each frame was calculated separately and the values of 9.5 and 13.6 were recommended for ultimate limit state and allowable stress design methods, respectively.
This paper proposes a Particle Swarm Optimization (PSO) algorithm, which is improved by making use of the Harmony Search (HS) approach and called HS-PSO algorithm. A computer code is developed for optimal sizing design of non-linear steel frames with various semi-rigid and rigid beam-to-column connections based on the HS-PSO algorithm. The developed code selects suitable sections for beams and columns, from a standard set of steel sections such as American Institute of Steel Construction (AISC) wide-flange W-shapes, such that the minimum total cost, which comprises total member plus connection costs, is obtained. Stress and displacement constraints of AISC-LRFD code together with the size constraints are imposed on the frame in the optimal design procedure. The nonlinear moment-rotation behavior of connections is modeled using the Frye-Morris polynomial model. Moreover, the P-${\Delta}$ effects of beam-column members are taken into account in the non-linear structural analysis. Three benchmark design examples with several types of connections are presented and the results are compared with those of standard PSO and of other researches as well. The comparison shows that the proposed HS-PSO algorithm performs better both than the PSO and the Big Bang-Big Crunch (BB-BC) methods.

Even todays, accurate and efficient algorithms for the large deformation analysis of elastoplastic frame structures lack due to the complexities of kinematics, material nonlinearities and numerical methods to cater for. The author suggests appropriate beam element based upon the incremental formulation from the 3D rod theory where Cauchy stress and engineering strain are variables to incorporate plasticity equations so that objectivity may be satisfied. A rectum mapping methods which can integrate and satisfy yield criteria efficiently is suggested and a continuation method which has global convergency and quadratic speed is developed as well. leading-unloading example problems are tested and the ideas are proved to be valuable.

This paper is to study elasto-plastic behavior of shear deformed braced frames. Two types of frames are considered , X-type and K-type. The slenderness ratio has been used in the parametric study. The stress-strain curve is assumed tri-linear model, and considered the strain hardening range. The finite difference method is used to solve the load-displacement relationship of the braced frames. For the elastic slope and maximum load, experimental results are compared with theoretical results and its difference remains less than 10%. Therefore suggested method in this paper is reasonable.
