Optimization of Analytical Methods for Ochratoxin A and Zearalenone by UHPLC in Rice Straw Silage and Winter Forage Crops (UHPLC를 이용한 볏짚 사일리지와 동계사료작물의 오크라톡신과 제랄레논 분석법 최적화)

  • The objective of this study was to optimize analytical methods for ochratoxin A (OTA) and zearalenone (ZEA) in rice straw silage and winter forage crops using ultra-high performance liquid chromatography (UHPLC). Samples free of mycotoxins were spiked with $50{\mu}g/kg$, $250{\mu}g/kg$, or $500{\mu}g/kg$ of OTA and $300{\mu}g/kg$, $1500{\mu}g/kg$, or $3000{\mu}g/kg$ of ZEA. OTA and ZEA were extracted by acetonitrile and cleaned-up using an immunoaffinity column. They were then subjected to analysis with UHPLC equipped with a fluorescence detector. The correlation coefficients of calibration curves showed high linearity ($R^2{\geq_-}0.9999$ for OTA and $R^2{\geq_-}0.9995$ for ZEA). The limit of detection and quantification were $0.1{\mu}g/kg$ and $0.3{\mu}g/kg$, respectively, for OTA and $5{\mu}g/kg$ and $16.7{\mu}g/kg$, respectively, for ZEA. The recovery and relative standard deviation (RSD) of OTA were as follows: rice straw = 84.23~95.33%, 2.59~4.77%; Italian ryegrass = 79.02~95%, 0.86~5.83%; barley = 74.93~97%, 0.85~9.19%; rye = 77.99~96.67%, 0.33~6.26%. The recovery and RSD of ZEA were: rice straw = 109.6~114.22%, 0.67~7.15%; Italian ryegrass = 98.01~109.44%, 1.65~4.81%; barley = 98~113.53%, 0.25~5.85%; rye = 90.44~108.56%, 2.5~4.66%. They both satisfied the standards of European Commission criteria (EC 401-2006) for quantitative analysis. These results showed that the optimized methods could be used for mycotoxin analysis of forages.

Effect of Grass Filter Strips on NO3-N in Runoff from Forage Cropland (사료작물 재배지에서 초지식생대를 이용한 NO3-N 저감효과에 관한 연구)

  • The performance of grass filter strips (GFS) in abating $NO_3$-N concentrations from the forage cropland was tested in an experiment on the 10% slope in Grassland and Forages Division, National Institute of Animal Science, Rural Development Administration (RDA) from October 2007 to September 2009. Forage croplands with rye-corn double cropping system applied with chemical fertilizer and livestock manure (LM) were compared in a natural condition. The plots were hydrologically isolated in a randomized block layout of 3 treatments $\times$ 2 factors $\times$ 3 replicates. Main plots consisted of the length of GFS, such as 0 m, 5 m, 10m and 15m. Sub plots consisted of the type of LM, such as chemical fertilizer (CF), cattle manure (CM) and swine manure (SM). Dry matter yields of rye and corn increased significantly in order of CF > CM > SM (p<0.05). Concentrations of $NO_3$-N in surface runoff water were reduced as the length of GFS increased. Especially, GFS with 10 m and 15m reduced $NO_3$-N concentrations significantly compared to that with 0 m and 5 m (p<0.05). The results from this study suggest that GFS improved the removal and trapping of manure nutrients from forage croplands.

Effects of Forage Cropping System and Cattle Slurry Application on Productivity of Rice and Forage Crops and Nutrient Movement in Paddy Land (녹비작물의 경작형태와 우분액비시용이 벼와 사료작물의 생산성 및 양분이동에 미치는 영향)

  • This study was conducted to investigate the effects of green manure crops and cattle slurry application on rice productivity and nutrient movement in paddy land. Cropping systems used in this study were consisted of five designs, such as mono-cropping rice applied with standard fertilizer (MRS), double-cropping whole crop barley following rice applied with cattle slurry (DWBRC), double-cropping whole crop rye following rice applied with cattle slurry (DWRRC), mono-cropping rice applied with following milk vetch (MRMV) and mono-cropping rice applied with following hairy vetch (MRHV). The field experiments were conducted on the clay loam at Backsanmyun, Kimje, Chunlabukdo province in Korea for three years (May 2006 to Apr. 2009). This study was arranged in completely randomized design with three replicates. Culm and panicle length of rice were higher in MRS than other treatments. Ripened grain ratio was higher in MRS than other treatments. Yield of Grain showed high in MRS, DWBRC and DWRRC than MRMV and MRHV. The yield of dry matter (DM) of whole crop barley in DWBRC increased significantly as compared with that of rye in DWRRC. The pH, and contents of T-N, $P_2O_5$ and organic matter (OM) and exchangeable cation (Ca, Na, Mg and K) in soil samples collected at the end of the experiment were remarkably higher than those at the beginning of the experiment. The concentrations of $NO_3$-N, $NH_4$-N and $PO_4$ in discharge water in DWBRC and DWRRC were higher than MRS.

