Effects of Formalin treatment on embryogenesis and larvae growing in fertilized eggs and hatched larvae of olive flounder, Paralichthys alivaceus were investigated. Fertilized eggs and hatched larvae were exposed to aqueous solution with 100, 200, 400, 800 and 1,000 ppm of nominal formalin concentration. They were kept in sea water of 16, 18 and 22$^{\circ}C$, respectively. Survival rate and hatchability of fertilized eggs were high in control groups more than formalin treated groups in 16, 18 and 22$^{\circ}C$, respectively (P<0.05). A similar effect that survival rate of hatched larvae was also observed for control and treatment groups. On the other hand, fertilized eggs treated with 400 ppm formalin, were all death in kept in 22$^{\circ}C$. These results suggest that high-dose of formalin in fertilized eggs and hatched larvae in olive flounder, Paralichthys olivaceus was inhibited the normal embryogenesis for fertilized eggs and growing for hatched larvae. Also, these inhibited effects was promoted in higher temperature.
This study was carried out to examine the effects of bis (tribytyltin)oxide (TBT), endocrine disrupting compounds (EDCs). on the changes of survival, growth and oxygen consumption rate in the oliver flounder, Paralichthys olivaceus. Oliver Flounders were exposed to sublethal concentration of TBT (0, 1.67, 3.20, 6.30 and 12.50 $\mu\textrm{g}$/L) during 6 weeks. Survival rate was decreased in a concentration and exposure period-dependent way and suddenly the reduction of more than 20% occurred at TBT concentration greater than 3.20 $\mu\textrm{g}$/L. after exposure 6 weeks. Growth rate and feed efficiency significantly decreased at concentration greater than 3.20 $\mu\textrm{g}$/L. Oxygen consumption rate was also decreased in a concentration-dependent way and significantly decreased to 17,48 and 67% than that of the control at the TBT concentration of 3.20, 6.32 and 12.50 $\mu\textrm{g}$/L, respectively. This study revealed that high TBT concentration ($\geq$3.20 $\mu\textrm{g}$/L) reduced growth and oxygen consumption rates of the juvenile oliver flounder suggesting potential influence on the natural mortality of Paralichthys olivaceus in the coastal areas.

Experiments were carried out to investigate the effects of lead exposure on survival, growth and oxygen consumption of juvenile oliver flounder, Paralichthys olivaceus for 6 weeks. Survival rate of the oliver flounder was significantly affected above 150 $\mu{g/L}$. Growth rate of the olive flounder exposed to ≥ 150 $\mu{g/L}$ of lead concentration were significantly decreased than that exposed to normal condition. Metabolic rate in the lead concentration ≥ 80 $\mu{g/L}$ was significantly reduced with increasing lead concentration then that in normal condition. This study revealed that high lead concentration (≥ 80 $\mu{g/L}$) reduced growth and metabolic rate of the juvenile olive flounder suggesting potential influence on the natural mortality of olive flounder in the coastal areas.
Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

Triplicate groups of fed and starved olive flounder, Paralichthys olivaceus (body weight: $119.8{\pm}17.46$ g), were examined over 42 days for physiological changes using hematological, biochemical, and non-specific immune parameters. No significant differences in concentrations of blood hemoglobin and hematocrit and plasma levels of total cholesterol, aspartate aminotransferase, alanine aminotransferase, glucose, and cortisol were detected between fed and starved groups at any sampling time throughout the experiment. In contrast, plasma total protein concentrations were significantly lower in starved fish than in fed fish from day 7 onwards. Moreover, plasma lysozyme concentrations were significantly higher in starved flounder from day 21 onwards. This result confirms that the response of olive flounder to short-term (less than about 1.5 months) starvation consists of a readjustment of metabolism rather than the activation of an alarm-stress response. The present results indicate that starvation does not significantly compromise the health status of fish despite food limitation.

