Nutritional and Biochemical Studies on the Pollen Loads -2. Amino Acid Composition of Sunflower Pollen Load and Its Effects on the Hepatic Alcohol Dehydrogenase(ADH) Activity in Rat- (화분립(花紛粒)의 영양생화학적(營養生化學的) 연구(硏究) -2. 해바라기 화분립(花紛粒)의 아미노산조성(産組成)과 RAT 간(肝) Alcoholdehydrogenase 활성(活性)에 미치는 영향(影響))

  • For the purpose of investigating the influence of pollen load un alcohol metabolism in rat, we analyzed quantitatively amino acids of pollen load, and investigated the changes of hepatic alcohol dehydrogenase(ADH) activity and hepatocyte morphology in rat administrated various concentrations of alcohol and various amounts of pollen load. 18 species of amino acids including phenylalanine in the sunflower pollen load were quantitatively analyzed, and it was found that the amount of phenylalanine, leucine, threonine, lysine are especially higher than that of the other amino acids. The liver ADH activity of experimental animals decreased with the proportion of ethanol concentration much more in ethanol administrated group than in control group, while increased in pollen load mixed with ethanol administrated group, but didn't increased as much as that in control group. In any case the less the degree of ethanol concentration was administrated, the higher the liver ADH activity increased. There was fat infiltration in the hepatocyte of ethanol administrated animals, and remarkably little fat infiltration in that of animals administrated pollen load mixed with ethanol.

Coffee intake can promote activity of antioxidant enzymes with increasing MDA level and decreasing HDL-cholesterol in physically trained rats

  • This study investigated the effect of coffee intake and exercise on the antioxidative activity and plasma cholesterol profile of physically trained rats while they were exercising. Forty eight rats were under either the control diet with water (C) or control diet with coffee (CF) and at the same time they were given physical training for 4 weeks. In terms of physical training, the rats were exercised on a treadmill for 30 minutes everyday. At the end of 4 weeks, animals in each dietary group were subdivided into 3 groups: before-exercise (BE); during-exercise (DE); after-exercise (AE). Animals in the DE group were exercised on a treadmill for one hour, immediately before being sacrificed. Animals in the AE group were allowed to take a rest for one hour after exercise. TG levels were significantly high in coffee intake group than in control group. Also TG level of AE group was significantly higher than that of BE group. Exercise and coffee-exercise interaction effects were significant in total cholesterol (P = 0.0004, 0.0170). The AE of coffee intake group showed highest total cholesterol levels. HDL-cholesterol was significantly lower in coffee intake group than in control group. Coffee, exercise, and coffee-exercise interaction effects were significant in SOD (P = 0.0001, 0.0001, and 0.0001). The AE and BE of coffee intake group showed higher SOD levels than the other four groups. Catalase activities were significantly higher in coffee intake group than control group. No significant main effect was found in GSH/GSSG. Coffee, exercise, and coffee-exercise interaction effects were significant in MDA levels (P = 0.0464, 0.0016, and 0.0353). The DE and AE of coffee intake group and the DE of control group showed higher MDA levels than the BE of control group. Therefore, coffee intake can promote activities of antioxidant enzyme but it also increases MDA and decreases HDL-cholesterol in physically trained rats.

Molecular Characterization of Fluoroquinolone Resistant Escherichia coli Isolates from Chickens in Korea (닭에서 동정된 플르오르퀴놀론 내성 대장균 균주의 분자생물학적 성상에 관한 연구)

  • An aim of current study was to investigate the prevalence and the mechanism of quinolone-resistance in E. coli isolates obtained from chicken cecum in Korea. In addition, multilocus sequence typing (MLST) was also performed for the molecular characterization of E. coli isolates. In an antimicrobial susceptibility test by the disk diffusion method, the 63.5% (54/85) of E. coli isolates showed the resistance to quinolone group of antimicrobial agents. All of the 54 E. coli isolates showing resistant to quinolone group had sense mutations in gyrA gene and point mutations at the $57^{th}$, $80^{th}$, or $84^{th}$ residues in parC gene were detected in 90.7% of the isolates. Interestingly, E. coli ST was closely related to amino acid substitutions in parE gene. Our results indicated that the long-term use of antimicrobial agents in food-producing animals was strongly associated with a prevalence of antimicrobial resistance in commensal Enterobacteriaceae, suggesting the need for continuous surveillance and monitoring of antimicrobial resistant determinants in bacterial isolates from food animals.

  • Currently, various probiotics are being used to improve the nutrition of companion animals. They are widely sold as additives in companion animal foods because of the numerous gastrointestinal and immune health benefits for dogs and cats. Therefore, extensive research is being conducted to improve quality and safety during manufacturing and to extend the shelf life of companion animal foods by adding probiotics. The manufacturing process must be conducted such that the characteristics and efficacy of probiotics added to food are optimally beneficial for companion animals. Therefore, this review aims to address the overall characteristics of the probiotic strains used and to examine the various methods through which probiotics are added to companion animal foods.

