Microbial Evaluation of Commercially Packed Kimchi Products

  • Commercially packed kimchi products from 6 different manufacturers, which are exported overseas as well as sold domestically, were analyzed to determine their microorganism distributions and presence of pathogenic bacteria. All samples showed decreasing pH levels (from 5.7-6.2 to 3.9-4.3) and increasing titratable acidities (from 0.3-0.4 to 0.8-1.2%) during 15 days of storage at $4^{\circ}C$. Total bacterial counts ranged from $2.1{\times}10^5-1.9{\times}10^6\;CFU/mL$ in the initial kimchi samples, and then increased to $1.1{\times}10^8-1.8{\times}10^9\;CFU/mL$. The coliform numbers decreased from approximately $2.5{\times}10^2-1.7{\times}10^4\;CFU/mL$ to zero. Major foodborne pathogens such as Salmonella spp., Staphylococcus aureus, Escherichia coli O157:H7, Listeria monocytogenes, Bacillus cereus, Yersinia enterocolitica, and Shigella spp. were not detected in any of the samples. However, 2 out of the 6 samples carried E. coli, emphasizing the need for improved hygiene practice. Interestingly, Hafnia alvei, belonging to the Enterobacteriaceae family, was isolated in all of the samples. Further study is needed on this newly reported bacterium in kimchi.

Physical, Mechanical, and Antimicrobial Properties of Edible Film Produced from Defatted Soybean Meal Fermented by Bacillus subtilis

  • In order to extend the shelf-life of packaged or coated foods, an antibacterial edible film was developed from soybean meal that had been fermented with Bacillus subtilis under the optimum condition of pH 7.0-7.5 and $33^{\circ}C$ for 33 h. The water vapor permeability of the fermented film ($86.0 mg/cm^2{\cdot}h$) was higher than those of normal soybean films ($66.9 mg/cm^2{\cdot}h$). Protein solubility of the fermented film was also higher than ordinary soy protein film at the pH range of 3 -10. The fermented soybean film had higher tensile strength and lower $\%$ elongation (elongation rate) than the ordinary soybean film, mainly because partial hydrolysis of proteins in the soybean film occurred during fermentation. Antimicrobial properties of the fermented film on foodstuffs were measured by placing the films on surime, jerked beef, and mashed sausage media; containing $10^2-10^3$ CFU/plate of foodborne pathogenic bacteria, and showed significantly higher inhibitory effects on the growths of all the indicating bacteria. The film could be used as a packaging material in the food industry. However, before direct application of the fermented film to the commercial food industry, its poor mechanical and antibacterial properties need to be improved.

Simple Evaluation of Listeria monocytogenes Pathogenesis Using Caenorhabditis elegans Animal Model

  • Listeria monocytogenes is a major cause of serious foodborne illness in the dairy foods. Although Caenorhabditis elegans model is well established as a virulence model of pathogenic bacteria, its application on L. monocytogenes is critically unclear. The objective of this study was to carry out an evaluation of L. monocytogenes toxicity using C. elegans nematode as a simple host model. We found that C. elegans nematodes have high susceptibility to L. monocytogenes infection, as a consequence of accumulation of bacteria in the worms' intestine. However, L. innocua, which is known to be non-toxic, is not accumulate in the intestine of worms and is not toxic similarly to Escherichia coli OP50 known as the normal feed source of C. elegans. Importantly, immune-associated genes of C. elegans were intensely upregulated more than 3.0-fold when they exposed to L. monocytogenes. In conclusion, we established that C. elegans is an effective model for studying the toxicity of L. monocytogenes and we anticipate that this system will result in the discovery of many potential anti-listeria agents for dairy foods.

