Proteomic Response of Alfalfa Subjected to Aluminum (Al) Stress at Low pH Soil

  • In order to reveal the aluminum (Al) stress tolerance mechanisms in alfalfa plant at low pH soil, a proteomic approach has been conducted. Alfalfa plants were exposed to Al stress for 5 days. The plant growth and total chlorophyll content are greatly affected by Al stress. The malondialdehyde (MDA) and $H_2O_2$ contents were increased in a low amount but free proline and soluble sugar contents, and the DPPH-radical scavenging activity were highly increased. These results indicate that antioxidant activity (DPPH activity) and osmoprotectants (proline and sugar) may involve in ROS ($H_2O_2$) homeostasis under Al stress. In proteomic analysis, over 500 protein spots were detected by 2-dimentional gel electrophoresis analysis. Total 17 Al stress-induced proteins were identified, of which 8 protein spots were up-regulated and 9 were down-regulated. The differential expression patterns of protein spots were selected and analyzed by the peptide mass fingerprinting (PMF) using MALDI-TOF MS analysis. Three protein spots corresponding to Rubisco were significantly down-regulated whereas peroxiredoxin and glutamine synthetase were up-regulated in response to Al stress. The different regulation patterns of identified proteins were involved in energy metabolism and antioxidant / ROS detoxification during Al stress in alfalfa. Taken together, these results provide new insight to understand the molecular mechanisms of alfalfa plant in terms of Al stress tolerance.

Study on Animal Production and Disease Affected by Different Varieties of Tall Feacue and White Clover in Mixed Grazing Pasture II. Effect on pasture production, nutrutuve value, botanical composition, and soil characteristics (혼파방목지에서 Tall Fescue와 두과목초 조합에 따른 가축생산성과 질병 비교 연구 II. 목초의 생산성 , 사료가치 , 식생구성율 및 토양특성에 미치는 효과)

  • This study was carried out with aim to find out a tall fescue cultivar which is palatable and resistant to hot climate, and a white clover cultivar which is less competitive with other grasses. Pastures with 3 treatments (TI: Fawn, tall fescue + Regal, white clover + orchardgrass + perennial ryegrass, T2: Roa, tall fescue + Regal, white clover + orchardgrass + perennial ryegrass, T3 : Roa, tall fescue + Tahora, white clover + orchardgrass + perennial ryegrass) were established by oversowing. Under continuous grazing, dry matter yield, soil characteristics, botanical composition and nutrient contents of forages were investigated during the grazing seasons 60m 1994 to 1996. No significant difference were found in establishment percents, physical and chemical characteristics of soil, and nutrient contents of forages between 3 treatments(P>0.05). Dry matter yield of TI, T2 and T3 were 24,188, 23,827 and 23,578kg/ha, respectively. Fawn and Roa, tall fescue cultivars occupied 6.1 and 4.8% of the total plants in 1994, 14.4 and 11.5 % in 1995, and 28.4% and 17.7% in 1996, respectively. These results indicate that Fawn, tall fescue dominantly occupied with time more than Roa, tall fescue. There were no differences in percentage of Regal and Tahora, white clover cultivars(P > 0.05), showing 4.2 and 2.4% in 1994, 16.1 and 17.3% in 1995, and 2.4 and 1.0% in 1996, respectively. The percentage of orchardgrass on mixed pasture decreased after summer season of high temperature, but percentage of Roa, tall fescue slowly increased with time firom 1994 to 1996.

Effects of Inoculation of Mycorrhiza and Rhizobium on N, P utilization and Vegetative Growth in Alfalfa/Perennial Ryegrass Intercropping (Mycorrhiza 및 Rhizobium 접종이 알팔파-페레니얼 라이그라스 혼파에 의한 질소와 인의 이용성 및 성장에 미치는 영향)

