Membrane and Virus Filter Trends in the Processes of Biopharmaceutical Production (바이오의약품 제조공정에서 분리막의 역할과 바이러스 필터 동향)

  • Membranes are used in most processes of biopharmaceutical production. It is used for pretreatment of other processes, separation of impurities in the process, virus removal, control of products concentration and buffer solution exchange. Virus filters play an important role in ensuring product efficacy and stability because viral contamination of biopharmaceuticals for humans is a sensitive issue that is directly related to serious clinical outcomes. Virus filters typically have complex multilayer structures made of various polymers such as surface-modified PVDF, PES, CRC. Depending on the manufacturer, filters have different pore structures and shapes, such as symmetric or asymmetric, and is used in the form of pleated membrane, flat sheets or hollow fibers. Virus filters are exclusively supplied by few foreign companies such as Asahi Kasei, Millipore, Pall and Sartorius. Replacing virus filters can be time consuming and expensive, including approval from regulatory agencies through validation. As localization has become important due to Japan's recent export regulations, it is necessary to increase the degree of technical independence.

Experimental determination of liquid entry pressure (LEP) in vacuum membrane distillation for oily wastewaters

  • In this study we investigate a laboratory scale vacuum membrane distillation system to produce pure water from model oil in water emulsion. Experimental determination of liquid entry pressure (LEP) of a commercial Durapore$^{TM}$ GVPH flat sheet membrane using model emulsions in various oil concentrations has been carried out. Two different methods of liquid entry pressure determination - a frequently used, so-called static and a novel dynamic method - have been investigated. In case of static method, LEP value was found to be 2.3 bar. No significant effect of oil content on LEP was detected up to 3200 ppm. In contrast, LEP values determined with dynamic method showed strong dependence on the oil concentration of the feed and decreased from 2.0 bar to a spontaneous wetting at 0.2 bar in the range 0-250 ppm, respectively. Vacuum membrane distillation tests were also performed. The separation performance is evaluated in terms of flux behavior, total organic carbon removal and droplet size distribution of the feed and final retentate. No significant effect of oil content on the flux was found ($5.05{\pm}0.31kgm^{-2}h^{-1}$) up to 250 ppm, where a spontaneous wetting occurred. High separation performance was achieved along with the increasing oil concentration between 93.4-97.0%.

  • Carbon dioxide was absorbed into water-in-oil (w/o) emulsion composed of aqueous 2-amino-2-methyl-1-propanol (AMP) droplets as a dispersed phase and benzene solutions of polybutene and polyisobutylene as a continuous phase in a flat-stirred vessel to investigate the effect of non-Newtonian rheological behavior on the rate of chemical absorption of $CO_2$, where the reaction between $CO_2$ and AMP in the aqueous phase was assumed to be a pseudo-first-order reaction. It was expressed that PIB with elastic property made the rate of chemical absorption of $CO_2$ accelerated by comparison of mass transfer coefficient of $CO_2$ in the non-Newtonian liquid with that in the Newtonian liquid.

  • The present study is devoted to the validation of a new method for in line electrokinetic characterisation of deposits on membrane surfaces. This method is based upon simultaneous measurements of transversal streaming potential and permeates flux at constant pressure before and during the deposit formation. Dead-end filtration experiments were conducted with negative flat membranes forming a narrow slit channel, negative hollow fiber membranes and mono-dispersed suspensions of (negatively charged) polystyrene latex and (positively charged) melamine particles at various concentrations. It was observed that the overall streaming potential coefficient increased in absolute value with the deposited latex quantity, whereas it decreased and changed of sign during the filtration of melamine suspensions. By considering a resistance-in-series model, the streaming potential coefficient of the single deposit ($SP_d$) was deduced from the electrokinetic and hydraulic measurements. The independence of $SP_d$ with respect to growth kinetics validates the measurement method and the reliability of the proposed procedure for calculating $SP_d$. It was found that $SP_d$ levelled off much more quickly when filtration was performed through the slit channel. This different behaviour could result from a non-uniform distribution of the deposit thickness along the membrane given that the position of measuring electrodes is different between the two cells.

Fabrication of Nanometer-sized Pattern on PMMA Plate Using AAO Membrane As a Template for Nano Imprint Lithography (AAO 나노기공을 나노 임프린트 리소그래피의 형틀로 이용한 PMMA 나노패턴 형성 기술)

  • PMMA light guiding plate with nano-sized pattern was fabricated using anodized aluminum oxide membrane as a template for nano imprint lithography. Nano-sized pore arrays were prepared by the self-organization processes of the anodic oxidation using the aluminum plate with 99.999% purity. Since the aluminum plate has a rough surface, the aluminum plate with thickness of 1mm was anodized after the pre-treatments of chemical polishing, and electrochemical polishing. The surface morphology of the alumina obtained by the first anodization process was controlled by the concentration of electrochemical solution during the first anodization. The surface morphology of the alumina was also changed according to temperature of the solution during chemical polishing performed after first anodization. The pore widening process was employed for obtaining the one-channel with flat surface and height of the channel because the pores of the alumina membrane prepared by the fixed voltage method shows the structure of two-channel with rough surface. It is shown from SPM results that the nano-sized pattern on PMMA light guiding plate fabricated by nano imprint lithography method was well transferred from that of anodized aluminum oxide template.

