Forage Quality Evaluation of Mutant Lines derived from Gamma-ray Treatments in Rubus fructicosus L. (감마선 조사 유래 블랙베리(Rubus fructicosus L.) 돌연변이 계통의 농업부산물 사료가치 분석)

  • We examined the forage quality to provide the basic information for the utilization of blackberry by-products (leaf and stem) among thirty-five mutant lines derived from gamma-ray. The leaves had higher crude protein (CP) contents than stems. In addition, the leaves showed broad ranges of variation in crude fat, crude fiber and crude ash. The overall forage quality of the stems were similar to those of the original variety. The mean of neutral detergent fiber (NDF) content of leaves and stems in mutant lines were 25.88% and 58.98%, respectively. Also, the mean of acid detergent fiber (ADF) content of leaves and stems in mutant lines were 15.43% and 49.27%, respectively. Forage quality grades of all blackberry leaf were special quality class. However, the blackberry stem ranked $1^{st}$ to $5^{th}$ in quality class. The stem diameter was negatively ($P{\leq}0.05$) correlated with the moisture and crude protein. Also, the fruit length was highly negatively ($P{\leq}0.01$) correlated with the RFV (relative feed value) in blackberry leaf. The one hundred fruit weight was highly negatively ($P{\leq}0.01$) correlated with the crude ash. Based on these results, the by-products of blackberry could provide high quality forage for feeding of live stock.

Studies on the Interspecific and Intergeneric Hybridization in Herbage Grasses II. Morphological physiological charateristics of Italian ryegrass X tall fescue hybrid (화본과목초의 종.속간 잡종에 관한 연구 II. Italian ryegrass X tall fescue F1 잡종의 형태 및 생리적 특성)

  • Tall fescue(Festuca amndinacea Schreb.) is a productive pasture grass that is widely adapted to the soil and climate of Korea. It is regarded, however, as relatively low in palatability and nutritive value. On the other hand, Italian ryegrass(lo1ium rnultz$om Lam.) is highly nutritious and palatable, but is essentially weak in cold tolerance. Numerous investigators have succeeded in producing hybrids between Lolium spp. and Festuca spp. but no improved varieties have resulted. Therefore, intergeneric hybridization of Italian ryegrass with tall fescue is employed in 1984 to improve the adaptive and cold resistance quality of Italian ryegrass. In the mean time, F1-hybrid plants obtained by crossing Italian ryegrass x tall fescue were observed in open field. The results are summarized as follows: 1. Leaf-size and silica cell hair were intermediate between the types of parents but the shape of auricle tended to show Italian ryegrass and auricle-cilia to show tall fescue. 2. Roots of $F_1$-hybrid plants were mainly distributed at top soil and did not produced florescence material. 3. Head type of hybrids was intermediate between the types of parents but ray length was somewhat less than the length of tall fescue ray. Typical Italian ryegrass or tall fescue-like phenotypes were not observed. 4. Anthers of $F_1$-hybrid plants were formed but pollen were not formed. Even though pollen were formed, they were all empty. 5. Hybrid plants were more winterhardy and summer persistency than Italian ryegrass. They showed a equivalent growth to Italian ryegrass in spring and to tall fescue in summer. 6. Chromosome of $F_1$-hybrids was allopolyploid(2n=4~=28) and all $F_1$-hybrid plants were sterile.

Fermentative products and bacterial community structure of C4 forage silage in response to epiphytic microbiota from C3 forages

