As problems such as water pollution and fish species depletion have become serious, a land-based fish farming is receiving a great attention for ensuring stable productivity. In the fish farming, it is important to determine the timing of shipments, as one of key factors to increase net profit on the aquaculture. In this paper, we propose a system for predicting net profit to support decision of timing of shipment using fish farming-related statistical data. The prediction system consists of growth and farm-gate price prediction models, a cost statistics table, and a net profit estimation algorithm. The Gaussian process regression (GPR) model is exploited for weight prediction based on the analysis that represents the characteristics of the weight data of cultured fish under the assumption of Gaussian probability processes. Moreover, the long short-term memory (LSTM) model is applied considering the simple time series characteristics of the farm-gate price data. In the case of GPR model, it allows to cope with data missing problem of the weight data collected from the fish farm in the time and temperature domains. To solve the problem that the data acquired from the fish farm is aperiodic and small in amount, we generate the corresponding data by adopting a data augmentation method based on the Gaussian model. Finally, the estimation method for net profit is proposed by concatenating weight, price, and cost predictions. The performance of the proposed system is analyzed by applying the system to the Korean flounder data.
Olive flounder, Paralichthys olivaceus is one of the most important cultured fish in Korea, its farming has been negatively impacted by viral, bacterial and parasitic diseases. Streptococcal infection was considered as a serious problem because of significant economic losses in olive flounder farm industry. The development and evaluation of vaccine for protection against infection by this agent were required. We evaluated the safety and efficacy of ${\beta}$-hemolytic Streptococcus (S.) iniae vaccine on olive flounder Three hundreds of flounders (weight $119.8{\pm}20.7g$, body length $22.6{\pm}1.4cm$) were reared in 0.5 tons aquaria in land-marine tank system. Seawater was provided from the sea of Inchon in Korea, and water temperature was set to $22^{\circ}C$ and $25^{\circ}C$ in the vaccination and challenge test, respectively. We used the formalin-inactivated ${\beta}$-hemolytic S. iniae (F2K) vaccine (M VAC INIAE; Kyoritsu seiyaku, Japan) originated in Japan. The vaccine was intraperitoneally administered to fish. Both of vaccinated group and control group were challenged with intraperitoneally injection by virulent S. iniae SI-36 isolates with $1.0{\times}10^7CFU/fish$ at 3 weeks after vaccination. Difference on mortality of control and vaccinated group (90.0 and 15.0%, 76.5 and 8.0% respectively) at two trials were found significant (p<0.05), and relative percent survival were 83.4% and 89.5%, respectively. The dead fishes were showed dark pigmentation of skin, abdominal extension, hemorrhagic ascites, and liver necrosis, and isolated the S. iniae strain from ascites, liver and kidney. We confirmed the safety and efficacy of ${\beta}$-hemolytic S. iniae vaccine by determinations of the optimal management condition and artificial challenge test in olive flounder.
본 연구는 연안 환경오염 및 양식용수의 이용과정에서 파생될 수 있는 철 오염이 넙치의 생리적 변화에 미치는 영향을 파악하기 위하여 12주 동안의 만성적 조건하에서 그들의 조직, 대사 및 생존의 변화를 검토하였다. 철에 노출시킨 넙치 아가미의 뚜렷한 조직학적 변화양상은 0.93mg/$\ell$ 이상의 농도에서 2주 후부터 조사개체 중에 $50\%$이상이 나타났다. 이들의 변화양상은 새판의 만곡, 새판 말단부의 곤봉-화, 상피세포의 파괴, 상피층의 박리현상, 괴사 및 새판의 융합 등으로 나타났으며, 이 같은 손상정도는 철 농도가 증가함에 따라 심하게 나타났다. 산소소비는 철 농도 0.42mg/$\ell$ 이하에서는 유의한 감소가 인정되지 않았으나, 0.93 mg/$\ell$ 농도에서는 실험 2주 후부터 유의한 감소가 관찰되었고, 12주 후의 0.93, 4.89 및 9.78mg/$\ell$농도에서 각각 37.1, 43.5 및 $53.2\%$가 감소하였다. 생존율은 0.42mg/$\ell$이하의 철 농도에서 $100\%$가 관찰되었고, 0.93mg/$\ell$농도에서는 $92.5\%$의 생존율을 보였으나, 유의한 감소는 철 농도 4.89mg/$\ell$에서 실험 5주 후, 9.78mg/$\ell$농도에서는 4주 후부터 관찰되었다. 이상의 결과는 연안해역 및 양식장 등에 0.93 mg/$\ell$ 이상의 철 농도가 존재할 경우, 넙치는 아가의 손상 및 대사활동의 저하 등으로 인해 성장에도 악영향을 받을 수 있을 것으로 사료된다.