가열 햄 및 소시지류 제조공장에서 식중독 세균에 대한 오염 분석 (Analysis of Foodborne Pathogenic Contamination of Cooked Hams and Sausages in Korean Processing Facilities)

  • 현재 국내 일부 햄과 소시지류 제조공장에서 식중독 세균에 대한 제어방안으로서 1차 가열살균 이후 제품포장 단계에서 2차 가열살균을 추가적으로 실시하고 있다. 이에 본 연구에서는 2010년 2월부터 11월까지 국내 10개 육가공제조회사에서 햄류 53개 품목과 소시지류 37개 품목의 동일 롯트에서 1차 가열 시료와 2차 가열까지 실시한 시료를 각각 6개씩 채취하여 축산물의 가공기준 및 성분규격에 따라 Salmonella spp., S. aureus, L. monocytogenes 및 C. perfringens 식중독 세균 오염 여부를 조사하였다. 햄과 소시지류 총 1,080건에 대한 검사 결과 S. aureus는 2개 제조회사에서 생산한 햄 4개 시료에서 검출되었으며, 그 중 3건은 1차 가열제품에서 나머지 1건은 2차 가열살균까지 처리한 제품에서 검출되었다. L. monocytogenes는 5개 제조회사에서 햄류 4건, 소시지류 8건을 포함하여 총 12건이 검출되었으며, 그 중 7건은 1차 가열 처리한 제품에서, 나머지 5건은 2차 가열까지 처리한 제품에서 각각 검출되었다. C. perfringens는 3개 제조회사의 햄류 1건과 소시지류 2건에서 검출되었으며, 1차 가열만 한 제품에서 1건, 2차 가열까지 한 제품에서 2건이 각각 검출되었다. 이에 반하여 Salmonella spp.는 한 건도 검출되지 않았다. 1차 가열살균 제품과 1차와 2차 가열살균 과정을 모두 처리했을 때를 비교하면 3가지 식중독 세균의 검출률에 있어서 차이가 없는 것으로 분석되었다(p<0.05). 또한, 제조 회사의 햄과 소시지류에서 분리한 L. monocytogenes 균주를 대상으로 유전적 다양성을 조사하기 위하여 PFGE를 실시한 바, 동일 가공장에서 분리된 균주들간 80% 이상의 높은 상동성을 가진 것으로 조사되었다. 이러한 결과에 비추어 볼 때 햄 및 소시지류 제조회사에서 식중독 세균의 제어를 위해서는 원료 및 제조단계에서부터 가공 과정에서의 교차 오염을 예방하기 위한 적절한 위생관리와 철저한 모니터링을 통하여 체계적인 식중독 세균에 대한 위생관리를 완성하여야 할 것으로 판단된다.

Mulberry Low-Fat Ice Cream Supplemented with Synbiotic: Formulation, Phytochemical Composition, Nutritional Characteristics, and Sensory Properties

  • For this study, we designed and produced mulberry low-fat ice cream supplemented with synbiotics (MLF-ISS). The sensory characteristics and physical, chemical, and microbiological qualities of MLF-ISS were then determined. Mulberry juice inoculated with or without probiotic (Lactobacillus plantarum TISTR 926 and Saccharomyces boulardii CNCM I-745) was also tested at 37℃ for 24 h to determine probiotic growth rate, pH, total anthocyanin content (TAC), total phenolic content (TPC), and antioxidant activity (AA). Only the TAC of mulberry juice inoculated with S. boulardii CNCM I-745 increased considerably (p < 0.05) among these parameters. MLF-ISS was produced with varied mulberry fruit concentrations (0, 10, 20, 30, or 40%) (w/w). The MLF-ISS prepared with 30% mulberry fruit (w/w) (30-MLF-ISS) had a higher score in appearance, color, and sweetness (p < 0.05) when sensory qualities were measured using the 9-point hedonic scale method. In the CIE lab system (L*, a*, b*), the color values of 30-MLF-ISS were 27.80 ± 0.26, 12.99 ± 0.59, and 1.43 ± 0.05, respectively. The 30-MLF-ISS was also subjected to a proximate analysis. The melting rate of 30-MLF-ISS was 0.29 ± 0.03 g/min and the time it took for the first drop to fall was 37.00 ± 7.00 min. TAC, TPC, and AA of 30-MLF-ISS were observed to alter significantly (p < 0.05) during varied intervals of storage at - 18℃ (0, 30, and 60 days). The viability of probiotics in 30-MLF-ISS slightly decreased after storage at -18℃ for 8 weeks, but remained about 6 log CFU/g. During storage at -18℃ for 0 and 120 days, no pathogenic bacteria were detected in 30-MLF-ISS. These findings show that 30-MLF-ISS has nutritional and functional value, is free of foodborne pathogenic bacteria, is safe for consumers' health, and is suitable for application in the ice cream and related food industries.