  • Objective: The observation that temperate C3 and tropical C4 forage silages easily produce large amounts of ethanol or acetic acid has puzzled researchers for many years. Hence, this study aimed to assess the effects of epiphytic microbiota from C3 forages (Italian ryegrass and oat) on fermentative products and bacterial community structure in C4 forage (sorghum) silage. Methods: Through microbiota transplantation and γ-ray irradiation sterilization, the irradiated sorghum was treated: i) sterile distilled water (STSG); ii) epiphytic microbiota from sorghum (SGSG); iii) epiphytic microbiota from Italian ryegrass (SGIR); iv) epiphytic microbiota from oat (SGOT). Results: After 60 days, all the treated groups had high lactic acid (>63.0 g/kg dry matter [DM]) contents and low pH values (<3.70), acetic acid (<14.0 g/kg DM) and ammonia nitrogen (<80.0 g/kg total nitrogen) contents. Notably, SGIR (59.8 g/kg DM) and SGOT (77.6 g/kg DM) had significantly (p<0.05) higher ethanol concentrations than SGSG (14.2 g/kg DM) on day 60. After 60 days, Lactobacillus were predominant genus in three treated groups. Higher proportions of Chishuiella (12.9%) and Chryseobacterium (7.33%) were first found in silages. The ethanol contents had a positive correlation (p<0.05) with the abundances of Chishuiella, Acinetobacter, Stenotrophomonas, Chryseobacterium, and Sphingobacterium. Conclusion: The epiphytic bacteria on raw materials played important roles in influencing the silage fermentation products between temperate C3 and tropical C4 forages. The quantity and activity of hetero-fermentative Lactobacillus, Chishuiella, Acinetobacter, Stenotrophomonas, Chryseobacterium, and Sphingobacterium may be the key factors for the higher ethanol contents and DM loss in silages.

Feed Value and Fermentative Quality of Haylage of Winter Cereal Crops for Forage at Different Growing Stages (사료맥류의 생육단계별 헤일리지 사료가치 및 발효품질)

  • This study was carried out to analyze winter cereal crops for harvest time, proper drying time and haylage quality in order to investigate the possibility of the production of good quality haylage substitute for hay. As a result, in case of productivity and feed value, dry matter yield and TDN yield was increased with late harvest and crude protein and NDF, ADF was decreased (p<0.05). To make haylage for whole crop barley, oats, triticale, whole crop wheat need more than 32 hour wilting time in the milk ripe stage, yellow ripe stage need about 8~24 hour, and wilting time did not need the full ripe stage. Rye does not suitable for making haylage because of difficulty pre-wilting time. In case of fermentative quality on haylage, pH was increased with late harvest on all winter cereal crop forage. Lactic acid content was decreased with late harvest (p<0.05), and highest in the milk ripe stage. Acetic acid was also decreased with late harvest, and butyric acid was not detected. In conclusion, to harvest sooner of winter cereal crops could be an efficient way when making haylage in terms of protein content and fermentation quality. Ray can used as fresh and silage, because difficulty for pre-wilting time in Korea. Whole crop barley, oats, triticale, wheat can be used haylage when harvested yellow ripen stage and pre-wilting time for 8~24 hours.

Effect of Application Level of Swine Slurry on Production and Nutritive Value of Rye (돈분액비 시용수준이 호밀의 수량 및 사료가치에 미치는 영향)

  • Livestock manure have been utilized as fertilizer, and trying to make resources natural circulation of organic materials. This experiment was carried out to investigate the effect of swine slurry on production and nutritive value of rye, and to determine optimal application level for 2 years from Oct. 2000 to May 2002 in the experimental field of Grassland and Forage Crops Division, National Livestock Research Institute. Summary of the results were as follows. The application of swine slurry 200% Plot showed the best regrowth after winter and lodging was severe in all plots except non-fertilized plot. Plant height was lower in lodging plots. The dry matter yield was not significantly difference between chemical fertilizer(CF) plot and swine slurry application plots, and swine slurry 100% + CF 50% plot showed the highest yield among manure application plots. The dry matter yield per input nitrogen was 80kg in CF, 81kg in swine slurry 100% application plot. Above the level, it was decreased severely by 41 ∼ 61kg. Conclusively, application to swine slurry 100% showed superior productivity considering the DM yield per input nitrogen, That level would be recommended to produce higher yield and to conserve environment.