Effects of Application Seasons and Levels of Cattle Slurry Nitrate Contents in Soils of Rye CuItivation (액상발효우분의 시용시기와 시용량이 호밀경작지 토양의 NO3- 함량에 미치는 영향)

  • This experiment was conducted to investigate the effects of application seasons and the level of cattle slurry on nitrate contents in soils of rye cultivation under climatic conditions of Korea. The main plots were appliaction seasons(100% application in spring, 100% application in autumn, 50% application in spring and autumnn after dividing cattle slurry into two equal parts) and subplots were the levels of cattle slurry-N application(0, 50, 100 and $200kgN\;ha^{-1}\;year^{-1}$). Cattle slurry was spred evenly over the soil surface in spring (end of March) and autumn(early of November). The experiment was done at Gongiam, Kwangju, Kyung-gi for 3 years, from 1994 to 1996. The results obtained are summarized as follows; 1. Nitrate content in soils was not influenced by application seasons of cattle slurry. 2. Nitrate content in soils increased as the level of cattle slurry application increased, and the average nitrate content became 3.6mg per liter. 3. Nitrate content in the soil depth from 0 to 20cm increased as the level of cattle slurry application increased, and the average nitrate content became 28.9mg per liter. However, nitrate content in the soil depth from 40 to 60cm was the lowest(7.5-8.3mg per liter).

The farm management analysis of the effect of yield increase and economic efficiency of cropping system on green manure crops - cabbage in the plastic house (녹비작물-양배추 작부체계에서 양배추 수량증대 효과 및 경영성과 분석)

  • This study was carried out to analyze the effect of the farm management and productivity increase of cabbages growing in the plastic house based on application of green manure crops such as silage corn, haussolgo (Sorghum bicolar L. Moench), hairy vetch, etc. According to the effect of green manure crop grown in summer season, the biomass amount of the silage corn was the highest; 7,630 kg per 10a, the next was haussolgo, 5,620 kg per 10a. In terms of the fertilizer ingredients, the first of forage soybean was 3.84% of nitrogen, whereas hairy vetch was 1.74% of phosphate and kalium, 4.74%. Productivity increase of cabbages was the highest in the haussolgo plot of which the yield of showed 10,090 kg per 10a and the farm household income would be worth 8,053 thousand won. By growing forage crops in the winter season, the biomass amount was the highest in the mixed sowing plot with rye (50%) and hairy vetch (50%) of 3,590 kg per 10a, whereas the productivity in the mixed seeding with rye (70%) and hairy vetch (30%) was highest, 6,249 kg per 10a and the farm household income would be worth 7,387 thousand won. Judging from these results, more practical on-farm research on applying different green manure crops as a basal fertilizer with cabbage in the plastic house should be considered to analyze the farm management and the farm household income at different sites.

Effects of Supplementing Whole Oilseeds to Alcohol-Fermented Feedstuff Based on Rye Hay on in vitro Rumen Fermentation Characteristics (호밀 건초 급여하의 By-pass 전지종실을 첨가한 알코올 발효사료의 in vitro 발효특성)

  • This study was conducted to determine the effects of supplementing whole oilseeds to the alcohol fermented feedstuff based on rye hay on in vitro rumen fermentation of Hanwoo steers. Experiments were arranged with four treatment groups, T1 (without any oilseeds), T2 (supplemented with cottonseed), T3 (supplemented with linseed) and T4 (supplemented with soybean). The pH decreased at 6 and 12h in vitro incubation in T2 and at 3 and 12h in vitro incubation in T4 (p<0.05). However, no decrease of the pH was observed at any in vitro incubation in T3 (p>0.05). Ammonia concentration increased at 6h in vitro incubation in T2 and at 6 and 9 h in vitro incubation in T4 (p<0.05). Concentrations of acetate, butyrate and propionate increased at 3, 9 and 12h in vitro incubation in T4 and at 3h in vitro incubation in T3 (p<0.05). This study indicates that the supplementation of whole oilseeds to alcohol fermented feedstuff based on rye kay can improve the in vitro rumen fermentation.