시판 생식에서 식중독균의 정량적 평가 (Quantitative Evaluation of Foodborne Pathogenic Bacteria in Commercial Sangshik)

  • 최근 생식제품에 대한 국민 선호도가 높아지고 판매량이 증가됨에 따라 안전하고 위생적인 생식의 유통을 유도하고, 위생 강화를 위한 미생물 규격 제정의 기초연구를 위하여 시판 생식 및 원재료에 대한 식중독균 분포실태를 조사하였다. 2002년부터 2004년까지 서울, 인천, 부산 등 전국 대도시를 중심으로 대형마트 및 통신판매에서 시판 중인 생식제품 191건을 구입하여 주요 식중 독균인 E. coli, Salmonella, Staphylococcus aureus, Bacillus cereus, Clostridium perfringens, Listeria monocytogenes, Campylobactey jejuni, Yersinia enterocolitica, E. coli O157:H7, Vibrio parahaemolyticus를 검사하였다. 그 결과 B. cereus는 29건(15.2%), C. perfringens는 21건(11.0%), E. coli는 1건(0.5%)에서 검출되었으나 그 외 식중독균은 검출되지 않았다. 정량적 평가 결과 일반세균수는 $1.2X10^3cfu/g{\sim}1.6X10^9cfu/g$, B. cereus는 100 cfu/g 이하${\sim}11X10^3cfu/g$, C. perfringens는 100 cfu/g 이하${\sim}2.2x10^3cfu/g$의 분포를 보였다. 생식의 미생물 오염원을 파악하기 위하여 9개 생식제조공장에서 사용되고 있는 현미, 당근, 율무, 옥수수 등 53건의 원재료에 대한 미생물 분포도를 조사하였다. 그 결과 일반세균수는 $1.0X10^3cfu/g{\sim}1.5X10^8cfu/g$의 분포를 보였고, B. cereus는 13건(24.5%)에서 검출되었는데 11건(84.6%)이 100 cfu/g 이하이었다. C. perfringens는 2건(3.8%)에서 검출되었는데 모두 100 cfu/g 이하의 낮은 균수를 보였다. 생식 완제품과 같이 E. coli, Salmonella, S. aureus 등 8종의 식중독균은 검출되지 않았다. 본 실험 결과 유통 생식은 일반적으로 식중독균에 의한 위해 가능성이 적은 것으로 추정되었다. 그러나 제조과정 및 유통과정 중 부적절한 보관 및 유통에서 기인될 수 있는 오염균의 증식이나 교차오염 예방을 위한 철저한 관리는 필요한 것으로 판단되었다.

인삼의 GAP 실천모델 개발을 위한 재배단계의 미생물학적 위해도 평가 (Microbiological Hazard Analysis of Ginseng Farms at the Cultivation Stage to Develop a Good Agricultural Practices (GAP) Model)

  • 본 연구는 인삼의 재배단계에서의 생물학적 위해요소를 조사하고 그 결과를 인삼 GAP 실천 모델의 개발을 위한 기초자료로 제공하기 위해 수행하였다. 충남 금산에 소재한 인삼 경작지 3곳에서 재배환경, 작물, 개인위생 항목에 대해 총 96점의 시료를 수집하여 위생지표세균, 병원성미생물, 그리고 곰팡이에 대해 분석하였다. 일반세균과 대장균군, 곰팡이의 오염도는 각각 1.3~6.0, 0.1~5.0 및 0.4~4.9 log CFU/g (or mL, hand, and $100cm^2$)으로 확인되었고, 대장균의 경우 C 농장의 농업용수에서 검출되었다. 병원성 미생물은 모든 시료에서 B. cereus만 0.1~4.9 log/g (or mL, hand, and $100cm^2$)범위로 검출되었으며, L. monocytogenes, E. coli O157:H7, Salmonella spp. 및 S. aureus는 검출되지 않았다. 이상의 결과 인삼 경작지 3곳은 미생물학적 위해요소에 대해서는 안전한 것으로 나타났으나, 주변 환경이나 작업자들에 의해 교차오염이 발생할 가능성은 항상 존재하므로 보다 안전성이 확보된 인삼을 재배하기 위해서 미생물학적 위해요소의 관리과 포함된 GAP 모델의 적용이 필요하다.