  • To develop transgenic birdsfoot trefoil (Lotus corniculatus L.) plants tolerant to environmental stress, Arabidopsis NDPK gene (AtNDPK) was introduced into birdsfoot trefoil plants using Agrobacterium-mediated transformation and expressed powerfully under the control of the E35S promoter. The expression vector, pEN-K was used for introduction of AtNDPK gene into birdsfoot trefoil plaits. The transformed calli were selected on kanamycin containing medium and then regenerated. The transformed birdsfoot trefoil plants were cultivated for 4 months on BOi2Y medium. Genomic DNA PCR and Southern blot analysis confirmed the incorporation of AtNDPK into the birdsfoot trefoil genome.

  • To develop transgenic orchardgrass (Dactylis glomerata L.) resistant to high temperature, a thermptolerance gene, DgP23, was introduced into orchardgrass using Agrobacterium - mediated transformation method. PCR and Southern blot analyses using genomic DNA showed specific DNA band on agarose gel and hybridization signal on X- ray film in transgenic orchardgrass harboring the recombinant DgP23 gene, but not in the wild type and empty vector control plants. RT-PCR and Southern blot analyses using total RNA also showed specific DNA band and hybridization signal. Transgenic orchardgrass did not showed ny morphological aberration both in the green house and field cultivation. Thermotolerance of transgenic plants was not detected in laboratory test. but may detected in field test.

  • Centipedegrass (Eremochloa ophiuroides) is a popular lawn grass in the southeastern USA. It has a naturally light green color and grows well on a wide range of soil types. Studies show limited morphological variation present in centipedegrass germplasm. To obtain the high morphological variation, plants were established from the irradiated seed at 10 Kr, allowed to interpollinate and harvested bulk seed, and then irradiated again for the next cycles. Morphological characteristics were measured in the 5 genetic varition lines (TC201 : cv. Common and non irradiated, TC202 : 4th cycles, TC241 : 6th cycles, TC306 : 8th cycles, and TC318 : 5th cycles) induced by recurrent gamma radiation. The ranges of variation of recurrently radiated centipedegrass lines < TC202, TC241 and TC306 except TC318(TifBlair) > for the stolons per plant, total stolon length per plant, longest stolon length, leaf length and width at top-most exposed internode were wider than those of non-irradiated line (TC201). Recurrent gamma radiation was very effective to enlarge the ranges of variation of morphological characteristics in reproductive organ like stolons of centipedegrass. The effect of quantity of gamma ray irradiation cycles on the means and ranges of variation in the morphological characteristics of centipedegrass was not regularly tended.

A Study on Hay Preparation Technology for Italian Ryegrass Using Stationary Far-Infrared Dryer (정치식 원적외선 건조기를 이용한 이탈리안 라이그라스 건초 조제 기술 연구)

  • This experiment was conducted to confirm the possibility of manufacturing artificial Italian ryegrass hay using far-infrared rays in Korea. The machine used in this experiment was a far-infrared ray dryer capable of adjusting temperature, airflow, and far-infrared radiation, and was conducted on Italian ryegrass harvested in May. Conditions for drying were performed by selecting a total of nine conditions, and each condition was set to emission rate of 42 to 45%, and the internal temperature was set to 65℃. The speed of the air flow in the machine was 40-60 m/s, and the overall drying time was 30 minutes for 42% radiation, 25 minutes for 43% radiation, and 20 minutes for 45% radiation. The final dry matter content according to each drying condition was 88.5% on average, and the dry matter content suitable for hay was shown in the all treatment. Looking at the power consumption according to the drying conditions, the lowest was found in the treatment that dried for 20 minutes at 45% radiation. In the drying rate, there was no difference in drying conditions 1 to 5, but a significantly low tendency was shown in conditions 6 to 7. In terms of feed value, CP and IVDMD were higher than raw materials in most drying conditions, and ADF and NDF contents were low, and tended to be high in drying conditions 4, 7, and 8. Through the above results, it was judged that drying conditions 7 and 8 were the most advantageous when considering drying speed, power consumption, and quality.