  • Analysis of Dynamic Characterisocs of Rectangular Plate by Finite Element Method. Dynamic characteristics of a rectangular plate with opening in it is studied by finite element method. To investigate these characteristics 12 degrees of freedom membrane finite element in used. The rectangular membrane finite elements are defined by specifying geometry, internal displacement functions and strain-displacement relations. Then, the governing equation for the finite element is derived by energy method. To derive the mass matrix and stiffness matrix of the element, expressions for strain and kineic energy in terms of the node displacement are generated. In constructing the overall structure matrix, the matrix of each elements are superposed and partitioned by applying the given boundary condition to obtain a nonslngular matrix. To find the natural freguencies and viration modes, the eigen values and the corresponding eigen vectors are computed by the computer using well known Jacobi power method. In order to verify the capability of the membrane finite element, a flat rectangular plate is analyzed first, and the result is compared with well known analytical results to show the good agreement. A rectangular plate with opening in It is analyzed with the same finite element. The results are presented in this paper. Unfortunately, the literature study could not provide with some results to compare, but the results reveal that the output of this research is phlslcally reasonable. And the results of this research are useful not only in practice but also for the future experimental research in comparison purpose.

An NMR Study on the Conformation of Substance P in Acidic Bicelles

  • The conformation of a neuropeptide, substance P (SP), in isotropic (q = 0.5) acidic bicelles was investigated using two-dimensional NMR techniques. By the nuclear Overhauser effect (NOE) cross peaks between SP and long-chain lipid molecules SP was probed to bind on the flat surface of the disc-like bicelles. Structural analysis of NMR data indicated that the helical conformation of SP extended to the C-terminal region of Leu10 as well as in the mid-region from Pro4 to Phe8. As compared with the conformations of SP bound on the sodium dodecylsulfate (SDS) or the dodecylphosphocholine (DPC) micelles with curved surfaces, the surface curvature of the membrane mimics was found to be one of the major factors inducing the biologically relevant conformation of SP. The negative surface charge of the membrane is also a key factor inducing both the binding of SP on the membrane and its biologically active structure.

The Effect of Filling Step on the Removal Efficiency and Filtration Performance in the Operation of Submerged Membrane-Coupled Sequencing Batch Reactor (침지형 막결합 연속회분식 반응기의 운전에서 폐수의 도입단계가 제거효율과 여과성능에 미치는 영향)

  • In the operation of submerged membrane-coupled sequencing batch reactor, the effect of filling step on the removal efficiency and filtration performance were investigated. Two sets of operation modes, the filling step located in the beginning of aerobic step (Mode-1) and the beginning of anoxic step (Mode-2), during 89 days were conducted. There was no wide difference in the COD removal and filtration performance between two sets of operation modes. But in the removal efficiency of nutrients (total nitrogen and total phosphorous), Mode-2 was more effective than Mode-1. In the case of Mode-2, average removal efficiencies of COD, total nitrogen, and total phosphorous were 99.1, 73.3, and 77.3%, respectively.

  • Separation characteristics of cutting oil-in-water emulsions were studied experimentally by using various kinds of flat-type microfiltration and ultrafiltration membranes. For ultrafiltration membranes the permeation behavior of cutting oil emulsions obeys the film model, whereas a significant deviation from the model was observed for ASYPOR microfiltration membranes. The experimental data obtained for all the membranes showed that the effect of operating pressure on the permeation flux of oil-in-water emulsions is not very significant. At low transmembrane pressures the permeation flux decreased gradually with increasing filtration time, whereas the permeation flux at high transmembrane pressures decreased steeply for early filtration time. However, every flux eventually reached a constant value that depends only on the applied transmembrane pressure. For the hydrophobic polycarbonate microfiltration membrane the permeation flux increased with the filtration time. The critical permeation pressures were also determined from the data obtained from unstirred cell experiments.

Anisotropic Wet Etching of Single Crystal Silicon for Formation of Membrane Structure (멤브레인 구조 제작은 위한 단결정 실리콘의 이방성 습식 식각)

  • We have studied micro-machining technologies to fabricate parts and sensors used in the semiconductor equipment. The studies were based on the silicon integrated circuit processes, and composed of the anisotropic etching of single crystal silicon to fabricate a membrane structure for hot and cold junctions in the infrared absorber. KOH and TMAH were used as etching solutions for the anisotropic wet etching for membrane structure formation. The etching characteristic was observed for the each solution, and etching rate was measured depending upon the temperature and concentration of the etching solution. The different characteristics were observed according to pattern directions and etchant concentration. The pattern was made to incline $45^{\circ}$ on the primary flat, and optimum etching property was obtained in the case of 30 wt% and $90^{\circ}C$ of KOH etching solution for the formation of the membrane structure.