  • While various foodborne pathogenic bacteria can be detected more rapidly via polymerase chain reaction than via conventional plating methods, it is impossible to distinguish between viable and dead cells in DNA-based assays. Hence, propidium monoazide (PMA) treatment has been introduced to detect living cells. The purpose of this study is to evaluate the applicability of the PMA treatment and real-time qPCR method for the detection of Cronobacter sakazakii and to compare it to that of plate counting. Based on our positive results, we suggest the use of PMA treatment and real-time qPCR for the detection of viable Cronobacter sakazakii in various food sources and an update of the Korean Food Code.

레토르트 곡물 두유 내 Bacillus cereus, Escherichia coli O157:H7, Listeria monocytogenes의 내열특성 (Thermal Resistance Characteristics of Bacillus cereus, Escherichia coli O157:H7, and Listeria monocytogenes in a Multi-grain Soy Milk Product)

  • This study determined the thermal resistance of Bacillus cereus, Escherichia coli O157:H7, and Listeria monocytogenes in multi-grain soymilk and proposes processing conditions that meet the national standard for retort food products in Korea. D and z values were calculated from thermal inactivation kinetic curves after heating at 55, 60, and $65^{\circ}C$. The D value for B. cereus at $55^{\circ}C$ was the highest (22.8 min), followed by that for E. coli O157:H7 (18.8 min) and L. monocytogenes (17.6 min). At $60-65^{\circ}C$, the order was L. monocytogenes ($D_{60-65^{\circ}C}=3.4-0.9min$), E. coli O157:H7 (3.0-0.3 min), and B. cereus (1.2-0.3 min). The z values for these species were 5.2, 5.5, and $7.7^{\circ}C$, respectively. The Korean national standard for retort food products was achieved by thermal processing at $124{\pm}2^{\circ}C$ for 0.3-2.2 min. This study provides useful data for ensuring both the microbiological safety and product quality of multi-grain soymilk products.

Inverse Correlation between Extracellular DNase Activity and Biofilm Formation among Chicken-Derived Campylobacter Strains

  • Campylobacter jejuni and Campylobacter coli are important foodborne pathogenic bacteria, particularly in poultry meat. In this study, the presence of extracellular DNase activity was investigated for biofilm-deficient Campylobacter strains versus biofilm-forming Campylobacter strains isolated from chickens, to understand the relationship between extracellular DNase activity and biofilm formation. A biofilm-forming reference strain, C. jejuni NCTC11168, was co-incubated with biofilm non-forming strains isolated from raw chickens or their supernatants. The biofilm non-forming strains or supernatants significantly prohibited the biofilm formation of C. jejuni NCTC11168. In addition, the strains degraded pre-formed biofilms of C. jejuni NCTC11168. Degradation of C. jejuni NCTC11168 biofilm was confirmed after treatment with the supernatant of the biofilm non-forming strain 2-1 by confocal laser scanning microscopy. Quantitative analysis of the biofilm matrix revealed reduction of extracellular DNA (16%) and proteins (8.7%) after treatment. Whereas the biofilm-forming strains C. jejuni Y23-5 and C. coli 34-3 isolated from raw chickens and the C. jejuni NCTC11168 reference strain showed no extracellular DNase activity against their own genomic DNA, most biofilm non-forming strains tested, including C. jejuni 2-1, C. coli 34-1, and C. jejuni 63-1, exhibited obvious extracellular DNase activities against their own or 11168 genomic DNA, except for one biofilm non-former, C. jejuni 22-1. Our results suggest that extracellular DNase activity is a common feature suppressing biofilm formation among biofilm non-forming C. jejuni or C. coli strains of chicken origin.