Turbot Scophthalmus maximus can be cultured at a higher density and has a faster growth rate than olive flounder Paralichthys olivaceus, making it an attractive alternative for flounder farmers in Korea. As it is a relatively new species to Korean farming environments, there is no general farming protocol, which takes multiple generations to develop, for turbot in Korea. Nevertheless, we applied breeding technology to develop a stock, which we considered first-generation turbot. Then we compared its growth rate (an important economic trait) to different populations (the original stock of Korean turbot, French turbot, and Chinese turbot). The four different populations were grown in similar culture conditions. First-generation Korean turbots grew about 28% faster than all others. We will continue to select for important traits such as color, disease resistance, and heat tolerance in this turbot broodstock; we believe that with the appropriate selection process, Korean turbot should become competitive in the markets, and will become a major farming species in the Korean aquaculture industry.
Despite its economic importance, olive flounder Paralichthys olivaceus aquaculture industry is facing a crisis with a continuous production decline. There have been many solutions to overcome the complicate predicament proposed. Increasing genetic diversity and discovering new commercial value through selective breeding are among them. The aims of the present study are to increase the selection power of the golden flounders. We examined the genetic diversity of the breeder population of golden flounders and developed selective markers for the golden flounder population. The 6 microsatellite (MS) markers were selected from melanogenesis-related genes, which are believed to be involved in the pigmentation of fish. All markers were polymorphic (except PO4) and 5 of them had PIC value of 0.6 or above. All makers had distinctive alleles indicating either normal or golden individuals. For examples, from PO4 marker, the frequency of an allele (316) in the golden population was 100% and in normal population was 0% (P<0.001). Although some more studies with more samples at the later generations should be performed to confirm this result, the 316 allele from PO4 marker could be a distinctive tool for decision of the colors in olive flounders at an early stage of the life cycle.
최근 2년간 동해안 지역 넙치 양식장의 양식 넙치에 피해를 일으키는 P. damselae 균을 분리하였으며, 분리된 P. damselae의 16s rRNA 염기 서열은 P. damselae subsp. damselae ATCC 33539와 99%의 상동성을 나타내었다. 분리 균주는 Pedersen et al.(1997)의 biotype과 비교한 결과, biotype No 8과 동일하게 나타났으며, P. damselae subsp. damselae ATCC 33539의 LPS와 동일한 단백질 패턴을 나타내었다. 넙치의 병어로부터 P. damselae와 Vibrio 속 세균의 감염 상태를 조사한 결과, P. damselae가 가장 높은 감염율을 나타내었고, 그 다음으로 V. anguillarum, V. splendidus, V. harveyi와 V. ordalii순으로 감염율이 나타났다.
Objectives: Formalin is used as an anthelmintic in farms where flounder are raised. In this study, we aim to identify formaldehyde exposure levels for aquaculture industry workers and provide basic data for managing formaldehyde exposure. Methods: Exposure levels of formaldehyde in the air, including formalin spraying operations, were assessed separately for personal and area samples. In addition, considering the formalin administration method, dermal exposure to the hands was estimated when administering the chemical, and dermal exposure to the legs during water tank work was estimated by collecting water in the water tank and evaluating the amount of formaldehyde remaining. Finally, the respiratory exposure level and the estimated dermal exposure level were added to derive the total exposure level and compared with the maximum allowable human dose. Results: As a result of the airborne evaluation, the formaldehyde concentration of the worker (1 person) who performed the formalin spraying and flounder sorting was 33.61 ppb, and the arithmetic mean of formaldehyde concentrations of the workers (3 people) who only performed the flounder sorting was 3.28 ppb (range: 2.25-4.89 ppb). In the case of dermal exposure, when spraying formalin once, the amount was estimated to be 0.33-2.62 mg when wearing protective gear and 3.27-26.12 mg when not wearing it. Conclusions: There was a difference in the formaldehyde exposure level of workers depending on their operation of handling formalin and whether or not protective gear was worn. In particular, because the level of formaldehyde exposure due to dermal exposure can be significant, there is a need to improve formalin administration methods in a way that avoids skin contact as much as possible.
