화학적 발암원이 Clone 9 세포의 알데히드대사 효소와 항산화 효소에 미치는 영향 (Effects of Chemical Carcinogens on the Aldehyde Metabolic Enzymes and Antioxidant Enzymes in Clone 9 Cell)

  • Chemical carcinogen-induced alteration of aldehyde metabolic enzymes were examined in clone 9 cell. Diethylnitrosamine (DENA), N-nitrosoethylurea (NEU) and N-nitrosomorpholine (NNM) were wed as model carcinogens. Changes in enzyme activities by repetitive treatment of DENA, NEU or NNM were analyzed in terms of specific activities and activity stainings of the enzymes on the gel. Upon treatment of DENA, lipid peroxide level increased upto 10 fold, indicating strong oxidative stress state of the cell. Notable enhancement of ADH and ALDH activity occurred after DENA treatment, while glutathione-S-transferase activity was slightly increased. Furthermore, about 2.5 fold higher superoxide dismutase (SOD) activity was detected during deactivation of catalase (CAT) activity by repetitive treatment of DENA. However in NEU-treated cell, about 2.3 fold higher ALDH activity was found while ADH activity was slightly increased. Notable increase CAT and SOD could also be found. In contrast, maximum 3.5 fold higher CAT activity occurred during SOD deactivation in NNM-treated cell. These results suggest that there might be different enzymatic responses in relation to cell protection against DENA, NEU or NNM.

Involvement of Caenohabditis elegans MAPK Signaling Pathways in Oxidative Stress Response Induced by Silver Nanoparticles Exposure

  • In the present study, toxicity of silver nanoparticles (AgNPs) was investigated in the nematode, Caenohabditis elegans focusing on the upstream signaling pathway responsible for regulating oxidative stress, such as mitogen-activated protein kinase (MAPK) cascades. Formation of reactive oxygen species (ROS) was observed in AgNPs exposed C.elegans, suggesting oxidative stress as an important mechanism in the toxicity of AgNPs towards C. elegans. Expression of genes in MAPK signaling pathways increased by AgNPs exposure in less than 2-fold compared to the control in wildtype C.elegans, however, those were increased dramatically in sod-3 (gk235) mutant after 48 h exposure of AgNPs (i.e. 4-fold for jnk-1 and mpk-2; 6-fold for nsy-1, sek-1, and pmk-1, and 10-fold for jkk-1). These results on the expression of oxidative stress response genes suggest that sod-3 gene expression appears to be dependent on p38 MAPK activation. The high expressions of the pmk-1 gene 48 h exposure to AgNPs in the sod-3 (gk235) mutant can also be interpreted as compensatory mechanisms in the absence of important stress response genes. Overall results suggest that MAPK-based integrated stress signaling network seems to be involved in defense to AgNPs exposure in C.elegans.

배추좀나방의 Fenvalerate에 대한 저항성 발달과 교차저항성 (Development of Fenvalerate Resistance in the Diamondback Moth, Plutella xylostela Linne (Lepidoptera : Yponomeutidae) and its Cross Resistance)

  • Pyrethroid계 살충제 fenvalerate를 공시하여 실내 감수성계통 배추좀나방(Plu tella xylostella L.)을 인위적으로 누대도태한 후 저항성 발달속도와 정도를 조사하고, 이 저항성계통의 타 살충제에 대한 교차저항성 유무와 정도를 검토하였다. 저항성 발달속도는 fenvalerate 24세대 도태계통에서 도태전에 비하여 66.2배 증가하였다. Fenvalerate 도태계통은 pyrethroid계 살충제인 deltamethrin에 145배의 고도의 교차저항성을 나타내었고, alphamethrin, cypermethrin, fenvalerate, permethrin, tetramethrin에 17.4 -45.0배의 교차저항성을 나타내었다. 유기인계 diazinon, dichlorvos, ethoprophos, EPN과 카바메이트계 BPMC, carbaryl, methorny I에서는 2.1-4.3배의 낮은 교차저항성을 나타내었다. 그러나, 유기인계의 acephate, fenitrothion과 카바메트계 carbofuran에 대해서 비교차저항성을 나타내었다.

  • A study on the traditional houses of North Korea(I) have been published in 1996. This paper is written to supplement the preceding paper. This paper aims at collecting new data of traditional house in North Korea. But still being prohibited for the researchers of South Korea to approach to the field, I had to depend on the memories and experiences of the immigrants from North Korea who are now living in Kangwon and Incheon Province. Through the questionnaire and drawings, they described vivid memory of their old houses. I was able to add new data of 70 cases, which are significant and valuable as much as those of the real field are. Those data, including the exisiting data, are enough for me to analize statstically the regional charateristics, the differnces among economical classes, and the periodical change. It opens the way for verfying the existing theory. Regional charateristics of house in North Korea can be described as followings: a. Enhanced Clavulanic Acid Production in Streptomyces clavuligerus NRRL3585 by Overexpression of Regulatory Genes

  • We constructed four recombinant plasm ids to enhance the production of clavulanic acid (CA) in Streptomyces clavuligerus NRRL3585: (1) pIBRHL1, which includes ccaR, a pathway-specific regulatory gene involved in cephamycin C and CA biosynthesis; (2) pIBRHL2, containing claR, again a regulatory gene, which controls the late steps of CA biosynthesis; (3) pGIBR containing afsR-p, a global regulatory gene from Streptomyces peucetius; and (4) pKS, which harbors all of the genes (ccaR/ claR/ afsR-p). The plasmids were expressed in S. clavuligerus NRRL3585 along with the $ermE^*$ promoter. All of them enhanced the production of CA; 2.5-fold overproduction for pIBRHL1, 1.5-fold for pIBRHL2, 1.6-fold for pGIBR, and 1.5-fold for pKS compared to the wild type.