  • Obesity, as defined by the World Health Organization (WHO), is excessive fat accumulation that can pose health risks and is a disorder of the energy homeostasis system. In typical westernized diets, ω-6 polyunsaturated fatty acids (PUFAs) vastly exceed the amount of ω-3 PUFAs, with ω-6/ω-3 ratios ranging from 10:1 to 25:1. ω-6 PUFAs, such as arachidonic acid, have pro-inflammatory effects and increase obesity. On the other hand, ω-3 PUFAs, including eicosapentaenoic acid and docosahexaenoic acid, have anti-inflammatory and anti-obesity effects. Linoleic acid (LA) and alpha-linolenic acid (ALA) are synthesized in almost all higher plants, algae, and some fungi. However, in humans and animals, they are essential fatty acids and must be consumed through diet or supplementation. Therefore, balancing LA/ALA ratios is essential for obesity prevention and human health. Monogastric animals such as pigs and chickens can produce meat and eggs fortified with ω-3 PUFAs by controlling dietary fatty acid (FA). Additionally, ruminant animals such as feeder cattle and lactating dairy cows can opt for feed supplementation with ω-3 PUFAs sources and rumen-protected microencapsulated FAs or pasture finishing. This method can produce ω-3 PUFAs and conjugated linoleic acid (CLA) fortified meat, milk, and cheese. A high ω-6/ω-3 ratio is associated with proinflammation and obesity, whereas a balanced ratio reduces inflammation and obesity. Additionally, probiotics containing lactic acid bacteria are necessary, which reduces inflammation and obesity by converting ω-6 PUFAs into functional metabolites such as 10-hydroxy-cis-12-octadecenoic acid and CLA.

Uterotrophic and Hershberger Assay for Butyl ρ-Hydroxybenzoic Acid

  • Butyl $\rho$-hydroxybenzoic acid (butyl paraben, BP) is a homologous series of parabens and is widely used as a preservative in cosmetic and pharmaceutical products. The purpose of this study was to investigate the estrogenic/antiandrogenic activities of BP in animals. For that, we performed an uterotrophic assay and a Hershberger assay in rats. In uterotrophic assay, BP was administered subcutaneously to immature female SD rats (18 days old) for 3 consecutive days. The wet and dry uterus weights were significantly increased in the groups treated with BP in dose­dependent manner. In case of Hershberger assay, BP significantly reduced the weight of seminal vesicle of castrated rats. And other accessory organ/glands - prostate, Cowper's glands, bulbocavernosus muscle and glans penis were also slightly decreased. The results of this study suggested that BP showed estrogenic and anti-androgenic activities in vivo.

Effects of Hovenia dulcis Thunberg Extract on Enzymes Related Reactive Oxygen Intermediate (헛개나무(Hovenia dulcis Thunberg) 추출물이 활성 산소종과 관련한 효소에 미치는 영향)

  • In order to investigate the effects of 70% EtOH extract obtained from Hovenia dulcis Thunberg on enzymes relating reactive oxygen intermediate, cancer-stricken animals induced by DEN (N,N-diethylnitrosamine) were recovered by administering the extract of Hovenia dulcis Thunberg. It showed that there was no effect on the generation of superoxide radical by the extract of Hovenia dulcis Thunberg. However, considering the increase of the activity of Cu, Zn-SOD and Mn-SOD in the tested animal class, the extract of Hovenia dulcis Thunberg could participate directly in removing of superoxides. The experimented-animals treated with the extract of Hovenia dulcis Thunberg showed an increase in the activity of the enzymes, catalase and glutathione peroxidase, which can eliminate hydrogen peroxide pertained in liver tissue. The extract of Hovenia dulcis Thunberg seemed to have some factors that accelerate the oxidation. Also, the extract of Hovenia dulcis Thunberg showed effects on the enzymes relating to the active oxygen toxicity which could be an indicator of aging and body toxicity.

The Change of Electrolytes Composition in Skeletal Muscles by Protein Restriction and Rehabilitation (단백질의 제한과 회복으로 인한 골격 근육내의 성분변화)

  • The objective of this study was to compare the electrolytes contents in skeletal muscles of two different groups of rats, one for control fed 20% casein diet, and the other experimental group recovered from 7% casein diet as protein restriction feeding for two weeks at the beginning of this experiment. Two different comparison criteria were used in this study, one for comparison between animals at the same age groups and the other between groups weighing same body weight as the control groups. Food consumption of animals for experimental and control groups were ad libitum. Following measurement and samples were collected: body weight, five different muscles-anterior tibialis (A.T.) extensor digitorium longus (E.D.L.) soleus, plantaris, gastrocnemius-for electrolytes and protein analysis. The experimental groups showed lower body weight than that of control group. Same trend was showen in protein content in five different muscles. Magnesium and potassium content in four muscles except gastrocnemius showed lower and sodium higher in experimental groups than those in control group at fourteenth day, after recovery from protein restriction, electrolytes content change showed differently in four muscles. Magnesium and potassium contents were increased as protein content in diet. Sodium content decreased as one week intake of recovery diet started, thereafter it was rather slowly increased. Phosphorus content in gastrocnemius at the second week in experimental group was slightly lower than that of control group, and it recovered rapider while at the same body weight group it was higher in experimental group.

  • Due to the increase in single-person households among the people who have companion animals, it is difficult to feed them to companion animals in the absence of the owner, so an automatic feeding machine was designed. In this paper, we propose an automatic feeding machine that has a drug distribution function as well as feeds using the Arduino platform. It is expected that in the proposed automatic feeding machine, users can access the food service through the website, and experience the convenient and extended food service function through the automatic dispensing system that combines the two materials.
