Proline and Ammonia Accumulation in the Zoysiagrass Infected with Large Patch (라지 팻치에 감염된 잔디에서 프롤린과 암모니아의 축적)

  • To investigate the response of proline and ammonia to pathogen infection, plant growth and relevant chemical component were examined in large patch-infected or healthy (control) zoysiagrass during 6 days after treatment. Pathogen-infection increased root mortality by 30% compared to control. Soluble protein was not significantly affected by pathogen-infection except in the leaf at day 6. Ammonia concentration also increased significantly in both leaves and roots of pathogen-infected plants. Proline concentration in leaves and roots increased to 3.4- and 4.5-fold, respectively, compared to those of control at day 6. These results suggest that proline accumulation may be a sensitive biochemical indicator representing the stress intensity caused by pathogen infection in zoysiagrass.

Effect of Dicyandiamide and Hydroquinone on Ammonia and Nitrous Oxide Emission from Pig Slurry Applied to Timothy (Phleum pretense L.) Sward

  • The objective of this study was to determine the effect of nitrification inhibitor dicyandiamide (DCD) and urease inhibitor hydroquinone (HQ) on ammonia ($NH_3$) and nitrous oxide ($N_2O$) emission from pig slurry applied to Timothy (Phleum pretense L.) sward. The daily emission of ammonia ($NH_3$) and nitrous oxide ($N_2O$) was monitored for 9 days in three different treatments; 1) control (only pig slurry application), 2) DCD treatment (pig slurry + DCD), and 3) HQ treatment (pig slurry + HQ). Most $NH_3$ emission occurred after 4~5 days in three treatments. Total $NH_3$ emission, expressed as a cumulative amount throughout the measurement time, was $1.33kg\;N\;ha^{-1}$ in the control. The DCD and HQ treatment decreased total $NH_3$ emission by 16.3% and 25.1%, respectively, compared to the control. Total $N_2O$ emission in the control was $47.1g\;N\;ha^{-1}$. The DCD and HQ treatment resulted in a reduction of 67.9% and 41.8% in total $N_2O$ emission, respectively, compared to the control. The present study clearly indicated that nitrification and urease inhibitor exhibited positive roles in reducing N losses through $NH_3$ and $N_2O$ emission.

Comparison on the Changes in Non-structural Carbohydrate Concentration with Regard to Cold Tolerance of Zoysiagrass and Creeping Bentgrass (월동 기간 중 Zoysiagrass와 Creeping Bentgrass의 비구조 탄수화물 농도의 변화 비교)

  • 월동기간 동안 zoysiagrass[Zoysia matrella(L.) Merr]와 creeping bentgrass(Agrostis palustris Hunds)의 저온 스트레스에 대한 탄수화물 대사 반응을 구명하기 위해 11월부터 이듬해 4월까지의 식물체내의 탄수화물 대사산물의 변화를 비교 분석하였다. Creeping bentgrass의 녹색도와 뿌리 성장은 온도변화와 거의 평행하게 변화하였으나 zoysiagrass는 뿌리 성장에 거의 변화가 없이 3월 말까지 담황색을 나타냈다. 11월부터 이듬해 1월까지 zoysiagrass에서 glucose와 sucrose 함량은 크게 높았고, 1월에서 4월까지 starch를 제외한 모든 탄수화물 화합물들의 감소의 정도가 현저하게 낮았다. Reeping bentgrass에서 fructan의 depolymerization이 2월 이후 단당류와 이당류의 감소와 일치했으나 zoysiagrass에서는 starch 가수분해가 11월부터 일찍이 시작되었다. 이상의 결과들은 creeping bentgrass와 비교 했을때 zoysisagrass 뿌리의 높은 비구조적 탄수화물의 농도가 저온 내성과 밀접한 관계가 있음을 잘 나타내어 주고 있다.