  • To investigate the effect of Mycorrhiza (Glomus intradics) and Rhizobium inoculation on the N, P utilization and growth response of Alfalfa (Medicago saliva L.) and Perennial ryegrass (Lolium perenne L.) in mixed sward, four treatments (non-inoculation, Control; Mycorrhiza inoculation: M; Rhizobium inoculation, R and Mycorrhiza and Rhizobium inoculation, M+R) were applied. The associated analyses were canied out on the early vegetative growth stage (DAS 56, 56 days after seeding) and on the early flowering stage (DAS 126). The decreased rate of total N and P content in soil, with advancing plant growth, was relatively higher in the M and M + R treatment than control. The content of availabie phosphorus in soil at DAS 126 increased by about 34 and 38 % in M and M+R treatment compared to control (189.2 mg PzOskg DM), while non-significant changes was observed in R treatment. Total N uptake and P uptake in the control at DAS 126 were 44.71 and 3.52 mglplant in mixed sward, respectively. About 71, 98 and 197 % of increases in total N uptake and 70, 72 and 11 1 % of increases in total P uptake were estimated in M, R and M+R treatment. Comparing to control, total dry matter yield significantly increased by 27, 33 and 53 %, and crude protein yield also by 78, 83 and 204 %, respectively, in M, R and M+R treatment. The present data indicated that mycorrhiza orland rhizobium inoculation improved N, P utilization of both alfalfa and perennial ryegrass plants, and consequently increased total yield (especially by dual inoculation, M+R). Effect of Copper on the Plant Regeneration from Seed Derived Callus of Orchardgrass (Dactylis glomerata L.) (오차드그래스의 종자유래의 캘러스로부터 식물체 재분화율에 미치는 Copper의 영향)

  • This study was conducted to investigate the effects of copper in the efficiencies of callus formation and plant regeneration of orchardgrass (Dactylis glomarata L.). Seeds were cultured on MS medium containing $2\;mg/{\ell}$ 2,4-D and different concentrations ($0.1-100\;{\mu}mol$) of copper sulfate. Plant regeneration was achieved on N6 medium containing $1\;mg/{\ell}$ NAA, $5\;mg/{\ell}$ kinetin and $0.1-100\;{\mu}mol$ of copper sulfate. Callus formation was not affected by copper incorporation into MS medium. However, the efficiency of plant regeneration was promoted by copper and the maximum efficiency was obtained when $70\;{\mu}mol$ copper was incorporated in the culture medium. The average number of regenerated plants from the seed-derived callus was also increased by copper.

Effects of pasture Type on Behavior pattern , Change of Body Type and Daily Gain of Korean native Goat (방목지 유형이 재래산양의 행동양식 , 체형변화 및 증체량에 미치는 영향)

  • This experiment was carried out to evaluate the effect of pasture type on behavior pattern, change of body type and daily gain of Korean native goat at the College of Natural Science of Kon-KuK University in Chungju. The results are summarized as follows : Relative coverage of native herbage at the pasture area was 62% in initial grazing period and that final grazing period decreased by 36%. while pasture plants increased from 17 to 42%. In brush area, Quercus serrata. Quercus dentutu, Prunus surgenrii and Rhus vemicrflua of high palatability decreased with increase in grazing period. but Rhododendron mucronulatum and Rhodendrorl schlippenbachii of low palatability tended to increase. Average crude protein content at the pasture area(12.3Q) during grazing period was higher than that at the brush area(10.2). However, ADF and NDF content was lower than brush area. Ca, Mg and Na of mineral content showed highly at the brush area during the grazing period. but Na showed highly at the pasture area. Eating time Korean native goat was not different between pasture area(369 min.) and brush area(361 min.), but ruminating time showed highly at the pasture area(441 min.). Average ratio of GTlRT at the pasture and brush area was not different m 0.88 and 0.87, respectively. All body type(withers height, body length, shin circum, hip height, hip width and chest girth) showed highly that pasture area compared to brush area during the grazing periods. The daily liveweight gain showed 60.6 and 48.98 at the pasture and brush area, respectively.

  • This experiment was conducted to investigate the morphological and structual characters and the dry weight of plant (DW/pl.) concerned vegetational structure of orchardgrass pastures by the times of year, from 1 year to 7 year previous, 1986 through 1980, alternately. this experiment was carried out on the experimental fields of Yonsei University. The results are summarized as follows: 1. Population density (PD) was extremely decreased by the times of year. 2. Pasture productivity was highest at 3 year old pasture, was lowest at 5 year old pasture and was recovered at 7 year old pasture. 3. Recovery of pasture productivity was followed the increase of dry weight of plant (DW/pl.) and number of tillers per plant (NT/pl.) 4. The population density of 7 year old pasture was 14 plants per square meter. 5. The number of tillers per plant (NT/pl.), dry weight of plant (DW/pl.) plant length (PL), and plant size (PS) was frequently showed the highest values by the times of established year. 6. The stubble diameter (DIA), stubble area (SB), plant size (PS), distance between neibour plant (DIS) and leaf area index (LAI) was increased according to the times of year. 7. The dry weight of plant (DW/pl.) was positively significant correlations with the number of tillers per plant (NT/pl.) all of the established pastures, but the dry weight of a tiller (WT) was tended to increase of correlations with the dry weight of plant (DW/pl.) by the times of year. 8. Differences between morphological and structual characters was recognized according to the times of year.