제주도내 양식넙치 병어에서 분리된 연쇄상구균 75균주의 항균제에 대한 감수성 경향 및 내성균 출현빈도를 조사하고 다재내성 균주로부터 전이성 R plasmid를 검출하였다. 넙치 병어에서 분리된 모든 연쇄상구균은 flumequine (AR)과 oxolinic acid (OA)에 대하여 저항성이었으며, 그 중 30균주 (41%)는 ampicillin (ABPC), doxycycline (DOXY), erythromycin (EM), norfloxacine (NFLX), oxyteracycline (OTC)의 약제에 대하여 다양한 조합으로 동시내성을 나타내었다. AR과 OA를 포함하여 4~6약제에 대하여 다재내성을 보인 균주는 26주였다. 연쇄상구균의 다재내성 전이에 관여하는 R plasmid인 pST9와 DNA구조가 유사한 plasmid의 분포를 알아보기 위하여 60 분리 균주에 대한 colony 혼성화를 실시하였다. 그 결과 13 분리균에서 혼성화 양성반응이 나타났으며 생사료의 원료로 사용되는 양치와 고등어에서도 양성반응이 나타남으로써 동일 DNA구조의 plasmid가 분포함을 시사하였다. 혼성화 양성반응을 보인 13균주로부터 전이성 R plasmid를 검출하기 위하여 conjugation을 실시하였다. OTC, DOXY, EM에 대한 항균제 내성 marker인 Otc, Doxy, Em은 R plasmid에 의하여 수용균인 Streptococcus sp.로 전이가 이루어졌다. 또한 이들 전이성 R plasmid를 보유하고 있는 연쇄상구균들이 동일 분류군에 속하는지를 알아보기 위하여 RAPD pattern을 분석하였다. 내성균주들의 RAPD pattern은 모두 유사하였으며, Streptococcus iniae type의 RAPD pattern (Kim and Kim, 2003)은 나타나지 않았다. 그러므로 pST9와 동일 DNA 구조의 R plasmid는 S. iniae에서의 출현빈도가 매우 낮을 것으로 예상되었다.
2000년도 2회에 걸쳐 제주도 일원 넙치 양식장을 대상으로 감염어에서 분리된 연쇄구균에 대한 특성시험 결과를 요약하면 다음과 같다. 감염어로부터 분리된 연쇄구균은 형태적으로 연쇄상의 gram 양성의 구균이었고, catalase와 oxidase에 음성이었으며, 용혈성 시험에서는 $\beta$-용혈성을 나타내었다. 감염어에서 각 장기별 생균수에서는 환부, 신장, 혈액 그리고 뇌순으로 연쇄구균이 검출되었다. 약제 감수성에서는 ampicillin, ciprofloxicin 둥에 감수성이 있었지만 erythromycin 등에는 감수성이 없었다. 복강에 주사하여 병원성 시험을 한 결과 10일 이내에 전량 폐사하는 강한 병원성을 보였다. 감염균의 용혈성에 대한 특성은 $50^{\circ}C$ 30분의 배양에서 용혈활성이 마르게 증가되어 최대값에 도달되었고 또한 $pH 5.5\~6.5$에서 높은 활성값을 보였다.
We evaluated the efficacy of $\beta$-hemolytic Streptococcus(S.) iniae vaccine on cultured olive flounder. Three hundred flounders(weight $50{\pm}5$ g) were obtained from two farm at Wando and Taean in the southern and western coast of Korea at May and June 2007, respectively. Twenty of flounders moved in 0.5 tons aquaria in land-marine tank system of National Veterinary Research and Quarantine Service. Seawater was transported from the sea of Inchon in western Korea, and water temperature maintained to $22^{\circ}C$ and $25^{\circ}C$ during the vaccination and challenge test, respectively. We used the formalin-inactivated $\beta$-hemolytic S. iniae vaccine produced by domestic manufacturers. The vaccine was intraperitoneally administered to fish. The vaccinated and control group were challenged with intraperitoneal injection by virulent S. iniae SI-36 isolates with $5.0{\times}10^8$ CFU/fish at 3 weeks after vaccination. We evaluated the vaccine efficacy by calculating numbers of dead fish, and observing of clinical signs, exterior and gross lesions, and examining bacteria isolation and identification. Thirty-four(25.2%) of 135 control and vaccinated group fish were dead with serious anemia, abdominal extension, and hernia of intestine during 3 weeks post vaccination. We isolated Neoheterobothrium hirame from the buccal cavity and Edwardsiella tarda from kidney of dead and diseased fish. When infected fish with these agents were challenged with S. iniae SI-36 isolates, the cumulative mortality of control and vaccinated group were 86.7, and 46.7%, respectively. However, significant differences(p<0.05) were observed on cumulative mortality between control(20.0%) and vaccinated group(95.0%) at second trials with 40 healthy, and relative percent survival(RPS) was 78.0%. We confirmed that the efficacy of $\beta$-hemolytic S. iniae vaccine on olive flounder were impacted by health condition such as bacterial and parasitic diseases.