Effects of Cattle Manure and Swine Slurry Acidification on Ammonia Emission as Estimated by an Acid Trap System

  • This study was conducted to assess the efficacy of slurry acidification in reducing ammonia emission from manure storage and application. The non-fermented cattle manure (NFC) and swine slurry (SS) were acidified by sulfuric acid and stored in an acryl chamber for 168 and 96 hours, respectively. Ammonia emitted from the chamber was collected using an acid trap system. The amount of ammonia emission was significantly reduced when the livestock manures were treated with sulfuric acid. The absolute amount of ammonia in NFC increased rapidly starting from 48 h and 72 h in the control (pH 8.6) and acidified NFC (pH 6.5), respectively. The absolute amount of ammonia was the highest at 96 h ($3.65g\;kg^{-1}h^{-1}$) in the control and at 144 h ($2.34g\;kg^{-1}h^{-1}$) in pH 6.5 NFC. The cumulative ammonia content in the control continuously increased until 96 h and was maintained until 168 h, whereas the increase rate of emission gas accumulation in acidified NFC was much less throughout the experimental period. Acidification of SS mitigated ammonia emission as proven in NFC. The cumulative amount of ammonia emission was decreased by 49.4% and 92.3% in the acidified SS at pH 6.5 and pH 5.5, respectively, compared to the control at 96 h after treatment. These results indicate that ammonia emission can be significantly reduced by sulfuric acid treatment of livestock manure during processing and the subsequent land application.

Molybdate Alters Sulfate Assimilation and Induces Oxidative Stress in White Clover (Trifolium repens L.)

  • Molybdenum (Mo) in rhizosphere influences sulfate assimilation as well as a number of other physiological aspects. In this study, the activity of key enzymes in sulfate assimilatory pathways, such as ATP sulfurylase (ATPs), adenosine 5'-phosphosulphate reductase (APR), as well as the responses of reactive oxygen species (ROS), were analyzed to elucidate the metabolic and physiological effects of external Mo supply to detached leaves of Trifolium repens L. Mo supply with a range from 1 mM to 40 mM depressed the activity of ATPs throughout the entire time course. In the leaves exposed to 1 mM Mo, a continuous decrease in the activity of ATPs was confirmed by Native-PAGE. The APR activity was also declined by Mo treatment. The accumulation of $H_2O_2$ and ${O_2}^{{\cdot}-}$ were not significant up to 10 mM Mo, whereas a remarked accumulation was detected under 40 mM Mo supply. The data suggest that an external supply of Mo has an inhibitory effect on sulfate assimilation, and induces oxidative stress only at an extremely high concentration.

Ammonification and NH3 emission in the Soil Amended with Different Animal Manures

  • Mineralization is an important biological process for conversion of organic nitrogen (N) to inorganic N which can be used by plants directly. To investigate the effect of different manures on soil mineralization, the soil amended with cattle (CtM), goat (GM), chicken manure (ChM) and pig slurry (PS) were incubated under in vitro condition and ammonium N ($NH_4{^+}-N$), ammonification rate and ammonia emission were determined for eighty-four days. $NH_4{^+}-N$ was the highest in PS-amended soil for the whole experimental period. $NH_4{^+}-N$ in PS-amended soil was gradually decreased until day 84, whereas it was rapidly decreased for the first 14 days and then slightly increased until 84 days in ChM-, CtM- and GM-amended soil. The ammonification rate showed negative value for the first 14 days in all treatments. From day 14, ammonification rate started to increase in CtM- and ChM-amended soil, whereas it was maintained in GM- and PS-amended soil until day 84. The daily ammonia emission was the highest in PS-amended soil ($41mg\;kg^{-1}d^{-1}$), followed by CtM-, ChM-, and GM-amended soil at day 1. It was gradually decreased until day 84 in all treatments. The total $NH_3$ emission was the highest in PS-amended soil with $0.6mg\;kg^{-1}$ for 84 days, while less than $0.1mg\;kg^{-1}$ in three other plots. These results indicate that different manures showed different soil ammonification rate and $NH_3$ emission.

