On-line Monitoring of Glucose and Acetate by Flow-Injection Analysis in Escherichia coli Fermentation Process (대장균 발효공정에서 흐름주입분석을 이용한 글루코스와 초산의 온라인 모니터링)

  • Flow-injection analysis (FIA) for on-line monitoring of glucose and acetate are described and employed in E.coli fermentation process. Glucose oxidase (GOD) for the detection of glucose is immobilized on epoxy polymer support, which is packed in a small cartirdge, and applied to a GOD-FIA system. The detection of acetate is based on the inhibition of acetate to the oxidation of sarcosine by sarcosine oxidase (SOD). SOD is also immobilized on epoxy polymer support and used for a SOD-FIA system. GOD-FIA system is characterized as well as SOD-FIA system by the investigation of the effects of pH, temperature and metabolites in samples on the peak height. GOD-FIA and SOD-FIA systems were also applied for on-line On-line measurements buy FIA measurements by FIA were in god agreement with off-line measurements.

  • This paper investigates CFD investigation on single phase supersonic nozzle flow and 2-phase subson ic flow prior to rocket nozzle supersonic 2-phase flow with water injection within the flame deflector. Numerical results of supersonic nozzle single phase flow showed no notable unrealistic behavior as it captures the usual shock cell structures. Three-dimensional 2-phase flow analysis has also been performed to verify whether the approach can grab the droplet behavior during cooling by water injection. It is expected these basic studies will enhance the cooling problem analysis of supersonic 2-phase rocket plume in the future.

  • When a cold HPSI (High Pressure Safety Injection) fluid associated with an design basis accident, such as LOCA (Loss of Coolant Accident), enters the cold legs of a stagnated primary coolant loop, thermal stratification phenomena will arise due to incomplete mixing. If the stratified flow enters a reactor pressure vessel downcomer, severe thermal stresses are created in a radiation embrittled vessel wall by local overcooling. Previous thermal-mixing analyses have assumed that the thermal stratification phenomena generated in stagnated loop of a partially stagnated coolant loop are neutralized in the vessel downcomer by strong flow from unstagnated loop. On the basis of these reasons, this paper presents the thermal-mixing analysis results in order to identify the fact that the cold plume generated in the vessel downcomer due to the thermal stratification phenomena of the stagnated loop is affected by the strong flow of the unstagnated loop.

  • Spectrophotometric properties of lanthanide complexes with methylthymol blue(MTB) and cetyltrimethylammonium bromide(CTAB) were studied and also lanthanide(III) ions were determined by flow injection analysis on the base of the above results. The absorption maxima of lanthanide(III)-MTB complexes in the presence of CTAB are 635nm with molar absorptivity of $4.51{\sim}6.11{\times}10^4Lmol^{-1}cm^{-l}$ at pH 5.8. The mole ratio of lanthanide(III) complexes with MTB is 1:2 in the presence of CTAB. The calibration curves of lanthanide(III) ions obey the Beer's law in the range of 0.1 to 0.4ppm under the optimum condition. The samples throughput was ca. $60hr^{-1}$. The interfering effect of some cations and anions was investigated. The ligand anions such as tartrate and citrate, many transition and rare earth elements interfered severely and must be removed before the determination of lanthanide(III) ions.

  • In the present study, a numerical analysis of an injection molding process was conducted for predicting the mold deformation considering non-Newtonian flow, heat transfer, and structural behavior. The accurate prediction of mold deformation during the filling stage is important to successfully design and manufacture a precision injection mold. While the local mold deformation can be caused by various factors, a pressure induced by the polymer melt is considered to be one of the most significant ones. In this regard, the numerical simulation considering both the melt filling and the mold deformation was carried out. A mold core for a 2D axisymmetric center-gated disk was used for the demonstration of the present study. The flow behavior inside the mold cavity and temperature distribution were analyzed along with the core displacement. Also, a Taguchi method was employed to investigate the influence of the relevant parameters including flow velocity, mold core temperature, and melt temperature.

  • This paper is the first of several companion papers, which investigate axial stratification process and its effects in an Sl engine. The axial stratification is very sophisticate phenomenon, which results from combination of fuel injection, port and in-cylinder flow and mixing. Because of the inherent unsteady condition in the reciprocating engine, it Is impossible to understand the mechanism through the analytical method. In this paper, the ports were characterized by swir and tumble number in steady flow bench test. After this, lean misfire limit of the engines, which had different port characteristic, were investigated as a function of swirl ratio and injection timing for confirming the existence of stratification. In addition, gas fuel was used for verifying whether this phenomenon depends on bulk air motion of cylinder or on evaporation of fuel. High-speed gas sampling and analysis was also performed to estimate stratification charging effect. The results show that the AFR at the spark plug and LML are very closely related and the AFR is the results of bulk air motion.

  • Renal failure patients have to operate arteriovenous graft for hemodialysis. Blood flow characteristics influence the patency rate of arteriovenous graft. Numerical investigation is performed with the arteriovenous graft according to injection of blood. As a result, when the injection is not applied to venous graft, the low wall shear stress region appears at venous anastomosis. It may cause intimal hyperplasia at venous anastomosis.

  • Developed is the new gas-solid flow ball valve where air injection purging concept is applied for the control and the reduction of particle fouling. CFD analyses are conducted for investigating the interaction between gas-sold and air injection streams and the fouling phenomena in valve, and the analysis results are reflected in the design of valve geometry and air injection condition. Through the actual tests on designed ball valve, the present valve is shown to be superior in fouling reduction.

  • In this paper, tile characteristics of flow resulting from the configurations of piston head and intake-port of the cylinder in a gasoline-direct-injection engine are investigated numerically. Calculations are carried out from intake process to the end of compression. GTT code which includes the third order upwind Chakravarthy-Osher TVD scheme and κ-ε turbulence model with the law of wall as a boundary condition. As a result, a piston head with a smaller radius of curvature and larger radius gives stronger reverse tumble. It is also shown that as the maximum tumble ratio increases by the configuration of the intake-port the tumble ratio at the end of compression stroke increases. It is concluded that flows at the end of compression stroke can be controlled by the optimum design of intake-port and piston head.

  • This paper presents a dynamic simulation of the unified power flow controller (UPFC) using a current injection method. Flexible AC Transmission System (FACTS) devices give more flexibility of control for security and economic operation of power systems. Diffculties of modeling UPFC in the conventional dynamic simulation programs arise from the fact that the injected voltage by the series inverter is superimposed on the shunt inverter side voltage. A solution can be a current injection method, in which a serial part of UPFC is converted to a parallel equivalent circuit using source transformation, and two current sources affect each other at every time step. To verify efficiency of this method, the proposed model is applied for the transient analysis of an example power system.