The complete nucleotide sequence of olive flounder (Paralichthys olivaceus) hsp70-related rDNA was determined by RT- and RACE-PCR methods. A full-length of hsp70-related cDNA has an open reading frame of 1.95 kb encoding 650 amino acids with a calculated molecular weight of 71.1 kD. A corresponding hsp70-related protein contains a number of conserved elements including an ATP-binding domain, a nuclear localization signal and the carboxyl terminal motif, EEVD, which may have a role in chaperone function. Comparison of nucleotide and predicted amino acid sequence between olive flounder hsp70-related gene and hsp/hsc70 genes of other species revealed a high similarity with the cognate form of these genes. These results indicated that we recovered likely to be a olive flounder cognate hsc70 gene.
Effect of dietary nutrient composition on growth and body composition of juvenile olive flounder Paralichthys olivaceus with different feeding strategy was determined. Twenty-five fish averaging 16 g were randomly distributed into 12, 180 L flow-through tank each. Four treatments in triplicates were prepared: fish were fed to satiation twice daily by the control diet for 8 weeks as the control group (Con) and fish were fed to satiation twice daily by the control and high nutrient diets for 6 weeks after 2-week fasting (2WS-6WFC, 2WS-6WFHN, respectively) and finally, fish were fed to satiation twice daily by the high nutrient diet for the consecutive 3 days after 4-day fasting for 8 weeks (4DS-3DFHN). No significant difference was found in either survival or weight gain of flounder among treatments. Feed efficiency ratio (FER) for fish in the 2WS-6WFHN treatment was significantly higher than that for fish in the Con and 2WS-6WFC treatments. Protein efficiency ratio (PER) of fish in the 2WS-6WFHN and 4DS-3DFHN treatments was significantly higher than that of fish in the 2WS-6WFC treatment. In conclusion, manipulation of dietary nutrient composition and/or feeding strategy can effectively improve growth of juvenile olive flounder without growth retardation at restricted feeding regime.
To develop a promoter capable of driving transgene expression in non-model fish, we identified and characterized the muscle-specific alpha-actin gene in olive flounder, Paralichthys olivaceus (PoACTC1). The regulatory region of PoACTC1 includes putative regulatory elements such as a TATA box, two MyoD binding sites, three CArG boxes, and a CCAAT box. Microinjection experiments demonstrated that the regulatory region of PoACTC1, covering from -2,126 bp to +751 bp, just prior to the start codon, drove the expression of red fluorescent protein in developing zebrafish embryos and hatching olive flounder. These results suggest that the regulatory region of PoACTC1 may be useful in developing a promoter for biotechnological applications such as transgene expression in olive flounder.
Quantitative analysis of a myxosporean parasite, Parvicapsula sp. in internal organs (kidney, intestine, spleen, brain and liver) from non-emaciated (farm-A) or emaciated (farm-B and farm-C) olive flounder Paralichthys olivaceus were performed by real-time PCR. The highest DNA copy number ($1.7{\times}10^7copies/mg$ tissue) was detected in kidney of the emaciated olive flounder from farm-C, while the DNA copy number was below detection limit in all the organs of the olive flounder from farm-B. There was not positive result in all of organs from olive flounder in farm-A. PCR and histopathological analysis were also performed using the same specimen and showed same results as those by real-time PCR.
This study was designed to analyze the effects of reducing water turnover in olive flounder Paralichthys olivaceus farms, focusing on olive flounder growth, decreasing electricity costs, and developing measures to ensure business stability. Daily water turnover was set at 18 in the control group and six in the experimental group. Juvenile fish were reared for 12 months. No significant differences in mean weight were observed between groups until five months. After five months, the mean weight of the control group grew significantly faster than that of the experimental group. Maintaining water turnovers leverl at six turnovers for the first five months after stocking juvenile fish and then increasing water turnover resulted in a 34.4% reduction in electricity costs compared to the control group. This approach presents a potential method to enhance the profitability of olive flounder farms and ensure stable productivity and profitability without sacrificing olive flounder growth.