Changes in Non-Structural Carbohydrate in Overwintering Creeping Bentgrass (Agrostis palustris) (크리핑 벤트그라스의 월동중 비구조적 탄수화물의 변화)

  • To investigate the physiological responses to naturally occurring winter freezing stress in creeping bentgrass, changes in carbohydrates were monitored during winter period. Turf quality and leaf growth was nearly parallel with temperature fluctuation. The concentration of glucose, fructose and sucrose in both shoot and root gradually increased from November to January, and then sharply decreased until April. Sucrose was the largest pool of soluble sugars. Fructan also slightly accumulated in both shoot and roots from November to February. Fructan hydrolysis in both organs was found to be much active between February to April. Shoot contained largely higher carbohydrate content in all compounds examined than roots did. Fructan was found to be a main carbohydrate storage form, showing the highest concentration (176.7 and 126.7 mg g-' DW for shoot and root in February). The depolymerization of fructan from February coincided with the high declines in mono- and disaccharide. These results suggest that the accumulation of non-structural carbohydrate until January could be associated with freezing tolerance, and the active decrease from February with shoot regrowth.

The Effect of Pre-Wilting Herbage on the Composition and Feeding Value of Silage (예건의 Silage의 품질에 미치는 영향)

  • Experiment were conducted to study effect of pre-wilting on the quality of silage. Herbage samples were taken on November 11, 1981. Italian ryegrass and rye plant at grazing stage were taken and wilted to adjust their dry matter contents to the range of 15.8 to 53.2% and ensiled in order to evaluate the effect of pre-wilting on the chemical composition and nutrient loss of silage. Both prewilted and unwilted grass material were ensiled in small polyetylene film bag and stored under room temperature. 1. The formation of total acid decreased by increasing DM content, however, lactic acid content of the total organic acid increased. 2. In the silage, the loss of DM declined as DM content increased. 3. The quality of silage estimated by organic acid constituent was improved as DM content in the herbage increased. 4. The production of $NH_3-N$ decreased by increasing degree of prewilting. 5. In vitro dry matter digestibility was not affected by prewilting.

Comparison of Sulfur Utilization among 10 Different Genotypes of Rape (Brassica napus) (다양한 유채품종에서의 황의 이용 효율 비교)

  • To investigate the sulfate utilization efficiency in different rape (Brassica napus) cultivars, sulfate uptake are analyzed under complete S-supply level (2.0mM ${SO_4}^{2-}$). This study used ten different genotypes of rape (Mokpo, Tamra, Youngsan, Naehan, Saturnin, Akela, Mosa, Capitol, Pollen and Colosse). For comparison of ${SO_4}^{2-}$ uptake among 10 cultivars, leaf number, leaf length and width, root length was also observed. Leaf length and width in all cultivars less variable among the cultivars examined. The longest root was shown in Saturnin (36.3 cm). ${SO_4}^{2-}$ uptake in Saturnin, Youngsan and Mokpo was significantly higher whereas that of Mosa and Pollen was relativety lower. Saturnin and Mokpo which have a high ${SO_4}^{2-}$ uptake exhibited a high ${NO_3}^-$ uptake.

Different Levels of N Supply Impacts on Seed Yield by Modulating C and N Metabolism in Brassica Napus

  • Oilseed rape is known to crop having low nitrogen use efficiency (NUE) but requires high levels of N fertilizer. NUE is associated with N remobilization from source to sink organ, consequently affects seed yield. Remobilization of leaf N is also related to transport of C/N metabolites in phloem. However, interaction between seed yield and phloem transport was not fully documented. In response to seed yield, N and C metabolites and their transport into seed from bolting to pod filling stage investigated in two contrasting genotypes (Capitol and Pollen) cultivated under ample (HN) or limiting nitrate (LN) supply. Seed yield was significantly reduced in N limitation and its reduction rate was much lower in Capitol than in Pollen compared to HN treated plants. Amino acid and protein content was higher in Capitol than in Pollen at bolting stage. They gradually decreased during plant development but not significant between two cultivars and/or two treatments. Glucose, fructose and sucrose content were 1.8-,1.6- or 1.25-fold higher in LN condition than in HN condition, respectively. Amino acid and sucrose content in phloem were largely higher in Capitol than in Pollen under LN condition. These results indicate that the higher seed yield might be related to greater transport ability of amino acid and sucrose in phloem under LN condition.
