  • Objective : The objective of this study was to investigate the antioxidative effects of Carthami Flos extract. Methods : Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Carthami Flos extract was examined far details of total phenolic content concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, the inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : TAC of Carthami Flos extract at the concentration of 5 mg/ml was 1.84 mM Trolox equivalent. 2. TAR of Carthami Flos extract, on the other hand, couldn't be determined due to interference from unidentified compounds. 3. Total phenolic content of Carthami Flos extract at the concentration of 5 mg/ml was 2.01 mM gallic acid equivalent. 4. Concentration of Carthami Flos extract at which DPPH radical scavenging activity was inhibited by 50% was 6.43 mg/ml as compared to 100% by Pyrogallol solution as a reference. 5. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeS04/ascorbic acid. Carthami Flos extract at the concentration of 10 ms/ml slightly but significantly decreased TBARS concentration. The extract continued to prevent lipid peroxidation in a dose-dependent manner. 6. The effect of Carthami Flos extract on reactive oxygen species(ROS) generation was examined using a cell-free system induced by hydrogen peroxide/FeS04. Addition of 1 mg/ml of Carthami Flos extract significantly reduced dichlorofluorescein(DCF) fluorescence. Carthami Flos extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the ektract significantly prevented ROS generation in vitro. Conclusion: : Antioxidant efffcts of Carffami ffor extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fellowed by inhibition of lipid peroxidation.

  • Objectives : This study is to investigate whether Carthami flos exhibits a synergistic effect on the apoptotic effect of doxorubicin on human gastric cancer cells. Methods : We used AGS, a human gastric cancer cell line. To investigate the apoptotic efficacy of doxorubicin and Carthami flos, MTT and CCK-8 methods were used. To confirm apoptosis, cell cycle and mitochondrial membrane potential changes were confirmed. To investigate the mechanism of apoptosis, the reactive oxygen species (ROS) experiment was performed. Results : 1. Doxorubicin or Carthami flos induced cell death in the human gastric cancer cell line AGS. 2. Carthami flos showed a synergistic effect of cell death by doxorubicin. 3. The cell cycle and mitochondrial membrane potential changes revealed that cell death was apoptosis. 4. Apoptosis was related to reactive oxygen species (ROS) generation. Conclusions : This result shows the anticancer synergistic effect of Carthami flos in gastric cancer cells, and is considered to be an important basis for the development of anticancer drugs for Carthami flos.

  • Objectives : The purpose of this study was to investigate the effects of Carthami flos on pacemaker potentials of small intestinal and colonic Interstitial Cells of Cajal (ICC). Methods : To dissociate the ICC, we used enzymatic digestions from the small intestine and colon in mice. In the ICC, the electrophysiological whole-cell patch-clamp configuration was used to record pacemaker potentials in the cultured ICC. Results : 1. The ICC generated pacemaker potentials in the murine small intestine and colon. 2. Pretreatment with a Ca2+ free solution and thapsigargin, a Ca2+-ATPase inhibitor in the endoplasmic reticulum, stopped the pacemaker potentials. In the case of Ca2+-free solutions, Carthami flos did not induce membrane depolarizations in the murine small intestine and colon. However, when thapsigargin in a bath solution was applied, Carthami flos induced membrane depolarizations only in the murine colon. 3. Pretreatment with 2-APB (transient receptor potential melastatin (TRPM) channel inhibitor) abolished the pacemaker potentials and suppressed Carthami flos-induced effects in the murine small intestine and colon. 4. However, pretreatment with T16Ainh-AO1 (Ca2+ activated Cl- channel; anoctamin 1 (ANO1) inhibitor) did not affect the pacemaker potentials and induced Carthami flos-induced effects only in the murine small intestine. Conclusions : These results suggest that Carthami flos can modulate the pacemaker activity of ICC and the mechanisms underlying pacemaking in ICC might be different in the small intestine and the colon.