Cancer Chemopreventive Potential of Scenedesmus spp. Cultured in Medium Containing Bioreacted Swine Urine

  • Scenedesmus spp. were cultured for 51 days in newly developed medium, KEP I (Kim and Ecopeace: initials of corresponding author and environmental company) made with Bacterio-Mineral-Water (3%, v/v) that had been bio-reacted with swine urine medium to 10% (v/v) Bold's Basal medium, and investigated for cancer chemopreventive potential by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST), and reduced glutathione (GSH), and inhibition of cytochrome P450 (CYP) 1A1 activity. The activitives of QR and GST of Scenedesmus spp. cultured in KEP I medium were increased by 3.0-fold and 1.5-fold, respectively. However, Scenedesmus spp. cultured in control medium (CT) increased the activitives of QR and GST by 1.8-fold and 1.3-fold, respectively. Scenedesmus spp. in KEP I medium strongly inhibited CYP 1Al activity. These results show that Scenedesmus spp. in KEP I medium has cancer chemopreventive potential and may be a candidate for further development as a chemopreventive agent.

A Transfer of Silver Nanoparticles from Pregnant Rat to Offspring

  • Silver nanoparticles (size: $7.9{\pm}0.95$ nm, dosage: 250 mg/kg) were orally administered to pregnant rats. At 4 days after parturition, four pups were randomly selected (one pup from one dam) and silver level in liver, kidney, lung and brain was determined by ICP-MS and electron microscope. As results, silver nanoparticles highly accumulated in the tissues of the pups. Silver level in the treated group was $132.4{\pm}43.9$ ng/g in the kidney (12.3 fold compared to control group), $37.3{\pm}11.3$ ng/g in the liver (7.9 fold), $42.0{\pm}8.6$ ng/g in the lung (5.9 fold), and $31.1{\pm}4.3$ ng/g in the brain (5.4 fold). This result suggested that the possible transfer of silver nanoparticles from pregnant dams to the fetus through mainly placenta.

  • Dysphonia is a medical terminology for voice disorders characterized by hoarseness, harshness, weakness, or even loss of voice ; any impairment in ability to produce voice sounds using the vocal organs, larynx, The causes of dysphonia can be classified into two groups, organic and functional. Functional dysphonia includes spasmodic dysphonia, muscle tension dysphonia, mutational dysphonia and conversion dysphonia, etc, The findings of laryngoscopy in these dysphonia are almost normal. Therefore, physicians should diagnosis these diseases from careful history taking and abundant understandings about the phonation pattern, Organic dysphonia is caused by anatomical problems in the larynx, especially on the vocal fold, Some lesions, however, are not easily found because these lesions are too small, or located on the lower lip of vibrating vocal fold. Laryngopharyngeal reflux induced laryngitis, vascular lesions, sulcus vocalis, vocal atropy including presbylaryngis, and mucosal tears are common lesions easily missed in laryngoscopy, Therefore, a high index of suspicion is necessary to avoid missing vocal fold mucosal lesions, and the strobovideolaryngoscopy is indispensable in making the diagnosis,

Immune-enhancement effect of JaSaengHwan

  • We investigated the immune enhancement effects of JaSaengHwan (JSH). The forced swimming test (FST) has been used as a screening model for new immune enhancement agents. We found that JSH (0.1 mg/ml) significantly reduced the immobility time in the FST compared to the control. Also, we investigated the effect of JSH on the proliferation of T cell and production of cytokines in human T-cell line, MOLT-4 cells and mouse peritoneal macrophages. JSH (1 mg/ml) significantly increased the cell proliferation by $46.78{\pm}6.41%$ (p<0.05) and also significantly increased the interleukin (IL)-2, IL-4 and interferon $(IFN)-{\gamma}$ production compared with media control (about 2-fold for IL-2, 3-fold for IL-4 and 1.5-fold for $IFN-{\gamma}$, p<0.05) at 24 h. In addition, JSH increased the production of IL-12 on the mouse peritoneal macrophages (by 3.6-fold for IL-12, p<0.05). In conclusion, these data indicate that JSH may have an immune-enhancement effect.

Immune Enhancing Effect by Orally-Administered Mixture of Saccharomyces cerevisiae and Fermented Rice Bran

  • The mixture (PM) of Saccharomyces cerevisiae and fermented rice bran on the activation of macrophage and bone marrow cell proliferation was studied in mice. PM stimulated not only the activation of macrophage (1.8-fold of saline) but also IL-6 production from macrophage (1.5-fold) at 2.0 g/㎏/day during 7 days of oral administration. By the culture supernatant of Peyer's patch cells from C3H/HeJ mice fed PM at 2.0 g/㎏/day for 7 days, the bone marrow cells significantly proliferated compared with that of mice receiving only saline (1.7-fold). In addition, the contents of GM-CSF and IL-6 in the culture supernatant of Peyer's patch cells from mice fed PM at 2.0 g/㎏/day were increased in comparison with those from the control (1.8 and 1.4-fold, respectively). These results revealed that oral administration of PM may modulate IL-6 production to induce the activation of macrophage, and also enhance secretion of hematopoietic growth factors such as GM-CSF and IL-6 from Peyer's patch cells.
