Loxoprofen-Na (sodium 2-〔4-(2-oxocyclopentylmethyl)pheny)propionate dihydrate) is a potent analgesic drug. We developed loxoprofen-Na plasters to extend duration time of analgesic activity and to reduce side effect on gastrointestinal tract. Analgesic effect of Loxoprofen-Na plasters was investigated. Loxoprofen-Na plaster had good analgesic effect in rat paw pressure test, Tail-flick latency test and acetic acid-induced writhing test. Also, it had anti-inflammatory effect on carrageenan-induced rat hind paw edema. In pharmacokinetic study of Loxoprofen-Na, plasters dosage form showed that plasma drug concentration was prolonged up to 14 hours. So, we can conclude that loxoprofen-Na plasters, when applied on skin, will be a new type of drug for controlling the various local pain or inflammation.
This study was performed to investigate the effects of glycine on the development of tolerance to and physical dependence on morphine. Repeated administration of morphine (10 mg/kg) developed tolerance and physical dependence. Glycine (100, 200 and 400 mg/kg) was administered intraperitoneally (i.p.) to mice for 7 days prior to the morphine injection. Analgesic effects were estimated by the tail flick methods. The inhibitory degree of the development of morphine-induced analgsic tolerance by i.p. administration of glycine was evidenced by the increase in analgesic response to morphine. Glycine inhibited the development of tolerance to morphine. In addition, we separately measured jumping response as the naloxone-precipitated withdrawal sign in mice that had received the same morphine. Glycine reduced the number of jumping behaviors in morphine dependent mice. These results suggest that glycine might be useful the prevention or treatment of morphine tolerance and physical dependence.
The ethanolic (50% v/v) extracts of Peristrophe bicalyculata Nees. (Acanthaceae) were examined for anti-inflammatory and analgesic activities in experimental animals. P. bicalyculata in doses of 50, 100 and 200 mg/kg caused a dose dependent inhibition of swelling caused by carrageenin equivalent to 12.25-24.49% protection and 16.62-39.44% in cotton pellet induced granuloma in rats. There was a significant increase in the tail-flick reaction time in mice (18.05-76.43% protection) and analgesy-meter induced pain in rats (14.49-56.85%) protection). The extract of P. bicalyculata resulted in an inhibition of stretching episodes and the protection range of 14.49-56.85% respectively in acetic acid induced writhing.
The present investigation was carried out to evaluate the antinociceptive and sedative - hypnotic effect of a vacuum dried methanol extract of aerial parts of Artemisia pallens. In the tail-flick method with Swiss albino mice the methanol extract at the doses of (1500 mg/kg, 2000 mg/kg, and 2500 mg/kg) showed significant antinociceptive activity. Significant potentiation of Pentobarbitone sodium - induced sleeping time was observed in mice on co-administration of the various doses of the methanol extract of Artemisia pallens.
Prostaglandins are generated through two isoforms of the enzyme cyclooxygenase, constitutively expressed cyclooxygenase(COX)-1 and COX-2, which is induced at sites of inflammation. Inhibition of COX-2 is desirable as this may avoid side effects seen with NSAIDs. We examined the effects of transcutaneous electrical nerve stimulation and cold therapy on the levels of muscle cycloooxygenase-2 mRNA in rats of carrageenan-induced inflammatory. The method of behavioral assessment were paw withdrawal latency(PWL) and tail flick test(TFT). The COX-2 mRNA levels were quantified by reverse transcription-polymerase chain reaction (RT-PCR). Following the transcutaneous electrical nerve stimulation and cold therapy, PWL and TFT were increased and COX-2 mRNA expression in gastrocnemius muscles were decreased. These results suggest that a transcutaneous electrical nerve stimulation and cold therapy were good therapy for a muscle pain.

This study examined the Influence of ginseng total saponins (GTS) on the analgestic action and tolerance development of pentazocine in mice. Pentazocine prolonged the latency to response in the tail flick rather than in the tail pinch test. The analgesic effect of pentaEocine was antagonized by naloxone and completely eliminated by pretreatment u·ith f-chlorophenylalanine (PCPA). GTS provented the pentasocine-incuced analgesia ann inhibited the development of tolerance to pentazocine. The antagonistic effect of GTS on the pentazocine-induced analgesia was abolished by 5-HTP, but not by L-DOPA. These results suggest that GTS inhibits the analgesic action of pentazocine by the interaction with serotonergic neuron.
Dichloromethane, ethanol, and boiling water extracts of the brown alga Ecklonia cava were examined in vivo for their antipyretic, analgesic, and anti-inflammatory activities in mice. These activities were evaluated by yeast-induced pyrexia, tail-flick test, and phorbol myristate acetate-induced inflammation (edema, erythema, and blood flow). Ethanol extract of E. cava (0.4 mg/ear) inhibited the inflammatory symptoms of mouse ear edema, erythema, and blood flow by 82.6%, 69.0%, and 65.4%, respectively. This extract also demonstrated potent analgesic activity. No acute toxicity was observed after p.o. administration of each extract (5 g/kg bw). These in vivo data are in agreement with the claims of the health care industry and indigenous medicine that E. cava is an effective remedy for inflammation-related symptoms.
We have investigated the antinociceptive efficacy of ginseng saponins in mice using l% formalin, which induce two phases of pain (acute and tonic pains) and is known to induce a clinically related pain. Ginseng total saponins (GTS) relieved both phases of pain with EDso of 162 mghg for acute and 92 mg/kg for tonic pain, respectively. Both protopanaxadiol (PD) and protopanaxatriol (PT) saponins did not attenuated acute phase of pain but relieved tonic phase of pain with EDso of 45 mg/kg for PD saponins and 105 mghg for PT saponins, respectively. Moreover, ginsenoside Rc, Rd, and Re among representative ginsenosides such as Rbl, Rc, Rd, Re and Rgl relieved slightly but significantly acute phase of pain and strongly attenuated tonic phase of pain but Rf relieved only tonic phase of pain. However, PD and PT saponins, and the individual ginsenosides tested except GTS did not greatly attenuate thermal noxious pain (tail-flick test). These results suggest that single ginsenoside or mixture of various ginsenosides mainly induce differential antinociception in mice.
To establish the usefulness of oxytocin (OT) as an analgesic for women in delivery, the pharmacokinetic parameters and blood-brain barrier (BBB) permeability of [$^3H$] OT were obtained using an intravenous injection technique or the internal carotid artery perfusion/capillary depletion (ICAP/CDM) method. Brain uptake of OT was similar to that of sucrose, plasma space marker, indicating that OT has a poor BBB permeability. Moreover, the analgesic effects of OT injected through the jugular vein on nociception were evaluated by the tail-flick method. The antinociceptive effects of OT injected at a dose of 0.2 ${m}g/kg$or 2 ${m}g/kg$ were dose-dependent. In addition, the analgesic effects of OT on the CNS were unaffected by naloxone, a m-receptor antagonist. In a similar manner to the opioid system, OT may play a modulatory role in antinociception.

This paper studies a more exact flicker evaluation method by detecting power quality disturbances and excluding the effects of power quality disturbances. Up to the present, power quality disturbances affect flicker evaluation index because power quality problems do not have been considered. However, flick index should represent only flicker without power quality disturbances. Thus, in this paper, we present the improved flicker evaluation method which removing the effects of power quality disturbances such as voltage sag and transient caused by fault and inverter/breaker switching. We detect voltage sag and transient using wavelet transform and remove the effects of power quality disturbances from flicker index.