Growth Characteristics and Productivity of Winter Crops After the Continuous Whole Crop Rice Cultivation in Paddy Field in Middle Region (중부지역 답리작에서 동계 사료작물의 조기파종 효과)

  • This study was conducted to determine the effects of seeding date on agronomic characteristics, productivity and feed value of Italian ryegrass, barley and rye during cropping after rice in Suwon, middle region of Korea, during 2004 and 2005. In general, early seeding (26 September) showed longer plant length and branch numbers per square meter than late sowing (11 October). Italian ryegrass had more early seeding effect than barley and rye. The yield of each plant was affected by sowing date in all experiments: the earlier seeding had higher yields than late seeding. In case of Italian ryegrass and barley as late-heading varieties showed more early seeding effect than rye. On the other hand, feed value tended to respond differently to yields. The feed value of each plant were based on liquid swine manure (12%) > composted cattle manure (7%) > composted swine manure (2%). The present results highlight the earlier planting obtained higher yield compared to those planted conventionally.

Effect of Application of Pig Manure Slurry to Rye on Rice Productivity in Paddy-land (답작지대에서 돈분액비에 의한 호밀 생산시 후작 벼의 생산성에 미치는 영향)

  • This study was conducted to examine the effect of application of pig manure slurry(PMS) for rye cultivation in Paddy-land on rice productivity The field experiments were carried out from 1999 to 2002 on well-grown rice in Paddy-land at Kimje, Chunbuk province in Korea. PMS was manufactured by the fermentation for 6 months in natural condition. Amount of PMS was adjusted equal to total nitrogen value relative to its value of commercial chemical fertilizers. The amount of PMS were spread evenly over the soil surface two times; first in the autumn (before sowing, end of November) and second in the spring(regrowth of rye, middle of March). The field plots were consisted of four kinds (PMS $100\%$ treatment, PMS $200\%$ treatment PMS $100\%$ treatment with half of CF (CF $50\%$), Full of CF treatment (CF $100\%$). The experiment was designed as a randomized complete block. Each treatment was replicated three times. Rices are customarily gown under tropical korean condition as annuals. The results obtained from this study were summarized as fellows. Panicle length of rice was shorter in PMS $100\%$ than other treatments. Ripening grain ratio was the lower in CF treatment. In treatment of PMS $100\%$ and PMS $200\%$, 1000 grain weight was a downward trend as compared to that of full of CF treatments, whereas number of grains per $m^2$ was increased as compared to other treatments. Number of panicles reveals that there is an increase in order; CF $50\% + PMS\;100\%>\;PMS\;100\%\;>\;CF\;100\%\;>\;PMS\;200\%$. Yield of milled rice showed hi인 in CF $100\%$ and $CF 50\%+ PMS 100\%$, but were was no significant difference among treatments. There were did not a regular tendency in the ratio of leaf sheath bright, however the ratio of lodging area was higher in $CF50\%+PMS\;100\%$ treatment than other treatments. Based on the results of this experiment, $CF 50\%+PMS\;100\%$ may be the most effective in rice productivity under application of PMS for rye cultivation in Paddy-land.

Effects of Nitrogen Level on Nitrogen Partitioning and Harvest Index in Brassica napus L.

  • To investigate the impact of nitrogen (N) mineral on reproductive potential of Brassica napus L, plants were treated with different levels of N treatment ($N_0$; $N_{100}$; $N_{500}$). The half of N content for each treatment were applied at the beginning of the early vegetative stage and the rest was applied at the late vegetative stage. Nitrogen content in plant tissues such as root, stem and branch, leaf, pod and seed was analyzed and harvest index (HI) was calculated as percentage of seed yield to total plant weight. Biomass and nitrogen content were significantly affected by different levels of N supply. Biomass was significantly decreased by 59.2% in nitrogen deficiency ($N_0$) but significantly increased by 50.3% in N excess ($N_{500}$), compared to control ($N_{100}$). Nitrogen content in all organs was remarkably increased with nitrogen levels. N distribution to stem and branches, and dead leaves was higher in N-deficient ($N_0$) and N excessive plants ($N_{500}$) than in control ($N_{100}$). However, nitrogen allocated to seed was higher in control ($N_{100}$) than in other treatments ($N_0$ or $N_{500}$), accompanied by higher HI. These results indicate that the optimum level of N supply ($N_{100}$) improve HI and N distribution to seed and excessive N input is unnecessary.