  • The conditions for callus formation and plant regeneration were confirmed in birdsfoot trefoil (Lotus corniculatus L.). Among SH (Schenk and Hildebrandt), MS (Murashige and Skoog) and N6 medium (Chu), SH medium was highest degree of efficiencies respectively in callus formation and plant regeneration. In this study, we determined volume of hormones and other compounds appended in media. For callus formation, only $3mg/{\ell}$ of 2,4-D (2,4-dichlorophenoxy acetic acid) was appended in their media. For plant regeneration, we used BOi2Y medium (Bingham et al.). We obtained birdsfoot trefoil plants from callus by regeneration, about sixty days later transfer calli to regeneration media.

Proline and Ammonia Accumulation in the Zoysiagrass Infected with Large Patch (라지 팻치에 감염된 잔디에서 프롤린과 암모니아의 축적)

  • To investigate the response of proline and ammonia to pathogen infection, plant growth and relevant chemical component were examined in large patch-infected or healthy (control) zoysiagrass during 6 days after treatment. Pathogen-infection increased root mortality by 30% compared to control. Soluble protein was not significantly affected by pathogen-infection except in the leaf at day 6. Ammonia concentration also increased significantly in both leaves and roots of pathogen-infected plants. Proline concentration in leaves and roots increased to 3.4- and 4.5-fold, respectively, compared to those of control at day 6. These results suggest that proline accumulation may be a sensitive biochemical indicator representing the stress intensity caused by pathogen infection in zoysiagrass.

  • Humic substances that can be obtained from coal resources such as leonardite in a bulk scale have been employed as crop stimulators and soil conditioners. The polymeric organics containing a variety of aromatic and aliphatic structures are known to activate plants in a multifunctional way, thus resulting in enhanced germination rate and abiotic stress resistance concomitant with induction of numerous genes and proteins. Although detailed structural-functional relationship of humic substances for plant stimulations has not been deciphered yet, cutting-edge analytical tools have unraveled critical features of humic architectures that could be linked to the action mechanisms of their plant stimulations. In this review article, we introduce key findings of humic structures and related biological functions that boost plant growth and abiotic stress resistance. Oxygen-based functional groups and plant hormone-like structures combined with labile and recalcitrant carbon backbones are believed to be critical moieties to induce plant stimulations. Some proteins such as HIGH-AFFINITY $K^+$ TRANSPORTER 1, phospholipase A2 and $H^+$-ATPase have been also recognized as key players that could be critically involved in humic substance-driven changes in plant physiology.

Nutritional Characteristics of Forage Grown in South of Benin

  • In order to provide recommendations on the most useful forage species to smallholder farmers, eleven grass and eleven legume forages grown in Abomey-Calavi in Republic of Benin were investigated for nutritive value (i.e. chemical composition and energy content) and fermentation characteristics (i.e. gas and volatile fatty acid production, organic matter degradability). The in vitro gas production technique was used, incubating the forages for 120 h under anaerobic condition with buffalo rumen fluid. Compared to legume, tropical grass forages showed lower energy (8.07 vs 10.57 MJ/kg dry matter [DM]) and crude protein level (16.10% vs 19.91% DM) and higher cell wall content (neutral detergent fiber: 63.8% vs 40.45% DM), respectively. In grass forages, the chemical composition showed a quite high crude protein content; the in vitro degradability was slightly lower than the range of tropical pasture. The woody legumes were richer in protein and energy and lower in structural carbohydrates than herbaceous plants, however, their in vitro results are influenced by the presence of complex compounds (i.e. tannins). Significant correlations were found between chemical composition and in vitro fermentation characteristics. The in vitro gas production method appears to be a suitable technique for the evaluation of the nutritive value of forages in developing countries.
